1.The prognostic value of plasma brain natriuretic peptide in patients with acute pulmonary embolism

The aim of the study is to investigate the prognostic value of plasma brain natriuretic peptide (BNP) in patients with acute pulmonary embolism. Plasma BNP concentrations were measured in 47 cases with acute pulmonary embolism. Plasma BNP levels in massive or sub-massive pulmonary embolism were higher than those in no-massive pulmonary embolism (P = 0. 001,0. 048) ; BNP levels in survival cases were higher than those in fatal cases ( P = 0. 027 ). The incidence of massive or sub-massive pulmonary embolism in patients with BNP >400 pg/ml was higher than that with BNP≤400 pg/ml ( 100% vs. 75%,P = 0. 023 ). Results indicate that plasma BNP concentration might be associated with risk stratification and prognosis of the disease.
2.Application analysis of thematic apperception test in the internet addiction

Objective To understand the potential internet addicts personality traits through the use of Thematic Apperception Test (TAT) and provide evidence for psychological therapy.Methods The experimental group included 34 outpatients defined as Internet Addiction Scale (IAS) ≥ 5 points,and the control group included 34 volunteers with the same age and the common cultural backgroud and defined as IAS < 5 points.Results Compared with the control group,the internet addiction group had a higher desire projects including antisocial aggression (4.83±2.16),substance destroy (2.56± 1.61),intraggression (3.40±2.43) and passivity desires (2.65± 2.29) (P<0.05).In contrast,the internet addiction group had a lower desire projects including achievements(2.31 ±2.14),domination (1.51±1.29),nurturance (2.30±2.34),succorance (1.80± 1.25) (P<0.05,P<0.01).Compared with the control group,the internet addiction group had a higher pressure projects include emotion (3.90±2.81),emotional language offensive attack (3.62±2.18),physical social aggression (2.73±2.23),physical antisocial aggressive (3.95± 1.88),substance destroy (3.33± 1.78),nuturanced (4.53 ± 2.57),rejected (2.18 ± 2.19),loss (4.01±2.41),physical danger pressure (2.65±2.37) and total pressure (49.49± 36.16) (P<0.05,P<0.01).Conclusion Internet addiction patients have potential personality with ultra-low in superego,lack in ego,possess stronger in id.Outpatient treatment with internet addicts should focus on psychotherapy,and improve interpersonal relationships.
3.The application research in the close-loop muscle relaxant injection system combined with auditory evoked potentials index for monitoring in elderly patients during general anesthesia

Objective To observe the clinical efficacy and safety of close-loop muscle relaxant injcction system (CLMRIS) combined with auditory evoked potentials index for monitoring in elderly patients during general anesthesia.Methods A total of 100 patients undergong abdominal operation with ASA Ⅰ-Ⅱ anesthesia in our hospital from February 2012 to October 2012 were selected.Patients were randomly divided into 2 groups:the traditional group (n=50) and the CLMRIS group (n=50).The traditional group was anaesthetized according to clinical experience,and the CLMRIS group was used with the muscle relaxant guided by the close-loop muscle relaxant injection system.The auditory evoked potential (AEP) index and the life index were observed,and the dose of muscle relaxant,the time of consciousness recovery,dynamic response rate during operation and the adverse reaction were recorded at the time points at entering the operating room,induction of anesthesia,tracheal intubation,incision,into the abdominal,exploration,closing abdomen,skin suture,consciousness recovery,tracheal extubation,out of room (T0-T10).Results The vital signs in all patients were stable.The dynamic response rate during operation was lower in the CLMRIS group than in the traditional group at each time points of T8-T10 (all P<0.05).There was no increase in mean artery pressure (MAP) and heart rate in groups at the time points of T0-T1,T8-T10 (all P>0.05).The time of tracheal intubation,tracheal extubation and consciousness recovery were shorter in the CLMRIS group than in the traditional group [(268±30.4) min vs.(172±23.7) min,(14±6.4) min us.(18±7.9) min,(8.8±2.9) min vs.(12.2±4.6) min,respectively,t=3.277,6.341,3.346,all P<0.05].There were no significant differences in AEP index between groups at the time of T0 (P>0.05),but at the time of T1,the AEP index was lower in CLMRIS group than in the traditional group (P<0.05).Conclusions The application of close loop muscle relaxant injection system can improve the safety of general anesthesia in the elderly and reduce the incidence of postoperative residual paralysis.The CLMRIS combined with the auditory evoked potential for monitoring depth of anesthesia can prevent and reduce the incidence of intraoperative awareness,has a strong controllability in general anesthesia with smooth induction,and can reduce the dose of anesthetic drugs to achieve the best anesthetic effect.
4.Analysis of the relationship between temporary or permanent amenorrhea and ovarian function after uterine artery embolization

