1.Preliminary investigation into clinical values of intravascular ultrasound in carotid artery narrowing of transient ischemia attack

ObjectiveTo investigate preliminary applic ation values of intravascular ultrasound(IVUS)in the carotid artery narrowing of transient ischemia attack (TIA) patients. MethodsIVUS has been used to 4 TIA patients with carotid artery narrowing identified by carotid artery echo, following carotid stenting and completion angiogram to detect evidence of inappropriate stent deployment. ResultsIVUS revealed the vessel wall structure, plaque morphology, measured accurately the narrowing degree of carotid artery. The narrowing percent identified by IVUS was larger than angiogram, and IVUS was more sensitive in detecting plaque than digital subtraction angiography(DSA). Four patients accepted stent deployment successfully.ConclusionsDSA underestimated the severity of the lesion. IVUS more accurately evaluates the extent and characteristics of the lesions than DSA, it is an important and useful component of carotid artery stent procedures.
2.Determination of Methotrexate in Human Serum by HPLC

OBJECTIVE:To determinate the serum concentration of methotrexate by HPLC. METHODS:The samples were separated on a Waters C18 column at a column temperature of 30 ℃. The mobile phase consisted of acetonitrile-phosphate buffer(pH 7.2,12∶88) at a flow rate of 1.0 mL?min-1. The detection wavelength was set at 303 nm. RESULTS:The linear range of methotrexate was 0.5~80 ?g?mL-1(r=0.999 4). The methodological recovery rates were all above 90%; the intraday RSD was 3.03 %~3.52 % and the inter-day RSD was 2.57%~4.05%. CONCLUSION:The method is proved to be simple,rapid,accurate and sensitive,and it is suitable for the determination of serum concentration and pharmacokinetic study of methotrexate.
3.RP-HPLC Determination of the Content of Ephedrine?HCl in Xiaoyan Zhike Plaster

Objective To establish a RP-HPLC method to determine the content of ephedrine?HCl in Xiaoyan Zhike Plaster. Method The sample was prepared according to 2005 edition of Chinese Pharmacopocia, the column of Inertsil-3 ODS (4.6 mm?250 mm, 5 ?m) was used. The mobile phase was composed of methanol-0.1% of phosphoric acid (5∶95). The flow rate was 1 mL/min, with UV detection at 207 nm. Results The linear range was 0.041~0.41 ?g (r=0.999 8). The lowest limit of detection was 0.025 6 ?g. The average recovery and the precision was 99.55% and 2.58%. Conclusion The method is simple, sensitive and accurate, and can be used for the quality control of Xiaoyan Zhike Plaster.
4.Optimization of Alcohol Refluxing Extraction Technique of Rhizoma Corydalis and Radix Angelicae Dahuricae in Compound Yuanhu Zhitong Patch

Objective To optimize the alcohol extraction technique of the medical herbs such as Rhizoma Corydalis and Radix Angelicae Dahuricae. Methods To optimize the alcohol extraction technique with L9(34) orthogonal test. The content of the tetrahydropalmatine in Rhizoma Corydalis,which was used as evaluating criteria,was determined by HPLC. Results The optimized extraction technique was as follows: the medicinal material was extracted for 3 times with 3 times amount of 75% alcohol,1.5 hours per time. Conclusions The optimized technique is reasonable and the extraction rate was high.
5.RP-HPLC Determination of Geniposide in Xiaoyan Tuire Patch

Objective To establish a RP-HPLC method to determine the content of geniposide in Xiaoyan Tuire Patch. Method The column of Agilent ZORBAX SB-C18 (4.6 mm?250 mm 5 ?m) was used. The mobile phase was composed of water-acetonitrile-phosphoric acid (86∶14∶0.01). The flow rate was 1 mL/min, with UV detection at 238 nm, and the temperature was 30 ℃. Results The linear range was 0.07~0.70 ?g (r =0.99997). The average recovery was 98.60% and the RSD was 1.96%. Conclusion The method is simple, sensitive and accurate, and can be used for the determination of Xiaoyan Tuire Patch.
6.Possibility of osteogenesis of partially deproteinised decalcified bone composited with osteoblast in vivo

