1.Increased expression of caspase-3 in ipsilateral neonatal hypoxic-ischemia encephalopathy model in mice

Objective To observe apoptotic cells and caspase-3-positive cells in ipsilateral neonatal hypoxic-isch?emia encephalopathy (NHIE) model in mice. Methods CD1 mice of age 7 days (n=30) were randomly divided into two groups: sham group (n=9) and model group (n=21). NHIE model was induced by right common carotid artery ligation fol?lowed by 8%oxygen hypoxia for 100 min. TTC staining was used to determine area of brain infarction. DAPI staining was used to detect pathological change in brains. TUNEL assay was used to detect apoptotic cells and fluorescence immunohisto?chemistry was used to detect caspase-3 expression in the ipsilateral brain. Results No infarct was detected in sham group. Cells were densely and orderly arranged in brain. TUNEL-positive cells (18.57±4.98) and caspase-3-positive cells (9.17± 2.14) in the ipsilateral brain were both less than those in the ipsilateral brain of mice in model group (209.57±41.27) and (63.33±16.22) respectively. Mice in model group presented infarct in the right hemisphere with more dead cells and wider in?terstitial space compared with sham group. Conclusion Brain injury in NHIE model might be related to the increasing cas?pase-3 expression thus leads to apoptosis.
2.Study of polymorphism on 15 STR loci of Luoba ethnic group from Tibet

Objective To select 15 short tandem repeat(STR) loci that are highly and steadily polymorphic,and study the genetic relationships between Luoba ethnic group,Tibetan living in Lassa and Changdu area,and other Asian populations by using 15 STR polymorphism.Methods The blood samples of unrelated individuals from Luoba ethnic group,Lassa Tibetan and Changdu Tibetan were collected.DNA templates were screened using AmpF/ STR Identifiler kit and ABI3100Avant DNA analyzer with Genescan and Genotype software.The STR data of Luoba,Lassa Tibetan,Changdu Tibetan,other 21 Chinese ethnic groups and six Asian populations were compared;phylogenesis tree was constructed;the genetic relationships among ethnic groups living in Tibet and other Asian populations were analysed.Results Eight Hans clustered,and five ethnic groups of Guangxi clustered firstly;then the two clustered with Luoba,and two groups of Tibetan secondly;and then clustered with other Chinese populations,and then the Chinese groups clustered with other six Asian populations and Chinese Uygur lastly.Conclusion The results of this study are basically consistent with geographic distribution,and histories of these ethnic groups.They can provide the genetic proofs for the origin,migration,formation and development of ethnic groups living in Tibet.
3.The diagnostic value of apparent diffusion coefficient in monitoring the development of cerebral infarction

Objective To study the values of apparent diffusion coefficient (ADC) changing with time and space in cerebral infarction,and to provide the evidence in defining the infarction stages.Methods 117 work-ups in 98 patients with cerebral infarction (12 hyperacute,43 acute,29 subacute,10 steady,and 23 chronic infarctions) were imaged with both conventional MRI and diffusion weighted imaging.The average ADC values,the relative ADC (rADC) values,and the ADC values or rADC values from the center to the periphery of the lesion were calculated.Results The average ADC values and the rADC values of hyperacute and acute infarction lesion depressed obviously.rADC values in hyperacute and acute stage was minimized,and progressively increased as time passed and appeared as "pseudonormal" values in approximately 8 to 14 days.Thereafter,rADC values became greater than normal in chronic stage.There was positive correlation between rADC values and time[ (174 ±3.47)% vs (58±6.75)%,t =2.03,P <0.05 ].The ADC values and the rADC values in hyperacute and acute lesions had gradient signs that these lesions increased from the center to the periphery.The ADC values and the rADC values in subacute lesions had adverse gradient signs that these lesions decreased from the center to the periphery.Conclusion The ADC values of infarction lesions have evolution rules with time and space.The evolution rules with time and those in space can be helpful to provide the evidence in guiding the treatment or judging the prognosis in infarction.
4.DLK1 affects T cell acute lymphoblastic leukemia cells by regulating Notch pathway

