1.Status and role of three-dimensional printing technology in spine surgery

BACKGROUND: Spine has a variety of morphological structures and complicated adjacent location, so the surgery to treat spinal abnormality is a difficult. Three-dimensional (3D) printing technology can transform 3D digital model into physical model, which has been widely used in the field of medicine, even popularized gradually in spine surgery. OBJECTIVE: To summarize the application and status of 3D printing technology in spine surgery, and to make aprospect. METHODS: The first author retrieved CNKI, Baidu Xueshu, Google Scholar, WanFang and PubMed databases for the articles published from 2010 to 2016 using the keywords of 3D printing technology, spine surgery, navigation template,spine physical model, pedicle screw, individualized implants, clinical teaching in Chinese and English, respectively.Finally 29 eligible literatures were enrolled for further analysis in accordance with the inclusion and exclusion criteria.RESULTS AND CONCLUSION: 3D printing technology has been extensively applied in spine surgery, and shows rapid development in preoperative diagnosis, intraoperative navigation, communication, teaching, graft production, implant customization, bone tissue engineering and other aspects. With the development of society, and medical imaging, 3D printing technology will be more promising in the field of medicine, especially in spine surgery
2.Stem cell transplantation for inflammatory bowel diseases

BACKGROUND:Stem cell transplantation can, in theory, rebuild the immune system and regulate immunologic function. In addition, the transplanted stem cells may differentiate into tissues or cells we want. Therefore, this intervention may restore gastrointestinal inflammatory lesion and exert therapeutic effect on inflammatory bowel diseases (IBD). OBJECTIVE:To review the application of different kinds of stem cells in IBD treatment. METHODS:A computer-based search of CNKI, PubMed and FMJS was performed for articles concerning stem cel transplantation and IBD published after January 2002. The key words were stem cells, inflammatory bowel diseases, Crohn's diseases, ulcerative colitis, cell therapy in Chinese and English respectively. At last, 49 articles were included in result analysis. RESULTS AND CONCLUSION: Embryonic stem cells and adult stem cells both have certain therapeutic benefits in IBD. But which kind of stem cell is the safest and most efficacious cannot be confirmed, and at the same time the most effective and the optimal dose and way of transplantation of such cells are still unclear. All in all, there is still a lot of work to do to remove the obstacles.
3.Expression and clinical signiifcance of Nusap1 in hepatical carcinoma

Objective:To investigate the expression and clinical significance of Nusap1 in hepatical carcinoma. Methods:The expression of Nusap1 protein in 61 specimens of hepatical carcinoma was examined by immunohistochemistry. Based on the levels of Nusap1 expression, the 61 specimens were divided into a high Nusap1 expression group and a low Nusap1 expression group. The correlation between Nusap1 expression with clinicopathologic features and prognosis of hepatical carcinoma was analyzed. Results:TherateofhighNusap1expressionwas54.1%inhepaticalcarcinoma.TherateofhighNusap1 expression was 21.3%in noncarcinoma, with signiifcant difference between the 2 groups (P<0.01).Nusap1 overexpression had signiifcant correlation with histological differentiation, tumor size, liver cirrhosis,lymphaticmetastasis,tumorthrombiandearlyrecurrence(P<0.05),butnotwithsex,age,AFP level,tumornumber,TNMclassificationandtumorencapsulation(P>0.05).Survivalanalysissuggested thatthe6monthand12monthnoncarcinomasurvivalratewassignificantlylowerinthehighNusap1 expression group [33.3%(11/33), 17.9%(5/33)] than that in the low Nusap1 expression group [89.3%(25/28), 53.6%(15/28);P<0.005]. Conclusion:Nusap1 is overexpressed in hepatical carcinoma and is a valuable prognostic factor for hepatical carcinoma.
4.Analysis of Clinical Pharmacists Particpating in the Clinical Drug Treatment Consultation in China

OBJECTIVE:To focus on the situation of clinical pharmacists particpating in the clinical drug treatment consulta-tion in China. METHODS:With the keywords of“clinical pharmacists”,“consultation”and others,retrieved from China Hospital Knowledge Database (CHKD),Wanfang Database and China BioMedical Literature Service System (SinoMed),the situation of clinical pharmacists particpating in the clinical drug treatment consultation in China was concluded and summarized. RESULTS:A total of 186 literatures were included,invloving 114 effective literatures. 2 606 cases were clinical pharmacists particpating in the clinical drug treatment consultation,2 257 cases were all or part of adoption of clinical pharmacists’suggestions,the execution lev-el of clinicians for clinical pharmacists’consultation suggestions reached 93.02%. After carrying out the clinical pharmacists’con-sultation suggestions,patients’condition improved or cured reached 2 290 cases,with the effective rate of 94.47%. CONCLU-SIONS:Clinical pharmacists participating in the clinical consultation has certain function,however,the working method and mode need to be further explored and standarized.
5.Efficacy Observation of Extracorporeal Shock Wave Lithotripsy Combined Use Paishi Decoction in the Treatment of Kidney Stone