BACKGROUND: Natural bilogical bone-derived materials processed with physical and chemical methods possess natural network pore system. They have good cellular compatibility, and help osteoblasts attach and grow on them, and can be used as the scaffolds of osteoblasts.OBJECTIVE: To observe the osteogenetic effect of partially deproteinised decalcified bone (PDDB) as scaffolds of osteoblasts in vivo.DESIGN: A randomized and controlled trial.SETTING: Central Laboratory of the First Hospital of Kunming Medical College.MATERIALS: Partially deproteinised decalcified bone; human embryonic periosteum-derived osteoblasts; twenty 4-week-old BALB/c nude mice,with the body mass of 25 to 28 g, of either gender.METHODS: This experiment was conducted at the Central Laboratory of the First Affiliated Hospital of Kuming Medical College from January to June 2003. PDDB composited with human embryonic periostea-induced osteoblasts were implanted into the nude mice after cultured for 1 week in vivo, 4 scaffolds in each nude mouse. Composite of scalfolds and cells implanted on the left side of the spine was set as experimental side and simple implanted material on the right side of the spine was set as blank control side. Then alkaline phosphatase activity and routine histological examination were performed 4 and 8 weeks after operation.MAIN OUTCOME MEASURES: General observation, alkaline phosphatase activity and routine histological examination were performed after the materials were taken out.RESULTS: Twenty nude mice entered the stage of result analysis. ① General observation of the implanted materials: No necrosis, ecpyesis, or fluidifying was found around the implanted materials, but ingrowth and enwrapption of soft tissues were found. ② Measurement of alkaline phosphatase activity: alkaline phosphatase activity after PDDB composited osteoblasts in vivo was stronger at week 8 than at week 4 [(22.854±6.018) nkat/g vs(11.286±4.268) nkat/g], and was much stronger than that of the simple implanted materials [(1.217±0.083) nkat/g vs (2.717±0.583) nkat/g]. ③ Results of routine histological examination: Cartilage formed at week 4 and part of cartilage formed new bone and marrow cavity at week 8 at the experimental side, cartilage and new bone formed much more as time went by, but there was no any cartilage or bone formation at the control side.CONCLUSION: Cartilage and bone form after PDDB composited with osteoblasts are implanted, and more cartilage and new bone form as time passes. PDDB can be used as the scaffolds of osteoblasts.
7.Clinical Study on 1 068 Cases of Acne Treated by Fire Needle Therapy

Objective: To investigate the clinical efficacies of fire acupuncture in treat various types of acne. Methods: An open test was designed. One thousand and eighty-six patients who visited for clinical service were collected as a sample. Results: The cure and marked effective rate was 78.5% for lung heat type, 80.8% for heat toxin type, 77.4% for disharmony of the Thoroughfare and Conception Vessels type and 80.1% for blood stasis and phlegm accumulation type. Statistical analysis showed no significant differences in therapeutic effect between different types (P>0.05). Conclusion: Fire acupuncture has an exact therapeutic effect on various types of acne without toxic side effects.
8.Epimedium combined with bone marrow mesenchymal stem cell transplantation for treatment of kidney disease in rats

BACKGROUND:A large number of studies have shown that bone marrow mesenchymal stem cels (BMSCs) with differentiation ability can be used to treat kidney injury. Epimedium has good anti-inflammatory and immune restoration ability.
OBJECTIVE:To observe the effect of epimedium with BMSC transplantation on rat’s kidney disease caused by adriamycin nephrosis.
METHODS:Fifty rats were equivalently randomized to receive no treatment in control group, tail vein injection of normal saline plus intragastric injection of normal saline in model group, tail vein injection of BMSCs plus intragastric injection of epimedium decoction in combined group, tail vein injection of BMSCs in BMSCs group, and intragastric injection of epimedium decoction in epimedium group. Rat models of adriamycin nephrosis were made in the latter four groups through tail vein injection of doxorubicin hydrochloride, while rats in the control group were only given the same volume of normal saline. Twenty-four hours after modeling, rats in the latter four groups began to be given the corresponding treatments. Epimedium decoction was given in the combined and epimedium groups at a dose of 1 mL/d, for consecutive 1 week.
RESULTS AND CONCLUSION:Compared with the model group, mitigated kidney injury and reduced urinary protein level were found in rats undergoing BMSCs and/or epimedium decoction treatment; the levels of plasma albumin and catalase were significantly increased in the combined group and epimedium group, while the levels of serum cholesterol, triglycerides and malondialdehyde decreased; the mRNA expression of transforming growth factor β1 in the kidney tissues was dramaticaly declined in the combined group and BMSC group. To conclude, the combined use of epimedium and BMSCs is superior to their use alone in the treatment of kidney disease.
9.Evaluation of storage performance of special plastic blood bags for apheresis platelets

Objective To evaluate the storage performance of storage bags for apheresis platelets produced by Shandong Weigao Group Medical Polymer Co .,Ltd ( experimental bags ) with Trima set platelet storage bags produced by the U .S. Gambro BCT as the control .Method One unit of apheresis platelets was divided into two equal parts , added to control blood bags and experimental blood bags respectively .All samples were stored at ( 22 ±2 )℃ with consecutive oscillation . The platelets′count, mean volume, aggregate activity (ADP, THR), pH, glucose, lactate concentration, lactate dehydro-genase concentration , hypotonic shock reaction , expression of CD62P and phosphatidyl serine on surface of cell membrane were detected at 0,3,5 and 7 d respectively.Results There was no significant difference in platelet quality after five days of storage between the experimental group and the control group (t-test, P>0.05).Conclusion Two types of platelet stor-age blood bags have similar storage performance for apheresis platelets .
10.Computation of mutual information in medical image registration based on mutual information.

Registration based on mutual information is a typical method in medical image registration. Mutual information is a common similarity measure in image registration, which has excellent robustness and accuracy, but large calculation amount makes it difficult to be applied to clinics. In this paper, a registration algorithm based on multi-resolution and hybrid optimization is adopted to implement 2-dimension monomodal and multimodal registrations of MRI and CT images of human heads with different numbers of gray bins. Results of experiments show that registration precisions have not notable change with 32, 64 gray bins, compared with 256 gray bins, whereas the computation costs decrease remarkably.
