1.Relationship between trefoil factor 1 expression and gastric mucosa injuries as well as gastric cancer

Objective To determine whether trefoil factor 1(TFF1) could be associated with healing of gastric mucosa and differentiation of gastric carcinoma. Methods TFF1 in normal and various pathologic gastric mucosa was assessed by immunohistochemical method and analyzed with Motic Images Advanced 3.0 software. Results Compared with normal mucosa, increased TFF1 was detected in gastritis, gastric ulcer and duodenal ulcer. TFF1 was in increased expression in multiple and compound ulcer than simple ulcer. There was no significant difference among gastric ulcer, duodenal ulcer, gastritis and simple ulcer. Increased TFF1 was detected in peripheral mucosa of the gastric adenocarcinoma as compared with normal mucosa. The expression of TFF1 in gastric adenocarcinoma was related to the differentiation of adenocarcinoma, that is, the more poorly differentiated, the lower expression of TFF1. Conclusion TFF1 may play a significant role in gastric mucosa protection and epithelial restitution. TFF1 may also contribute to the differentiation of gastric adenocarcinoma.
2.Effects of febuxostat on the levels of IL-1β, TNF-αand COX-2 in serum of patients with gout

Objective To observe the effect of febuxostat on IL-1β, TNF-αand COX-2 in gout patients.Methods 82 patients with gout in our hospital from August 2015 to August 2016 were divided into the groups of 40 mg/day of febuxostat and 80 mg/day of febuxostat randomly, 41 cases respectively.Select the same period in our hospital health examination of 41 cases.The levels of serum uric acid were measured before and after one week, two weeks and four weeks after treatment.Serum levels of IL-1β, TNF-αand IL-1βin serum were measured by enzyme-linked immunosorbent assay (ELISA) COX-2 levels.Results The levels of serum uric acid in the group of 40 mg/day vs 80 mg/day of febuxostat were (570.4 ±70.5) μmol/L and (572.8 ±71.2) μmol/L, significantly higher than the normal group (296.5 ±9.7) μmol/L (P<0.05), serum uric acid level decreased gradually with the time of administration, and the serum uric acid levels of the two dosages of fosobutamol at one,two,four weeks were significantly lower than before treatment (P<0.05), but not to below the normal level.The IL-1βof 40 mg/day and 80 mg/day of febuxostat was (40.5 ±6.8) pg/mL and (41.7 ±7.2) pg/mL, COX-2 were (15.5 ±1.7) pg/mL and (15.8 ±1.8) pg/mL respectively, significantly higher than the normal group (8.8 ± 1.7) pg/mL (P<0.05), The expression of IL-1βwas significantly decreased at the second week of IL-1βdecreasing as the time of IL-1βdecreased gradually (P<0.05); TNF-αin 40 mg/day group and 80 mg/day group was significantly lower than that before treatrment (P<0.05), but not to below the normal level.Conclusion Febuxostat can significantly reduce serum uric acid levels in patients with gout, and may reduce IL-1β, TNF-αand COX-2 and other cytokine levels.
3.Management of recurrent inguinal hernia;the value of tension-free repair

Objective To evaluate tension-free repair in the management of recurrent inguinal hernia. Methods From 1/1993 to 12/2002, 163 patients with recurrent inguinal hernia underwent reoperation, clinical data were reviewed. Results The male: female ratio was 138: 25, age from 34 to 76 years. The primary surgery had been traditional hemiorrhaphy without prothesis in 142 cases (87. 1 % , including 65 cases of Bassini ,35 cases of McWay and 42 cases of Shouldice) , a tension-free procedure in 12 cases(7. 4% , including 3 cases of laparoscopic mesh repair) and unknown techniques in 9 cases(5. 5% ). The average interval from the initial surgery to recurrence was 36?14 months (range from 3 months to 10 years). Recurrent hernias were treated with a Shouldice repair in 71 cases(43. 6% ) , a tension-free repair in 92 cases(56. 4% , including a laparoscopic mesh repair in 3 cases). All these 163 cases(100% ) were followed up for 56 ?1 months (range from 30 months to 12 years) after the second surgery. It was found that after reoperation the recurrence rate of Shouldice procedure and tension-free repair was 16. 1% (11/71) and 2. 2% (2/92) respectively (x2 = 8. 327 ,P
4.Clinical study of Danshen Shuizi Capsule in preventing restenosis after stent placement intervention

