1.A study on the expression of MTSS1 in hepatocellular carcinoma

Objective To explore the expression and significance of MTSS1 ( metastasis suppressor 1) in hepatocellular carcinoma.Methods MTSS1 expression was detected by immunohistochemistry in hepatocellular carcinoma, liver cirrhosis and normal liver tissues.Single-factor analysis was used to study the relationship with clinicopathological factor.Correlations between MTSS1 expression and TNM stage were analyzed with Spearman rank correlation analysis.Postoperative 5-year survival was evaluated using Kaplan-Meier survival curve analysis.Results The expression of MTSS1 in hepatocellular carcinoma was higher than normal liver tissue ( U = 168.000, P < 0.05), and liver cirrhotic tissue ( U = 106.000, P < 0.05); MTSS1 expression was correlated with TNM stage of hepatocellular carcinoma patients, lymph vascular invasion and tumor capsule ( separately U = 259.000, 258.500, 202.000, all P < 0.05).MTSS1 expression in hepatocellular carcinoma was not correlated with patients age, gender, tumor size, AFP level, and hepatitis B surface antigen.MTSS1 expression and TNM stage of liver cancer patients was negatively correlated ( rs = - 0.383 , P < 0.05 ).Postoperative 5-year survival of hepatocellular carcinoma patients with MTSS1 positive expression was significantly poorer than patients with negative and weakly positive expression (respectively 34.1% and 52.3% , x2 =6.386, P < 0.05).Conclusions MTSS1 high expression may play an important role in the early hepatocellular carcinoma progression, indicating a poor prognosis.
2.Detection of West Nile Virus Using SYBR GreenⅠ Fluorescent Quantitative PCR Assay

OBJECTIVE To establish SYBR Green Ⅰ fluorescent quantitative PCR assay for the detection of West Nile virus(WNV),which could be used for early laboratory diagnosis.METHODS A fragment of WNV gene was amplified by PCR,then cloned into pMD-18 T vector.The combinant plasmid was sequenced and analyzed by means of BLAST program,and used as the positive DNA in place of WNV.The SYBR GreenⅠfluorescent quantitative PCR assay was established based on positive plasmid.The sensitivity and specificity of the assay were performed.RESULTS The combinant plasmid was confirmed by sequencing and the fragment belonged to WNV.Ten copies of WNV RNA were detected by SYBR GreenⅠfluorescent quantitative RT-PCR assay.Results of the other members of Flaviviridae were negative,which indicated this assay was specific for WNV.CONCLUSIONS SYBR GreenⅠfluorescent quantitative PCR assay established in this study is highly sensitive and specific,and so it can be used for early diagnosis of WNV infection.
3.Effect of SIRT6/NF-kappa B signal axis during hematopoietic stem and progenitor cell senescence induced by tert-butylhydroperoxide in vitro

BACKGROUND:SIRT6/NF-κB is the important signal axis to cellsenescence, but the effect of SIRT6/NF-κB signal axis to hematopoietic stem and progenitor cell(HSC/HPC) senescence is unclear. OBJECTIVE:To induce (t-BHP) in vitro and to investigate the role of SIRT6/NF-κB signal axis in Sca-1+HSC/HPC senescence induced by tert-butylhydroperoxide in vitro. METHODS:Sca-1+HSC/HPC was isolated and purified by magnetic activated cellsorting. Sca-1+HSC/HPC senescence was induced by 100μmol/L tert-butylhydroperoxide in vitro. The senescence-associatedβ-Galactosidase staining, cellcycle analysis and culture of mixed hematopoietic progenitor cellwere used to investigate the biological effects of tert-butylhydroperoxide on Sca-1+HSC/HPC senescence. The expression of senescence associated SIRT6, NF-κB mRNA and protein was examined by real-time fluorescence quantitative PCR and western blot assay. RESULTS AND CONCLUSION:Compared with control group, the percentage of positive cells expressing SA-β-Gal and cells in G0/G1 phase increased and the number of forming colony of mixed hematopoietic progenitor decreased in the aging group. It showed lower expression of SIRT6 and higher expression of NF-κB in the aging group. The SIRT6/NF-κB signal axis may play a key role in the Sca-1+ HSC /HPC senescence inducted by tert-butylhydroperoxide.
4.Effect of YunKe on bone metabolism of patients with rheumatoid arthritis

