1.Relationship between Serum Von Willebrand Factor and Diabetic Nephropathy in Type 2 Diabetic Patients

Objective To investigate the change of serum von willebrand factor(vWF)level in the different stages of diabetic nephropathy and its significance. Methods 50 healthy subjects and 150 patients with type 2 diabetes mellitus were enrolled in this study. Diabetic patients were further divided into three subgroups according to their urinary albumin excretion rate(UAER): 56 patients with normal UAER,49 patients with microalbuminuria and 45 patients with clinical proteinuria. Serum vWF concentration was measured with emzyme linked immunosorbent assay (ELISA) method. Results vWF concentration was significantly higher in patients with type 2 diabetes mellitus than that in normal controls. Serum vWF concentration increased with the elevation of UAER. Serum vWF level was positively related with UAER, blood creatine and course of disease independently. Conclusion Serum vWF concentration increased in the patients with type 2 diabetes mellitus and the increased extent of vWF was consistent with the severe degree of diabetic nephropathy.
2.Application and effect of platelet-rich plasma and its relevant factors for tissue repair and regeneration

Platelet-rich plasma (PRP) is a concentrated source of autologous platelets after centrifugation, being used widely in plastic surgery and oral-maxillofacial surgery. Recent studies have showed that PRP can improve bone repair, vasculogenesis, and soft tissue repair. Composite of PRP and stem cells is applied in the field of orthopedics and tissue engineering. As a matter of fact, PRP has become a hot research topic. In addition, PRP plays an important role in the healing of chronic wound. This paper briefly introduces the main components of PRP and its clinical application, the mechanism for bone healing and soft tissue repair, as well as the latest research achievement and discussion in the shortcoming of PRP in clinical application.
3.MRI Manifestations of MELAS Syndrome

Objective To investigate MR imaging characteristics of MELAS syndrome.Methods The clinical data and MRI features of 13 cases with MELAS syndrome were retrospectively analyzed.Results Multiple hypointense on T1WI and hyperintense on T2WI lesions were showed predominantly in the gray matter of the temporal, parietal and occipital lobes. The lesions crossed vascular boundaries. No stenosis and occlusion of main artery were displayed by MRA.Basal ganglia calcifications and cerebral atrophy with widening of ventricles were also frequent found. Increased ADC value of the leison on DWI and appearance of Lac peak on1H-MRS were useful in diagnosis.Conclusion There are some characteristics of MRI in patients with MELAS syndrome.Combined clinical informations with neuroimaging and muscle biopsy, the diagnosis of MELAS syndrome can be made correctly.
4.Action of Staphylococcal Exfoliative Toxin A on Pemphigus Antigens Expressed on Human Keratinocytes

Objective To study the action and mechanism of staphylococcal exfoliative toxin A(E-TA)on pemphigus foliaceus antigen(PFA)and pemphigus vulgaris antigen(PVA)expressed on cultured human keratinocytes.Methods Stratified human keratinocytes were incubated with ETA and then stained with sera from patients with pemphigus foliaceus or pemphigus vulgaris as the first antibodies and FITC-la-beled sheep anti-human IgG as the second antibody.Total protein was harvested from the cells pretreated with ETA and run on SDS-PAGE for Western blot with the same antibodies.Simultaneously,supernatants of the keratinocytes before and after ETA treatment were collected for detection of the levels of IL-1?,IL-6with ELISA kits.The caseinolytic activities of the supernatants were tested by spectrometry in which casein was used as a non-specific substrate.Results Down-expression of PFA was shown after ETA treatment while no change of PVA expression was found.The high intensity and continuous linear appearance of fluo-rescent staining before ETA treatment became weak and discontinuous after ETA treatment,which were re-covered gradually in24hours.The degradation of proteins recognized by PF sera after ETV treatment was revealed by Western blot.The decreasing tendency of IL-1?concentration was found in the supernatants of cell culture after ETA treatment,but IL-6level was too low to be detected.Increased caseinolytic activities were found in the supernatants,and declined36hours after ETA treatment.Conclusions ETA acts on PFA expressed on keratinocytes in vitro,which is reversible along with withdrawal of ETA.The mechanism of E-TA act on PFA may be related to proteolytic action instead of promoting cytokine secretion.
5.Effect of GRP94 silencing on proliferation and apoptosis of human breast carcinoma MCF7 cells