Objective:To evaluate the preventive effect of Danshen Shuizi Capsule on vascular restenosis after stent implantation.Methods:48 patients with endovascular stent implantation were randomly assigned to control group(n=25) (treated with western medicines)and treatment group(n=23)(treated with Danshen Shuizi Capsule)after endovascular stenting implantation.The intraluminal diameter of stenting segment was measured by QCA at 6 months after stent implantation.Results: The diameter of reference vessel in two groups was not significantly different.The intraluminal diameter of stent segment in the treatment group was not larger than that in the control group(2.22?0.21)mm vs(2.18?0.25)mm.The index number of vessel diameter reduction in treatment group was not less than that in the control group(0.50?0.16)mm vs(0.51?0.11)mm.The rate of restenosis(18.4%)and reintervention(13.1%)in treatment group were not lower than those in the control(20%,12%).However,the complication in treatment group was significantly lower than that in control group.Conclusion:These findings indicated that endovascular was useful for the prevention of endovascular restenosis after stent implantation with fewer complications
5.Correlations of hyperuricemia and matrix metalloproteinase-9 level with left ventricular remodeling in patients with type 2 diabetes and the clinical effect of intervention for hyperuricemia

Objective To investigate correlations of hyperuricemia and matrix metalloproteinase -9 level with left ventricular remodeling in patients with type 2 diabetes and the clinical effect of intervention for hyperuricemia.Methods 100 type 2 diabetic patients with hyperuricemia in our hospital from June 2011 to June 2012 were selected as hyperuricemia group,50 type 2 diabetic cases with normal uric acid were considered as DM group.Levels of blood glucose,blood lipids,glycosylated hemoglobin and matrix metalloproteinase-9 were detected.All patients underwent echocardiography.Patients in hyperuricemia group were randomly divided into treatment group (receiving allopurinol and alkaline urine-alkalify therapy added to hypoglycemic treatment,n=50) and control group (receiving hypoglycemic treatment,n=50).After 6 months of treatment,levels of blood glucose,blood lipids,glycosylated hemoglobin,matrix metalloproteinase-9 and echocardiography were rechecked.Results The levels of triglyceride (TG),blood uric acid,MMP-9,left ventricular posterior wall thickness (LVPWT),interventricular septal thickness (IVST) and left ventricular mass index (LVMI) were increased in hyperuricemia group versus DM group[(2.3±0.5) mmol/L vs.(1.6±0.30) mmol/L,(498.9±32.5) μmol/L vs.(322.8±35.6) μmol/L,(765±38) g/L vs.(420±26) pg/L,(11.0±1.1) mm vs.(9.2±0.8) mm,(11.5±1.6) mm vs.(10.1±1.1) mm,(138.7±35.6) g/m2 vs.(115.0±10.4)g/m2,t=4.945,5.124,4.895,3.985,3.965,3.989,respectively,all P<0.05].Levels of TG,blood uric acid,MMP,LVPWT,IVST were decreased in treatment group after treatment versus before treatment [(1.6±0.5) mmol/L vs.(2.3±0.6) mmol/L,(323.0±30.5) μmol/L vs.(496.6±32.6) μmol/L,(766±34) pg/L vs.(410±25)pg/L,(9.1±0.8) mm vs.(11.0±1.1) mm,(9.2±1.1) mm vs.(11.6±1.5) mm,(116±10.5)g/m2 vs.(138.8±35.2) g/m2,t=3.386,4.525,4.865,3.256,3.895,3.985,all P<0.05].Correlation analysis showed that LVMI had positive correlations with levels of blood uric acid,TG,MMP-9 (r=0.435,0.348,0.356,respectively).Conclusions Hyperuricemia involves in left ventricular remodeling,and therapeutic intervention on the hyperuricemia can reverse this remodeling.
6.Study on Volatile Ingredient and GC-MS Fingerprint of Bu-yang Huan-wu Decoction

This study was aimed to establish GC-MS fingerprint of Bu-yang Huan-wu (BYHW) decoction. The 5HP-MS quartz capillary column was used. The temperature program carrier gas was He. The velocity was 1.0 mL·min-1. The injection volume was 1 μL. The split ratio was 1:20. The MS conditions were electron impact (EI) ion source with the transmission line temperature at 280oC, ion source temperature at 230oC, and quadrupole temperature at 150oC. The mass scan range was m/z 50~550. And the results were contrasted with ChemStation and NIST 05a. The results showed that nine common peaks were obtained in the fingerprint of eleven batches of BYHW decoction by peaks matched with the Similarity Evaluation System for Chromatographic Fingerprint (2004 A Version). The similari-ty of fingerprints was calculated more than 0.93. It was concluded that fingerprinting can be used as a method of quality control and provide evidence for the research of volatile ingredient of BYHW decoction.
7.The value of NT-probrain natriuretic peptide in predicting the severity of the coronary lesions in old patients with non-ST elevation acute coronary syndromes

