1.Establishment of experimental femoral pseudoaneurysm and arteriovenous fistula model in rabbit and its related study

Objective:To explore the possibility of establishment of experimental femoral pseudoaneurysm (PA)and arteriovenous fistula (AVF) models in rabbits and to make the related study.Methods:Experimental PA and AVF models were established in rabbits with microsurgical technique.Models were analyzed by blood gas analysis,checked by color Doppler flow imaging(CDFI),examined by computed tomography angiography(CTA),magnetic resonance angiography (MRA),digital subtraction angiography(DSA) and histological observation.Results:16 of 20 femoral pseudoaneurysms and 20 arteriovenous fistulae were successfully anastomosed and were confirmed by related examination methods.Conclusion:It is possible to establish experimental model in rabbit with microsurgery technique and each of examination methods has its own characteristics.
2.Application of ViabahnTM serf-expanding covered stent in endovascular repair of femoral pseudoaneurysm caused by intravenous drug injection: report of nine cases

Objective To discuss the feasibility,safety and validity of ViabahnTM self-expanding covered stent in endovascular repair of femoral pseudoaneurysm caused by intravenous drug injection.Methods The clinical data of 9 patients with femoral pseudoaneurysm caused by intravenous drug injection,who were admitted to authors' hospital during the period from April 2014 to April 2015,were retrospectively analyzed.The pseudoaneurysms were located at the common femoral artery in 6 patients and at the superficial femoral artery in 3 patients.Endovascular repair with ViabahnTM self-expanding covered stent was performed in all the 9 patients.The intraoperative angiographic findings,the sizes of the implanted stents,and the manifestations of the angiography performed immediately after stent implantation were recorded,after operation,the patients were kept under close observation to evaluate the improvement of clinical symptoms,the effect of pseudoaneurysm repair,and the occurrence of endoleak,stent infection,in-stent stenosisand fracture of stent.Anti-infection treatment was employed both before and after the stent implantation;after the stent implantation debridement and/or drainage for soft tissue infection area of inguinal region was adopted.Results Successful implantation of ViabahnTM self-expanding covered stent was accomplished in all 9 patients,with a technical success rate of 100%.Angiography performed immediately after stent implantation showed that complete repair of pseudoaneurysm cavity was obtained in all 9 patients,the distal segment of the parent artery was patent,and no endoleak occurred.The patients were followed up for 6-12 months,and limb salvage was successful in all patients,no intermittent claudication or muscular atrophy occurred.CT angiographyreexamination showed that no endoleak,stent infection,stent fracture or in-stent stenosis occurred.Conclusion Under the conditions of adequate anti-infection,local debridement and drainage,the use of ViabahnTM self-expanding covered stent can promptly close the break of femoral pseudoaneurysm to save the patient's life.Because of the excellent qualities,such as flexibility,adhesion and patency,of ViabahnTM self-expanding covered stent,the postoperative occurrence of intermittent claudication,stent fracture and in-stent stenosis can be effectively reduced.It is expected that ViabahnTM self-expanding covered stent may become one of the vascular reconstruction methods for femoral pseudoaneurysm caused by intravenous drug injection,although its long-term curative effect needs to be further observed and verified.
3.Effects of low frequency vibration on the biological characteristics of human osteoblasts