Objective:To determine the effects of silencing glucose regulated protein ( GRP94 ) on the proliferation and apoptosis of breast carcinoma MCF7 cells. Methods:Chemically synthesized siRNA targeting GRP94 gene and transfected into MCF7 cells used by Liopfectamine RNAIMAX. The mRNA and protein expression levels of GRP94,cyclinD1,Bax and Bcl-2 were detected by Real-time PCR and Western blot. CCK8 assay was used to detect the effect of specific GRP94 siRNA on cell proliferation and the effect on cell cycle and apoptosis were analyzed by flow cytometry and Hoechst 33258 staining. Results:Compared with the siRNA-NC cells, the expression of GRP94 was significantly down-regulated in MCF7 cells. Knockdown of GRP94 in MCF7 cells decreased cell proliferation and promoted cell apoptosis. The expression of cyclinD1and Bcl-2levels were significantly reduced, and Bax level was increased in siRNA-GRP94 MCF7 cells. Conclusion: The siRNA-mediated GRP94 silence significantly inhibits MCF7 cell proliferation,promoted cell apoptosis by down-regulating cyclin D1,Bcl-2 expression and up-regulating the Bax expression in MCF7 cells.
6.Clinical study and analysis of pathogenic bacteria of pulmonary infection in patients with terminal cancer

Objective To analyze of pathogenic bacteria of pulmonary infection in patients with terminal cancer.Methods The clinical data were retrospectively analyzed including the general cultivation of pathogenic bac-teria in sputum and the results of drug sensitivity of hospitalization of patients with lung cancer from July 2013 to May 2014.Results 132 strains of pathogenic bacteria were cultured from120 cases of patients with pulmonary infection, including gram-negative bacilli accounted for the vast majority of 27.3%,accounted for 59.8% of Gram-positive coc-ci,Ranked in the top 5 pathogenic bacteria were Streptococcus pneumoniae,Staphylococcus aureus,Candida albicans, Klebsiella pneumoniae,there arevarying degrees of resistance to commonly used antibiotics in bacteria.Conclusion Selecting and using the antimicrobial agents according to the results of pathogen detection in patients,could improve the effectiveness of antibiotic treatment and reduce bacterial resistance.
7.Unilateral olfactory functional deprivation model in the left peripheral nostrils by electric cautery injury:olfactory bulb neurogenesis and transformation

BACKGROUND:Olfactory bulb neurogenesis, transformation and maturation have been considered the hot topic. Olfactory bulb experience and nervous activity can influence olfactory bulb neurogenesis. However, no study reports that olfactory bulb functions can affect olfactory bulb neurogenesis in guinea pigs.
OBJECTIVE:To investigate the effect of unilateral olfactory functional deprivation on doublecortin, calbindin and parvalbumin expression in olfactory bulb of juvenile guinea pigs.
METHODS:Total y 24 guinea pigs were randomly divided into two groups, which were kil ed after establishing olfactory functional deprivation model through electric cautery injury at the left peripheral nostrils. At 3 and 6 weeks after modeling, the specimens were harvested. The expression change of doublecortin, calbindin and parvalbumin in two sides’ olfactory bulb of juvenile guinea pigs was detected by immunohistochemistry.
RESULTS AND CONCLUSION:The number of doublecortin, calbindin and parvalbumin positive cells in olfactory bulb at the un-deprived side was significantly higher than that at the deprived side at 3 and 6 weeks (P<0.05). This finding indicates that olfactory neural activities can affect neurogenesis and transformation in guinea pigs.
8.Primary lymphoma of thyroid gland

OBJECTIVE To analyze the reasons of misdiagnosis and surgical complications of primary lymphoma of thyroid gland(PLT). METHODS The clinical data of 10 patients with PLT in our hospital from 1992 to 2006 were retrospectively analyzed. RESULTS All of 10 patients were misdiagnosed,the major surgical complication was hoarseness. CONCLUSION The misdiagnosis of PLT is often encountered. Mastering the clinical features,using fine-needle biopsy,rapid biopsy combining immunocytochemical method can diagnose the primary lymphoma of thyroid gland.
9.Karyotype Analysis/Genetic Testing in Children Suspected with Hereditary Disease

Objective To apply karyotype analysis/genetic testing in children suspected with hereditary disease. Methods From July, 2014 to July, 2016, a total of 47 cases in our department were tested using G-banding karyotype analysis or selected the relevant genetic package, for screening the related diseases. Results 38 cases received karyotype analysis, in which three cases were abnormal, and one case was diagnosed definitely. And nine cases received related genetic testing, in which seven cases were abnormal, and four cases were diag-nosed definitely. Totally, the positive rate was 21.28%, and the diagnosis rate was 10.64%. Conclusion Karyotype analysis/genetic testing is an etiological diagnosis method for highly suspected hereditary disease in children.
10.Effect of Acupuncture on Motor Function of Children with Spastic Cerebral Palsy

Objective To observe the effect of acupuncture on motor function of children with spastic cerebral palsy. Methods 66 children with spastic cerebral palsy were randomly divided into control group (n=33) and observation group (n=33). They were evaluated with Gross Motor Function Measure (GMFM) before and after treatment. Results The scores of GMFM improved significantly in both groups after treatment (P<0.001), and improved more in the observation group than in the control group (P<0.05). Conclusion Acupuncture combined with rehabilitation training can further improve the motor function of children with spastic cerebral palsy.
