1.Determination of Gentiopicrin in Qianggan Capsules by HPLC

OBJCTIVE:To determine the content of Gentiopicrin in Qianggan capsules by HPLC.METHODS:The sample was separated on Diamonsil TM C18 (200 mm?4.6 mm,5 ?m) with mobile phase consisted of methanol-water (23∶77) at a flow rate of 1.0 mL?min-1.The detection wavelength was set at 254 nm.RESULTS: The linear range of Gentiopicrin was 0.018 59~0.232 40 ?g(r=0.999 6) with an average recovery of 96.59%(RSD=1.35%,n=6).CONCLUSION: The meth-od developed is accurate,stable and reproducible,and it provides basis for the improvement of the quality standard of Qianggan capsules.
2.Effects of RNA interference on PIDD expression and drug resistance of HT-29 cells

Objective To explore the distributive changes of PIDD protein in HT29 cells stimulated by 5-fluorouracil(5-FU),and the influence of the PIDD protein interfered by small interference RNA(siRNA) on the drug resistance of HT29 cells in vitro.Methods siRNA was used to interfere the PIDD expression,and HT-29 cells were treated with 5-FU.Western blotting was employed to detect the PIDD expression before and after interference.The distribution of PIDD in nucleus and cytoplasm after 5-FU treatment was also detected.Changes in drug sensitivity of HT-29 cells to 5-FU were determined with MTT assay and IC50 values were evaluated.Results PIDD protein was expressed mainly in the cytoplasm before 5-FU stimulation,and it migraded into the nucleus after stimulation.Western blotting showed that the total PIDD expression and the expression both in cytoplasm and nuclei decreased significantly after RNA interference,and no increase of the total PIDD and that migrated into nucleus was found even after 5-FU stimulation.It was found that in the transfection group(cells were incubated 12h after being transfected with PIDD-360),normal group(cells without treatment) and control group(cells were incubated 12h after being transfected with negative transfection reagent),the IC50 values of cells treated with 5-FU were 0.23?0.06?g/ml,2.71?0.70?g/ml and 2.78?1.03?g/ml,respectively.The IC50 value declined significantly in transfection group compared with that of both the normal group and control group(P
3.Comparison of immediate PTCA and elective PTCA on infarct related artery after spontaneous recanalization

Objective To compare the clinical outcomes of immediate PTCA and elective PTCA on infarct related artery after spontaneous recanalization. Methods One hundred and eighteen patients with SR between January 1996 and March 2001 were enrolled. According to TIMI flow, the patients were divided into 2 study populations. The two study populations were divided into 2 groups according to immediate PTCA or elective PTCA, respectively. Results Mortality and reinfarction were similar between the 2 groups two weeks after heart attack. However, recurrent ischemic events were more frequent in elective PTCA group than in immediate PTCA group according to TIMI flow grade 2 (1/24 vs 4/12,P=0.034). Objective Low-dose naloxone has been shown to reduce side-effects of morphine while morphine analgesia is not antagonized and may even be enhanced. The purpose of this study was to evaluate the effects of low-dose naloxone infusion (50 ng? kg-1? h-1 ) on morphine analgesia and plasma levels of opioid peptides in patients after abdominal hysterectomy.Methods Forty-two ASA Ⅰ- Ⅱ patients aged 36-50 yrs, weighing 55-67 kg undergoing abdominal hysterectomy under combined spinal-epidural anesthesia were enrolled in this study. Spinal puncture was performed at L2-3 interspace. The patients received intrathecal 0.75% ropivacaine 2.0-2.6 ml. 2% lidocaine was used for epidural injection. The block height was maintained at T8-6 . For postoperative analgesia the patients were randomized to receive either intravenous morphine infusion at 10 ?g?kg-1 ?h-1 (group M, n = 21) or IV morphine infusion (10 ?g?kg-1?h-1)+ naloxone infusion at 50 ng?kg-1?h-1 (group MN, n = 21). Pain was assessed using VAS (0-10) with 0 representing no pain and 10 representing the worst possible pain. Blood samples were taken from peripheral vein before anesthesia (T0), at the end of surgery (T1) and at 6, 24, 48 h (T2,3,4) after operation for determination of plasma levels of ?-endorphin (?-EP), dynorphin A1-13 (Dyn) and leu-enkepholin (L-EK) .Results The patients were comparable with respect to, age, weight, occupation and duration of operation between the two groups. Two patients (1 patient in each group) were excluded from the study because of their refusal to have repeated blood samples taken. The analgesia was significantly better in group MN than that in group M in terms of VAS scores. Objective To evaluate the effect and safety of use of tirofiban in percutaneous coronary interventional therapy (PCI) in elder patients with acute ST segment elevation myocardial infarction (STEMI). Methods Sixty-five patients with acute STEMI were divided into two groups: 31 patients in the elderly group (≥70 years) and 34 patients in the younger group (0.05). Conclusion It is safe and effective to administer tirofiban to elderly patients with acute STEMI undergoing PCI.

