1.Effect of Sanazole in Combination with ?-ray Irradiation on In Vivo CFU-GM from Bone Marrow in Mice

OBJECTIVE:To examine the effects of sanazole and of sanazole combining with ?-ray irradiation on the colony formation ratio of granuloid/macrophage committed progenitor cell(CFU-GM)from mouse bone marrow in vivo.METHODS:(1)After a series of doses of sanazole solution(50,100,200mg/kg)was given to mice intravenously,CFU-GMs were separated from mouse bone marrow for evaluating their colony formation ratio.(2)To evaluate the radiosensitization of sanazole,a series of doses of sanazole solution(50,100,200mg/kg)was given to mice intravenously 30min before the mice were exposed to ?-ray irradiation in different doses(5,10,20Gy)respectively.After each treatment,CFU-GMs were separated from mice marrow for counting the ratio of colony formation.RESULTS:(1)Sanazole especially in large doses didn’t showed considerable cytotoxicity to CFU-GM colony formation in mice in vivo.(2)The inhibiting effect of a series of doses of sanazole solution combined with a series of doses of radiation (5,10,20Gy)on colony formation of CFU-GM from mice bone marrow was the same as that of radiation alone.CONCLUSION:Large doses of sanazole showed no obvious effect on the colony formation of CFU-GM of mouse bone marrow in vivo.No radiosensitization to the colony formation of CFU-GM was found in vivo when a series of doses of sanazole in combination with radiation were used.
2.Retrospective study on relationship between expression of insulin-like growth factor receptor and prognosis of patients with osteosarcoma

Objective to observe the correlation between the expression and prognosis of osteosarcoma Insulin-like growth factor receptor(IGF1R),explore the factors affecting the prognosis of patients with osteosarcoma.Methods between January 2005 and April 2014 menstrual standardized comprehensive treatment of 60 cases of patients with stage ⅡB osteosarcoma were retrospectively analyzed.Using immunohistochemical method,the detection of osteosarcoma tissue and IGF1R expression rate in normal tissue adjacent to tumors and to analyze its correlation with survival of patients with osteosarcoma and factors affecting the prognosis.Results IGF1R in the positive expression rate of osteosarcoma tissues was 80.0% (48/60),expression in normal tissue adjacent to tumor rate was 4/8,has a statistically significant difference between (P =0.032).IGF1R positive expression in osteosarcoma tissues group median disease-free survival time and the median overall survival time were significantly lower than the negative expression group (P =0.023,P =0.040) respectively.IGF1R expression level and pathological necrosis rate of preoperative chemotherapy,the time of first visit to the lung metastasis,and the survival prognosis of patients have significant correlation (P < 0.05),and age,tumor location,tumor size,the serum LDH and ALP levels,is urgent to make a diagnosis and give treatment time,factors such as operation,preoperative chemotherapy times no significant correlation (P > 0.05).Condusion IGF1R positive expression rate in stage Ⅱ B osteosarcoma tissue significantly negatively correlated with the prognosis of patients with osteosarcoma,is the independent prognostic factors of patients with osteosarcoma.
3.Super-early Minimal Invasive Evacuation of Hematoma via Keyhole for Treating Hypertensive Basal Ganglionic Hemorrhage

Objective To estimate the efficacy of super-early minimal invasive evacuation of hematoma via keyhole for treating hypertensive basal ganglionic hemorrhage. Methods 36 patients with hypertensive basal ganglionic hemorrhage(9 cases in grade Ⅱ, 17 in grade Ⅲ and 10 in grade Ⅳ) were treated by minimal invasive evacuation of hematoma via keyhole at super-early stage, and their efficacy was statistically compared with 35 patients with hypertensive cerebral hemorrhage underwent conservative therapy or lateral ventriculopuncture. Results In the 36 patients underwent minimal invasive evacuation of hematoma via keyhole, 22 cases had a good prognosis(61.1%), 9 had a poor prognosis(25.0%), and 5 died(13.9%). Among the 35 patients underwent conservative therapy or lateral ventriculopuncture, 5 cases had a good prognosis(14.3%), 22 had a poor prognosis(62.9%), and 8 died(22.9%). 4.Effects of femoral isthmus on cementless total hip replacement