Objective To evaluate the relationship between NT-probrain natriuretic peptide (NT-proBNP) levels and the degree of coronary artery stenosis in elderly patients with non-ST elevation acute coronary syndromes (NST-ACS).Methods The levels of NT-proBNP were determined in 258 elderly patients with NST-ACS divided into 3 quartile groups based on the degree of coronary artery stenosis,and 62 normal controls.And NT-pro BNP were compared among 4 groups.Results The serum levels of NT proBNP were increased in the NST-ACS patients with single,two or three-artery lesion compared with normal controls [(197.3±80.2)ng/L,(381.7±73.5)ng/L,(496.5± 99.8) ng/L vs.(68.2 ± 36.1) ng/L].The level of NT proBNP was enhanced along with increasing severity coronary artery disease (all P<0.01).And with aging,the NT-proBNP levels were rising [aged>60-69 years:(182.34±69.13) ng/L; aged≥70-79 years:(302.68±87.51)ng/L; aged≥80 years:(482.09±82.2)ng/L] (all P<0.01).Conclusions The NT-proBNP level is enhanced along with aging and increasing severity of coronary artery stenosis in elderly NST-ACS patients.
8.Relationship of plasma homocysteine and gene polymorphism of homocysteine metabolism related enzyme with diabetic peripheral neuropathy

Sixty cases of type 2 diabetes with diabetic peripheral neuropathy (DPN), 46 cases of type 2 diabetes without DPN and 50 healthy persons were recruited. Plasma homocysteine (Hcy), serum folic acid, vitamin B 12 levels and the mutations of Hcy metabolism related enzyme-methyhenetetrahydrofolate reductase (MTHFR) were assayed in individuals in this study. The results showed that high Hcy, low folic acid and vitamin B 12 levels are related with DPN in the patients with type 2 diabetes. The mutation of MTHFR C677T is associated with diabetes, but not with DPN.

Objective Moderate autophagy helps improve the viability of neurocytes.This study aims to investigate the effect of SP600125 on the autophagy and loss of nerve cells in the hippocampus in rats with subarachnoid hemorrhage (SHA).Methods Forty healthy male SD rats were equally randomized into a sham operation, an DMSO group, an SAH model, and an SP600125 group.The SAH model was established by vascular puncture and the rats of the SP600125 group were injected with 10 μL of SP600125 (3 μg/μL) into the lateral cerebral ventricle at 30 minutes before modeling.Sham group and SAH group were injected with equal volume of normal saline, DMSO group was injected with the same amount of DMSO.The animals were sacrificed at 24 hours after modeling for observation of the changes in the morphology and the number of neurons in the hippocampus by HE staining and qualitative and quantitative determination of the expressions of the p-JNK protein and the autophagy markers beclin-1 and LC3-II by immunohistochemistry and Western blot.Results Compared with the sham operation group, the neurons exhibited a disordered arrangement and the cells were polygonal and decreased in number in the hippocampus of the SAH models, while milder neuronal injury and more cells were observed in the rats of the SP600125 group than in the SAH models.The mean optical density values of Beclin-1, LC3-II and p-JNK in the hippocampus were significantly higher in the SAH models (14.66±4.40, 12.62±3.46, and 12.82±3.68) and DMSO (13.85±3.85、11.59±4.52、13.03±3.53), and the SP600125 group (9.86±3.14, 6.78±2.56, and 5.60±2.42) than in the sham operation group (1.56±0.28, 1.60±0.30, and 1.58±0.32) (P<0.05), but markedly lower in the SP600125 than in the SAH model group (P<0.05).The expressions of Beclin-1, LC3-II and p-JNK were remarkably increased in the SAH models (0.474±0.122, 0.668±0.130, and 0.496±0.124) and DMSO (0.432±0.102、0.628±0.113、0.416±0.094) and the SP600125 group (0.264±0.106, 0.332±0.113, and 0.219±0.104) than in the sham operation group (1.56±0.28, 1.60±0.30, and 1.58±0.32) (P<0.05), but significantly decreased in the SP600125 group as compared with the SAH models (P<0.05).Conclusion SP600125 has a protective effect on the neurocytes in the hippocampus of SAH rats, which may be associated with SP600125 moderately activating neuronal autophagy by inhibiting the activity of the JNK signaling pathway.
