1.Comparison of Two Methods for Determination of Cyanide in Quality-Controlled Water Sample

Objective To explore an easy, rapid and suitable method for determination of cyanide with lower concentration in quality_controlled water sample. Methods To overcome the shortcomings in determination of low concentration cyanide in water by iso_niacin_pyrazolone spectrophotometry and pyridin_barbituric acid spectrophotometry, the iso_niacin_barbituric acid spectrophotometry was studied for determining the lower concentration cyanide in water, the data obtained were compared with those obtained from the standard method iso_niacin_pyrazdone spectrophotometry. Results The results showed no statistically significant differences between these two methods. The precision, accuracy and other indexes all accorded with the analysis criteria. Conclusion The iso_niacin_barbituric acid spectrophotometry was easier to operate, more rapid and lower cost, the conditions of colorimetry were easier to be controlled compared with those of iso_niacin_pyrazolone spectrophotometry.
2.Establishment of a Bacterial Endotoxin Test for Monitoring Pyrogen in Wilsonii Injection

OBJECTIVE:To establish a bacterial endotoxin test for monitoring pyrogen in wilsonii injection METHODS:Bacterial endotoxin test procedure and guiding principle in Chinese Phamacopoeia were used RESULTS:The test showed that the reaction with limulus agent was not interfered by wilsonii injection with a sensitivity of 0 25Eu/ml CONCLUSION:Limulus lysate method can be used to take the place of rabbit method for monitoring pyrogen in wilsonii injection
3.Preparation and Quality Standard Establishment of Qingrejieduhuazhuo Tablets

OBJECTIVE:To prepare Qingrejieduhuazhuo tablets and establish the standard of quality control.METHODS:The content of baicalin in the product was determined with HPLC method.RESULTS:The linear range of baicalin concentra?tion was0.128~0.642?g/ml and correlation coefficient was0.9997.The average recovery of sample was98.31%with a RSD of0.82%(n=6).CONCLUSION:The method is suitable for the quality control of Qingrejieduhuazhuo tablets.
4.Effects of Storage Condition on Quality of Semen Armeniacae Amarum

Objective To observe the effects of storage condition on the quality of Semen Armeniacae Amarum. Methods Orthogonal design was adopted. The storage condition was optimized with temperature,humidity and storage time as the observation factors,with the content of amygdalin,and the content,acid value and peroxide value of fatty oil as the indexes.The content of fatty oil,acid value and peroxide value were measured according to the methods provided by 2005 edition of Chinese Pharmacopoeia. The content of amygdalin was measured by HPLC. Results Temperature was the main influencing factor of amygdalin content,then came humidity and storage time. Temperature was also the main influencing factor of the concent and the acid value of fatty oil,and the humidity was the second. For peroxide value,humidity was the main factor,then came temperature and storage time. Conclusion The optimal conditions of storing Semen Armeniacae Amarum as follows:storage in dry place with the temperature of 2~ 8 ℃,and in short storage time.
5.Application of swine iPS cell transplantation in porcine myocardial infarction model

Objective To observe the therapeutic effect of porcine myocardium against porcine myocardial infarction (MI) by porcine myocardial infarction model with connexin-43/zonula occludens-1 (CX43/ZO-1) double gene induced pluripotent stem cells (iPS) transplantation.Methods Porcine myocardial infarction model was established by balloon occlusion.One week later,1640 culture medium,CX43 gene ips cells and CX43/ZO-1 gene ips cells were injected into the different coronary arteries.After 3 months,electron microscopy and color Doppler ultrasonography were used to evaluate the curative effect.Results Compared to ips group of CX43 and 1640 culture group,in CX43/ZO-1 gene ips group,electron microscope showed a large number of island-like cardiomyocytes,blood vessels and intact intercalated discs in the infarct area and normal myocardial junction area structure,color Doppler echocardiography showed left ventricular end diastolic diameter reduced,and ejection fraction was significantly increased (P < 0.05).Conclusions Transfection of CX43/ZO-1 double gene ips cells can promote cardiomyocyte regeneration and cardiac function recovery in porcine myocardial infarction model.
6.Determination of the Contents of Berberine Hydrochloride in Spring Rain Burns Gels

