1.Barrier function of porcine retinal pigment epithelial cells cultured in vitro

Objective To investigate the blood-retinal barrier(BRB)function of porcine retinal pigment epithelial(RPE)cells cultured in vitro. Methods Primary porcine RPE cells were cultured,and the third generation were inoculated in a microporous filter with the filter membrane of polyvinylpyrrolidone(PVP)-free polycarbonate membrane.After 1,2,3 and 4 weeks of culture,the surface of filter membrane was observed by light microscope,and after 2 weeks of culture,the section of filter membrane was observed by light microscope and transmission electron microscope.Transepithelial electrical resistance(TER)was detected and the permeability was measured with fluorescein sodium(FS)and horseradish peroxidase(HRP). Results Primary porcine RPE cells were cultured successfully.RPE cells converged1week after inoculation; 2 and 3 weeks after inoculation,the density of RPE cells did not changed obviously; 4 weeks after inoculation,the density of RPE cells decreased.The characteristics of polarized growth of monolayer were found in RPE cells on the surface of filter membrane; 2 weeks after inoculation,the TER of RPE cells was(97.44?11.36)?/cm~2 which maintained till the 3rd week after inocubation.After incubated for 30 minutes,only 0.27% of FS and 0.17% HRP reached the inferior filter membrane,and the permeability rate of SF with low molecular weight was higher than which of HRP with high molecular weight. Conclusions The filter with PVP-free polycarbonate membrane may be used to set up the model of RPE cells with polarized growth of monolayer and investigate the barrier function of RPE cells.
2.Wedge-shaped autograft reconstructing deformity of severe varus knee in primary total knee arthroplasty

[Abstrct] Objective The deformity of severe varus knee (varus angulation > 15°) can be treated by many ways in primary total knee arthroplasty , but the efficacy and cost are different. The topic of this study provides a new surgery: autograft (wedge grafts) implant and substitute medial tibial bone defects. Methods From Jan. 2006 to Jan. 2014 , a retrospective study was conducted in the First Affiliated Hospital of BaoTou Mendical College. Out of 687 patients, 721 primary knees were operated. Bone defects in proximal tibia (varus angulation >15° ) were encounted in 16 patients in 21 knees in the pattern of non-inclusive slope bone defects in the mean varus degree of 27°. Their IBM was 31 kg/m2. The average age of patients was (63 ± 5) years. The score of clinical knee society score (KSS) before operation was 18 on average. Among of them, the pain score were 12 points, the average range of motion (ROM) were 12 points. the average funtional score were 20 points. During operation,the slope bone defect of the medial tibial plateau was tailored into wedge-shaped ladder bone defect by osteotomy,and then the defect was filled with the resected tibial plateau autograft in which the thickness and shape were matched completely. The high-intensity cortical part of the autograft was placed in the rim to sustain the tibial prosthesis. Three months after operation ,patients started to walk without the help of crutch. Results The average followup period was 5.3 y , We observed that an average postoperative scores of clinical KSS from 42 points to 88 points with the score of functional KSS from 57 points to 92 points. Fusion was observed by trabeculae crossing from the graft to the tibial metaphysis on plain films at least 6 mouths without collapse, stress fracture, untight and bone ununion. Confirmation of vascularization of the graft was observed in bone scans of nuclide. showing increased uptake in the operated area. Inflammatory makers were normal. Conclusion Autograft (wedge grafts) reconstruction of medial tibial bone defects, is of high rate of healing in implant bones, good restoration of the function and low cost.
3.Expression and signifance of TRIM59,Twist and E-cadherin in hepatocellular carcinoma

Objective To investigate the expressions of TRIM59,Twist and E-cadherin in hepatocellular carcinoma and their clinical significance. Methods The expressions of TRIM59,Twist and E-cadherin protein were tested by immunohistochemistry in 80 cases of hepatocellular carcinomas and the adjacent paracancerous tissues. Results The positive rate of TRIM59 in hepatocellular carcinoma was significantly higher than that in the adjacent paracancerous tissue(76.3%vs. 8.0%,P<0.05). Significant difference was also observed in the expres-sion rate of Twist between the hepatocellular carcinomas and the paracancerous tissue(66.3%vs. 6.0%,P<0.05). The positive rate of E-cadherin in hepatocellular carcinoma was significantly lower than that in the adjacent para-cancerous tissue(27.5%vs. 90.0%,P<0.05). The differences of the expression of TRIM59 in hepatocellular carci-noma of pathological grading,tumor differentiation,vascular invasion and clinical TNM stage were significant(P<0.05). The differences of the expression of Twist in hepatocellular carcinoma of pathological grading ,differentia-tion,vascular invasion and clinical TNM stage was also significant(P<0.05,respectively). The differences of the expression of E-cadherin in hepatocellular carcinoma of pathological grading,differentiation,vascular invasion and clinical TNM stage were also significant(P<0.05,respectively). Significantly positive correlation was also found between TRIM59 and Twist by using spearman correlation analysis(P<0.05). Negative correlations were observed between TRIM59 and E-cadherin(P < 0.05),and between Twist and E-cadherin(P < 0.05). Survival analysis showed that TRIM59 expression was an independent prognostic factor in hepatocellular carcinoma. Conclusion TRIM59,Twist and E-cadherin protein expression might be associated with the development,invasion,and metas-tasis of hepatocellular carcinoma. TRIM59 may become a new target gene for the treatment of human hepatocellular carcinoma.
4.Effects of levodopa on nerve growth factor in visual cortex of kitten

