1.Determination of Calycosin-7-?-O-D-glucopyranpside in Danggui Buxue Dropping Pills by HPLC

Objective To determine the content of calycosin-7-?-O-D-glucopyranpside in Danggui Buxie Dropping Pills.Methods HPLC method was used. The chromatographic column was YMC-C18(150?4.6 mm, 5 ?m), the mobile phase was methanol and water(40 ∶60), the detection wavelenth was 235 nm, column temperature at 20 ℃and flow rate was 1.0mL/min.Results Calycosin-7-?-O-D-glucopyranpside was separated well with other components and was in good linearity in the range of 0.02~0.80 ?g. The average recovery was 97.62 %and RSD=1.06 %.Conclusion The method is rapid and accurate and can be used for quality control of Danggui Buxie Dropping Pills.
2.Expression and localization of CB2 (cannabinoid receptor) in rat skin

Objective To study the expression and distribution of CB2 in rat skin tissue. Method Immunohistochemistrial technics were employed to study the expression and distribution of CB2 in rat skin tissue. Results CB 2 receptors mostly presented in suprabasal layers of the epidermis and hair follicles in dermis by CB2 like immunoreactivity with a N terminal anti CB2 receptor antibody. The positive expression of CB2 appeared in rat spleen and a negative result occured in liver. Conclusion The CB2 receptors distributed mainly in the rats, epidermis and hair follicles, may involve in some of the dermic physiological and pathological process, such as the communication between central neural system and skin.
3.Effects of high energy shock wave on oxidative stress and apoptosis in the renal cells of rabbits

Objective To investigate the relationship between oxidative stress and cell apoptosis in the kidney of rabbit induced by high energy shock wave(HESW).Methods Forty healthy male rabbits were surgically managed to the mono-nephron models and randomized into 4 groups consisting of ten each: control group,1 000 HESW group,2 000 HESW group and allopurinol pretreated plus 2 000 HESW group.The rabbit kidney tissues were obtained.The activity of superoxide dismutase(SOD) and malondialdehyde(MDA) levels in the renal tissue were measured 24 hours after HESW application.The cell apoptosis was detected by TdT-mediated dUTP nick end labelling(TUNEL) and flow cytometry.Results Compared with the control group,the MDA level and the rate of apoptosis in the HESW groups increased,and the activity of SOD reduced significantly(P
4.Pathogens distribution and drug resistance from cerebrospinal lfuid culture from 2007 to 2014

Objective To explore the common pathogens distribution and the drug resistance pattern in vitro from cerebrospinal lfuid culture in children. Methods The results of cerebrospinal lfuid culture in hospitalized children from January 2007 to December 2014 were retrospectively analyzed. Bacteria identiifcation and antimicrobial susceptibility were assayed by Vitek system from Bio Mrieux Company. Some bacterial strains were tested by disk diffusion method. Results Cerebrospinal lfuid from 23099 patients were tested at least one time during research period. 671 strains of bacteria were isolated with positive rate of 2 . 9%, among which there were 579 strains of gram positive bacteria ( 86 . 3%) and 92 strains of gram negative bacteria ( 13 . 7%). The top ifve bacteria were coagulase negative Staphylococcus ( 399 strains, 58 . 9%), Micrococcus ( 37 strains, 5 . 5%), Streptococcus pneumoniae (34 strains, 5.1%), Escherichia coli (32 strains, 4.8%), and Enterococcus faecium (26 strains, 3.9%). The positive rates of cerebrospinal lfuid culture in 8 consecutive years showed a downward trend (χ2?=?10 . 410 , P=?0 . 001 ). The positive rates of coagulase negative Staphylococci showed annual decline trend (χ2?=?31 . 200 , P0 . 001 ). However, annual positive rates of Escherichia coli showed an upward trend (χ2?=?4 . 786 , P=?0 . 029 ). The resistant rate of coagulase negative Staphylococcus to oxacillin was 79 . 8%, and no coagulase negative Staphylococcus resistant to linezolid and vancomycin was found. Micrococcus sensitive to vancomycin was 100%. The resistant rate of Streptococcus pneumoniae to penicillin was 77 . 8%, while the sensitive rate of Streptococcus pneumoniae to vancomycin was 100%. The sensitive rate of Escherichia coli to piperacillin/tazobactam and meropenemwas 100%. Conclusions The positive rate of cerebrospinal fluid culture is low. The main strains are coagulase negative Staphylococcus, Micrococcus, Streptococcus pneumoniae, Escherichia coli, and Enterococcus faecium. The antibiotics should been rationally chosen according to the results of bacterial strain culture and drug sensitive test.
5.Clinical analysis of 408 children with positive blood culture of Escherichia coli