Objective To evaluate the effect of YunKe on bone metabolism of patients with rheumatoid arthritis (RA). Methods A total of 108 RA patients were divided into YunKe group (n = 72), non-YunKe group (n = 36), and 54 controls. The bone mineral densities (BMDs) of the wrists, hips and lumbar spines were measured by dual-energy X-ray absorptiometry. Moreover , the expression of cross-linked N-telopeptides of type I collagen (NTX) in the serum was detected for statistically analyzing the difference between YunKe group and non-YunKe group. Results In RA group, the BMDs of wrist, hip and lumbar spine declined compared with that of control group (P < 0.001, respectively). In YunKe group, the BMDs of wrist, hip and lumbar spine after 6 months all were improved with statistically significance (P < 0.01, respectively); The differences also were statistically significant when compared with those in non-YunKe group (P < 0.01, respectively). In YunKe group, NTX level in the serum declined after 3 months of the treatment. The difference was statistically significant when compared with that in non-YunKe group (P < 0.001), but the difference disappeared 6 months later. Conclusions The BMD of wrist joints is low and declines in RA patients significantly. The treatment withYunKe improves the BMDs of the wrist , hip and lumbar spine and decreases the serum NTX. This study proposed that YunKe could inhibit the chronic loss of bone mass induced by RA.
5.Effect of family supporting system on psychological emotions and quality life of leukemia children′s parents

Objective To investigate the effect of family supporting system on psychological emotion and quality life of leukemic children′s parents. Methods Fifty-eight parents of children with leukemia hosptalized from Juanary to December 2013 were set as control group, which received traditional education. Fifty-nine parents of children with leukemia hospitalized from Juanary to December 2014 were set as observation group which received family supporting system. The survey was done separately before intervention and 6 months after treatment by depression self rating scale (SDS), anxiety self-assessment scale (SAS) and SF-36 (the MOS 36 items short form health survey). Result There were statistical significances in the scores on SDS, SAS and SF-36 as compared those with the pre-intervention (P<0.05). Conclusion The leukemic children parents supporting system can effectively reduce the parents′depression and anxiety and improve their quality of life.

7.Imaging appearances of pulmonary mucosa-associated lymphoid tissue type lymphoma

Objective To assess the value of CT or PET-CT with fluorine-18-labeled fluorodeoxyglucose (FDG) for the diagnosis of pulmonary mucosa-associated lymphoid tissue (MALT) type lymphoma. Methods The CT or FDG PET-CT findings in 14 patients with pathologically proved pulmonary MALT lymphoma were retrospectively analyzed. Results Lung lesions were unilateral in 7 patients and bilateral in 7 patients. Lesions presented as a single mass in 3 patients, as a single consolidation in 3 patients, as a nodule in 1 patient, as multiple nodules in 1 patient, as multiple patchy consolidations in 4 patients, as a mass with multiple nodules and patchy consolidations in 1 patients, as diffuse interstitial change in 1 patients. Air bronchogram was found in 9 patients and CT angiogram sign in 5 patients. On PET-CT, lesions showed heterogeneous FDG uptake in 2 patients, maximum standard uptake value was higher than 2. 5. Conclusion Imaging characteristics of pulmonary MALT lymphoma are single or multiple nodules or consolidations with air bronchogram on CT, and heterogeneous high FDG uptake on PET-CT.
8.Association of peptidylarginine deiminase Ⅳ gene polymorphisms and its expression with rheumatoid arthritis in Chinese patients