Objective To detect the proliferation and the expression levels of downstream target genes of Notch pathway of T-ALL CCRF-CEM(CEM)cell line treated with exogenous DLK1 protein,in order to investigatethe effects of DLK1 protein on the Notch pathway in CEM cells.Methods CCK-8 assay was performed to examine the proliferation of CCRF-CEM cells,which were treated with various concentration(0.5,1.0,1.5 μg/ml)DLK1 for various time(24,48,72 h).RFQ-PCR was applied to assess the mRNA expression level of Notch1 receptor and downstream target genes of Notch pathway in CEM cells,which were treated for various time(24,48,72 h).Results DLK1 protein stimulated the proliferation of CCRF-CEM cells,and the proliferation rates with different concentrations of DLK1 were 0.14±0.03,0.17±0.04,0.55±0.01 in 72 hours,respectively,there were statistic differences between that in the experimental group and that in the control group(P<0.05).DLK1 protein up-regulated the Notch1 receptor and its downstream target genes HES1,c-myc and NF-κB.The relative transcript levels of target genes HES1 in 72 hours,c-myc in 48 hours and NF-κB in 72 hours were 3.2551±0.3100,1.6086±0.0941,2.0515±0.3453 respectively,and there were statistic differences between that in the experimental group and that in the control group(P<0.05).Conclusion DLK1 protein stimulates the proliferation of T-ALL CCRF-CEM cells by up-regulating Notch1 receptor and downstream target genes HES1,c-myc and NF-κB of Notch pathway.
5.Effect of Notch1 gene expression on the proliferation and apoptosis of T-cell acute lymphoblastic leukemia SupT1 cells and the mechanism

Objective To explore the relationship between T-cell acute lymphoblastic leukemia and the Notch signaling pathway.Methods Human T-cell acute lymphoblastic leukemia SupT1 cells were infected with the lentiviral vector made up specific Notch1-shRNA gene and nonspecific Notch1-shRNA gene.The inhibitive rate of SupT1 cells was detected by CCK-8.The rates of early apoptotic cells (Annexin V+/ 7-AAD-) and late apoptotic cells (Annexin V+/7-AAD+) were analyzed by flow cytometry and the expression levels of Notch1 receptor gene and downstream target genes were assessed by quantitative reverse transcription and polymerase chain reaction (QT-PCR).Results The cell inhibition rates of Notch1 interference group,control group and empty vector group at 96 h were 0.902±0.013,0,and 0.486±0.084,respectively,and it was increased obviously in Notch1 interference group (both P < 0.05).The cell early apoptosis rates of the three groups were (15.27±0.31) %,(5.57±0.25) %,(5.80±0.20) %,respectively.The cell early apoptosis rate of Notch1 interference group was increased obviously compared with the control group and empty vector group (both P < 0.05).While the cell late apoptosis rates had no significant difference among the three groups (P > 0.05).The mRNA expression levels of Notch1 receptor gene and its target genes (Hesl,c-myc,NF-κB) at 48 h,72 h and 96 h were higher than those in the control group and empty vector group (all P < 0.05).Conclusions The specificity of Notch1-shRNA can effectively decrease the Notch1 mRNA expression,and reduce the expression level of downstream target genes.Notch1 cut can inhibit the proliferation of SupT1 cells,and promote the early apoptosis.
6.Analysis of the relation between dental arch size and upper airway morphology in patients with obstructive sleep apnea hypoventilation syndrome

Objective:To study the anatomical correlation between dental arch and the volume of upper airway in patients with obstruc-tive sleep apnea hypoventilation syndrome(OSAHS). Methods: Dental arch architecture and upper airway volume were measured by cone beam CT(CBCT) in the subjects with OSAHS(n=22) and without OSAHS(n=19). The correlation between dental arch and the supper airway volume in OSAHS patients was analyzed. Results:The length of the upper dental arch and the height of palate in OSAHS patients were larger than those of the controls(All, P<0. 05). Cross-sectional area of nasopharynx and retropalatal and the total volume of upper airway were negatively correlated with the palatal height and upper dental arch length(P<0. 05), while positively correlated with upper dental arch of molar regions(P<0. 05). Conclusion:The abnormal shape of upper dental arch is related to the airway vol-ume of nasopharynx and retropalatal region in patients with OSAHS.
7.Struma Ovarii: Correlation of CT Appearances and Pathological Findings

Objective To analyzed the CT features of struma ovarii(SO).Methods The clinical data and CT features of eleven patients with pathologically proved SO were retrospectively analyzed.CT features were compared with pathological results.Results(1)All tumors were unilateral.On non-enhanced CT,the lesions presented as well-defined irregular masses,which were cystic-solid(n=6,54.5%)or cystic(n=5,45,5%).(2)The cystic portions presented as well-defined,multiple,various size,and there were entire cystic walls with smooth inner wall.The tumors(63.6%)showed a high at attenuation lesions in the cyst portion of the mass on precontrast scans and the attenuation ranged from 72.3 to 113.5 Hounsfield units(HU)in 7 cases.The solid portions showed irregular tissue density,and were often distributed in the cysts.After contrast administration,the cystic portions showed no enhancement,the solid portions marked enhancement,and the cystic walls demonstrated no moderate,or marked enhancement.(3)The tumors(72.7%)showed stippled calcification in solid portions and(or)cystic wall in 8 cases.(4)The tumors(27.3%)accompanied with mature cystic teratomas,and showed fat density in 3 cases.(5)The tumors(27.3%)accompanied a great of ascites and pleural liquid.Conclusion CT characteristics of SO might be of great value for the diagnosis.
8.A primary study of the relationship between apparent diffusion coefficient value of rectal adenocarcinoma on DWI and its pathological grading

