1.Dose of expression and study of the theory for adverse reactions of cefamandole nafate for injection- Gold Zone Method

Objective Inaccurate dosing is an important cause of medical malpractice, how to adjust the medication dose to overcome the excessive deviation will be discussed in this paper. Methods Gold zone method was proposed on the basis of the probability theory and mathematical statistics for handling inaccurate dosing problems. Investigation and study in large sample size was carried out on adverse reactions of Cefamandole Nafate for Injection by adopting the method. Results The conclusions from the super large sample size test indicate that there is highly significant difference of total adverse reaction rates between taking values within and outside golden zone of dosage, and the adverse reaction rate of taking values within golden zone is significantly lower than that of outside golden zone. It is noteworthy that when dosage was lower than golden zone,the adverse reaction rate was significantly increased,which is more than 4 times of that within golden zone. Conclusion It is effective measures and simply and easy to apply that taking values within golden zone of dosage reduce adverse drug reactions,probability within golden zone less than 1/4 range of experience that contains the actual demand of the body up to 70%.
2.Callus induction and cultivation of Phyllanthus urinaria

Objective To establish culture method for Phyllanthus urinaria. Methods To study the possible effective factors of culture condition by comparing with different explants,sucrose,plant growth substance,and its ratio. Results The inductivity of stem was the highest about 55.56%,but callus of leaves could not be induced.6-BA was the most important factor on callus induction,followed by 2,4-D NAA and sucrose. Conclusion The optimal medium to induce and subculture is MS+2,4-D 0.5 mg/L+6-BA 1.0 mg/L+NAA 0.1 mg/L+sucrose 10 g/L.
3.Effect of hypoxia and reoxygenation on the expression of a disintegrin and metalloprotease10,?-amyloid precursor protein cleaving enzyme mRNA in SH-SY5Y cells

Objective To study the effects of hypoxia and reoxygenation on expression of a disintegrin and metalloprotease10(ADAM10),?-amyloid precursor protein cleaving enzyme(BACE1) mRNA in SH-SY5Y cells.Methods SH-SY5Y cells were grouped into hypoxic and control groups.The hypoxic cells were incubated in hypoxic condition(95%N_2,5%CO_2) for 12 hours and 24 hours,and then cells were reoxygenated for 0 hours,12 hours and 24 hours.The expression of ADAM10,BACE1 mRNA were tested respectively at different time points after reoxygenation by RT-PCR.Control groups were incubated in normal conditions,seeded and treated at the same time with the hypoxic cells.Results The expression of ADAM10 mRNA was down-regulated by 19.8%,41.4% and 64.6%(P=0.005,0.038,0.001) at different time after reoxygenation with 12 hours hypoxia and down-regulated by 30.1%,75.9% and 86.5%(P=0.009,0.005,0.043)after reoxygenation with 24 hours hypoxia.The expression of BACE1 mRNA was up-regulated by 31.5% and 35.1%(P=0.028,0.005)only at 12 hours and 24 hours points after reoxygenation with 24 hours hypoxia.Conclusion Hypoxia and reoxygenation might alter the expression of ADAM10 and BACE1,which demonstrates that the vascular factors should make the amyloid precursor protein easy to be processed by ?-secrease pathway,thus to involve the pathology of Alzheimer's disease.
4.Emotion, Personality in Students with Menstrual Disorders

Objective: To investigate the incidence of menstrual disorder and the relationship between menstrual disorders and emotion, personality et al psychosocial factors in students. Methods: The study adopted stratified sampling method. 521 students were subjected to questionnaire of MHQ, STAI, CED-S and EPQ. Results: The rate of menstrual disorder was 44.7%. The average score of anxiety and depression in stress was significant higher than usual. The increasing trend of incidence of menstrual disorder accompanied with age, reaction degree of anxiety and depression and the level of trait anxiety in stress. Conclusion: The psychosocial factors such as emotion and personality play an important roles in the process of resulting in menstrual disorder. The doctors should pay more attention to strengthen the psychological interventions.
5.Mutation of the ret proto-oncogene in two Chinese families with multiple endocrine neoplasia type 2A(MEN2A).

