1.Clinical Efficacy and Safety of Bianyan Oral Liquids in the Treatment of Acute Pharyngitis

Objective:To observe the clinical efficacy and safety of Bianyan oral liquids in the treatment of acute pharyngitis. Methods:The clinical data of 200 patients with acute pharyngitis in the outpatient department of traditional Chinese medicine in our hospital from September 2010 to June 2012 were analyzed. They were divided into the control group with 100 cases and the observation group with another 100 cases. The control group was given Jinlianhua granules, while the observation group was received Bianyan oral liquids. After 5 days, the symptom score, effective rate, changes in accompanied symptoms and safety index were observed and com-pared. Results:The symptom score and effective rate of the observation group were better than those of the control group (P<0. 05), and the improvement in defecation was better than that of the control group (P<0. 05) as well. There were two cases in the observation group suffered nausea and stomachache, and one case in the control group suffered diarrhea while no untoward effect emerged. Conclu-sion:Bianyan oral liquids with the effects of heat-clearing and detoxicating, nourishing yin and fluid and treatment both manifestation and root cause show significant effect in the treatment of acute pharyngitis.
2.Differentiation of human umbilical cord mesenchymal stem cells into hepatocyte-like cells induced by liver homogenate supernatants

BACKGROUND:Preliminary experiments have demonstrated that rat liver homogenate supernatants can induce the morphological changes of human umbilical cord mesenchymal stem cells. However, little is known about whether the induced cells have some phenotypic and functional features of hepatocytes.
OBJECTIVE:To investigate whether human umbilical cord mesenchymal stem cells have some phenotypic and functional characteristic of hepatocytes after being induced by liver homogenate supernatants.
METHODS:Passage 3 human umbilical cord mesenchymal stem cells were used and divided into control group (cells were cultured in basic culture medium) and liver homogenate supernatant group (cells were cultured in liver homogenate supernatants for 3, 5, 7 days). Meanwhile, positive control group (QSG-7701 human liver celllines) and negative control group (simple liver homogenate supernatants) were set up. The protein and mRNA level of hepatocyte markers, alpha-fetoprotein, cytokeratin 18 and tryptophan 2,3-dioxygenase enzyme, were detected at different time points.
RESULTS AND CONCLUSION:After inducement, the stem cells of fusiform shape began to lose their sharp edges and progressively shrunk, and then they changed into hepatocyte-like cells with the morphous of triangle, polygon and anomalism shape. Compared with the control group, the protein and mRNA level of alpha-fetoprotein, cytokeratin 18 and tryptophan 2,3-dioxygenase enzyme significantly increased time dependently after inducement with liver homogenate supernatants (P<0.01). This study demonstrated that human umbilical cord mesenchymal stem cells are able to differentiate into hepatocyte-like cells in vitro that possess some functions of liver cells.
3.The Mutation Analysis of Common Deafness Genes Using SNPscan Technology in Nonsyndromic Hearing Loss Patients of Minority Ethnicities in Qinghai Province

Objective This study aims to investigate the mutation spectrum and frequency of GJB2 , mtDNA12SrRNA,and SLC26A4 genes in Hui people,Tibetan,Tu nationality,and Mongolian patients with non-syndromic hearing loss in Qinghai province.Methods Peripheral blood samples were obtained from a total of 211 minority patients with nonsyndromic hearing loss in Qinghai province to extract genomic DNA.Three genes of GJB2,mitochondrialDNA12SrRNA,and SLC26A4 were screened for mutations in our study cohort using SNPscan technology.Results Among these 211 patients,5 Tu patients and 1 Mongolian patient were found to carry the ho-moplasmic mtDNAA1555G mutation.The GJB2 mutations detection rates were 11.38%,4.55%,5.88%,and 10%in Hui people,Tibetan,Tu nationality,and Mongolian patients,respectively.No statistically significant differences in the GJB2 mutations detection rates were found among all four ethnicities (P>0.05).c.235delC was the most prevalent mutation in both Tu patients and Mongolian patients.The allele frequency was 2.94% and 5%,respec-tively.While for Hui patients,c.299 300delAT was the most prevalent mutation with the allele frequency of 4.47%.The mutations detection rates of SLC26A4 were 6.5%,4.55%and 2.94%in Hui people,Tibetan,and Tu nationality patients,respectively.No statistically significant differences in the SLC26A4 mutations detection rates were found among all three ethnicities (P>0.05).c.235delC was the most prevalent mutation in Hui patients,the allele frequency was 2.44%.While for Tibetan patients,c.1226G>A was the most prevalent mutation with allele frequency of 2.27%.Conclusion A total of 10.9% of deaf patients have inherited hearing impairment caused by GJB2,SLC26A4,and mtDNAA1555G mutations.The mutation spectrum of GJB2 and SLC26A4 genes has the eth-nic specificity in nonsyndromic hearing loss patients of minority ethnicities in Qinghai province.