Objective To investigate the clinical manifestations and the antibiotics resistance patterns in children with positive blood culture of Escherichia coli. Methods The clinical data of children with positive blood culture of Escherichia coli were retrospectively analyzed from Jan.2007 to Dec.2014. Results In a total of 154774 children who had blood culture in the study period, 8446 children were positive, among whom 408 (4.83%) children were isolated Escherichia coli. The children with the positive blood culture of Escherichia coli mainly were under one year old (51.72%), of which 36.77%was neonates. There were 275 children had underlying diseases, and the most common disease was Leukemia. 199 (48.77%) Escherichia coli strains were producing extended spectrumβ-lactamase (ESBLs) and 85.23%were resistant to ampicillin. All strains were susceptible to meropenem. Conclusions Septicemia caused by Escherichia coli is usually occurred in children with leukemia or in neonates. Since blood infections of Escherichia coil had high rate of ESBLs, the use of carbapenem antibiotics should be cautious.
6.Expressions and correlation of Glil and β-catenin in the colorectal carcinoma

Objective To investigate the expression of Glil and β-catenin,which are central molecules of Hedgehog and Wnt signaling pathway,in colon cancer tissues and cell lines,and evaluate their relationship.Methods RT-PCR and Western blot were used to determine the expression of Glil and β-catenin in colon cancer tissues and cells.Interaction between endogenous Glil and endogenous 3-catenin was examined using immunoprecipitation method.Results Compared to corresponding normal tissues,the expression of Gli1 and β-catenin raised raising with one accord,the average growth rates were 42.69,72.11 respectively (P < 0.05).In colon cancer cell lines,the expression levels of Glil and β-catenin increased,which were 52.54,17.23,5.54 and 5.30,6.34,2.78 (all P < 0.05).Both of them were enhanced in three Wnt aberrant active cells visibly.The immunoprecipitation result indicated that there was an interaction between Glil and β-catenin in colon cancer cells.Conclusion The expression of Glil and β-catenin rise congruously in both colon cancer cells and tissues,and there is an interaction between them.This interaction between two key components of Hedgehog and Wnt signaling may suggest the possible crosstalk manner in these pathways.
7.Localization of dopamine- and cAMP-regulated phosphoprotein in mouse kidney

Objective To investigate the expression and localization of dopamine- and cAMP-regulated phosphoprotein of 32 000 (DARPP-32) in mouse kidney tissue. Methods The cellular localization of DARPP-32 in mouse kidney tissue was detected by immunoblot and immunohistochemistry. Results DARPP-32-like immunoreactivity was detected in the medullary thick ascending limb of the loop of Henle, the cortical proximal convoluted tubule, and collecting ducts in medullary rays. The renal tubules were enriched of Na+, K+-ATPase for sodium reabsorption. Conclusion The participation of DARPP-32 is a likely crucial step of the signal-transduction pathway of dopamine regulation on sodium reabsorption in renal tubule cells.
8.Preparation and transdermal resorption of tanshinone ethosomes in vitro