Objective To detect ret mutations in two Chinese families with multiple endocrine neoplasia type 2A(MEN2A).Methods In our study,9 members from these two families including 3 patients were recruited.Polymerase chain reaction(PCR)and direct gene sequencing of PCR products by an automated DNA sequencer were applied to scan the entire 21 exons of ret proto-oncogene in the leukocyte DNA of the subjects.Results Two missense mutations were detected in exon11 of the ret proto-oncogene.One was C634R in the 2 patients from a family and the other was C634Y in 2 subjects from another family(one was the patient).Two heterozygous variants(A45A and L769L)in these 4 subjects were revealed.Conclusion Two mutations(C634R and C634Y)are detected in two families with MEN2A.Direct DNA sequencing analysis can diagnose MEN2A at gene level,which is helpful in making clinical management of the disease and in diagnosing earlier in their offspring.
6.Effect of fluctuant high glucose to pancreatic β-Cell lines INS-1

Objective To investigate the damage mechanism of fluetuant high glucose on the INS-1 cells (pancreatic β-cell lines).Methods The cells were divided into five groups:the control groups (A group:5.5 mmol/L of glucose),the continuing high glucose group (B group:16.7 mmoL/L of glucose),the fluctuant glucose group ( C group:16.7 mmol/L of glucose for cultivation for 2 h,then the concentration changed to 5.5 mmol/L for cultivation for 3 h,which was repeated 3 times per day;the ceils were kept in the medium containing 5.5 mmol/L of glucose during night time for 9 h),the continuing high glucose plus NAC ( 1.0 mmo/L) group ( D group),the fluctuant glucose plus NAC group ( E group).The intracellular reactive oxygen species (ROS) was observed by the flow cytometry.The activity of glucose-6-phosphate dehydrogenase (G6PD) was estimated by the tetrazolium linked cytochemical method.Results 72 h after intervention,the levels of ROSwere 37.77±2.31,86.97±7.97,124.27±10.04,60.92±2.61 and 51.47±3.36,respectively,in A~E group;the activities of G6PD were 1.25±0.03,1.09±0.02,1.03±0.01,1.12±0.02 and 1.21±0.01,respectively;the levels of NADPH were (0.123±0.003) mmol/mg prot,(0.112±0.004) mmoL/mg prot,(0.099±0.002 ) mmol/mg prot,( 0.116±0.005 ) mmol/mg prot and ( 0.120±0.002) mmol/mg prot,respectively.The level of ROS in the cells of the fluctuant glucose group were significantly higher than that in the continuing high glucose group ( P < 0.01 ).The G6PD activity and NADPH was significantly lower in fluctuant high glucose group than those in the continuing high glucose group (P <0.01 ).NAC co-cultivation decreased the extent of cell's change.Conclusions Exposure of INS-1 to high glucose lead to increased oxidative stress, possible mechanism included decreased G6PD activity and subsequent imbalance between oxidation and reduction.
7.Comparison of gelatin and loctated Ringer's solution on the efficacy of volume expansion during hemorrhagic shock in dogs by volume kinetics

Objective To compare the time course of distribution and elimination of gelatin and lactated Ringer's solution (LR) by volume kinetics and mass balance analysis during hemorrhagic shock in dogs, and try to design and optimize fluid therapy in a more scientific manner. Methods Twenty dogs were randomly divided into 4 groups: CL group, CG group, BL group, and BG group. Each animal was subjected to two randomly ordered experiments that separated for at least 1 week. In the first phase, plasma volume expansion was studied in the state of anesthesia, animals received 30 mL/kg of LR (CL group) or 10 mL/kg of gelatin (CG group) over 30 min. In the second phase, plasma volume expansion was studied in the state of hemorrhagic shock, animals received 30 mL/kg of LR (BL group) or 10 mL/kg of gelatin (BG group) over 30 min. Hb concentration and Hct were measured every 5 min during and after infusion for 90 min. Hemodynamic parameters were recorded at the same time. The distribution and elimination of infused fluid were studied by volume kinetics, based on serial analysis of hemoglobin dilution in arterial blood, and by mass balance that incorporated volume calculations derived from volume kinetic analysis and measurements of urinary volumes. Results When a one-volume kinetic model was fitted to the data, the value of V and Kr in CG, BL, and BG group were significantly smaller than those in CL group (P<0.05), which could be found from the computer-generated curves.When a two-volume kinetic model was fitted to the data, the value of V1, Kr, Kt in BL group were significantly smaller than those in CL group (P<0.05). The calculations based on mass balance corresponded to the predicted based on volume kinetics. The change of central volume (CCV) in BL, BG, and CG group was significantly greater than those in CL group (P<0.05). The VEE in BG and CG group was significantly higher than that in BL and CL group. The value of VEE in BL group was significantly higher than that in CL group (P<0.05). Conclusions Both of the efficacy of lactated Ringer's solution and gelatin increased significantly in the state of hemorrhagic shock, and the former increased more.
8.Study on polymorphism in the apolipoprotein A5 gene in patients with premature coronary heart disease