C1q /TNF-related proteins (CTRPs)belong to the same category of C1q /TNF-related protein family,mainly secreted by adipose tissue and widely expressed in human and rodent.Their function turned out to be metabolic modulation,cardiovascular system protection and suppression of inflammation.But for metabolic modulation,enhancing the synthesis of glycogen,promoting fatty acid uptake and oxidation,in-creasing insulin sensitivity is the major role of CTRPs playing in governing the metabolism.Each CTRP has its own unique tissue expression profile and nonredundant function in regulating sugar and /or fat metabo-lism.In this review we focus on the recent progress about the metabolic function of CTRPs.
5.Application of Antibacterials in Orthopedic Clean-operations During Perioperative Period:A Survey and Analysis

OBJECTIVE To understand the situation of application of antibacterials in perioperative period in Department of Orthopedics,in order to find evidence for strengthening the rational use and standard management of antibacterial.METHODS A total of 308 cases with orthopedic clean-operations were randomly selected for the analysis of rationality of application of antibiotic prophylaxis in perioperative period.RESULTS All of the 308 patients were given antibiotics in perioperative period.And 18 kinds of drugs from 7 categories were used,the average kinds used were(2.07?0.75).The patients started antibiotic administration within 0.5-2 hours before operation accounted for 84.74%.The average time of antibiotic using was(6.07?2.64) days.72.40% patients in medication period used more than one kind drugs.CONCLUSIONS There are some problems in application of antibacterials in orthopedic clean-operations during perioperative period,such as too long-term use of antibiotics,frequent changes and so on.It is necessary to strengthen the management and improve the rationality of application of antibacterials.
6.Determination of Palmatine Hydrochloride and Berberine Hydrochloride in Baishi Shuitiao Powder by HPLC

Objective:To establish an HPLC method for the determination of palmatine hydrochloride and berberine hydrochloride in Baishi Shuitiao Powder simultaneously. Methods:A reversed phase ion pair HPLC method was adopted. An Agilent Zorbax SB-C18 (150 mm × 4. 6 mm,5 μm)column was used, the mobile phase consisted of acetonitrile-0. 1% phosphoric acid (50 :50) (0. 2 g do-decyl sodium sulfate added to 100 ml solution), the flow rate was 1. 0 ml·min-1, the detection wavelength was at 345 nm, the injec-tion volume was 10 μl, the column temperature was at room temperature. Results: A good linear correlation was observed within the range of 1. 11-22. 16μg·ml-1 for palmatine hydrochloride (r=0. 9996) and 2. 11-42. 24μg·ml-1 for berberine hydrochloride (r=0. 9998). The average recovery was 98. 94% with RSD of 2. 43% for palmatine hydrochloride and 101. 3% with RSD of 2. 05% for berberine hydrochloride (n=6). Conclusion:The method is rapid, simple and accurate, and can be used for the content determina-tion of palmatine hydrochloride and berberine hydrochloride in Baishi Shuitiao Powder.
7.Effects of recombinant epidermal growth factor receptor antisense adenovirus combined with irradiation on breast cnacer cells

Objective To investigate the effects of a recombinant antisense adeno virus for epidermal growth factor receptor (EGFR) combined with irradiation on b reast cancer cells.Methods Human EGFR cDNA fragment was subcloned in the oppos ite orientation to the cytomegaloviral promoter and inserted into a E1/E3-delet e d type 5 adenoviral vector to obtain AdE5 construct which expresses EGFR antisen se RNA. Combined with ?-ray irradiation, its effects on clonogenicity and cell cycle phase distribution were studied in a human breast cancer line MDA-MB-231 . Results EGFR protein expression was dramatically inhibited in MDA-MB-231 cell s after AdE5 infection. The post-irradiation clonogenicity was reduced by AdE5 in a viral and irradiation dose-dependent manner. Further cytometric analysis show e d that AdE5 infection at a?MOI of 300?pfu/cell induced a cell cycle progre ssion from radio-resistant G 0+G 1 phases to radiosensitive G 2+M phases, resultin g in a synergistic effect after combination of these two treatments. Conclusions The t ransduction of EGFR antisense RNA by adenoviral vector is effective for antisens e strategy targeting EGFR, and increases the cell-killing effect of ionizing radiation on breast cancer cells.
8.The clinical application study on 64-MSCT virtual endoscopy in the examination of colon lesions