Objective The current study is aimed to investigate the association of single nucleotide polymorphisms and gene expression in peptidylarginine deiminase Ⅳ (PADI4) with rheumatoid arthritis (RA).Methods The gene expression of PADI4 was examined using real-time quantitative reverse transcription-polymcrase chain reaction (RT-PCR) in 70 patients with RA and 81 healthy controls. Four exonie SNPs of the PADI4 gene (PADI4_89, _90, _92, _104) were genotyped using DNA sequencing and TA cloning.Results The distribution of PADI4_89, _90, _104 SNPs in RA was different from that of healthy controls. The increased RA susceptibility was significantly associated with minor alleles. When haplotypes were construe -ted with 4 SNPs, two major haplotypes, ACCC and GTGT were found in all samples, and GTGT haplo-type (carrying only the minor alleles) was significandy associated with increased RA susceptibility (P<0.01)in comparison with the reference haplotype ACCC. There was over expression of PADI4 in RA than controls(P<0.05). C, enotypes carrying the minor alleles had higher expression level of PADI4 in RA and controls than those with the common alleles. Conclusion PADI4 SNPs and haplotypes are associated with RA susceptibility in Chinese. PADI4 is over-expressed in the blood of RA patients, and there is significant association between the haplotypes and expression level of the PADI4 gene.
9.Design,preparation and screening of siRNA of proliferating cell nuclear antigen

AIM: To design,prepare and screen out functional small interfering RNA(siRNA) for specifically silencing proliferating cell nuclear antigen(PCNA) gene expression and effectively inhibiting cell proliferation on human hepatoma cell line SMMC-7721,human gastric carcinoma cell line SGC-7901 and human colorectal carcinoma cell line Caco2.METHODS: PCNA siRNA was designed based on previous studies about design guidelines and synthesized in vitro by T7 Mega short script reaction kit according to the manufacturer's instructions.After purification,the integrities of siRNA were identified through 19% denaturing polyacrylamide gel electrophoresis,and the concentrations of the generated siRNA were measured by testing the absorbance at 260 nm using a spectrophotometer.Four synthesized double-strand siRNA were transfected into three types of carcinoma cell lines by LipofectamineTM2000 reagent,respectively.WST-8 assay was employed to examine the proliferative inhibitions of these three cell lines.The subsequent alterations on PCNA mRNA and protein levels were determined by semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR) and immunocytochemistry,respectively.RESULTS: 3 sequences,No.2,No.3 and No.4 PCNA siRNA showed effective inhibitions on tumor cells among the 4 candidate siRNA,and a single dose of 50 nmol/L of No.2,No.4 PCNA siRNA showed the most effective inhibitory rates as more than 62% at 48 h after transfection.50 nmol/L of No.2 and No.4 PCNA siRNA transfection caused 72% decrease of PCNA mRNA level and almost completely loss of the protein in human Caco2 cells.CONCLUSION: No.2 and No.4 PCNA siRNA have been screened out in this study,which show the capability to effectively down-regulated PCNA expression and significantly inhibit the proliferation of carcinoma cells.The optimal concentration is 50 nmol/L and satisfactory effects are achieved 48 h after transfection in vitro.
10.Diagnosis of BI-RADS 4 breast lesion using contrast-enhanced ultrasonography

Objective To assess the value of contrast-enhanced ultrasonography (CEUS ) in diagnosing BI-RADS 4 breast lesion.Methods The CEUS findings of 79 breast lesions of BI-RADS 4 were analyzed,and the CEUS diagnosis was made according to our preliminary study results.Histology results were served as golden reference.Results Of all the 79 breast lesions that were confirmed by histopathology results,36 were malignant and 43 were benign.The differences of morphologic features of area variety, margin,heterogeneous or homogeneous distribution between benign and malignant lesions were significant (P < 0.05 ).The area under ROC curve of correction BI-RADS classification in the diagnosis of breast cancer was 0.938,and it was significantly higher than that of the BI-RADS classification (0.889,Z =2.209, P =0.0272).Conclusions CEUS can improve the diagnostic accuracy of breast lesions preoperatively.
