1.Angle of acetabular cup in total hip replacement affects the safety of joint flexion and extension

BACKGROUND:Major complication after total hip replacement was instability in the form of dislocation. The probability of above complications has a great relationship with the mistakes of the angle of acetabular cup prosthesis. OBJECTIVE:To explore the influence of angle of acetabular cup in total hip replacement on the safety of joint flexion and extension. METHODS: A total of 60 patients, who underwent total hip replacement in the Wujin Hospital Affiliated to Jiangsu University from January 2012 to December 2014, were enroled in this study. Three dimensional directional monitoring device, CT scanning and image processing tools were used to preset the angle of the acetabular cup and to set abduction angle and anteversion angle. Test data were recorded and subjected to statistical analysis. RESULTS AND CONCLUSION:Acetabular cup was implanted under five different conditions: abduction angle 45°, anteversion angle 15°; abduction angle 60°, anteversion angle 15°; abduction angle 30°, anteversion angle 15°; abduction angle 45°, anteversion angle 5°; abduction angle 45°, anteversion angle 25°. According to statistics, in the patients with anteversion angle of 15°, when the acetabular cup was placed, the bigger the abduction angle, the bigger the range of abduction angle of the acetabular cup induced by pelvic tilt was. If the anteversion angle increased, the anteversion angle of the acetabular cup was reduced. With the increased pelvic tilt angle, the anteversion angle of the acetabular cup was big. Moreover, in patients with anteversion angle of 15°, the anteversion angle of the acetabular cup should keep consistent. When the abduction angle of the acetabular cup increased, the range of anteversion angle of the acetabular cup induced by pelvic tilt was diminished. These data showed that the abduction angle and anteversion angle of the acetabular cup have mutual restriction. When the acetabular cup was placed, big anteversion angle (15±10)° can wrap the femoral head, reduce the incidence of dislocation, and make flexion and extension safe.
2.Purification of anti-HBcAg monoclonal antibodies using immobilized metal ion affinity chromatography.

Anti-HBcAg monoclonal antibodies from mouse ascites were purified by using immobilized metal ion affinity chromatography. We optimized the conditions of sample loading and elution. The results showed that when the pH stepwise elution was used, the best solution for sample loading was 20 mmol/L phosphate buffer containing 0.5 mol/L sodium chloride at pH 8.0 and the mAb was eluted at pH 5.0. The purity of obtained mAb was more than 85% and recovery reached 80%. When the adsorbed proteins were eluted by using gradient elution of an imidazole, the best solution for loading condition was 20 mmolL phosphate buffer containing 5 mmol/L imidazole at pH 8.0. The purity and recovery of antibody were up to 95%.

3.Effect observation and nursing of different doses of simvastatin in the treatment of acute myocardial infarction

Objective To observe the effect of different doses of simvastatin in the treat-ment of acute myocardial infarction,and to explore the nursing countermeasures.Methods A total of 86 patients with acute myocardial infarction in our hospital were as the research objects,and were randomly divided into 2 groups,and all the patients used simvastatin on the basis of conventional treatment.A group with 43 cases was given simvastatin dosage for 20 mg,B group with 43 cases was given simvastatin dosage for 40 mg.Both 2 groups during the treatment were given comprehen-sive nursing intervention.The treatment effect of 2 groups was compared.Results After treat-ment,the total efficiency in A group was 95.4% and 81.4% in B group,the difference in the total efficiency between 2 groups was significant (P <0.05).Before treatment,there was no significant difference in TC,TG,LDL-C and CRP (P >0.05)and the index of group B after the treatment was better that that in the A group (P < 0 .0 5 ).The incidence rate of cardiovascular events was 25.6% in A group and 9.3% in B group,and the incidence rate of cardiovascular events in B group was significantly less than that that in A group (P <0.05).Adverse reaction rate was 11.6% in A group and 16.3% in B group,there was no significant difference between 2 groups (P >0.05). Conclusion The curative effect of higher dose simvastatin in adjuvant treatment of acute myocar-dial infarction is better and it can reduce cardiovascular adverse events,so it is worthy of applica-tion.

5.Nerolidol inhibits malignant biological behaviors of melanoma cells by regulating the Wnt-β-catenin pathway

[摘 要] 目的:探讨橙花叔醇通过Wnt-β-catenin通路抑制黑色素瘤A-375和WM-115细胞恶性生物学行为的分子机制。方法: 体外培养黑色素瘤细胞A-375和WM-115,用不同浓度的橙花叔醇处理,采用SRB法和克隆形成实验、FCM术、Transwell实验和细胞划痕实验、DCFH-DA染色法、qPCR和WB法分别检测橙花叔醇对A-375和WM-115细胞的增殖能力、细胞周期和凋亡、迁移能力、活性氧(ROS)水平和Wnt-β-catenin通路及其下游相关基因和相关蛋白表达的影响。利用ULCAN和GEPIA2数据库分析黑色素瘤中Wnt-β-catenin通路的激活与患者预后的关系。结果:与对照组比较,橙花叔醇处理组A-375和WM-115细胞的增殖能力受明显抑制(均P<0.01)、细胞周期阻滞于G2/M期(P<0.05或P<0.01)、细胞凋亡率增加(均P<0.01)、迁移能力降低(P<0.05或P<0.01)、ROS水平升高(均P<0.01)、Wnt-β-catenin通路被抑制而其下游基因和蛋白表达明显上调(均P<0.01)。数据库数据分析显示,WNT1基因高表达患者OS低于低表达患者(P<0.01)。结论:橙花叔醇通过上调A-375和WM-115细胞中ROS水平影响Wnt-β-catenin通路,从而抑制其恶性生物学行为;Wnt-β-catenin通路可能是黑色素瘤治疗的潜在靶点。