Objective Nerve growth factor (NGF) is thought to be one of regulating factors of cellular function during the visual plasticity period. Levodopa is a neurotransmitter in central nerve system. Its mechanism of improving the visual function in amblyopia is unclear. Present study was to observe the role of levodopa in treating monocular deprived amblyopia and explore the effects of levodopa on the expression of NGF in visual cortex. Methods The animal models of monocular form deprived amblyopia were established in the right eyes of 12 4-week-old domestic cats by suturing the eyelid for 2 weeks and then the left eyes were reverse sutured. The 6 cats from models received 20 mg/kg of levodopa by oral administration. Other 6 normal cats were as normal controls. Pattern VEP (P-VEP) was recorded to check the visual acuity of the cats. The cell number density of NGF expression in visual cortex area 17 was detected and calculated by immunochemistry and expressed as positive number/field. Results The amplitude of P_(100) of P-VEP was significantly declined in monocular deprived eyes compared with fellow eyes and normal eyes in 2 weeks after monocular deprivation (P <0. 01) . However, after administration of levodopa, the amplitude of P_(100) recovered to normal 2 weeks later, presenting a insignificant difference between levodopa treating group and normal group (P> 0. 05). The latency of P_(100) was followed the same pattern between two groups. The NGF positive cells were decreased in monocular deprived group compared with normal group and levodopa group (80. 23 ± 9. 54 vs 111. 83 ± 7. 49, 80. 23 ± 9. 54 vs 118. 06 ± 12. 37, P < 0. 01), but no significant difference in NGF positive cells was found between normal group and levodopa group (111.83 ±7. 49 vs 118. 06 ± 12. 37, P = 0. 94). Conclusion Levodopa is supposed to accelerate the recovery of visual function in amblyopic cat by enhancing the NGF expression in visual cortex area 17 during the critical period of visual development.
5.Effects of Different Acupuncture Therapies on Serum BACC and F-Trp in High-intensity Military Training Soldiers

Objective To investigate the effects of different acupuncture therapies on serum branched-chain amino acids (BAC and free tryptophan (F-Trp) in high-intensity military training soldiers. Methods Ninety healthy soldiers were randomly allocated to groups A, B and C, 30 soldiers each. Group A received treatment with electroacupuncture at Zusanli; group B, treatment by cupping on Zusanli;group C, no treatment. The three groups did a high-intensity 5 km running exercise after two weeks of treatment. Serum BACC and F-Trp contents were measured in the three groups before and at the end of treatment and at five days after treatment. Results It can be seen from table 2 that there were no statistically significant pre-treatment differences in serum BACC and F-Trp contents between the three groups of soldier subjects (P>0.05); there were statistically significant pre-/post-treatment differences in serum BACC and F-Trp contents in the three groups of soldier subjects (P<0.05). There was a statistically significant post-treatment difference in serum BACC content between group A or B and group C (P<0.05). There was a statistically significant difference in serum F-Trp content between group A or B and group C at the end of treatment and at five days after treatment (P<0.05). Conclusion Acupuncture therapy can effectively improve bodily anti-fatigue ability and reduce high-intensity exercise induced muscular injury in military training soldiers.
6.Homeobox genes CDX1 and CDX2 reduced malignancy of esophageal adenocarcinoma cells

Objective To study the effect of homeobox genes CDX1 and CDX2 on the phenotype of esophageal adenocarcinoma cells.Methods Esophageal adenocarcinoma cell line SEG-1 was transfected with CDX1 or CDX2 cDNA.The morphology,growth rate,division index and tumorigenicity were analyzed.Results The expression of CDX1 or CDX2 leaded to occurring adeniform-like manifestation.The growth rate,division index and tumorigenicity were reduced,especially by CDX2.Conclusion CDX1 and CDX2 all could increase differentiation of esophageal adenocarcinoma cells and reduction of its malignancy.
7.High expression of PIK3CB is an independent prognostic predictor for gastric adenocarcinoma