Objective: To evaluate the efficiency and safety for single-incision laparoscopic appendectomy (SILA) and conventional laparoscopic appendectomy (CLA). Methods: The literature of randomized controlled trials (RCTs) concerning SILA versus CLA was retrieved by searching the electronic databases from the inception date to May 2014. Methodological quality of the included trials was assessed by using the Cochrane Reviewers’ Handbook criteria, and the data were extracted and subjected to Meta-analysis by using RevMan 5.1 analysis sotfware. Results: A total of 1183 patients were ifnally selected atfer the screening. Among them, 582 cases underwent SILA and 601 cases underwent CLA. Meta-analysis results demonstrated that SILA was associated with higher conversion rate (RR=4.38, 95% CI 1.96–9.79,Z=3.59,P=0.0003), longer operative time (RR=4.83, 95% CI 1.57–8.09,Z=2.90,P=0.004), shorter length of hospital stay (WMD=?0.11, 95%CI ?0.21–?0.01,Z=2.02,P=0.04), and better scores for the postoperative appearance of incision (WMD=0.94, 95%CI 0.49–1.40,Z=4.06,P<0.001) compared with CLA; no signiifcant difference was observed in postoperative complications and postoperative pain scores (RR=1.03, 95% CI 0.74–1.45,Z=0.18,P=0.86; WMD=?0.19, 95%CI ?0.59–0.20,Z=0.95,P=0.34) between the 2 groups. Conclusion: For patients under the conditions of the strict selection, SILA is a safe and effective procedure for the treatment of adult acute appendicitis, especially apply to those who care about incision appearance.
9.Improvement of immunoprecipitation/immunoblotting of endogenous protein by TrueBlot antibody

Objective:To improve the immunoblotting of immunoprecipitated proteins and decrease the interference of immunoprecipitation antibody in the interaction of endogenous proteins. Methods:Transient transfect cells with fusion protein expression vector containing the targeted S5b gene and the FLAG tag, the transfected cells or untransfected cells were harvested to study the exogenous or endogenous protein interaction. The total cell lysate was immunoprecipitated by specific antibody. Then the eluted immunocomplex was separated by SDS-PAGE, and the TrueBlot antibody or conventional antibody was used as the secondary antibody for immunoblotting detection of S5b and its partner (Rpt1 and Rpt2). Results:Clear immunoblotting bands for S5b, Rpt1 and Rpt2 were obtained. Conclusion:TrueBlot antibody prefers the immunoblot antibody to immunoprecipitation antibody, and decreases the interruption of immunoprecipitation antibody to display clear protein band.
10.Observation on Preventive Effect of Ca/Mg Infusion Combined with Glutathione and Carbamazepine on Oxaliplatin-induced Neurotoxicity

OBJECTIVE:To observe the preventive effect of Ca/Mg infusion combined with glutathione and carbamazepine on oxaliplatin-induced neurotoxicity.METHODS:65patients treated with oxaliplatin,5-fluorouracil and leucovorin for gas-trointestinal cancer were divided into treatment group undergoing treatment with Ca/Mg infusion combined with glutathione and carbamazepine,and control group without any preventive treatment.RESULTS:At the third cycle,7patients in the treat-ment group but17patients in the control group showed clinically evident neurotoxicity,which demonstrated the statistical difference between the two groups(P=0.036).After12cycles,neurotoxicity of grade2to3was observed in2patients in the treatment group but in9patients in the control group,which demonstrated the statistical difference between the two groups(P=0.022).CONCLUSION:Ca/Mg infusions combined with glutathione and carbamazepine can prevent oxaliplatin-induced neurotoxicity.