BACKGROUND:The template method used in cementless total hip arthroplasty is influenced by many factors,so the accuracy is poor.Studies demonstrated that femoral isthmus plays an important role in total hip arthroplasty,which affect femoral prosthesis.OBJECTIVE:To discuss the effects of femoral isthmus on cementless total hip arthroplasty.METHODS:The images of 4 cadaver femoral isthmus were obtained by computer tomography.The relations between femoral component and cortical bone contact were observed on coronal slices by sawing manully.The longest and the smallest medullary canal dimensions and the cortical thickness of femoral isthmus were measured by CT films from 50 patients (55 hips) prepared forprimary total hip arthroplasty.The size of prosthesis was predicted based on the smallest medullary canal dimension of isthmus.And the conventional preoperative template was served as control.RESULTS AND CONCLUSION:There were a press fit and good contact between the femoral component and cortical bone of isthmus in 3 cadaver femoral bones.In 50 patients,the longest medullary canal dimensions of femoral isthmus were 8.2-22.4 mm(mean 14.2±3.1 ram).The smallest medullary canal dimensions of femoral isthmus were 6.1-17.9 mm[mean (10.2±2.9) mm],and the cortical thickness of femoral isthmus were 2.7-12.7 mm[mean (7.5±1.8) mm].The results of CT-isthmus measurement method predicted sizes were excellence in the 50.9% cases.There was statistically significant difference of predicted sizesbetween the excellence rate of CT-isthmus measurement method and the rate of control (P<0.05).Particularly,the excellence rate of CT-isthmus measurement method was 63.9% in the group aged below or equal to 65 years.When the ratio of the longest medullary canal dimensions and the smallest medullary canal dimensions of femoral isthmus was large than or equal to 1.4,the excellence rate of CT-isthmus measurement method was 70%.The press fit and good contact between the femoral component and femoral isthmus plays a very important role in stabilizing of femoral prosthesis in total hip arthroplasty.The predicted sizes of femoral prosthesis depending on the medullary canal dimensions of femoral isthmus measured by CT film has better accuracy than the conventional preoperative template.
5.Interleukin-17 and interleukin-23 mRNA expressions in peripheral blood of patients with psoriasis vulgaris and their correlations with disease severity

Objective To compare the mRNA expressions of interleukin (IL)-17 and IL-23 in peripheral blood of patients with psoriasis vulgaris versus healthy individuals,assess the relationship of these parameters with psoriasis area and severity index (PASI) score,and to investigate the therapeutic mechanisms of total glucosides of peony (TGP) for psoriasis vulgaris.Methods Fifty patients with psoriasis vulgaris and 40 healthy individuals were enrolled in this study.Of these patients,42 were treated with TGP of 600-900 mg twice a day for 8 weeks.Blood samples were obtained from all the healthy individuals,50 patients before treatment,42 patients after 4-week treatment,and 23 patients after 8-week treatment.Real-time fluorescence-based quantitative reverse transcription PCR (RT-PCR) was used to determine the mRNA expression levels of IL-17 and IL-23 in the blood samples.The severity of psoriasis was evaluated using PASI score before and after the treatment.Statistical analysis was done by t test,rank sum test,and Pearson correlation analysis using the SPSS16.0 software.Results The IL-17 and IL-23 mRNA expression levels (given in △Ct value) in the patients before treatment were significantly higher than those in the healthy controls (IL-17,-5.32 ± 0.80 vs.2.79 ± 0.76,t =47.71,P < 0.05; IL-23,-5.43 ± 0.68 vs.-3.77 ± 0.86,t =10.38,P < 0.05),and positively correlated with the PASI score (r =0.61,0.52 respectively,both P < 0.05).A significant decrease was observed in the mRNA expression levels of IL-17 and IL-23 as well as PASI score in the 42 patients after 4-week treatment with TGP compared with those before treatment(IL-17,-2.24 ± 0.61 vs.-5.30 ± 0.78,t =20.40,P < 0.05; IL-23,-1.97 ± 0.74 vs.-5.44 ± 0.68,t =21.69,P < 0.05; PASI,5.8 ± 2.7 vs.9.4 ± 4.2,t =4.68,P < 0.05),and in the 23 patients after 8-week treatment compared with those after 4-wek treatment(IL-17,-1.51 ± 0.78 vs.-2.21 ± 0.59,t =3.50,P < 0.05; IL-23,-1.27 ± 0.81 vs.-1.89 ± 0.72,t =2.70,P< 0.05; PASI,3.8 ± 1.8 vs.7.3 ± 2.5,t =5.47,P< 0.05).Conclusions It seems that both IL-17 and IL-23 are involved in the pathogenesis of psoriasis vulgaris,and TGP treatment can reduce the mRNA expression levels of IL-17 and IL-23 as well as PASI score in patients with psoriasis vulgaris.
6.The Evaluation of Cranial and Cervical Arteries and the Brain in TIA Patients with MRA and DWI

