BACKGROUND:Researches have found that minocycline plays a neuroprotective effect by inhibiting the microgliacel proliferation and activation and suppressing glial cels to release cytokines and chemokines. OBJECTIVE:To investigate the influence of minocycline on glial cel line derived neurotrophic factor, NTN and gene expression in substantia nigra and corpus striatum in Parkinson’s disease model rats.
METHODS:144 rats were randomly divided into four groups, with 36 rats in each group. In the normal control group, no intervention was given. In the model and experimental groups, 6-hydroxydopamine was injectedin the right substantia nigra pars compacta and ventral tegmental area to establish Parkinson’s disease models. In the sham surgery group, vitamin C was injected in the two points. In the experimental group, after model establishment, rats were intragastricaly given 4.5 g/L minocycline 45 mg/kg. From then on, additional 22.5 mg/kg minocycline was added every 12 hours. The last group was normal control group. Immediately, 12 hours, 1, 7, 14 and 21 days after model induction, SP immunohistochemistry was used to detect the expression of glial cel line derived neurotrophic factor and NTN expression in the substantia nigra and corpus striatum. RT-PCR was used to identify glial cel line derived neurotrophic factor and NTN mRNA expression in the substantia nigra and corpus striatum.
RESULTS AND CONCLUSION:Both in the substantia nigra and corpus striatum, the positive cel number and relative gene expression of glial cel line derived neurotrophic factor and NTN were lower in the model group than in the normalcontrol and sham surgery groups (P< 0.05). Glial cel line derived neurotrophic factor-and NTN-positive cel number and relative expression were higher in the experimental group than in the model group (P< 0.05). These findings suggest that minocyclinecan delay the process of Parkinson’s disease pathogenesis by promoting glial cel line derived neurotrophic factor protein and gene expression.

Objective To investigate the effect of acidic serine protease ASPNJ on the expression of heat shock protein HSP90, 60 and 27 in human chronic myeloid leukemia K562 cells, in order to reveal the related mechanism of anti leukemic effects of ASPNJ. Methods K562 leukemia cell lines were cultured in vitro and treated with ASPNJ alone or in combination with chemotherapeutic agents. Western blot and RT-PCR were used to detect the changes of HSP90, 60 and 27 gene expressions in levels of total protein and membrane protein, as well as in mRNA levels. Results ASPNJ showed different effects on the expression of HSPs in total protein and membrane protein levels and had some modified effect on HSPs in total protein or membrane protein levels. Effects of ASPNJon expression of HSPs mRNA were not apparent, but HSPs mRNA were apparently lower in the ASPNJ and doxorubicin combination group than that in the ASPNJ alone or doxorubicin alone groups. Conclusion The mechanism of ASPNJ on the inhibitory effect of leukemia cells proliferation and the promoting effects on chemotherapeutic drugs may involve some complicated correlations with the effect of ASPNJ on the expression of HSPs and the modification of HSPs proteins.

Objective:
To investigate mental health and its associated factors in college students during COVID-19 confinement in campus, and to provide a scientific basis for mental health education.
Methods:
A general questionnaire, the Patient Health Questionnaire 9 (PHQ-9) and the Generalized Anxiety Disorder- 7 (GAD-7) were administered. A total of 1 816 college students under COVID-19 confinement in campus in Lanzhou City were surveyed from October 18 to November 18, 2021. Data were analyzed by using the ordinal Logistic regression method.
Results:
The overall incidence of depressive emotions was 38.76%, and the incidences of mild, moderate to severe depression emotions were 31.33% and 7.43%, respectively. About 16.36% of students showed anxiety, with mild, moderate to severe anxiety being 13.33% and 3.03%, respectively. Multivariate analysis showed that poverty( OR =1.29), daily schedule (basically normal OR =0.33, normal OR =0.18), adaptability of online learning (moderate adaptation OR =0.45, high level of adaptation OR =0.25), concerns about the infection of oneself and family members (some concerns OR =1.73, considerable concerns OR =2.09),male( OR =0.78), and the isolation mode( OR =1.70). The music listening (sometimes OR =0.44, often OR =0.41), daily schedule (basically normal OR =0.36, normal OR =0.19), adaptability of online learning (moderate adaptation OR =0.42, high level of adaptation OR =0.28), and concerns about the infection of oneself and family members (some concerns OR =1.87, considerable concerns OR =3.27) were primary factors associated with high level of anxiety among college students( P <0.05).
Conclusion
The incidence of depression and anxiety among college students increased following COVID-19 confinement and centralized isolation for medical observation in campus. Universities and relevant departments should take timely and precise measures for psychological counseling.

Chinese Journal of Radiation Oncology 2023;32(6):551-556
Objective:To investigate the effect of heat shock protein 90 (Hsp90) inhibitor PU-H71 combined with X-ray on radioresistant human cervical cancer cells. Methods:The expression levels of Hsp90 gene between cervical cancer tissues and adjacent tissues were analyzed by bioinformatics. Radioresistant cervical cancer cell lines HeLa RR and SiHa RR were obtained by fractional irradiations (2 Gy per fraction, 30 fractions). The cell lines were divided into the control group (treated with dimethyl sulfoxide), irradiation alone group, PU-H71 group (treated with 0.5 μmol/L PU-H71), and PU-H71+irradiation group (irradiation at 24 h after treatment with 0.5 μmol/L PU-H71). Cell survival was detected by clonal formation assay. Immunofluorescence assay was used to detect γH2AX foci at 1, 6, and 24 h after cell treatment. The expression level of Rad51 protein at 1, 2, 6, 12, and 24 h after cell treatment was detected using Western blot. The expression level of phosphorylated DNA-dependent protein kinase catalytic subunit (p-DNA-PKcs) was measured at 2 h after cell treatment. Cell apoptosis at 48 h after cell treatment was assessed by flow cytometry. Results:PU-H71 enhanced the sensitivity of radioresistant cervical cancer cells to X-ray. Compared with the irradiation alone group, the radiation sensitization ratios (SER) of HeLa RR and SiHa RR cells at 10% survival were 1.36 and 1.27, and the apoptosis rates were increased by approximately 72.1% and 63.1% in the PU-H71+irradiation group, respectively. PU-H71 delayed the duration of γH2AX foci induced by X-ray, inhibited the phosphorylation of DNA-dependent protein kinase catalytic subunit (DNA-PKcs), thus preventing non-homologous end joining (NHEJ) repair and delaying homologous recombination repair.Conclusion:PU-H71 increases the radiosensitivity of radioresistant cervical cancer cells by inhibiting the repair pathway of DNA double-strand break, which is expected to be a radiosensitizer to enhance the efficacy of radiotherapy for cervical cancer.