OBJECTIVE:To observe clinical efficacy of extracorporeal shock wave lithotripsy combined with Paishi decoction in the treatment of kidney stone. METHODS:160 patients with kidney stone were selected and randomly divided into observation group and control group,with 80 cases in each group. Control group only received HB-ESWL-VG extracorporeal shock wave litho-tripsy;observation group additionally received Paishi decoction 300 ml,tid,for consecutive 1 week,on the basis of control group. Clinical efficacy was observed in 2 groups;the levels of serum inflammatory factor IL-2,IL-6,TNF-α,T lymphocyte subset CD3+,CD4+,CD8+ and CD4+/CD8+,erythrocyte immune function indicator C3b receptor rosette(C3bRR),immunity complex ro-sette(ICR)and tumor erythrocyte rosette(TER)level were detected in 2 groups before and after treatment. RESULTS:The effec-tive rate of observation group(98.75%)was significantly higher than that of control group(91.25%),with statistical significance (P<0.05). After treatment,the levels of IL-2,IL-6 and TNF-α in observation group was significantly lower than in control group, the levels of CD3+,CD4+,CD4+/CD8+,C3bRR,ICR and TER were significantly higher than control group,with statistical signifi-cance(P<0.05). Comparsion of CD8+between 2 groups was not statistically significant(P>0.05). CONCLUSIONS:The extracor-poreal shock wave lithotripsy combined with Paishi decoction can relieve inflammation reaction,enhance immune function and im-prove therapeutic efficacy.
6.Construction and expression of trefoil factor 3 expressing vector for tandem affinity purification

BACKGROUND:As a novel growth factor, human intestinal trefoil factor (TFF3) can promote cel growth and migration, and increase cel resistance to apoptosis, and it plays a great role in maintaining the mucosa integrity, mucosa protection and repairing the injured mucosa, also it has been closely related to the tumor growth and progression. With the function of mucosa repair, and as the tumor biomarker, TFF3 has a promising clinical application, but its definite interacting protein and molecular mechanism is stil unclear. OBJECTIVE:To construct and express the TFF3 recombinant protein with the tandem tag of StrepII-6×His in the target cel s for further purifying its interaction protein in the native condition based on the tandem affinity purification technique. METHODS:The DNA sequence for the tag (StrepII-TEV-6×His) and TFF3 as template was got by chemical synthesis and PCR amplification respectively. They were fused by the restriction enzyme XbaI site, and the tag sequence was located at the C terminus of TFF3 protein. TFF3-tag fusion gene was cloned into the pCDNA3.0 using EcoRI+HindIII, thus the TFF3-tag expressing vector pTFF3-C-StH was constructed and transfected transiently into gastric cel AGS by lipofectin. The recombinant TFF3-tag protein was expressed and detected by western blot assay. RESULTS AND CONCLUSION:The expressing vector pTFF3-C-StH for tandem affinity purification was constructed successful y, and was confirmed further by restriction enzyme analysis and sequenced. The recombinant TFF3-C-StH protein of TFF3-tag was expressed in the AGS cel , and showed specific antigenicity by western blot assay. Thus this work provides experimental base for further purification of the TFF3 interacting proteins.
7.Precise Prediction of Deoxyribonucleic Acid sizes with Transient Isotachophoreis-Capillary Gel Electrophoresis Analysis on a Microchip

The migration-time in transient isotachophoresis (tITP) separation is affected by sample salinity as the dependence of ITP time on sample-zone conductivity.The sample-to-sample variation of migration-time in microchip tITP-CGE analysis is an undesired factor for precise DNA sizing.In this work, a DNA sizing method that based on relative migration-time proportion (RMP) was developed to eliminate the effect of sample salinity on sizing precision.RMP was defined as the ratio of the migration-time difference between the target fragment and the lower marker to that between the upper marker and the lower marker.The RMP values were tested to be reproducible in microchip tITP-CGE separations irrespective of sample salinity.Size of a target DNA was predicted by fitting its RMP value to the equation derived from RMPs of standard DNA ladder vs.DNA sizes.The precision and reproducibility of the sizing method were validated testing multiple standard PCR amplicons.Experimental results showed that the RMP method is simple and reliable, thus well suited to precise DNA sizing with microchip tITP-CGE analysis.
8.Expression of nucleolar spindle-associated protein 1 in hepatocellular carcinoma and its clinical significance

