1.High level prokaryotic expression of neuron protective protein TAT-Bcl-XL and preliminary detection of its anti-apoptosis activity

Objective:To prepare the recombinant neuron protective protein TAT-Bcl-XL in E.coli expression system and to determine its anti-apoptosis activity.Methods:DNA fragment of TAT-Bcl-XL was obtained by RT-PCR using primers that were specific for Bcl-XL gene. Prokaryotic expression vector(pTBTOPO) was constructed by insertion of TAT-Bcl-XL DNA fragment into pCRT7/CT-TOPO vectors, and the recombinant protein was detected by SDS-PAGE and Western blot analysis using anti-V5 tag epitope antibody as the primary antibody. The recombinant fusion protein was purified by affinity chromatography, and the anti-apoptosis function of the purified protein was determined by flow cytometry.Results:A molecular weight of 30 000 protein was detectable in both SDS-PAGE and Western blot analysis. Immunofluorescent staining showed the fusion protein was distributed in cell plasma after incubation with 200 nmol/L fusion protein. Flow cytometry data indicated that the recombinant protein could enhance cell survival by 40% when 293T cells were treated with 250 ?mol/L zinc chloride as a supplementation in cell culture media.Conclusion:High level expression of the reported neuron protective protein was obtained in E.coli expression system, and the purified recombinant proteins remained its anti-apoptosis activity in our preliminary characterization.
2.Advances of community-acquired methicillin-resistant Staphylococcus aureus

Methicillin-resistant Staphylococcus aureus (MRSA) is one of the major pathogens of the hospital infection.Its clinical features and drug-resisetance situations have always been concerned.But since the late 1990s,another class of MRSA has become a major concern worldwide as an emerging pathogen in the community.This new class of MRSA is called community-acquired MRSA (CA-MRSA).With the rapid development of the infection of CA-MRSA in 20 years,especially in the latest 3 years,CA-MRSA may be replacing the hospital-acquired MRSA strains(HA-MRSA) as one of the major pathogens in the hospital and the community as well.The characteristics of CA-MRSA are very different from those of HA-MRSA.This review summarizes the current studies of CA-MRSA on the epidemiology and the molecular characteristics.

4.The detection meaning of D-dimer and fibrinogen in patients with coronary disease

Objective To explore the detection meaning of D-dimer(D-D) and fibrinogen(Fg) in patients with coronary disease.Methods 150 patients with coronary disease were randomly divided into UA group,SA group and AMI group,the D-D was detected by ELASA double resistance to clip method and Fg was detected by CLIUSE methods.Results The D-D in SA group,UA group and AMI group showed significant difference between two groups (SA:UA,t =9.340;SA:AMI,t =18.334; UA:AMI,t =10.240.all P <0.01 ).The Fg in SA group,UA group and AMI group showed significant difference between each two groups ( SA:UA,t =11.376; SA:AMI,t =11.730; UA:AMI,t =3.121.all P <0.01 ).D-D between one coronary artery disease and two coronaries artery disease,three coronary arteries disease showed significant difference( t =7.017,15.603,all P < 0.01 ) ; and between two coronaries artery disease and three coronary arteries disease showed significant difference,too ( t =8.662,P < 0.01 ).The Fg between one coronary artery disease and two coronaries artery disease,three coronary arteries disease,showed significant difference( t =8.687,11.855,all P < 0.01 ) ; and between two coronaries artery disease and three coronary arteries disease showed significant difference,too( t =4.379,P < 0.0 1 ).Conclusion The D-D and Fg showed obvious difference between different types of coronary disease,and the levels increase along with the disease degrees of coronaries
5.Nosocomial Infection Control and Management in Operating Room

OBJECTIVE To discuss effective methods of nosocomial infection management in operating room to control the nosocomial infection and improve quality of medical care.METHODS To strengthen and improve the environment of operating room,and the management of medical staff,room cleaning and articles,as well as operation classification in there,to establish the effective nosocomial infection management organization in whole hospital,particulerly in operating room,to study technology of hospital infection,and to enhance the consciousness in control nosocomial infection.RESULTS It is very important guarantee that strengthen management of operating room work in hospital could improve the medical safety and prevent the nosocomicl infection.CONCLUSIONS To actualize scientific management in operating room,and control nosocomial infection.
6.Antimicrobial Resistance of Nonfermentative Bacteria in Lower Respiratory Tract Infection

OBJECTIVE To investigate the epidemic situation of nonfermentative bacteria in lower respiratory tract infection and guide rational use of antimicrobials.METHODS API system was applied for bacterial identification.K-B disk diffuse method was used in susceptibility testing.WHONET 5.3 software was used for data analysis.Pseudomonas aeruginosa,Acinetobacter baumannii and Stenotrophomonas maltophilia isolates were included in this study.RESULTS Less than 20% of P.aeruginosa isolates were resistant to amikacin,imipenem or cefoperazone/sulbactam.For A.baumannii,the most active compounds tested were imipenem and cefoperazone/sulbactam with resistance rates of 4.8% and 7.3%,respectively.26.6% And 27.4% of A.baumannii isolates were resistant to piperacillin/tazobactam and ticarcillin/clavulanic acid.The resistance rate of S.maltophilia to cefoperazone/sulbactam,ticarcillin/clavulanic acid and piperacillin/tazobactam was 0,2.7% and 10.8%,respectively.CONCLUSIONS The isolation rate of nonfermenters is increasing in the lower respiratory tract infection,and the ways to improve the cure rate are to avert nosocomial infection and choose antimicrobials properly.

China is the second largest country of diabetes, which is a great challenge for the Chinese diabetes management. We need a guideline suitable for Chinese patients to guide the physicians. In 2007, Chinese Diabetes Society published "Chinese guideline for type 2 diabetes mellitus", according to the Chinese patients' characters, the guideline gives somewhat different suggestions from ADA and IDF.
9.Experimental study of the effects of PEDF on penetrating corneal graft of VEGF in rats

During the Qin-Han Dynasty,the system of terms of Traditional Chinese Medicine(TCM) has been initially established.With the progress of TCM subject and increasing of academic exchanges,one word having two or more meanings,one meaning with two or more words,and development of meanings of words have come into sight,following with the appearance of new concepts and terms.TCM terms have their distinction.The characteristics of TCM terms were: characteristic of history; ②characteristic of culture;qualitative description;using concrete word to describe the abstract concept.During the new century,TCM needs modernization.Clear concept and standard terms are important for modernization of TCM.Meanwhile,there was confusion on translation of terms on the internationalization of TCM.Standard terms were precondition of the academic exchange and popularizing TCM.In a word,the standardization of terms of TCM became one of the keys for the modernization and internationalization of TCM.
