1.Determination of 10-Hydroxy-2-decenoic acid in Chongcaojing Wangjiang Capsule by HPLC

To determine the content of 10-Hydroxy -2-decenoic acid in Chongcaojing Wangjiang Capsule by HPLC.The column was Discovery C 18 .The detective wavelength was at 209 nm. The mobile phase was acetonitrile: 0.5% phosphoric acid (25∶75) with the flow rate being 1.0 mL/min and the column temperature at 25℃.There was a good linearity (r=1) winthin the range of 0.02144 ~ 0.38592 ?g. The average recovery rate was 101.8%, RSD= 1.6%. [Conclusion] This method can be used for the quality control standard of Chongcaojing Wangjiang Capsule.
2.Quantitative determination of Ophiopogonin D in Zhe Ophiopogon japonicus(Thunb.)Ker-Gawl. and Chuan Ophiopogon japonicus(Thunb.) Ker-Gawl. by HPLC-ELSD

HPLC-ELSD was used to determine the content of Ophiopogonin D in Zhe Opiopogon japonicus(Thunb.) Ker-Gawl. and Chuan Ophiopogon japonicus(Thunb.) Ker-Gawl. The chromatographic conditions were as follow: Alltima C18 column (250mm?4.6mm) and the column temperature at 30℃, drift tube temperature at 98 ℃with Gas flow rate being 2.8 L/min and acetonitrile : water (55∶45) as mobile phase with the flow rate being 1.0 mL/min. There was a good linearity (r=0.9992) within the range of 0.2565~6.4125 ?g. The average recovery rate was 102.0%and RSD=2.1%.
3.Late course accelerated hyperfractionation radiotherapy and concurrent chemothe rapy on esophageal carcinoma

Objective To compare the treatment effects and toxicity of late co urse accelerated hyperfractionation radiotherapy (LCAFR), LCAFR plus concurren t chemotherapy (LCAFR+C) and conventional fractionation radiotherapy(CFR) on esop hageal cancer. Methods 150 patients with squamous carcinoma of thoracic esophag us were divided randomly into three groups: 1.CFR group, patients were irradiate d 2.0?Gy/f, 5 times a week, to a total does of 60?Gy. 2. LCAFR group, patients wer e first irradiated with CFR to 30?Gy, then followed by 1.5?Gy/f bid, at more t han 6 hours' interval, to the total dose of 60?Gy. 3.LCAFR+C group: The radiotherap y technique was the same as the LCAFR group, but weekly 20 mg DDP and 500 mg 5-Fu wer e added simultaneously for 5 weeks. Results All three groups completed their tre atment course. Of CFR, LCAFR and LCAFR+C groups, the 1-,2-,3- and 4-year sur viva l rates were 54%, 30%, 18%, 18%; 76%, 56%, 44%, 42% and 82%, 62%, 50%, 44%. The 1-,2-,3- and 4-year local control rates were 40%, 32%, 26%, 24%; 72%, 60%, 5 6%, 54% and 78%, 66%, 60%, 56%, with obvious better results in the latter two groups (P0.05). The acute toxic effect was severer in the LCAFR+C g roup than in the other two, with the difference significant between the LCAFR+C and CFR group, bu t not between the LCAFR and CFR group. The tolerance of the patients to LCAFR wa s better than that of LCAFR+C group. There were no significant differences in la te complications and causes of death between the three groups. The main cause of death was local recurrence and uncontrolled primary disease, which were signifi cantly lower in the LCAFR and LCAFR+C groups than in the CFR group. Conclusions Both late course accelerated hyperfractionation radiotherapy and late course acc elera ted hyperfractionation radiotherapy plus chemotherapy can significantly improve the local control and survival of esophageal cancer, but the latter has increase d toxicity. Concurrent small dose chemotherapy can not lowered the remote metas tatic rate.
4.Observations on the Efficacy of Acupuncture at Huatuo Jiaji Points plus Point Shixuan Bloodletting in Treating Cervical Spondylotic Radiculopathy

Objective To investigate the clinical efficacy of acupuncture at Huatuo jiaji points plus point Shixuan bloodletting in treating cervical spondylotic radiculopathy.Methods One hundred patients with cervical spondylotic radiculopathy were randomly allocated to treatment and control groups, 50 cases each. The treatment group received acupuncture at Huatuo jiaji points plus point Shixuan bloodletting and the control group, acupuncture at Huatuo jiaji points alone. Plasma viscosity was measured in the two groups before and after treatment. The clinical therapeutic effects were compared between the two groups.Results There was a statistically significant pre-/post-treatment difference in plasma viscosity in the two groups (P<0.05). There was a statistically significant post-treatment difference in plasma viscosity between the treatment and control groups (P<0.05). The total efficacy rate was 94.0% in the treatment group and 84.0% in the control group; there was a statistically significant difference between the two groups (P<0.05).Conclusion Acupuncture at Huatuo jiaji points plus point Shixuan bloodletting is an effective way to treat cervical spondylotic radiculopathy.
5.MR line-scan diffusion imaging in ischemic epiphysis of femoral head:preliminary experimental study in piglets

