1.EFFECTS AND MECHA NISMS OF HUMAN UMBILICAL-CORLD BLOOD-DERIVED INTERFERON-? AND ASTRAGALUS MEMBRANACEUS ON NATURAL KILLER CYTOTOXICITY

Natural Killer(NK)cytotoxicity of human PML coulcb be markedly increased by human umbilical-corld-blood-derived interferon-? (P-IFN) and Astragalus membranacesu (AIMember.)but slightly inhibited by high concentration of P-IFN (105u/ml) or A-Membr. (10mg/ml) .P-IFN and A.Membr., on the other hand, could protect target cells from NK cytotoxicity.However, this effect was lower than that of their enhancement of NK cytotoxicity.Moreover, P-IFN and A.Membr .could enhance each other.NK cytotoxicity might be increased by 5 to 6 times when the both agents were com-bined ia treatment of effector cells. A.Membr could induce interferon-r which mediated the enhancing effect of A.Membr NK cytotoxicity.There was a relationship be-tween enhancing effect of P-IFN on NK activity and large molecular metabolism of effector cells. It was supposed that interferon, except for activating NK cytotoxicity directly, might stimulate lymphocytes producing other kinds of factors by which interferon might further amplify its enhancing effect on NK cytotxicity.
2.Content Determination of the Principal Agent in Phenoxymethylpenicillin Potassium Tablets by HPLC

OBJECTIVE:To establish HPLC method for the determination of the principal agent in phenoxymethylpenicillin potassium tablets.METHODS:The determination was performed on Diamonsil C 18 column with column temperature at(26?1)℃.The mobile phase consisted of acetonitrile-0.02mol/L sodium acetate(70∶30)with a flow rate at1.0ml/min.The detection wavelength was set at268nm;the sensitivity was0.01AUFS and the sample size was20?l.RESULTS:Good linear correlation with peak area was noted when the detection concentration range was10.0~120.0?g/mL for phenoxymethylpeni?cillin potassium(r=0.9999,n=5).The average recovery was100.2%(RSD=0.42%).CONCLUSION:The method was rapid,simple and accurate and it can be used to determine the content of the principal agent in phenoxymethylpenicillin potas?sium tablets.

Calponin is an actin filament-associated regulatory protein expressed in smooth muscle and many types ofnonmuscle cells.Three calponin isoforms 1,2,and 3 are encoded by three homologous genes CNN1,CNN2 and CNN3 in vertebrate species with cell type-specific expression and functions.Besides modulating the activity of smooth muscle myofilaments and contractility,calponin also regulates the functions of actin cytoskeleton in non-muscle cells during cell proliferation,adhesion,migration,differentiation,phagocytosis and fusion.This review focuses on the evolution,tissue and cell type-specific expression,structure-function relationships,transcriptional regulation and relevant mechanisms.
5.Genetic Marker of Integron,Transposon in Escherichia coli Isolated from Elderly Patient

OBJECTIVE To investigate the existance of class Ⅰ gene integron and transposon(merA and tnpA) gene marker in Escherichia coli isolated from the elderly patients.METHODS Antibiotic sensitivity was performed by microdilution test.Integron gene IntⅠ1 and transposon marker genes were detected by polymerase chain reaction(PCR).The PCR products were purified and sequenced.RESULTS E.coli resistant to amikacin accounted for 60%,to sulfamethoxazole 95%,and to third-generation cephalosporin 100%(all ESBLs-producing strains).The resistance was related closely to the class Ⅰ integron.The positive rate of integron intⅠ1 was 80%(16/20),whereas the positive rate of gene merA was only 10%(2/20).Gene tnpA was not tested out.CONCLUSIONS The drug resistance of E.coli has close relation with transmission of integron and transposon.The transposons of Tn21/Tn501 are all firstly reported in China.
6.Analysis of the Use of Antihistamine Agents in 18 Hospitals of Hangzhou During the Period 2000~2002

OBJECTIVE:To survey and objectively evaluate the present situation and trend of the antihistamine agents used in 18 hospitals of Hangzhou.METHODS:To make a survey of the use of above mentioned drugs in 18 hospitals of Hangzhou during the period 2000~2002 in respect to the sum of money for drugs and manufacturing factories.RESULTS:In comparison with 2001,the sum of money for consumption of antihistamine agents increased 17.89%in 2000,and decreased 4.58%in 2002.Joint venture and imported products accounted for 70%of share of market.Lortadine and Cetirizine were still the first choice in therapy of allergic reactions.CONCLUSION:Consumption of antihistamine agents decreased,but kinds of drugs had no obvious change.
7.To Promote Rational Drug Use of Antibacterials by Developing Whole-ranged Pharmaceutical Care

OBJECTIVE:To explore ways of promoting the rational use of antibacterials.METHODS:To analyze and e-valuate the effect of whole-ranged pharmaceutical care on promoting rational use of antibacterials.RESULTS:The whole-ranged pharmaceutical care was the efficient way to ensure the rational drug use of antibacterials.CONCLUSION:The whole-ranged pharmaceutical care should be carried out in hospital pharmacy so as to promote the rational use of antibacterials and to make the utmost use of the limited resource of Antibacterials.
8.Discussion on astronomy background of yin-yang and five phases theory in traditional Chinese medicine