Objective To establish an HPLC method for determination the contents of berberine hydrochloride in spring rain burns gels. Methods Analytical column was Kromasil C18 (4.6 mm?250 mm, 5 ?m) column with a mobile phase of Acetonitrile-1%H3PO4-Triethylamine (24∶76∶0.76), the detective wavelength was 345 nm, the flow rate was 1.0 mL/min and column temperature was 30 ℃. Results A good linearity was obtained in the range of 0.088~0.440 ?g (r =0.999 7), and the average recovery was 98.13% with RSD=0.77% (n=6). Conclusion The method is rapid and reliable, and can be used for the quality control of spring rain burns gels.
7.Novel inhibitors against the bacterial signal peptidase I.

New antibiotics with novel modes of action and structures are urgently needed to combat the emergence of multidrug-resistant bacteria. Bacterial signal peptidase I (SPase I) is an indispensable enzyme responsible for cleaving the signal peptide of preprotein to release the matured proteins. Increasing evidence suggests that SPase I plays a crucial role in bacterial pathogenesis by regulating the excretion of a variety of virulent factors, maturation of quorum sensing factor and the intrinsic resistance against beta-lactams. Recently, breakthrough has been achieved in the understanding of three-dimensional structure of SPase I as well as the mechanism of enzyme-inhibitors interaction. Three families of inhibitors are identified, i.e. signal peptide derivatives, beta-lactams and arylomycins. In this article, we summarize the recent advance in the study of structure, activity and structure-activity relationship of SPase I inhibitors.
8.A Rapid SNP-based Identification Method for Distinguishing the Three Origins of Fructus Amomi

Medicinal plants of the Fructus Amomi containing three species (A momum villosum, A momum longiligu-lare, Amomum villosum var. xanthioides)are well-known, which are widely used as traditional medicines. The mor-phological characteristics of the three origins are very similar, especially in the form of seed. In this study, 60 sam-ples of Fructus Amomi were co llected, and 34 sequences of the Fructus Amomi and their adulterants from GenBank were analyzed. Single nucleotide polymorphisms (SNPs) were detected. All the ITS2 sequences here (including our ex-periments and GenBank data)were examined for SNPs at the interspecies level. Results from the study revealed that two stable bases at position 135 bp and 199 bp were found, which could be used as a unique marker to distinguish the three origins of Fructus Amomi. The two SNPs in the ITS2 were found to exist stably between the three species, and all the GenBank sequences of the Fructus Amomi. Our findings indicated that SNP-based DNA barcoding could be used as an efficient method for the rapid and accurate identification of the three origins of Fructus Amomi.

The pharmacognsy of Rhinacanthus nasutus (L.) Linclau was studied by microscopic ob-servation and UV spectroscopy to provide a scientific basis for the identification, development and utiliza-tion of its resources. As a result, obvious characteristics for its identification were revealed, which can beused to identify twigs and leaves of R. nasutus.
10.Relationship between pre-transjugular intrahepatic portosystemic stent shunt(TIPS) hepatic hemodynamics and incidence of post-TIPS hepatic encephalopathy

Objective To study the relationship between pre transjugular intrahepatic portosystemic shunt(TIPS) liver hemodynamics and the incidence of post TIPS hepatic encephalopathy(HE).Methods Hepatic blood dynamics was evaluated in 41 cases of liver cirrhosis before TIPS and one month later.The patients were divided into two groups according to Doppler findings before TIPS.Group 1 composed of patients with prograde portal flow,group 2 composed of patients with hepatofugal or back forth portal flow.The clinical characteristics,incidence of post TIPS HE and pre/post TIPS hepatic arterial resistant index(RI) of the groups were compared.The independent prognostic value of pre TIPS variables for the onset of HE after TIPS was tested with a multiple regression analysis.The variables included age,pugh score,presence of HE before TIPS,the pattern of portal flow.Results The two groups did not differ significantly in their age,etiology of liver disease,indications of TIPS placement, HE incidence before TIPS and portacaval gradient before and after TIPS.But liver failure was more severe in group 2(P