Objective To evaluate 64-MSCT virtual endoscopy in the examination method,displaying ability and clinical application of colon lesions.Methods Compared the results of 49 cases of patients examined with 64-MSCT with that got from Coloscopy,and statistical analysis was conducted.Results A total of 19 cases of adenocarcinoma and 2 cases of colonic diverticula as well as 44 cases of adenomatous polyp were detected.The display rate of polyps was 100% in which was larger than 10mm,73% in which was range of 5 ~ 10mm in size and 50% in which was smaller than 5mm.Conclusion As an relatively noninvasive examination method,64-MSCT virtual endoscopy is concordant with onventional colonoscopy in the aspect of detectable rate and revealing the lesion morphologic and can be used as an important examination measure in the diagnosis for colonic diseases.
9.Effects of a strong pulsed magnetic field on the growth-related gene expression of human bladder cancer BIU-87 cells

Objective To estimate any influence of strong pulsed magnetic fields on the expression of growth-related genes in human bladder cancer BIU-87 cells. Methods Human BIU-87 cells were cultured in vitro and randomly divided into a magnetic field group and a control group. Each group was further divided into 24 h, 48 h and 72 h sub-groups. The magnetic field group cells were exposed to an 8 T magnetic field pulsed at 15 Hz for 2 h every day. The control group cells also placed on the same environment, but not exposed to any strong, pulsed magnetic field. The expression of B cell lymphoma/leukemia gene-2 (Bcl-2) mRNA, Bax mRNA and caspase-3 mRNA was measured with RT-PCR, and flow cytometry was used to evaluate the expression of the Bcl-2, Bax and caspase-3 genes of the tumor cells in vitro. Results The expression of Bax mRNA and protein was significantly higher in the cells exposed to the magnetic field than in the control groups. The expression of Bcl-2 mRNA and protein was significantly less. The expression of caspase-3 mRNA and protein in the two groups showed no significant differences.Conclusions A strong, pulsed magnetic field can inhibit the growth of bladder tumor BIU-87 cells and promote their apoptosis. The mechanism is probably related with the magnetic field promoting Bax mRNA and protein expression and inhibiting Bcl-2 mRNA and protein expression.
10.Clinical study of comprehensive Traditional Chinese Medicine treatment for limb dysfunction after ischemic stroke

Objective:To evaluate the clinical effect of Limb dysfunction after the comprehensive treatment of ischemic stroke by Traditional Chinese Medicine (TCM).Methods:A total of 160 patients with limb dysfunction after ischemic stroke. Who in line with the inclusion criteria, were randomly divided into 2 groups by random number table, 80 cases each. These patients were treated in Multicenter Union Hospital from June 2017 to Janunry 2019. The patients in the control group were given basic Western medicine treatment and rehabilitation training, while the patients in the observation group were given a comprehensive treatment combined with traditional Chinese medicine based on the control group (Chinese herbs, herbal fumigation and ear point press). Both groups were treated for 4 weeks and follow-up for 2 months. The Fugl-Meyerscale were used to evaluate the degree of limb dysfunction and balance dysfunction, the National Institutes of Health Stroke Scale were used for assessing the damage extent of nerve function, the Barthel index for evaluating the mobility of daily life. The clinical efficacy was evaluated according to the method of Brunnstrom assessment.Results:After treatment, the Limb strong spasm (1.57 ± 0.36 vs. 1.98 ± 0.53, t=5.724), Hemianesthesia (1.37 ± 0.31 vs. 1.80 ± 0.36, t=8.096), Inhibited bending and stretching (1.31 ± 0.25 vs. 1.84 ± 0.46, t=9.055) in the observation group were significantly lower than those in the control group ( P<0.01). The Fugl-Meyer activity and balance rating in the observation group were significantly higher than those in the control group ( t values were 2.739, 4.705, respectively, P<0.05 or P<0.01). The total effective rate of observation group was 93.3% (78/80), the control group was 73.3% (71/80), and there exist statistical significance ( χ2=4.783, P=0.028) in the two group’s comparative difference. Conclusion:The comprehensive TCM treatment can improve the hemiplegia syndrome of ischemic stroke patients and their limb activity and balance function, promote their neural functional recovery, enhance the activity of their daily life and clinical efficacy.