Objective To analyze the correlation between the expression of phosphoinositide-3 kinase,catalytic subunit beta (PIK3CB) and the clinicopathologic characteristics in gastric adenocarcinoma tissues.Methods Immunohistochemistry was used to determine PIK3CB expression in human gastric adenocarcinoma tissues and matched adjacent tissues.The correlation between PIK3CB expression and the patients' clinicopathological characteristics and prognosis was statistically analyzed.Results PIK3CB is highly expressed in gastric adenocarcinoma tissues (positive rate 63.8%) compared with matched adjacent tissues (positive rate 45.7%) (x2 =9.139,P =0.003).PIK3CB expression is significantly correlated with tumor size (x2 =8.677,P =0.003),infiltration (x2 =9.306,P =0.025),and clinical stage (x2 =8.242,P =0.041) ; Level of PIK3CB expression has a significant impact on prognosis (x2 =25.644,P =0.000).Multivariate analyses showed that high PIK3CB expression is an independent poor prognostic factor for overall survival (HR =15.785,95% CI:1.943-7.805,P =0.000).Conclusions Upregulated PIK3CB is correlated with poor postoperative prognosis of gastric carcinoma.
8.Study of the Antifatigue Mechanism of Point Zusanli Acupuncture in High-intensity Military Training Soldiers

Objective To investigate the effect of point Zusanli acupuncture on serum creatine kinase (CK) and creatine kinase isoenzyme (CK-MB) in high-intensity military training soldiers and explore its possible antifatigue mechanism.Methods Sixty healthy soldiers were randomly allocated to groups A and B, 30 each. Group A received acupuncture at point Zusanli and group B, no treatment. After two weeks of treatment, two groups of soldiers separately did 5 km high-intensity running training and fulfilled the requirement of army examination. CK and CK-MB were measured in two groups before and after training and five days after training.Results In the two groups, there were statistically significant differences in serum CK and CK-MB contents between before training and after or five days after training (P<0.05). After or five days after training, there were statistically significant differences in serum CK and CK-MB contents between groups A and B (P<0.05).ConclusionsAcupuncture at point Zusanli can effectively improve bodily antifatigue ability and reduce myocardial and skeletal muscle injury caused by high-intensity exercise in military training soldiers.
9.Effects of Transcutaneous Electric Acupoint Stimulation at Zusanli on Fatigue after Training

Objective To observe the effects of transcutaneous electric acupoint stimulation(TEAS) at zusanli on fatigue after training.Methods 40 soldiers with exercise fatigue after marine training were divided into 2 groups.The soldiers in treatment group were treated with TEAS on Zusanli(ST36).Those in control group recovered naturally.Results The fatigue symptoms were improved,the consistency of the hemoglobin(HGB) were significantly increased,and the levels of lactic acid(LA) were significantly decreased in treatment group(P<0.01),which seemed better than that of the controls(P<0.05).Conclusion TEAS can facilitate the recovery from fatigue after training.
10.Expression and clinical significance of Survivin and COX-2 in human ampullary carcinoma

Objective To study the expression of Survivin and COX-2 in ampullary carcinoma and their clinical significance.MethodsThe expression of Survivin and COX-2 proteins were tested by EnVision immunohistochemistry in 40 cases of ampullary carcinomas,and 8 cases of normal ampulla of vater as the controls.ResultsThe positive rate of Survivin in ampullary carcinonas was significantly higher than that of the controls(82.5% vs 0,P < 0.01 ). The expression of Survivin in ampullary carcinoma was correlated with duodenal invasion,pancreatic invasion and lymph node metastasis ( P < 0.05 ).Significant difference was also observed in the expression rate of COX-2 between the patients with ampullary carcinoma and the normal controls (67.5% vs 0,P < 0.01 ).The expression of COX-2 in ampullary carcinoma was correlated with duodenal invasion,pancreatic invasion and lymph node metastasis (P < 0.05). Significantly positive correlation was found between the expression of Survivin and COX-2 by using spearman correlation analysis ( r =0.383,P =0.015).ConclusionThe specific up-regulation of COX-2 gene and Survivin gene may play an important role in the genesis and development of ampullary carcinoma.COX-2 and Survivin may be used as early diagnosis markers and potential therapeutic targets in ampullary carcinoma.