Objective To evaluate magnetic resonance angiography(MRA) and diffusion weighted imaging(DWI) features of transient ischemic attacks(TIA).Methods 40 patients with TIA were chosen as study subjects,and 34 healthy adults and 36 cases with brain infarction were randomly chosen as control subjects.MRA and DWI was performed with a superconductive GE 1.5T system.The relationship between TIA and the degree and distribution of stenosis of cranial and cervical arteries,the development of the vessels,DWI findings of the brain was analyzed.Results There were significant differences among the three groups in the number and the degree of stenosis of cranial and cervical arteries(P=0.006).The stenosis and occlusion of arteries were severe in the patients with TIA and brain infarctions than that in healthy adults(P=0.004,0.015),but there were no significant differences between TIA and brain infarctions(P=0.148).The incidence of dysplasia of unilateral vertebral arteries(VA) in TIA was higher than that in the normal subjects(P=0.039).The internal carotid artery(ICA)system supplying to the regions of brain tended to show more ischemic focus on DWI(4/8,50%) in the cases with TIA.Conclusion Severe stenosis or occlusion and irregular filling defects of cervical and cranial arteries in patients with TIA might be one of the risk factors for severe ischemia stroke.The hypoplastic VA was one of predisposing factor for TIA.
7.The clinical characteristics of interstitial lung disease in patients with dermatomyositis and polymyositis

Objective To analyze the clinical features,and prognosis of the interstitial lung disease (ILD) in patients with dermatomyositis (DM) and polymyositis (PM) by chest X-ray,chest high-resolution CT scan (HRCT) and pulmonary lung function.Methods Thirty-three patients hospitalized with DM/PM associated ILD were retrospectively analyzed.Results Thirty-three patients with ILD were confirmed by HRCT.Abnormal pulmonary function tests were available in 82% of patients.Clinical-imaging analysis revealed that the pathological features of ILD were non-specific interstitial pneumonia (NSIP,57%) and unusual interstitial pneumonia (UIP,25%).UIP types showed a poor prognosis and high mortality (70%).Conclusion This study shows that HRCT is more sensitive for the diagnosis of ILD than lung function tests and chest X-ray.Combined HRCT and chest X-ray with lung function tests and blood gas analysis have shown that the major pathological types of ILD are NSIP and UIP,in which UIP are associated with high mortality and poor prognosis.
8.MRI and CT representations of intracranial dermoid cyst