Objective:To investigate the association between the apolipoprotein A5(APOA5) -1131T/C polymorphism and premature coronary heart disease in northern Chinese Han population. Methods: Using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and polyacrylamide gel electrophoresis (PAGE), we analyzed the genotype and allele distribution in 140 patients with premature coronary heart disease diagnosed by coronary angiography and 156 healthy controls. The levels of serum lipid profiles were also studied by biochemical methods. Results: The allele frequency of APOA5-1131T/C polymorphism in the premature coronary heart disease group was significantly higher (43.2% vs. 33.0%, P=0.011) than that in the control group. Compared with TT homozygotes, CC homozygotes exhibited a 2.809-fold (95% CI 1.331-5.927) increased risk of developing premature coronary heart disease. Logistic regression analysis found that this correlation was independent of sex, age, body mass index (BMI), smoking history as well as serum total cholesterol(TC), high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C) levels; In premature coronary heart disease group, the triglyceride(TG) level in CC homozygotes was significantly higher than those in TC heterozygotes or TT homozygotes. Conclusion: The APOA5-1131T/C polymorphism has influence on serum TG level, and the APOA5-1131C allele is associated with the development of premature coronary heart disease in northern Chinese Han population.
9.Research Progress in Medicinal Plant Echinops Latifolius Tausch

Echinops latifolius Tausch is a plant of compositae echinops, whose dried roots are used as traditional Chinese medicine Yuzhou Loulu, and dried inflorescence is used as Mongolian medicine LanCitou. The studies on Chinese medicine Yuzhou Loulu and Mongolian medicine LanCitou in recent years were systemized and compared with each other in the paper, including chemical composi-tion, process research, quality control, pharmacological effects and so on. The results can provide scientific basis for the further studies and utilization of Echinops latifolius Tausch.
10.Role of XBP1 signal transduction pathway in the ischemia/reperfusion injury of graft liver in rats

Objective To explore the regulation mechanism of X box binding protein 1 (XBP1)signal transduction pathway for TNF-α and effective approach in ischemia/reperfusion (I/R) injury of liver transplantation for short hairpin RNA (shRNA) interference used to gene therapy in liver graft.Methods Male Sprague-Dawley rats were divided into three groups: the cold ischemia transfection group (CIT), the in vivo transfection group (IVT) and the control group. Experiments of orthotopic liver transplantation were performed by two cuff method. The rats in CIT were perfused with XBP1-shRNA plasmid (pSIXBP1) during cold ischemia phase, those in IVT received the equivalent volume (2 ml) of pSIIRAK 4 after portal vein inoculation, and those in the control group were not subjected to any treatment. Rats were killed at 60 or 180 min after restoring reperfusion of hepatic portal vein.Histopathological damage degree of graft liver was observed by light microscope. The expression levels of XBP1 gene and protein were detected by RT-PCR and Western blotting. The activities of NF-κB and the serum TNF-α level were detected by ELISA. Results All the indexes including the degree of histopathological damage, the expression levels of XBP1 mRNA and protein and the TNF-α level were significantly decreased in CIT as compared with IVT and control group (P<0. 05). However,there was no significant difference in NF-κB activity among the three groups (P>0. 05). Conclusion The role of XBP1 pathway in TNF-α gene regulation and that of NF-κB pathway in rat liver I/R injury are two relatively independent aspects, and the depression of XBP1 expression with XBP1 shRNA through portal vein perfusion during cold ischemia phase could effectively alleviate graft hepatic I/R
