Cullinl is a member of the cullin genome.The cullin1 assembly complex has typical ubiquitin ligase activity,and mediates cell cycle proteins,especially cancer-associated protein degradation process,an then effectively regulates the cell cycle progression.Cullin1 play an important role in the genesis and development of malignant tumors.

Objective To establish the high performance liquid chromatography(HPLC) fingerprint of wuzi yanzong pills. Methods HPLC was performed on Agilent Extend C18 column (250 mmí4. 6 mm,5 μm) with a gradient elution system using acetonitrile:methanol (101)-0. 4% phosphoric acid as the mobile phase. The column temperature was set at 30 ℃ and the flow rate was 1. 0 mL·min-1 . The eluate was detected at the wavelength of 254 nm. Chromatographic peaks were identified by LC-MS method. Results Nine common peaks in wuzi yanzong pill samples were identified by comparing their LC-MS data with those of reference compounds and related reference reports. The HPLC fingerprint of wuzi yanzong pills was finally developed based on the analysis of sixteen batches of samples and their similarities were above 0. 93. Conclusion This method has high precision,stability and repeatability. This study could be used for overall quality assessment of wuzi yanzong pills.

Objective To investigate the possibility of anti-liver fibrosis of 13-estradiol nanoparticle prepared by interfacial polymeri-zation method with butylcyanoacylate as carrier material (E2-PBCA-NP) and its effect on the expression of transforming growth factor β1 and connective tissue growth factor in pig serum induced animal fibrotic model. Methods Male Sprague-Dawley (SD) rats were random divided into five groups. Except normal control group, other four groups were all given intraperitoneal injection with pig serum. Therapeutic drugs were administered to rats from the ninth week after injection of pig serum. All rats were killed at the end of the twelfth week. Several experi-ments were done as below, the tissues of liver were observed by Masson staining, and the mRNA of TGF-β1 and CTGF of liver samples were detected by RT-PCR. Meanwhile, the expression of TGF-β1 and CTGF protein were detected by immunohistochemistry. Results It showed that both E2 and E2-PBCA-NP treatment groups had lower stage of liver fibrosis, according to the observation of pathology by Masson staining (P < 0.05). The anti-liver fibrosis effect of E2-PBCA-NP treatment group was better than that of E2 treatment group (P < 0.05). The mRNA and protein level of TGF-β1 and CTGF were markedly reduced by E2 and E2-PBCA-NP treatment, compared with liver fibrotic model groups (P <0.01). There was no statistical difference between E2-PBCA-NP and E2 treatment (P >0. 05), while no significant change was observed in blank nano -particle group (P > 0.05). Conclusion Both E2-PBCA-NP and E2 had anti-liver fibrosis activity. E2-PB-CA-NP has stronger anti - liver fibrosis activity than E2, which could be resulted from the inhibition of TGF-β1 and CTGF expression.

Objective To evaluate the helical CT diagnostic value of primary retroperitoneal neoplasm(PRN). Methods 32 cases of PRN confirmed by operation and pathology were retrospectively analyzed. Plain and enhanced CT scan were perfomed in 28 cases,and only 4 cases underwent plain CT scans. Results Of 32 cases,15 were benign tumor and 17 cases were malignant tumor.Among them ,16 cases were mesenchymal tissue-origin(11 cases were malignant neoplasm), 10 cases were nervous tissue-origin(3 cases were malignant neoplasm),3 cases were rudimental embryonal tissue-origin(all benign), and the source of unknown-origin were 3cases(all malignant neoplasm).To be correctly localized was 28 cases(87.5%) and correctly qualitative diagnosis of the tumor was 20 cases (62.5%) by CT before operation. Conclusion PRNs have many typies, helical CT provides informations in both position and characteristics before operation.