Objective To investigate the relationship between apparent diffusion coefficient (ADC)value of rectal adenocarcinoma on DWI and its pathological grading.Methods The ADC values of 46 rectal adenocarcinomas were measured and compared with their histopathological grades.Results The 46 rectal adenocarcinomas included well differentiated adenocarcinomas in 14,moderate-ly differentiated ones in 20,and poorly differentiated ones in 12.The ADC values of well,moderately and poorly differentiated ade-nocarcinomas were (1.125±0.103)×10 -3 mm2/s,(1.030±0.098)×10 -3 mm2/s and (0.922±0.091)×10 -3 mm2/s,respective-ly,exhibiting a statistical difference (χ2 =1 7.35 1,P =0.000).Mann-Whitney U test showed that difference in ADC value between different histopathological grades was statistically significant.Conclusion ADC value of rectal adenocarcinoma can be used as a bio-marker for cell grading to guide treatment decision and prognosis assessment.
9.Role of HIF-1αin reduction of apoptosis in cortical neurons of rats by sevoflurane preconditioning:the relationship with Slit2∕Robo signaling pathway

Objective To evaluate the role of hypoxia inducible factor?1α ( HIF?1α) in reduction of apoptosis in cortical neurons of rats by sevoflurane preconditioning and the relationship with Slit2∕Robo signaling pathway. Methods Primary cortical neurons obtained from neonatal Sprague?Dawley rats were seeded in 6?well (2 ml∕well) or 96?well plates (100 μl∕well) at a density of 1×106∕ml, and randomly divided into 4 groups ( n=24 each ) using a random number table: control group ( C group ) , anoxia?reoxygenation ( A∕R ) group, sevoflurane preconditioning group ( SP group ) and HIF?1α inhibitor 2?methoxyestradiol group ( H group ) . The neurons were subjected to O2?glucose deprivation for 90 min followed by restoration of O2?glucose supply for 24 h. In group SP, the neurons were exposed to 2%sevoflurane for 2 h followed by 5 min washout with phosphate buffered saline for 3 times, and then sevoflurane preconditioning was performed immediately. In group H, sevoflurane preconditioning was performed with 5μmol∕L 2?methoxyestradiol at 72 h of incubation. The apoptosis in neurons was assessed using AnnexinⅤ?FITC∕PI assay, and apoptosis rate ( AR) was calculated. The amount of lactic dehydrogenase ( LDH) released was measured using colorimetric method. The expression of Slit2, Robo1 and Robo4 mRNA and protein was detected by fluorescent quantitative real?time polymerase chain reaction or Western blot. Results Compared with group C, the amount of LDH released and AR were significantly increased, Silt2 and Robo1 mRNA and protein expression was up?regulated, and no significant change was found in Robo4 mRNA and protein expression in A∕R group. Compared with group A∕R, the amount of LDH released and AR were significantly decreased in SP and H groups, and Silt2 and Robo1 mRNA and protein expression was up?regulated, and no significant change was found in Robo4 mRNA and protein expression in SP group. Compared with group SP, the amount of LDH released and AR were significantly increased, and Silt2 and Robo1 mRNA and protein expression was down?regulated in H group. Conclusion HIF?1α mediates reduction of apoptosis in cortical neurons of rats by sevoflurane preconditioning, and the mechanism is associated with Slit2∕Robo1 signaling pathway, but not with Slit2∕Robo4 signaling pathway.
10.Effects of IL-8 on the tight junction of vascular endothelial cells

Objective To investigate the influence of IL-8 on the tight junction of vascular endothelial cells.Methods Immunofluorescence was used to observe the modality and the distribution of three tight junction proteins (occludin,claudin-5 and ZO-1) of the EA.hy926 cells treated with IL-8 under different concentrations and different times.RT-PCR was used to measure the mRNA expression of these three proteins.Results The results demonstrated that IL-8 could change the distribution of occludin,claudin-5 and ZO-1 in EA.hy926 cells,and the mRNA expression of occludin,claudin-5 and ZO-1 decreased with the increase of IL-8 concentration and treated time.Conclusion The effects of IL-8 on the distribution and the expression of occludin,claudin-5 and ZO-1 are dose and time-dependent.