Objective To observe diffusion changes of epiphysis of femoral head with ischemia of difference phases by line-scan diffusion weighted imaging(LSDWI),and determine whether LSDWI can provide temporal information and severity about ischemia of epiphysis.Methods lschemia was surgically induced in one hip of each piglet(n=25)and the other hip served as a normal control.Piglets were imaged before surgery and at 3 hours,72 hours and 1,3 and 6 weeks after surgery by using LSDWI.Apparent difrusion coefficients(A DC)in epiphysis of the femoral heads were calculated.Significant difierences in ADC values between ischemia group and control group were found by using paired t-test.After scan at individual time points,5 piglets were sacrificed for histological study each time.Results The ADC value in the ischemic femoral heads f(1.22±0.37)×10-3 mm2/s]decreased significantiy at 3 hours after surgery (t=3.914,P<0.01),compared to that in control[(1.73±0.33)×10-3mm2/s},and increased at 72 hours[(2.15±0.32)×10-3mm2/s versus(1.70±0.22)×10-3 mm2/s](t=3.348,P<0.01).Then ADC valne kept increasing until 6 weeks after surgery[(1.61±0.27)×10-3mm2/s in ischemia side vs (1.11±0.45)×10-3mm2/s in the control](t=4.136,P<0.01).rrhe percentage change of the ADC value significandy increased at 3 hours,72 hours,1 and 3 week(s)after the surgery(P<0.01),compared to that at the prior neighboring time point.No significant increase in the percentage change of ADC value was found between the 3rd week and the 6th week after the surgery(t=2.29.P>0.05).Histological examinations revealed abnormal thickening within epiphyseal cartilage,and cartilaginous islands within ossified tissues.Growth disturbante wag found in form of focal growth plate disruption.Conclusions Dynamic changes of ADC values were found with the prolonged ischemia of the femoral head by LSDWI.It could serve as a useful marker for evaluating duration and extent of ischemic epiphyseal disruption.
6.Analysis of the genetic polymorphisms of 15 autosomal short tandem repeat in a Han population of Shandong.

OBJECTIVETo obtain population genetic data at 15 short tandem repeat(STRs) in Shandong Han population.
METHODSFifteen STRs were genotyped in 200 unrelated individuals from Shandong Han population using Identifiler(TM) Kit.
RESULTSIn this Shandong Han population, the observed heterozygosities of the 15 loci ranged from 0.605 to 0.882, and the expected heterozygosities were 0.625-0.862, the polymorphic information contents ranged from 0.57 to 0.85, the discrimination powers ranged from 0.795 to 0.958, and the exclusion probabilities ranged from 0.297 to 0.758. The non-differentiation exact P values at TPOX loci were more than 0.05 between Shandong and Han population of Henan, Jilin, and Jiangsu. The non-differentiation exact P values between Shandong Han population and others populations were more than 0.05 at others loci. The P value at D13S317 was more than 0.05 between Shandong Han population and Ewenki population.
CONCLUSIONThese loci are polymorphic in Shandong Han population, and the discrimination powers and exclusion probabilities were high in the 15 STRs except TPOX and TH01.

7.Analysis of linkage disequilibrium and linkage for 12 short tandem repeat loci on chromosome X.

OBJECTIVETo analyze linkage disequilibrium of 12 short tandem repeat loci on chromosome X (X-STR) among an ethnic Han population from Guilin, Guangxi, and to study the genetic linkage and haplotype distributions of such loci in 2 linkage groups.
METHODS12 X-STR loci including DXS8378, DXS10159, DXS10162, DXS10164, DXS981, DXS6789, DXS7424, DXS101, DXS7133, GATA165B12, GATA31E08 and DXS7423 were genotyped using an AGCU X12 STR PCR Amplification kit. A total of 119 pedigrees were analyzed for linkage and linkage disequilibrium.
RESULTSTwo mutations were found at DXS7424, and 1 mutation was found at DXS10164. A total of 93 haplotypes of DXS10159-DXS10162-DXS10164 were constructed for 261 unrelated males and females, in addition with 167 haplotypes of DXS6789-DXS7424-DXS101-DXS7133. The values of recombination fraction between DXS10159 and DXS10162, DXS10162 and DXS10164, DXS6789 and DXS7424, and DXS7424 and DXS101 were 0.0269, 0.0236, 0.0505 and 0.0438, respectively.
CONCLUSIONLinkage disequilibrium of X-STR does not only depend on physical and genetic distances. There was incomplete linkage relationship between loci on DXS10159-DXS1016-DXS10164 and DXS6789-DXS7424-DXS101 linkage groups.
