Objective 17β-estradiol is known to have a neuroprotective effect. The aim of this study was to investigate the effects of 17β-estradiol on propofol-induced neuroapoptosis in primarily cultured cortical neurons. Methods Rat cortical neurons were primarily cultured for 7 days and randomly divided into groups A ( vehicle control) , B, and C, treated with equal volume of 20%intralipid, 500 μmol/L propofol, and 500 μmol/L propofol +0.1 μmol/L 17β-estradiol, respectively. At 12 hours after treatment, the morphology of the neurons was observed under the microscope, their survival rate calculated by MTT, their apoptosis was deter-mined by FCM assay, and their mitochondrial membrane potential measured by fluorescent dye rhodamine 123. Results Compared with group A, group B showed a significantly reduced number of neurons, lack of 3-dimensional appearance, unclear contour, and fractured neuron axons, but a remarkable improvement was observed in the propofol-induced morphological damage in group C.The survival rate of the neurons and the mitochondrial membrane potential were markedly decreased in group B ([52.3 ±5.2]% and [59.1 ± 5.3]%) as compared with groups A ( [99.9 ±3.6]%and [99.6 ± 5.8]%) and C ([90.1 ±7.2]%and [89.2 ±7.1]%) (both P<0.01 ) , while the rate of neuroapoptosis significantly increased in group B ([43.4 ±4.6]%) in comparison with A ([3.1 ±0.2]%) and C ([22.3 ±3.2]%) (both P<0.01). Conclusion 17β-es-tradiol can protect against propofol-induced apoptosis of primarily cul-tured neurons by inhibiting the reduction of their mitochondrial membrane potential.

Chinese Journal of Experimental Traditional Medical Formulae 2023;29(1):105-112
ObjectiveTo observe the effect of modified Zuoguiwan combined with perindopril tert-butylamine tablets on early diabetic kidney disease (DKD) of Qi-Yin deficiency with blood stasis syndrome. MethodA total of 77 patients with early DKD of Qi-Yin deficiency with blood stasis syndrome, who were treated in the Ruikang Hospital Affiliated to Guangxi University of Chinese Medicine from september 2020 to november 2021, were randomized into the control group (38 cases) and observation group (39 cases). The control group received basic treatment and perindopril tert-butylamine tablets, and the observation group was treated with modified Zuoguiwan and perindopril tert-butylamine tablets in addition to the basic treatment. The treatment lasted 3 months for both groups. The symptom scores, blood glucose [fasting blood glucose (FBG), 2 h postprandial blood glucose (2 h PBG), glycosylated hemoglobin (HbA1c), time in range (TIR)], serum creatinine (SCr), urea, estimated glomerular filtration rate (eGFR), cystatin-C (Cys-C), serum beta 2-microglobulin (Sβ2-MB), and urinary microproteins [24 h urinary microalbumin (24 h UmAlb), 24 h micrototal protein (24 h MTP), urine beta 2-microglobulin (Uβ2-MB)] were observed in two groups. ResultThe two groups showed no significant difference in the various indexes before treatment. After treatment, the total effective rate of the control group was 84.2% (32/38), as compared with the 94.9% (37/39) in the observation group (Z=6.03, P<0.05). After treatment, the symptom scores were lower than those before treatment in both groups (P<0.01) and lower in the observation group than in the control group (P<0.01). TIR level was higher (P<0.05) and levels of FBG, 2 h PBG, and HbA1c (P<0.01) were lower than those before treatment in both groups. After treatment, the TIR level in the observation group was significantly increased as compared with that in the control group (P<0.01). After treatment, the level of Cys-C was lower than that before treatment in both group (P<0.01) and lower in the observation group than in control group (P<0.01). The level of Sβ2-MB in both the observation group (P<0.05) and control group decreased as compared with that before treatment. The 24 h UmAlb and Uβ2-MB of two groups were decreased after treatment as compared with those before treatment (P<0.05). Moreover, 24 h MTP in observation group (P<0.05) was lower than that before treatment. After treatment, the levels of 24 h UmAlb, 24 h MTP and Uβ2-MB in observation group were lower than those in control group (P<0.01), and levels of SCr and eGFR in observation group did not change significantly as compared with those in the control group. ConclusionModified Zuoguiwan combined with perindopril tert-butylamine tablets can reduce blood glucose fluctuations and urinary albumin excretion in early DKD patients of Qi-Yin deficiency with blood stasis syndrome. The efficacy is better than that of basic western medicine.

Objective:To cultivate human-derived prostate cancer (PCa) cells via conditional reprogramming cell (CRC) technology, and establish individualized cell bank for PCa research in vitro. Methods:We obtained three fresh PCa tissue samples from different patients between January 2019 and April 2019. Then each sample was divided into two parts. One was used for cancer nature confirmation by intraoperative biopsy. Another part was sent to the laboratory and digested into single primary cancer cells with 0.25% EDTA enzyme for CRC technology. The details were described as followed: 1. The primary PCa cells were co-cultured with 3T3-J2 cells irradiated by 30 Gy (feeder cells) in conditioned medium, and observed for the growth of cell clones, 2. The feeder cells were removed by 0.25% EDTA trypsin for 1 minute before primary PCa cells digested for passage. All primary PCa cells were validated by multiple experiments such as immunofluorescence, immunohistochemistry, immunoblotting and fluorescence in situ hybridization (FISH).Results:Total three cases of human-derived PCa cell lines were successfully established during 15days through CRC technology. All those primary PCa cells could be steadily and continuously passaged, which also expressed AR, CK5, CK18, P504s and PSA. FISH demonstrated that each cell line harbored≥1.6% TMPRSS2/ERG fusion and conformed to the features of PCa.Conclusion:CRC technology can be used for stable and continuous PCa cell culture in vitro.