Objective To evaluate the effects of different concentrations of sevoflurane on the cognitive function in neonatal rats.Methods Twenty-four neonatal Sprague-Dawley rats of both sexes,aged 7 days,weighing 12-16g,were randomly divided into high concentration sevoflurane group (group Sev1),low concentration sevoflurane group (group Sev2) or control group (group C),with 8 rats in each group.Sev1 and Sev2 groups were exposed to 2％ and 4％ sevoflurane for 6h,respectively,and C group inhaled gas mixture composed of 30％ oxygen.At 2 and 8 weeks after exposure to sevoflurane,cognitive function was assessed using the open field test and Morris water maze test.Results In the open field test,no significant differences were observed among the three groups (P ＞ 0.05).Morris water-maze test showed that the escape latency was significantly prolonged,and the frequency of crossing the original platform and the time spent in the third quadrant were decreased at 2 weeks after exposure to sevoflurane in Sev1 group and at 2 and 8 weeks after exposure to sevoflurane in Sev2 group as compared with group C (P ＜ 0.05).Morris water-maze test showed that the escape latency was significantly prolonged,and the frequency of crossing the original platform and the time spent in the third quadrant were decreased at 2 and 8 weeks after exposure to sevoflurane in Sev2 group compared with Sev1 group (P ＜ 0.05).Conclusion Exposure to 4％ and 2％ sevoflurane for 6h both can induce cognitive decline and the decreasing effect is enhanced with the increasing concentration in the neonatal rats.

Objective To study the human telomerase reverse transcriptase (hTERT) and spleen tyrosine kinase (SYK) activity expression and molecular margin in the tumor center and peripheral tissues of patients with colorectal cancer, and to explore the security of tumor surgical margins. Methods The expressions of hTERT and SYK were examined by SABC and Supervisan immunohistochemistry in 20 specimens of colorectal cancer and tissues 1 cm, 2 cm, 3 cm, 4 cm and 5 cm, away from proximal and distal margin of the tumor respectively. Results The positive expression rate of hTERT in colorectal cancer was 80.00 %, the rate in tissues 3 cm, 4 cm and 5 cm away from the tumor margin was 0. The positive expression rate of hTERT in tissues from the cancer center to 3 cm away from the tumor margin was gradually diminishing (P<0.01). The positive expression rate of SYK in colorectal cancer was 10.00 %, the positive expression rate of SYK in tissues from the cancer center to 3 cm away from the tumor margin was gradually increased (P<0.05). Conclusion There is molecular margin around colorectal cancer tissues. Considering hTERT and SYK expressions, 3 cm can be defined as the safe molecular margin for the radical operation of colorectal cancer.

Objective:To observe the expression of chemokine receptor CCR7 and CXCR4 in human colorectal carcinoma and its significance,so as to assess their expression with the metastasis and prognosis of colorectal carcinoma. Methods:Immunohistochemistry was used to detect the expression of chemokine receptor CCR7 and CXCR4 in 110 patients with colon cancer.The relationship between the CCR7 and CXCR4 expression and the clinic pathological characters was statistically analyzed. Results:CCR7 and CXCR4 expression were positively expressed in 59.1% and 45.5%of the patients. The high expression rate of CCR7 and CXCR4 in lymph node metastasis positive cases was 81. 5% and 60.0% respectively,but the without lymph node metastasis positive cases were 26.7% and 24.4% respectively. Objective To discuss the therapeutic options for treatment of subaxial cervical dislocation combined with facet locking. Methods There were 49 patients with cervical dislocations including 7 patients with dislocation at C3,4, 15 at C4,5, 14 at C5,6 and 13 at C6,7. Eleven patients were with old dislocation, with duration of dislocation ranging from 2 hours to 61 days. Neurologic status of the patients according to Frankel scale was graded A in 14 patients, grade B in nine, grade C in 10 and grade D in nine. All patients were treated surgically after closed reduction with skull traction. Results The successful reduction rate was 63% for fresh dislocation, with average improvement of 0.65 grade for spinal cord function. All bone grafts got fusion at four months after operation. Conclusion Therapeutic options are based on fresh or old dislocations, paraplegia or not, intervertebral disk injury severity, and reduction or not through traction for patients with lower cervical dislocations.
