Gastrointestinal cancers are the most common types of malignant tumors in Human Beings.The main treatment remains surgery resection,in addition to which,adjunctive therapies are far not satisfactory for the target of which are not specific.Dendritic cells cancer vaccine is one of the most promising immunotherapy ways developed in the decades and some results have showed that the vaccines may help in the treatment of gastrointestinal cancers.Till now,there is no recognized standard in the design of the vaccine and no positive correlations between the immunologic effect and clinical prognosis are showed,but we believe that with the further research in the immunological mechanism and the use of new designed methods,dendritic cells cancer vaccines will play a greater role in the treatment of malignant tumors.

[Objective]To sum up pro. Lian Jianwei’s clinical experience in treating acne. [Method] By proved recipe analysis, it shows pro. Lian Jianwei ’s treatment based on differentiation of signs from different angles and the clinical thought of flexibly applying ancient formulae on acne. [Result] Pro. Lian Jianwei is good at revising ancient formulae for treating acne with good effect, according to patients ’pulse signs and mechanism;if the food is stagnated in the gastric-intestine, which wil transform into heat and up along the channel, Baohe Pil shal be used to remove stagnation and heat; if the liver is de-pressed with heat, deficient blood and weak spleen, Xiaoyan Powder shal be used for regulation and stretching the wood; use Wenqing Drink to nourish blood and remove toxin for deficient blood and hot liver, toxic heat implicated in spleen and stomach; apply Zisheng Pil to regulate and tonify middle part of the body for deficient spleen and stomach with turbidness and dyspepsia, etc. [Conclusion] Pro. Lian Jianwei has rich clinical experience, his clinical thought of “treatment based on differentiation of signs, with flexible methods and formulae, coping with shifting diseases by sticking to a fundamental principle”, worthy our deep experiencing and study.
4.Application of PDCA Cycle in Joint Management of Cooperative Research Projects

It has proved to be difficult for administrators of scientific research to improve process management,especially to manage cooperative research projects accomplished by various institutions.In this paper,we took the process management of Scientific Research in Public Interest (SRI-PI) based on a Grade 3A hospital and other 19 institutions as an example,to analyze the application of PDCA cycle in projects management.It is concluded that the comprehensive quality management of research projects should follow the scientific procedure of PDCA cycle.
5.Study on the mechanism of brain damage caused byStaphylococcus epidermidis in mice

ObjectivesTo study the mechanism of brain damage caused byStaphylococcus epidermidis (SE) in mice. Methods A total of 80 neonatal mice of postnatal day 1 (PND1) were divided into SE group (48 mice), normal saline (NS) group (16 mice) and control group (16 mice). Mice in SE group were intravenously injected with 50 μl SE (108/ml). Mice in NS group were given 50 μl NS. Mice in control group were not intervened. At different time points after SE injection (6 h, 24 h, 72 h, 5 d, 7 d), the CFU of brain, blood, and spleen were calculated. Serial sections of parafifn-embedded brain tissue were used for detection of ionized calcium-binding adaptor moleculor1 (Iba-1) by immunohistochemical staining. The positive cells were calculated. ELISA was used to measure the levels of tumor necrosis factor alpha (TNF-α), interleukin-1β (IL-1β), interleukin-5 (IL-5), interleukin-6 (IL-6) of brain at 6 h and 24 h after SE injection.Results There was no SE in brain in different time points. The CFU was at the highest level at 6 h and then decreased after 24 h in blood and spleen. The Iba-1 positive cells in SE group were signiifcantly increased compared to NS group and control group at 24 h and 72 h (P<0.05). There was no difference of Iba-1 positive cells be-tween 24 h and 72 h after SE injection (P>0.05). The levels of TNF-α, IL-1β, IL-5, and IL-6 were signiifcantly higher in SE group than those in NS and control at 6 h and 24 h (P<0.05). The levels of TNF-α, IL-1β, IL-5, and IL-6 were signiifcantly lower in SE group at 24 h than those in SE group at 6 h (P<0.01).Conclusions It is suggested that cytokines produced by microglias may be the mediators of SE-caused brain damage.
6.The influence of Staphylococcus epidermidis on the neonatal mice of different ages

Objectives To study the influence of Staphylococcus epidermidis (SE) on the neonatal mice of different ages. Methods A total of 60 neonatal mice including postnatal day 1(PND1) and postnatal day 3(PND3) were divided into SE group, normal saline (NS) group and control group, with 20 mice each. Mice in SE group were intravenously injected with 50μl SE (108/ml). Mice in NS group were given 50μl NS and mice in control group was not intervened. On postnatal day 14, the brain, liver and spleen obtained from mice were weighted. Serial sections of paraffin-embedded brain tissue were used for the detec-tion of microtubule associated protein-2 (MAP-2) and myelin basic protein (MBP) by immumohistochemical staining, and then the areas and volumes of grey and white matter were calculated. Result The mortality of PND1 mice in SE and NS group was 60.0%and 40.0%, respectively, and there was no difference between two groups (P>0.05). The mortality of PND3 mice in SE and NS group was 10.0%and 0.0%, respectively, and there was no difference between two groups (P>0.05). There were no dif-ferences in body weight, body weight gain, spleen and liver weights and organ coefficient between PND1 and PND3 mice (P>0.05). In PND1 mice, the areas and volumes of grey and white matter were significantly smaller in SE group than those in NS group (P<0.05). However, in PND3 mice, there was no differences in areas and volumes of grey and white matter between SE and NS group (P>0.05). Conclusions SE infection can result in brain injury in PND1 mice, but has no effect on brain tissues of PND3 mice.

