Objective: To sum up characteristics of hotline counseling about mental health problems in Shanghai. Method: All records of hotline counseling from 1990 to 2000 were input into computer. Retrospective analysis was done. Results: In the past 10 years, the main issues in hotline counseling were associated with love affairs (18.1%), emotional troubles (15.8%), psychosis (11.3%), and interpersonal relationship (8.1%), which were also related to the help-seekers background, such as gender, age, education levels, occupation and marriage status.
3.Tests of Reliability and Validity of Short-form Arthritis Impact Measurement Scales 2(AIMS2-SF)

Objective To evaluate the reliability and validity of Short-form Arthritis Impact Measurement Scales 2(AIMS2-SF).Method AIMS2-SF not including the role component was used to evaluate Quality of Life(QOL) of 51 arthritis patients,then its test-retest reliability and the internal consistency reliability were tested,and factor analysis was applied to determine its construct validity.Results The Intra-class correlation coefficients(ICCs) of test-retest reliability of AIMS2-SF ranged from 0.60 to 0.80,and the Cronbach's ? coefficient for internal consistency reliability ranged from 0.65 to 0.83.Furthermore,the factor analysis confirmed the construct validity for AIMS2-SF.Conclusions The study suggested that the AIMS2-SF had satisfactory reliability and validity,then it is applicable for the evaluation of QOLs of Chinese patients with arthritis.
4.The role of endogenous cystathionine beta synthase/hydrogen sulfide and heme oxygenase-1/carbon monoxide in a rat model of global cerebral ischemia-reperfusion

Objective To investigate the role of endogenous cystathionine beta synthase (CBS)/hydrogen sulfide (H2S) and heme oxygenase-1 (HO-1)/carbon monoxide (CO) during global cerebral ischemia-reperfusion (I/R).Methods Thirty male Wistar rats weighing 180-220g were anesthetized with intraperitoneal (IP) 3% pentobarbiral 40 mg?kg-1. Global cerebral I/R was produced by 4-vessel occlusion. Bilateral vertebral arteries were cauterized and bilateral common carotid arteries were occluded with atraumatic clamps for 20 min. The clamps were then released to allow reperfusion. The animals were randomly divided into 5 groups (n=6 each) : group Ⅰ sham operation(C); groupⅡ I/R; in group Ⅲ (Z+I/R),Ⅳ(H+I/R) andⅤ(Z + H + I/R) zinc protoporphyrin (HO-1 inhibitor) 45 ?mol?kg-1 or/and 1 ml of 5 mmol?L-1 hydroxylamine (CBS inhibitor) were given IP 30 min before I/R. The animals were killed at 6h of reperfusion. Brains were removed immediately for determination of H2S, CO, GSH, MDA level and SOD activity and expression of CBS mRNA and HO-1 mRNA in hippocampus and histologic examination with electron microscope. Results H2S, CO, MDA content and CBS mRNA and HO-1 mRNA expression were significantly higher while GSH content and SOD activity were significantly lower in I/R group than in control group (sham operation) . CO content and HO-1 mRNA expression were significantly lower while H2S, GSH content and CBS mRNA expression were significantly higher in Z + I/R group than in I/R group. H2S, GSH content and CBS mRNA expression were significantly lower and CO content and HO-1 mRNA expression were significantly higher in H + I/R group than in I/R group. In Z + H + I/R group H2S, CO, GSH content, SOD activity and expression of HO-1 mRNA and CBS mRNA were significantly decreased whereas MDA content was significantly increased as compared with I/R group. Mitochondria in hippocampal neurons were severely damaged in group Ⅱ-Ⅴ and the damage was worst in group Z + H + I/R. Conclusion CBS/H2S and HO-1/CO systems can protect brain against I/R injury.
5.Chtonic diseases and the MOS 36 item short from health survey score in community residents older than 60

This study was to investigate the relationship between chronic diseases and quality of life among community residents beyond 60.From September 2005 to November 2005,1731 residents aged 60-89 were surveyed with a general information form and the the MOS 36 item short from health survey (SF-36).The results showed that the subjects with no chronic diseases seemed to have higher scores,while SF-36 values dropped to some extent when the number of diseases increased.In comparison with diabetic or hypertensive subjects,those with heart disease had lower SF-36 scores for all items,except for general conditions and physical capability.
6.Survey on depression status of elderly residents in a Shanghai community

Surveys on depression status of elderly population in a community of Shanghai Jing'an District were carried out in 2005 and 2008 respectively. The cluster sampling method was used in the study,1731 and 2315 community residents aged 60 -89 were selected in two surveys. The depression status was evaluated with geriatric depression scale (GDS). During the first survey 297 out of 1731 people were identified as depression with a prevalence rate of 17. 2%; during the second survey 374 out of 2315 people were found to have depression with a prevalence rate of 16. 2%. The factors related to depression status included family support, hobby, ability of self-taking care, family finance and chronic diseases.
7.A CPLs vector-based RNAi technology to inhibit the expression of rat CⅡTA and MHCⅡ genes