The corresponding astronomy implications of yin-yang and five phases of traditional Chinese medicine are investigated based on the modern astronomy,universal biology,and literature research.It is found that the principles of yin-yang in traditional Chinese medicine actually refers to the strengths and the directions of gravitational forces,electromagnetic waves,high energy particle flows of celestial bodies to human.The yin-yang in the human body is a result of selective evolution from protobiont under the influence of yin-yang of celestial bodies in hundreds of millions of years.The revolution of seven luminaries,which includes the Sun,Moon,Mercury,Venus,Mars,Jupiter and Saturn,creates the day and night yin-yang from the self rotation of the Earth,season yin-yang from the revolution of the Earth around the Sun,the yin-yao from the rotation of the Moon around the Earth,and the yin-yao from the rotations of the Mars and the Jupiter.The 10 years cyclic movement of the Mercury displays the states of jiayi,bingding,wuji,gengxin,rengui,which have the allelopathic characteristics of five phases-wood,fire,earth,metal and water,respectively.With the addition of yin-yang from the Mars,the states of Mercury are divided into yang five phases including jia,bing,wu,geng and ren,and yin five phases including yi,ding,ji,xin and gui.The yin-yang and five phases of human visceras is the result of the selective evolution under the influence of yin-yang and five phases of celestial bodies in hundreds of millions of years.The 5 years periodic movement of the Venus shows the five phases characteristics of the five zhongyun.With the addition of the yin-yang from the Mars,every zhongyun shows the yin-yang characteristics,which alternate once every other year.In a cycle of sixty years,the Saturn,Mars,Venus,Mercury and Jupiter show 15 pairs of independent yin-yang states,which is corresponding to the 15 pairs of yin-yang nayin five phases.
9.Expression of CD14 in Kupffer′s cells induced by Lipopolysaccharide

Objective To investigate the expression of LPS receptor-CD14(CD14) on the membrane of Kupffer′s cells (KCs) induced by Lipopolysaccharide (LPS )and its role in activation of KCs and production of cytokines. Methods KCs were isolated by collagenase perfused Wistar rats and routinely cultured in 24-well dishes for 12 h. Cells were harvested and adjusted to a concentration of 1×106/ml/well and were devided into two groups. Group of LPS: KCs were induced with different concentration of LPS (0, 100 ng/ml, 1 μg/ml and 100 μg/ml). Group of PI-PLC: KCs were pre-incubated for 30 min with one unit of phophatidy linositol specific phospholipase C (PI-PLC) before different concentrations of LPS were added. KCs were cultured for 30 and 60 min respectively. Supernatants were then collected for measuring the level of TNFα and IL-6. Cells were stained by indirect immunofluorescent method ( rabbit anti-CD14 antibody and goat anti-rabbit IgG conjugated with FITC ) and analyzed with flow cytometer (FCM). The percentage and mean fluorescence intensity (FI) of CD14-positive cells were taken as the indexes. Results In LPS group, after incubation of cells with increasing concentration of LPS, a significant increase in the percentage of CD14 positive KCs were found and the mean FI was stronger when compared with the control points or the group of PI-PLC. The levels of TNFα and IL-6 in supernatant also increased (P<0.01). In group of PI-PLC, decreased percentage of CD14 positive KCs and weakened mean FI were found when compared with group of LPS. The increasing production of TNFα and IL-6 slowed down in the group of PI-PLC. Conclusion CD14 expression of KCs might be up-regulated by LPS with increase of some cytokines. The production of cytokines in KCs induced by LPS is partially inhibited by PI-PLC.
10.Correlation between clinical features at admission and recent prognostic outcome in patients with acute ;myocardial infarction

Objective:To analyze the correlation betWeen clinical features at admission and recent prognostic outcome in patients With acute myocardial infarction (AMI).Methods:The data of 108 patients admitted because of primary AMI Were retrospectively analyzed.According to occurrence of major adverse cardiovascular events (MACE)or not after discharge,they Were divided into event group (n=61)and non-event group (n=47).Clinical features such as Gender,age etc.of tWo groups Were collected,analyzed and compared.After one-year folloW-up,correlations betWeen clinical feature indexes and MACE incidence Were analyzed.Results:Compared With non-event group, there Were significant increase in mean age,mean arterial pressure (MAP),left ventricular end-diastolic dimension (LVEDd),percentage of> Killip class 2,Grace score and multi-vessel coronary disease,and significant decrease in left ventricular ejection fraction (LVEF)in event group,P<0.05~<0.01. Single factor correlation analysis indi-cated that age,MAP,LVEF,percentage of > Killip class 2,Grace score and multi-vessel coronary disease rate Were correlated With MACE incidence rate (P<0.05~<0.01);multi-factor Logistic regression analysis indicated that age (OR=0.827),LVEF (OR=0.624),Grace score (OR=0.589)and multi-vessel coronary disease rate (OR=0.461)Were risk factors for MACE in AMI patients,P<0.05 all.Conclusion:Advanced age,loW left ven-tricular ejection fraction,higher Grace score and more multi-vessel coronary disease are high risk factors for recent occurrence of major adverse cardiovascular events in patients With acute myocardial infarction.