AIM: To prepare ethosomes and study on transdermal resorption of tanshinone ethosomes in vitro. METHODS: Tanshinone ethosomes were prepared by thin-film ultrasonic method.The different dosage regimens and volume on the transdermal resorption of tanshinone were evaluated through excised mice skin in vitro using modified Franz-diffustion cells. RESULTS: The mean particle diameter of ethosomes was 79.7 nm,and the encapsulation efficiency was about 81.2%.Cumulative penetration amount of tanshinone from ethosomes was 1024? 104.0 ng/cm2 and skin residual amount was 345.2?58.0 ng/cm2.Cumulative penetration amount increased with ethosomes volume.Liposomes failed to promote the penetration of tanshinone,and its skin residual amount was 625.3 ?124.5 ng/cm2,water-ethanolic suspension also could not promote the penetration of tanshinone,and skin residual amount was 765.0?60.00 ng/cm2. CONCLUSION:Ethosomes can improve significantly the transdermal delivery of tanshinone.
9.Anxiety Status of Male Partner Involved in Induced Abortion and Realted Influencing Factors Analysis in One Hospital in Kunming

Objective To investigate the anxiety status of male partner involved in induced abortion and to analyze influencing factors related.Methods Male partners of the patients undergoing induced abortion in the gynecology and obstetrics department of a hospital in Kunming in April and May in 2015 were selected and a questionnaire survey was conducted.Self-rating Anxiety Scale (SAS) was used to measure the anxiety status and SPSS 17.0 was used for statistical analysis.Results A total of 53 out of 205 male partners were found to have anxiety symptoms (25.9%),with 44 cases of mild anxiety (21.5%),8 cases of moderate anxiety (3.9%) and 1 case of severe anxiety (0.5%).The main related risk factor was times of pregnancy (OR=3.35) while the protective factors were level of education (OR =0.60),times of induced abortion (OR =0.20) and age (OR =0.32) Conclusion When abortion occurs,the male partner's anxiety appears,mainly with mild and moderate one,and female's emotion are also affected.It is important to strengthen health education and popularize contraceptive knowledge so as to reduce the incidence of anxiety.
10.Effects of Pantoprazole on Stress-Induced Gastric Mucosal Lesions in Rats

Objective: To investigate the effects of Pantoprazole on the expression of TFF1 in stress-induced gastric mucosal lesions in rats, and the mechanism thereof. Methods: Fifty-six rats were randomly divided into seven groups, normal group, model groups (3 groups) and model therapy groups (3 groups). The rat model of water immersion- restraint stress (WRS) was established in model groups, model group1(the immediately after establishing models), model group 2 (4 h after establishing models) and model group 3(8 h after establishing models). The model therapy groups were divided into model therapy group 1 (immediately after establishing models), model therapy group 2 (4 h after establishing models), and model therapy group 3 (8 h after establishing models). The ulcer index (UI) and histological changes were observed after WRS in rats. The expression of TFF1 was detected by immunohistochemistry. Results: After WRS, the gastric mucosa was widely damaged in rats. UI were increased and the expression of TFF1 was decreased in model groups. After intervention with Pantoprazole, UI was lower in model therapy group than those in model groups (model group 1 vs model therapy group 1,69.13±1.97 vs 23.38±1.30, P < 0.01; model group 2 vs model therapy group 2, 57.50±8.81 vs 10.38±3.02, P < 0.01; model group 3 vs model therapy group 3, 43.50±6.76 vs 5.88±1.25, P < 0.01). The staining scores of TFF1 were increased (model group 1 vs model therapy group 1, 0.55±0.11 vs 0.92±O.15, P< 0.01; model group 2 vs model therapy group 2, 0.76±0.24 vs 1.36±0.21, P< 0.01; model group 3 vs model therapy group 3, 1.12±0.16 vs 1.65±0.11, P < 0.01). Conclusion: TFF1 may participate in the protection of gastric mucosa and promote ulcer recovery. Pantoprazole may participate in the defense of gastric mucosa through mediating the up-regulation of TFFI expression.