8.Bleeding of children's uper-digestive tract treated with endoscope metal clamp

Objective To explore the application technology and effect of endoscope hemostatic metal clamp in children's uper-digestive tract with non-variciform acute large bleeding.Methods Type-Olympus-HX-51R-1 or type-MD-850 endoscope metal clamp were used to hold and put in the pusher.Results For 28 cases with acute large bleeding of uper-digestive tract,hemostasis rate was 100 per cent.Among them one case reoccured bleeding.Totally 43 badges of metal clamps were put inside,at an average of 1.6 badges each.No complications and adverse reactions occurred.Conclusion As a safe and easily used method of treatment,endoscope metal clamps can achieve high hemostasis rate quickly and prevention of reoccurrence of bleeding.
9.Prenatal diagnosis for trisomy 18——analysis of 9 cases

Objective To assess the effectiveness of prenatal screening with maternal serum markers and the sonographic characteristics of the foetus for detecting trisomy 18. Methods Amniotic fluid or the fetal blood samples were obtained and cultured in women with positive results in serum screening (n=36) and with abnormal findings in sonography (n=92) at 18~32 weeks of gestation, and then fetal karyotyping was performed. Results Nine cases of trisomy 18 and 2 cases of other abnormal karyotype were found among the 128 pregnant women (8.59%, 11/128). Four cases were diagnosed as trisomy 18 among the 36 women with positive screening results. The rate of abnormal karyotype among those women with abnormal sonographic findings was 7.61%(7/92,5 cases of trisomy 18 and 2 cases of other abnormal karyotype). Two cases of trisomy 18 had both positive result in screening and abnormality in sonography. Conclusions Prenatal screening for maternal serum markers combined with sonographic examinations for the fetus is effective in detecting foetus with trisomy 18.
10.Application and evaluation of a probe melting analysis-based assay for detection of Isoniazid-resistant mutations in Mycobacterium tuberculosis

Objective To validate the performance of a PMA-based assay for detection of INH-resistant mutations in MTB and investigate the mutation characteristic of INH-resistance.Methods The MTB standard strain H37Rv was from National Tuberculosis Reference Laboratory,1 wild-type strain and 1 katG S315T ACC mutant strain were from Xiamen CDC,7 MTB INH-resistant strains with known INH resistance mutations were from Shenzhen Center for Chronic Disease Control,Henan CDC,No.309 Hospital of PIA and Xiamen CDC.707 MTB clinical isolates were from Xiamen CDC,Xiamen No.1 Hospital and Zhangzhou CDC,126 sputum samples were from Xiamen Tongan CDC.The genomic DNA of the MTB standard strain H37Rv,7 MTB INH-resistant strains and 833 clinical samples,were extracted with Xiamen Zeesan Mycobacterium tuberculosis Isoniazid-resistance Mutation Test Kit using the thermal lysis method.The genomic DNA of 1 wild-type strain and 1 katG S315T ACC mutant strain were extracted with AxyPrep Bacterial Genomic DNA Miniprep Kit.The mutations were discriminated by the △Tm between the samples and wild type control in katG315 codon,inhA promoter -17 to -8 region,ahpC promoter region -44 to -30 and - 15 to 3,and inhA94 codon.A 10-fold dilution series of MTB DNA from 3 × 105 to 300 copies/ reaction obtained from a wild-type strain and a katG S315T ACC mutant strain,respectively,were prepared to determine the analytical sensitivity.Seven MTB INH-resistant strains with 9 predetermined mutations were used for the analytical specificity assay,and 5 mutants of which were used for the repeatability assay.The clinical detection performance of PMA assay were confirmed by the sequencing method in 833 samples.Results Results could be obtained within 3 hours from DNA extraction to PMA assay,including 46 samples in a standard 96-well real-time PCR instrument simultaneously.The analytical sensitivities of PMA were 300 copies/reaction for both the wild-type strain and katG S315T ACC mutant strain.Nine INH-resistant point mutations could be discriminated and 5 of which had standard deviations of melting temperature less than 0.5 ℃.Fully concordant results of mutant locus between PMA assay and sequencing were obtained in all 162 mutant samples.INH-resistant mutations in the four loci were found in 19.4% (162/833) samples by PMA assay in Xiamen and Zhangzhou.Among the 14 lNH-resistant mutant types detected,katG S315T ( AGC→ACC),inhA promoter - 15C→T and katG S315N (AGC→AAC) accounted for 83.3% (135/162) of the overall mutations.
