1.Comparative Proteomics Analysis on Splenic Lymphocytes in Migraine Rat Model with Hyperactivity of Liver-yang

Objective To explore the molecule mechanisms of migraine with hyperactivity of liver-yang related proteins. Methods Animal model of migraine with hyperactivity of liver-yang was established through electrical trigeminal ganglion stimulation,and by administering orally with Fuzi decoction. The total proteins of splenic lymphocytes in the rats were separated by immobilized pH gradient-based two-dimensional gel electrophoresis. With coomassie blue staining,the 2-DE images were analyzed by PDQuest 7.0 software. Matrix-Assisted Laser Desorption/Ionization-Time of Flight Mass Spectrometry (MALDI-TOF-MS) and database were used to identify the differential proteins. Result Ten differential spots protein were identified,and they were chloride intracellular channel 1,thioredoxin peroxidase Ⅰ,thioredoxin peroxidase Ⅱ,sequence 7 from patent WO9709348,Rho GDP dissociation inhibitor (GDI) alpha,actin related protein 2/3 complex neurofilament light polypeptide,proteasome subunit beta type and heat shock protein-27,annexin A1. Conclusion These protein may be involved in the development of migraine with hyperactivity of liver-yang. These data provide useful clues for elucidating the mechanisms of migraine with hyperactivity of liver-yang.
2.Clinical Observation of Xiaocuo Prescription for Acne Vulgaris:A Report of 325 Cases

[Objective] To investigate the effect of Xiaocuo Prescription (XP), a herbal medicine compound with the actions of clearing away heat and toxins, activating blood and removing blood-stasis, on acne vulgaris. [Methods] Four hundred and forty six cases of acne vulgaris were randomized to two groups: group A (325 cases) treated with oral use of XP (mainly composed of Flos Lonicerae, Herba Hedyotis Diffusae, Herba Taraxaci, Flos Chrysanthemi, Radix Scutellariae, Cortex Mori, Radix Salviae Miltiorrhizae, etc.) and external application of Acne Vulgaris Liquid (composed of 3% metronidazole and 2% erythromycin), group B (121 cases) treated with oral use of isotretinoin and external application of Acne Vulgaris Liquid. Skin lesions on the face as well as the side effects were observed before and after treatment. [Results] Ridit analysis showed that curative effect in group A was better that in group B ( u = 2.204 6, P .05). No obvious side effects were found in group A but in 5 cases of group B side effects such as skin dryness, nausea and vertigo were found. [ Conclusion ] XP is effective in treating acne vulgaris . Further research is needed for its therapeutic mechanism.
3.Effects of Qilantangzhining Capsule on Hepatocyte Apoptosis and Expression of Bax and Bcl-2 in Rats with Diabetes Mellitus and Hyperlipemia

Objective: To study the mechanism of Qilantangzhining Capsule in treating diabetes mellitus complicated with hyperlipemia. Method: Rats were fed with high sugar and fat food for 2 months, and then 2% streptozocin (STZ, 30mg/kg) was injected intraperitoneally to induce hyperglycemic and hyperlipemic model. Thereafter, the model rats were administrated the drug. Flow cytometry (FCM) were used to detect effects of the capsule on hepatocyte apoptosis and the expression of related regulating genes, Bax and Bcl-2. Result: The rate of cell apoptosis remarkably increased among the model rats, the expression of Bax was significantly enhanced and that of Bcl-2 was decreased. Qilantangzhining Capsule could remarkably decrease the rate of cell apoptosis, decrease the expression of Bax, and increase that of Bcl-2. Conclusion: Qilantangzhining Capsule can prevent hepatocyte apoptosis by adjust the expression of Bax and Bcl-2, and accordingly provide evidences for its role in adjusting the metabolic disorders of sugar and fat.
4.Long-term result and prognostic analysis of 18FDG PET/CT positioning three-dimensional conformal radiotherapy for stage Ⅲ non-small cell lung cancer