Objective To investigate the inhibitory effects of cationic polymeric liposomes (CPLs) vector-based RNA interference (RNAi) technology on the expression of rat MHCⅡ transactivator ( CⅡTA) and MHCⅡgenes .Methods According to the genetic information of CⅡTA downloaded from GenBank, three short hairpin RNA (shRNA) sequences targeting CⅡTA sequences were designed .CPLs vectors were constructed and coupled to shRNA plasmid vectors to form pCⅡTA-CPLs vectors .Six groups including control group , CPLs control group , pHK-CⅡTA control group and three pCⅡTA-CPLs groups were set up.Rat dendritic cells (DCs) were transfected in vitro.Real time PCR and flow cytometry analysis were used to detect the expression of CⅡTA and MHCⅡat mRNA and protein levels in DCs after transfec-tion.Results The pCⅡTA-CPLs vectors were successfully constructed .Compared with control groups ,the transcription level of CⅡTA and MHCⅡand the expression of MHCⅡat protein level were significantly in-hibited in all pCⅡ TA-CPLs groups ( P<0 .01 ) .The strongest inhibitory effects of pCⅡTA-CPLs on the ex-pression of CⅡTA and MHCⅡgenes were observed in the second pCⅡTA-CPLs group.There was a positive correlation between the expression of CⅡTA and MHCⅡ.Conclusion CPLs vectors were effective gene carriers.The constructed pCⅡTA-CPLs vectors significantly inhibited the in vitro expression of rat CⅡTA and MHCⅡ, which provided evidences for further investigation on pCPLs-CⅡTA vectors in vivo.
8.The clinical observation of advanced primary liver cancer under the chemotherapy of hyperthermia combined with EOF approach

Objective To discuss the clinical efficacy of advanced primary liver cancer under the chemother-apy of hyperthermia combined with EOF approach.Methods 75 patient samples who had advanced primary liver cancer in total were chosen and were divided into the two groups randomly.The control group containing 37 patient samples had advanced primary liver cancer which would be applied with EOF approach.The treatment group contai-ning 38 samples had advanced primary liver cancer.The treatment strategy was to apply EOF approach combined with high -frequency hyperthermia in vitro.Results The total rate of efficiency on the treatment group and control group was 73.68% vs 48.6%.The DCR rate was 92.1% versus 56.8%,1 year survival rate was 57.8% versus 45.9%and 2 years survival rate was 10.5% versus 5.4% which showed that the results of the treatment group was obviously better than that of the control group.Conclusion The method that combining the chemotherapy of hyperthermia with EOF to treat advanced primary liver cancer can obviously improve clinical efficacy and survival period.What′s better is that this approach will not increase the toxicity of chemotherapy.
9.HGPAEs vector-based RNAi inhibits the expression of gene encoding MyD88 in rat liver tissues

Objective To investigate the inhibitory effects of histidine grafted poly (β-amino es-ter) ( HGPAEs) vector-based RNA interference ( RNAi) on the expression of gene encoding myeloid differ-entiation factor 88 (MyD88) in rat liver tissues.Methods The sequence of small hairpin RNA (shRNA) was designed based on the genetic information of MyD 88.HGPAEs vector was constructed and coupled with shRNA plasmid targeting MyD88 to construct pMyD88-HGPAEs vector.Rats were divided into five groups including control group , HGPAEs treatment group , pHK-HGPAEs treatment group , shRNA treatment group and pMyD88-HGPAEs treatment group .The rats in each group were transfected with the corresponding inter-ventions through portal vein injection .Real-time PCR and Western blot assay were performed to detect the expression of MyD88 in liver tissues 3 days after transfection .Results The pMyD88-HGPAEs vector was successfully constructed .The expression of gene encoding MyD 88 was inhibited in rats from shRNA treat-ment group and pMyD88-HGPAEs treatment group (P<0.05).Significantly decreased expression of gene encoding MyD88 at mRNA and protein levels were observed in rats from pMyD 88-HGPAEs treatment group as compared with those from other groups (P<0.01).Conclusion HGPAEs vector might be used as a po-tential gene carrier .The expression of gene encoding MyD 88 in rat liver tissues could be significantly inhibi-ted through portal vein injection of pMyD 88-HGPAEs vector .This study provided evidences for further re-search on pMyD88-HGPAEs vector in a high responder model of rat orthotopic liver transplantation .
10.PI3K/Akt/NF-κB regulate ABCB1/P-glycoprotein-mediated multidrug resistance in colon carcinoma cells

Background and purpose: Multidrug resistance (MDR) is the dominating obstacle to the chemotherapy. There is strong evidence that the phosphoinositide 3-kinases (PI3Ks) signaling pathway is involved in MDR phenotype, however, the mechanism of MDR occurrence is still unknown. This study tended to investigate the regulating effect of PI3K/Akt signaling pathway and its downstream target genes in P-glycoprotein (P-gp) (ABCB1 gene encoding)-mediated MDR in human colon carcinoma HCT-116/L-OHP cells. Methods:Pretreatment with PI3K selective inhibitor LY294002 (20μmol/L) for 2 h, the sensitivity of L-OHP was evaluated by the CCK-8 (cell counting kit-8) assay in HCT-116/L-OHP cells, and the expressions of P-gp, LRP, MRP-2, Akt, p-Akt, IκB and p-IκB were evaluated by Western blot. The activity of ABCB1 promoter was evaluated by chromatin immunoprecipitation analysis (CHIP). Results: After inhibiting the activity of PI3K/Akt signaling pathway, the IC50 value of L-OHP decreased from(157.48±16.73)μg/mL to (53.68±3.18)μg/mL in HCT-116/L-OHP cells, and the reversal index was 2.93 (P<0.01). The expressions of P-gp, p-Akt and p-IκB were down-regulation compared with the concrol group (P<0.01), but the expressions of LRP, MRP-2, Akt and IκB didn't change signiifcantly. CHIP result has conifrmed that NF-κB protein could bind to the region of ABCB1 gene promoter in HCT116/L-OHP cells. Conclusion:Blocking of PI3K/Akt/NF-kB signal pathway could increase the drug sensitivity to MDR cells, inhibit the phosphorylation of p-Akt and p-IκB, and reversing ABCB1/P-glycoprotein-mediated multidrug resistance in colon carcinoma cells.