Objective To evaluate the MRI and CT representations and their valuation of intraeranial dermoid cyst. Methods 23 cases who suffered from intracranial dermoid cyst confirmed by pathology were included retrospectively. The MRI and CT representations, and the pathological and clinical characteristics were analyzed.All the cases were performed with MRI, and 20 cases were performed with CT at the same time. Results 22 cases showed inhomogeneous high signal intensity in T1WI and T2WI, 1 cases showed iso-high signal intensity in T1WI and low signal intensity in T2WI. 8 cases showed plaque-like inhomogeneous high signal intensity in DWI. 19 cases showed homogeneous low density in CT scans with standard window width and level ,amt showed inhomogeneous low density with large window width and low window level. One case showed high density in CT scans with standard window width and level. Fat drop in subrachnoid space and bilateral cerebral ventricle and interstitial brain edema were found in 8 cases with MRI and CT. Conclusion Intracranial dermoid cyst always have typical MRI and CT representations. Distinctive representations could be found when the rupture of dermoid cyst exist, and correct diagnosis could be made before operation. MRI is better than CT in the diagnosis of dermoid cyst.
9.Effects of continuous venoveHous hemodiafiltration on organ function and eytokines in dogs with multiple organ dysfunction syndrome

Objective To investigate the effects of continuous vennvenous haemodiafihration(CVVHDF)on major organ fimction and plasma cytokine levels in MODS dogs.Methods Fifteen Beagle dogs were subjected to hemorrhagic shock plus resuscitation and endotoxemia to establish MODS model,then dogs were randomly divided into 2 groups:CVVHDF group(n=8)and MODS group without CVVHDF (n=7).CVVHDF was performed for 12 hours after endotoxin injection in CVVHDF group dogs.Serum ALT,AST,Scr,BUN and blood gas were routinely measured,and plasma TNF-α,IL-6 and IL-10 coneentrations were measured by ELISA at different time points. Results Plasma levels of IL-6 and IL-10 were significantly decreased in the CVVHDF group at 3 h,6 h,9 h and 12 h after endotoxin injection as compared with MODS group (P<0.01).TNF-α levels were similar in both groups(P>0.05).IL-6 and TNF-α were detected in the ultrafihrate.The sieving coefficients (SC) for IL-6 and TNF-α were 0.27±0.13 and 0.1±0.1 respectively.IL-10 could not be detected in the ultrafiltrate.In CVVHDF group,function of major organs was improved.The beneficial effect of CVVHDF on the hypotension of MODS dogs was particularly striking at 6 h,9 h,and 12 h alter endotoxin injection(P<0.01).Mean PaO2 in CVVHDF groups at 3 h,6 h,9 h,and 12 h after endotoxin injection was significantly higher than that in MODS group(P<0.01). Conclusion CVVHDF effectively removes IL-6 and IL-10from the cireulation,attennates endotoxin-induced hypotension and improves arterial oxygenation in MODS dogs.
10.Purge-trap Gas Chromatography and Mass Spectrometric Method for Analysis of Isoprene in Natural Waters

Gas chromatography with mass spectrum detector ( GC-MSD) coupled with purge-and-trap system was set up to analyze the concentration of isoprene in natural waters. The best experimental conditions were established, including purge gas flow rate ( 50 mL/min ) , purge time ( 15 min ) , the optimum capillary column ( Rt-Alumina BOND/KCl) and the appropriate condition of temperature programming. When analyzing isoprene in natural waters, the precision was <4% (n=6), the detection limit was 0. 5 pmol/L and the recovery was 91%-102%. The preservative experiment showed that there was no obvious variation in sample concentrations of isoprene within 60 days. The concentrations of isoprene measured with the method ranged from 60 . 8 to 278 . 7 pmol/L in the Jiaozhou Bay and its adjacent river estuaries and from 44 . 7 to 77 . 2 pmol/L in Yellow River estuary, which was in good accord with those results reported in literatures in other coastal waters. In conclusion, the analytical method could meet the requirements of the analysis of concentration of isoprene in natural waters.