Objective To analyse the long-term result and prognosis of 18 FDG PET/CT positioning three - dimensional conformal radiotherapy ( 3 DCRT ) for stage Ⅲ non - small cell lung cancer. Methods Sixty-four cases with stage Ⅲ non-small cell lung cancer (clinical stage Ⅲa- Ⅲb ) were randomly divided into two groups: PET/CT positioning three-dimensional conformal radiotherapy group (PET/CT group) and the conventional CT positioning three-dimensional conformal radiotherapy group (conventional CT group). In the PET/CT group, the target volume and critical organs were sketched according to PET/CT after fusion of the PET and the CT images; the treatment plan was worked out, then conventional fractionated 3DCRT ( total dosage around 40 Gy) followed by field-shrinked radiotherapy to a total dose of 65 Gy or sowas performed ;in the conventional CT group, the target volume and critical organs were sketched according to CT and 3DCRT were performed to the same total dose; All cases were treated with the TP scheme (paclitaxel 175 mg/m2,d1 ,cisplatin 40 mg,d2-4) adjuvant chemotherapy for 6 cycles after the radiotherapy. Results The followup rate was 100%. The number of patients who completed the 1-,2-and 5-year follow-up were 40,20 and 11 respectively ;The number of patients of the PET/CT group and conventional CT group were 23 and 17,11 and 9,7 and 4 respectively. Target volumes of 13 cases in the PET/CT group were changed. The complete remission and partial remission rates of the two groups were 13% 、66% and 19% 、53% (x2 = 0. 33, P =0. 564), respectively. The 1-,2-and 3-year local control rates of the PET/CT group and conventional CT group were84 % 、66% 、53 % an d72% 、59% 、44% ( x2 = 2.36, P = 0. 124 ) respectively. The1 -, 2-and 3-year survival rates were 72% 、34% 、22% and 53% 、28% 、13% (x2 =2. 46,P =0. 117) respectively. The level-1 and level-2 lungs' and trachea's late radiation injury of the PET/CT group and the conventional CT group were 28% and 53% ( x2 = 4. 14, P = 0. 042 ), respectively. The hilar and mediastinal lymph node recurrence rates of the PET/CT group were lower than those of the conventional CT group, were 3% ,25%(P = 0. 026) and 6%, 28% ( P = 0. 042 ), respectively. The main reason for treatment failure was distant metastasis both in the PET/CT group and conventional CT group,56% and 47% (x2 = 0. 56,P = 0. 453 ),respectively. Conclusions PET/CT, as a method of sketching the target of stage Ⅲ non-small cell lung cancer, can improve the radiation treatment plan, reduce the recurrence rate of hilar and mediastinal lymph nodes, meanwhile it can not improve the long-term survival rate; Distant metastasis was the main reason of failure.
5.Effects of ping-gan qianyang therapy on adrenal gland proteome in migraine rat model with hyperactivity of Hver-yang

Objective To explore the molecule mechanisms of ping-gan qianyang therapy on migraine rat model with the hyperactivity of liver-yang.Methods An animal model of migraine with hyperactivity of liver-yang was established through electrical trigeminal ganglion stimulation and administering orally with Fuzi decoction.The total proteins of adrenal#and in the rats were separated by immobilized PH gradient-based two-dimensional gel electrophoresis, and the 2-DE images Was analyzed by PDQuest 7.0 software.Matrix-Assisted Laser Desorption/Ionization-Time Of Flight Mass Spectrometry(MALDI-TOF-MS)and SWISS-PORT and MSDB database were used to identify differential proteins.Result The well-resolution and reproducible 2-DE patterns of rat adrenal glands in normal group,model group and therapy group were obtained.A total of 8 differentially expressed proteins were identified.Conclusion Ping-gan qianyang therapy may effect on migraine by regulating the expression of proteins related to energy metabolism and ubiquitin.
6.The Effects of Hairpin RNA on the Expression of Human Papillomavirus E6 Gene

Objective To investigate the effects of hairpin RNA on the expression of human papillomavirus E6 gene. Methods Plasmids expressing hairpin RNA, sense RNA and anti-sense RNA were constructed. The recombinants were transfected into cervical cancer cell line, CaSKi. Expression of E6 was detected with semi-quantitative RT-PCR and Western blot. Results Hairpin RNA could significantly reduce the expressions of E6 mRNA and protein, which was more efficient than traditional anti-sense RNA did. Conclusion The RNA interference through hairpin RNA can effectively inhibit the expression of target gene, and can be potentially useful in gene therapy of HPV related tumors.