100 u/L 24 months later and another 38 year-old patient underwent surgical operation of ovaries bilaterally before UAE. Conclusions Temporary amenorrhea may not mean ovarian dysfunction or failure. The probability of ovarian dysfunction or failure after UAE is low, which mainly occur in the peirmenopausal patients. Consideration of the probability of inducing early ovarian failure, caution should be taken for UAE on the patients having ovarian surgery before. (J Intervent Radiol, 2006, 15: 472-475)

6.Manufacture of Complementary DNA Arrays on Amino-modified Slides

The use of rnicroarrays of oligonucleotides or cDNA is considered to be a promising approach for DNA and RNA sequence analysis, diagnostics of genetic diseases, gene polymorphism studies and analysis of gene expression. To manufacture cDNA microarrays the samples were printed onto glass microscope slides treated with poly-L-lysine, and then the slides were processed by heat and UV light treatment to attach the cDNA sequence to the glass surface. But the immobilization efficiency of cDNA on the glass surface was low. A simple procedure for manufacture cDNA microarrays on a slide treated with 3-aminopropyltrimethoxysilane is described. The efficiency for attaching cDNA to the amino-modified slides is greater than that to the slides treated with poly-L-lysine. The cDNA microarray made by the amino-modified slides is stable for use in 80℃, 75 % humidity, 3 600Lx light, exposure in air, respectively.
7.Rapid Detection of Mutations of Neisseria gonorrhoeae gyrA Gene Using DNA Chip

Objective To develop a new method-DNA chip to be used for rapid detection of mutations of Neisseria gonorrhoeae gyrA gene. Methods Probes were designed according to the sequence of Neisseria gonorrhoeae gyrA genes, and DNA chip was fabricated accordingly. DNA fragment which contains gyrA gene mutation was amplified using PCR technique, labeled with Cy5 fluorescence, and then hybridized with DNA chip. Results of DNA sequencing were used as the control. Results All of the 50 urogenital swab specimens were detected using DNA chip. The consistency between the DNA chip and drug sensitivity test, and between the DNA chip and DNA sequencing were 100% and 98%, respectively. Conclusions DNA chip is a rapid technique with high sensitivity and specificity for the detection of mutations of Neisseria gonorrhoeae gyrA gene. This method can be used for the detection of drug resistance in clinical practice.

Objective To investigate the relationship of OPRM1 C354A mutations with the clinical efficacy and toxicity of Morphine in the treatment of cancer pain.Methods We recruited 100 patients with moderate-severe cancer pain treated at our department from January 2016 to December 2016,and divided them into a CA(wild type homozygotes) group,a CG(mutated heterozygotes) group,and a GG (homozygous homozygous) group,according to the allele type of OPRM1 C354A.Regular analgesic doses of Morphine were given to all groups,and VAS scores and adverse reactions at 2,4,8,16,24,aud 48 hours after analgesia were collected and compared among the groups.Results The (OPRM1 C354A mutation groups(CG+GG)had lower VAS scores at every time point,compared with those of the non-mutation group(CA) (2 h:3.61±0.39 vs.4.04±0.52;4 h:3.88±0.41 vs.4.20± 0.15;8 h:3.95±0.32 vs.4.37±0.24;16 h:3.81±0.38 vs.4.33±0.15;24 h:3.84±0.25 vs.4.42± 0.18;and48 h:3.86±0.20 vs.4.41± 0.14) (t=4.648,5.261,7.461,8.454,13.389,and 16.030,respectively,each P=0.000).The incidences of constipation(23.08% vs.6.25%)and vertigo(25% vs.8.33%)in the OPRM1 C354A mutation groups(CG+GG)were significantly higher than those in the non-mutation group(CA) (x2 =5.543 and 4.914.P=0.019 and 0.027,respectively).Conclusions Polymorphism of the (PRM1 C354A gene is associated with the clinical efficacy and toxicity of morphine in the treatment of cancer pain.
10.Influence of labeled primer and labeled dUTP assays on the signal intensity of the chip for the detection of HBV gene polymorphism.

BACKGROUNDTo evaluate the influence of assays with primer labeled with fluorochrome (Cy5) and dUTP labeled with Cy5 on the signal intensity of the chip for detection of hepatitis B virus (HBV) gene polymorphism.
METHODSThe P-region and pre-C/C-region of HBV gene were amplified by polymerase chain reaction (PCR) with Cy5 labeled primer or Cy5 labeled dUTP. The amplicons of the two assays were hybridized with chips, scanned and analyzed by computer software for the detection of HBV gene polymorphism.
RESULTSThe signal intensity of assay with Cy5 labeled dUTP was slightly higher than that of assay with Cy5 labeled primer, but non?specific signal intensity of the assay with Cy5 labeled dUTP was higher. The result of 42 samples showed that there was no significant difference between the two assays, and that both had a good repeatability and CV value (15%-20%).
CONCLUSIONSThe assay with Cy5 labeled primer may replace the assay with Cy5 labeled dUTP as a routine method to detect HBV gene polymorphism, and it is simpler and cheaper.