BACKGROUND: The reaction of osteoblasts is important role in the reconstruction of osteocytes, and the mechanism of bone remodeling can be known from the cytological level through analyzing the mechanical reaction of osteoblasts under different loading.OBJECTIVE: To analyze the effects of low frequency vibration on the proliferation and differentiation as well as the matrix excretion of cultured human osteoblasts.DESIGN: A comparative observation.SETTING: Department of Orthopaedics, Nanfang Hospital of Southern Medical University.MATERIALS: The experiment was carried out in the laboratory of Nanfang Hospital from January to December in 2002. Human osteoblasts were isolated from the iliac cancellous bone of adults.METHODS: The osteoblasts were cultured for 48 hours, and then treated with low frequency vibration of 0.1, 0.2, 0.5, 2 and 5 Hz. The proliferation of the osteoblasts was detected with flow cytometry, and the activity of alkaline phosphatase (ALP) and content of osteocalcin were assayed by spectrophotometric methods and radioimmunoassay respectively.MAIN OUTCOME MEASURES: ① Effects of low frequency vibration on the ALP activity and osteocalcin excretion of osteoblasts; ② Effects of low frequency vibration on the proliferation of osteoblasts detected with flow cytometry.RESULTS: ① 0.2 and 0.5 Hz vibrations could markedly increase the ALP activity of osteoblast (P < 0.01), but that of 5 Hz obviously decreased the ALP activity (P < 0.01). 0.1 and 2 Hz vibrations had no obvious effects on the ALP activity of osteoblasts. ② 0.2 and 0.5 Hz vibration could significantly increase number of osteoblasts at S phase from 10.4% to 12.45% and 16.12%, and the proliferative index increased from 20.14% to 26.21% and 28.75%; 0.1 and 2 Hz vibration has not obvious effects in osteoblast proliferation index; 5 Hz vibration obviously decreased the proliferative index value to 13.22% (P < 0.05). 0.1 and 2 Hz vibrations had no obvious effects on the proliferative index of osteoblasts (P > 0.05). ③ 0.2 and 0.5 Hz vibrations could obviously increase the amount of osteocalcin excretion to 1.87 μg/L and 2.47 μg/L(P < 0.05), but 2 and 5 Hz vibrations decrease the amount of osteocalcin excretion (P < 0.05), and that of 0.1 Hz had no obvious effect on amount of osteocalcin excretion (P > 0.05).CONCLUSION: Low frequency vibrations of 0.2-0.5 Hz could accelerate the proliferation and differentiation as well as the excretions of active materials of osteoblasts, and it plays an instructive role for low frequency vibration to treat fracture.
4.The method of directly assessing the immune-protective activity of HPV16L1 antibody

Objective:To observe whether the HPV16L1VLP Hela-cell binding-inhibition experiment can assess the immune-protective activities of HPV16L1 antibody in HPV16 prophylactic vaccine or other fields.Methods:C57BL/6 mice were divided into three groups,including pcDNAL1 plasmid,HPV16L1VLP protein,pcDNA3.1 plasmid.Each group of mice was immunized intramuscularly or subcutaneously with certain above antigens,accompanying three times and interval 3 weeks.After 14 days of 3 rd immunization,take blood to make serum for immunochemistry.Results:Hela cell neutralized by the serum IgG of experimental mice can be stained negatively,while the cells of control and non-inoculation groups can be stained positively (brown-yellow color).Conclusion:HPV16LIVLP Hela-cell binding-inhibition assay may be the better way to reflect the neutralizing-protective activities of HPV16L1 antibody in many fields including HPV16 prophylactic vaccine.

Objective To optimize the formulation of the Garlic oil ?-CD sustained-release tablet. Methods The preparation formulation of the sustained-release tablet was optimized by orthogonal design which took the released rate as the index. Result The optimized formulation contained 9% HPMC, 15% lactose, 10% PVP, 10% microcrystalline cellulose. The ingredient of Garlic oil sustained-release tablet by this formulation was released slowly in 8 h. Conclusion The optimized formulation is rational and stable, the tablet could be released slowly.
7.Education research:an exploration of tracking type internship with poisoning case-based learning

Objective To explore the teaching effect of acute poisoning course among students majoring in rescue medicine. Methods From March to June 2013, 30 students majoring in rescue medicine were randomly divided into two groups for the clinical practice. Case-based learning was used in experimental group and cases was tracked from one department to another in chronological order or in reverse order to explore the cause of poisoning and the development of disease and to ob-tain the complete medical records of patients. Traditional rotating internship was used in control group. Poisoning cases in the department were taken as the starting point and end point and the medical records of patients were obtained. The clinical teaching effect was compared between the two groups through questionnaire survey and final examination. The data were calculated by SAS 9.2 software, mean comparison between the two groups was conducted by t test and enumeration data were analyzed by chi-square test. P<0.05 stands for statistically significant difference. Results The number of cases in the experimental group was more than that in the control group during the same internship period ((15±3) vs. (9±5),P<0.01). Skill score of experimental group was significantly higher than that of the control group((79.1±6.9) vs. (66.2±6.7), P<0.01). Theory examination results between two groups had no obvious difference((88.4±5.7) vs. (87.8±6.3),P>0.05),however, theory examination results were increased significantly in this year compared with those of previous year((88.1±6.0) vs. (79.3±12.8), P<0.01). Questionnaire survey demonstrated that teachers and students' satisfaction were 95.3%(143/150) and 93.3%(140/150) respectively regarding the ten advantages of the track-ing type internship. Conclusions Tracking type internship with poisoning case-based learning is a good combination of lecture-based learning and problem-based learning. It is helpful to cultivate and exercise students' ability to apply the knowledge.
8.Morphological study of uveoscleral pathway following topical Prazosin in free-sympathetic rabbit eyes