8.Case report of cervical intraspinal primitive neuroectodermal tumor and literature review

Objective To investigate the clinical characters and therapy of primitive neuroectodermal tumor(PNET).Methods A retrospective analysis was conducted.A 36-year-old female patient was showed pain and numbness of the right upper limb and back for 6 months.The cervical spine MRI showed a spindleshaped intradural mass right ventrolateral of spinal cord at C5-7 with in homogeneously enhancing.Surgery and pathologic examination confirmed that was PNET.Combiled with a series of literatures to analyse the clinical characters Results Surgery was performed to remove the tumor and decompression combined with radiotherapy.The pathologic examination and immunohistochemical analysis revealed that it was PNET.MRI identified local recurrence in spinal canal at 3 month later after surgery.Conclusion Spinal PNET is an uncommon intraspinal tumor with poor prognosis.Histopathology is the evidence of diagnosis.Optimal therapy has not yet been found.Surgical resection with the combination of chemo-radiotherapy or radiotherapy might get the better outcomes.Multidisciplinary treatment should be further clinical required.
9.Effect of Quetiapine on Brain-derived Neurotrophic Factors in Patients with First-episode Schizophrenia

Objective To investigate the effects of quetiapine on serum levels of brain-derived neurotrophic factors ( BDNF) and the correlation between BDNF and psychiatric symptoms and cognitive function in patients with first-episode schizophrenia. Methods Eighty patients with first-episode schizophrenia ( treatment group) were treated with quetiapine orally for 4 weeks,at initial dose of 100 mg·d-1 and average dose of (580±120) mg·d-1 . The psychiatric symptoms were evaluated by using the positive and negative syndrome scale ( PANSS) . The cognitive function was assessed by using Wisconsin cards sort test ( WCST) . The serum BDNF level was detected with enzyme-linked immunosorbent assay ( ELISA) . Results The serum level of BDNF was markedly lower in schizophrenic patients before[(13. 72±8. 79) ng·mL-1,P<0. 01] and after treatment[(18. 02±9.06) ng·mL-1,P<0.05]in comparison with normal controls(23. 67±10. 13) ng·mL-1]. After treatment,the PANSS total scores and subscale scores decreased,WCST number of categories and the number of correct answers increased,and the number of wrong answers reduced. There was a positive correlation between the serum BDNF and negative symptoms ( SANS) ( r= 0. 54, P=0. 032),and the number of correct answers. Conclusion The quetiapine significantly increases serum level of BDNF in schizophrenia patients,which correlates positively with improvements in symptoms and cognitive function.
10.Application of microvascular device in free-tissue flap reconstruction of head-and-neck defects

Objective To evaluate the application of microvascular coupler device (MVCD) in the free-tissue flap transfer for head-and-neck defects reconstruction.Methods Its a retrospective study of the clinical data of 743 patients who received 763 free-tissue flap transfer for head and neck defects reconstruction in Hunan Cancer Hospital from January,2014 to January,2016.For microvascular anastomoses,413 were done manually (the manual group) and 350 done with MVCD (the MVCD group),of which the latter included end-to-end venous anastomosis in 159,endto-side venous anastomosis in 190 and end-to-end arterial anastomosis in 1.The time for anastomosis,rate of blood leakage from the anastomosis and venous thrombosis,and the survival rate of flaps were compared between the manual group and the MVCD group.The impacts of types of anastomosis of end-to-end vs end-to-side (both with MVCD),on the formation of venous thrombosis and survival of flaps were also recorded.The analysis was performed under t-test and chi-square test using SPSS software 19.0 with P < 0.05 for differences with a statistical significance.Results Time for anastomosis was significantly shorter in the MVCD group (4.43±0.51min) than in the manual group (14.75± 2.43min,P<0.05).The rates of anastomosis leakage were 0 in the MVCD group and 9.7% in the manual group(P<0.01),flap necrosis were 0.57% and 1.69% (P=0.15),the rate of venous thrombosis were 1.15% and 3.63%(P=0.02),respectively.There was no significant difference in the time for anastomosis,the rate of venous thrombosis and the survival rate of flaps with end-to-end anastomosis and end-to-side anastomosis with MVCD.Conclusion The application of microvascular coupler device is valuable in the reconstruction of head-and-neck defects with free-tissue flaps because it can significantly shorten the time for anastomosis,decrease the ocurrence of anastomosis leakage and venous thrombosis,reduce the risk of flap failure.