Objective To investigate the expression of nucleolar spindle-associated protein 1(NuSAP1) in hepatocellular carcinoma (HCC) and in non-cancerous tissues,and to study the correlation between NuSAP1 and early recurrence and prognosis of HCC.Methods The expression of NuSAP1 in 61 cases of HCC and non-cancerous tissues were assessed by RT-PCR,quantitative PCR and immunohistochemistry.The relationship between the expression and the clinicopathological features was studied.Results The levels of mRNA and protein in HCC were higher than the non-cancerous tissues (P<0.05).On univariate analysis,the expression levels of NuSAP1,mRNA and protein in HCC were significantly associated with TNM classification,lymphatic metastasis,early recurrence,tumor thrombosis and histological differentiation (P<0.05).Multivariate analysis showed early recurrence was associated with the expression of NuSAP1 protein in HCC (P<0.05).Overexpression of NuSAP1 protein was correlated with poor outcome of the patients with HCC (x2=15.846,P<0.001).Conclusions NuSAP1 was overexpressed in hepatocellular carcinoma.Overexpression of NuSAP1 was associated with early postoperative HCC recurrence and bad prognosis.
9.Intraoperative lymphatic mapping guided D2 lymphadenectomy in advanced gastric cancer

Objective To compare the number of lymph node dissected by intraoperative lymphatic mapping guided D2 gastrectomy and that by standard D2 gastrectomy plus lymphadenectomy in patients of advanced gastric cancer. Methods In this study 20 advanced gastric cancer cases received intraoperative peritumor injection of carbon nanoparticles suspension ( group 1 ) and D2 lymphadenectomy was guided by the black-stained lymph nodes. 21 cases undergoing standard D2 lymphadenectomy served as controls (group 2). The number of lymph nodes removed and the condition of lymphatic metastasis in two groups, blackstained lymph nodes in group 1, and postoperative complications were compared. Results The average lymph nodes dissected in group 1 (35. 1 ± 13.4) were higher than in control group (26.2 ±7.8). The differences were statistically significant (t =2. 126, P =0. 034). The number of removed N2 and N3 lymph nodes in group 1 were more than that in control group. The total black-stained ration of lymph nodes was 52. 7% in group 1. The positive rate of lymph nodes was higher in black-stained lymph nodes (27.6%) than in unstained lymph nodes ( 10. 8% ) in group 1 and in control group ( 16. 9% ). The differences were also statistically significant ( x2 = 6. 034, P = 0. 016; x2 = 5. 142, P = 0. 023 ). Postoperative afferent loop obstruction developed in one case in group 1. Conclusions Lymphatic mapping guided D2 radical gastrectomy plus lymphadenectomy increases the number of lymph nodes dessected and improves the efficiency of positive lymph nodes excision for patients of advanced gastric cancer.
10.Effect of Umbilical Cord Blood Stem Cell Transplantation on Axon Regeneration in Spinal Cord-injured Rats

Objective: To explore the effect of umbilical cord blood stem cell transplantation on axon regeneration in spinal cord injury (SCl)in rats. Methods: The umbilical cord blood was collected and prepared into suitable concentration of CD34 positive stem cells. Thirty SD rats were divided into two groups randomly. One group served as control, another one was the treatment group. The models of spinal cord contusion injury were made by Allen's weight dropping method. One week later,the treatment group was transplanted with 10x105 umbilical cord blood stem cells with Hamilton micro-syringe at the sites of rostral and caudal to the lesioned zone respectively, while control group received just the same volume of PBS injection. Five rats in each group were sacrificed at 1 w, 2 w and 6 w after this operation. Histological and immunohistochemieal examinations including GAP-43 and NF200 were used to evaluate axon regeneration. Meanwhile, BBB motion scoring and inclined plane test were performed to assess the motion function changes of hindlimbs. Results: Compared to the control group, the area of cavity in the lesioned spinal cord region decreased significantly and the expressions of GAP-43 and NF200 increased markedly in cell transplantation group. Also the motion function had better restoration in the treatment group. Conclusion: Transplantation of umbilical cord blood stem cell may achieve both morphological and behavioral improvement for the injured spinal cord.