ObjectiveTo observe the morphology of the uveoscleral pathway following topical Prazosin(PZ) in rabbit free-sympathetic eyes.MethodsThe one lateral superior cervical ganglionectomy(SCG) were preformed on each rabbit.PZ drops were installed only on the SCG eyes on 14 days Postoperationly.Microamounts of fluorescein isothiocyanate-bovine serum albumin(FITC-BSA) was infused into anterior chamber of rabbit eyes as the teacer,two rabbits in each group were killed at 2,4,6,8,10,and 12h after PZ instillation.Degress of fluorescent intensity of ciliary body,suprachoroidal space,anterior and posterior sclera,and choroid were observed under fluorescence microscopy.ResultsThe IOP of the SCG eyes was lower after installation of PZ.But the reduced extent is below that of normal rabbit eyes;After installation of PZ,the fluorescent intensity of the ciliary body,suprachoroidal space,anterior and posterior sclera,and choroid was less stronger than the control eyes,but fader than at the same area of the normal eyes obviously.ConclusionThe outflow effects of PZ drops on increasing uveoscleral pathway were obviously decreased in the SCG eyes.It depends on the activity of sympathetic nerve system partially.Besides,there are other mechanisms.
9.Total auricular reconstruction with single big expander at dual plane through incision at remnant ear.

OBJECTIVETo investigate the feasibility and advantages of total auricular reconstruction with single big expander at dual plane through incision at remnant ear.
METHODS52 patients with microtia were treated. Through incision at remnant ear, one big expander (150 ml) was implanted in the post-auricular area beneath the superficial facial in the hairless area, and above the superficial fascia in the area with hair. In the second stage, total auricular was reconstructed with autologous cartilage framework and expanded skin. Skin graft was not necessary. Another 32 paitents with single big expander above the superficial fascia were used as control. The downward shift distance of expanders, the appearance of auriculocephalic sulcus and complication were recorded in order to assess the effect of these two methods.
RESULTSThe downward shift distance of expanders in the dual plane group (0.7 ± 0.3) cm were lower than that in the control group (1.3 ± 0.4) cm, showing significant difference (P < 0.05). After a follow-up period of 6-12 months, shallow auriculocephalic sulcus was presented in 5 patients of the dual plane group and 12 patient of the control group. The shallow auriculocephalic sulcus was cut to make it deeper and covered with full skin graft.
CONCLUSIONSDual plane skin expansion could reduce the downward shift distance of expander. Adequate skin on the upper part of expander could make satisfactory auriculocephalic sulcus.

10.Lymphangiogenesis factor in the lymph node metastasis of head and neck cancer

Early lymph node metastasis is a common clinical phenomenon in many human cancers, which is associated with both aggressive disease and poor prognosis. Clearly, treatments to specifically block dissemination through the lymphatic network would be desirable either as independent therapies or as adjuncts to existing chemotherapy. At the present stage the target vessels for invasion by lymph-metastasizing tumor cells include preexisting tissue lymphatics abutting the tumor mass through either coincidence or mutual chemoattraction, as well as new lymphatic vessels that proliferate either within or around the tumor as a result of lymphangiogenesis. The lymphangiogenesis is regulated primarily by the lymphangiogenic growth factors VEGF-C and VEGF-D generated by tumor cells or host cells including tissue macrophages. Nevertheless, it remains controversial as to whether this metastasis-enhancing effect simply results from an increase of the number of target vessels due to mitogenesis or from other properties of the newly dividing tumor lymphatics.
