1.Combinational effects of bcl-2 antisense oligodexynucleotide with arsenic trioxide on malignant lymphoma cell lines

Purpose:To investigate the effects of bcl 2 antisense phosphorothioate oligodeoxynucleotides (ASODN) combined with arsenic trioxide on the malignant lymphoma cell lines.Methods:Apoptosis, cellular DNA contents and bcl 2 expression were determined by cell stain, TdT mediated dUTP Nick end Labeling (TUNEL) and flow cytometry. Results:10~40?mol/L bcl 2 ASODN and 0.5~2.0 ?mol/L As 2 O 3 could inhibit Raji cell growth and induce apoptosis significantly. Number of Sub G 1 cells significantly increased and Bcl 2 protein expression in Raji cells significantly was downregulated by flow cytometry assay. The relationship between level of bcl 2 protein and concentrations of As 2 O 3 and bcl 2 ASODN showed a dose and time dependent effect; so did the number of Sub G 1 cells. There is much more effective inhibition of growth and induction of apoptosis by their combination than alone. Conclusions:It is found that bcl 2 ASODN could significantly inhibit the growth of B lymphoma cell lines and induce their apoptosis when combined with arsenic trioxide.
2.Effects of pulmonary resection on perioperative right ventricular function

Objective To investigate the changes of perioperative right ventricular function after pulmonary resction. Methods 45 cases of pulmonary resection were divided into four groups.Group Ⅰwas wedge resection(n = 10), GroupⅡ was lobectomy(n = 19), Group Ⅲ was double lobectomy(n=7)and Group Ⅳ was pneumonectomy(n=9). The changes of CVP, PAMP, PEP/ET, Sa, VTIs, RVSP, Ea/Aa, and Tel were evaluated by traditional ultrasonic and tissue Doppler ultrasonic techniques at different time inter- vals (pre-operation, 5-7 days orland 1 month post-operation), Results Compared with the pre-operation CVP, the indexes of all groups have no significant changes post-operatively. Wedge resection didn' t obviously influence right cardiac after load and systolic function/diastolic function. No evidet changes detected in Tei pre-and post-operatively. However, the indexes (PAMP, RVSP and PEP/ET ratio) reflecting right cardiac afterload evidently increased at post-pneumonectomy or lobectomy group5- 7 days post-opera- tive. All these reflect the decrease of Tei, which was more obvious in pneumonectomy than in lobectomy group. Right cardiac after- load, systolic/diastolic function and Tel index recovered to pre-operative level 1 month post-operation in single lobectomy group. But the above indicators, especially the Tel, were still high in double lobectomy and pmeunonectomy groups 1 morth post-operation.Tei index is positively correlated with PAMP and is weakly correlated with PET/ET ratio and Ea/Aa ratio. Conclusion Pulmonary wedge resection doesn't evidently influence right cardiac function. However, right cardiac diastolic function evidently decreases temporarily at lobectomy group. The systolic function and diastolic funetions decrease after double-lobectomy and pneumonectomy and it's more evident in pneumonectomy group. Though the right cardiac afterloads of lobectomy, double lobectomy and pneumonectomy groupa all increase significantly post-operativlye, only the former recover to pre-operative level 1 month after surgery.
3.Effects of Swallowing Training Combined with Real-time Electrical Stimulation on Dysphagia after Stroke in Patients Aged over 80

ObjectiveTo investigate the effects of swallowing training combined with real-time electrical stimulation on dysphagia after stroke. Methods17 patients, older than 80 years old, with dysphagia after stroke, were treated with Vocastim-Master Physiomed Elektromedizin. They were assessed with the Watian drinking water test, swallowing disorder evaluation, swallowing ability evaluation, and α. ResultsAll the assessment significantly improved in the 17 patients (P<0.001). ConclusionSwallowing training combined with real-time electrical stimulation can significantly improve the swallowing capacity of the patients older than 80 after stroke.
4.Responsiveness of 3 Scale for Swallowing Disorder on Elderly Dysphagic Patients

ObjectiveTo determine the responsiveness of 3 assessment for swallowing disorder on elderly people. Methods40 elderly cases with were assessed with drinking test, classification of swallowing disorder, and swallowing ability evaluation before and 3 weeks after treatment. ResultsThe effect size was 1.04 for drinking test, 1.74 for classification of swallowing disorder and 2.06 for swallowing ability evaluation. The standardized response mean was 2.49 for drinking test, 2.35 for classification of swallowing disorder and 2.78 for swallowing ability evaluation. There was very significant difference before and after treatment in the score of all the scales (P<0.001). ConclusionDrinking test, classification of swallowing disorder, and swallowing ability evaluation are responsive for swallowing disorder in elderly people receiving rehabilitation.
5.Differentiation character of adult mesenchymal stem cells and transfection of MSCs with lentiviral vectors.

This study examined the differentiation character and pluripotency of mesenchymal stem cells (MSCs) under different conditions. Adult MSCs were initially isolated from the bone marrow of rats, cultured in vitro and identified by flow cytometry. After MSCs were transferred to osteogenic and adipogenic medium respectively, the morphological characterization of induced cells was observed. The expression of marker genes was detected by RT-PCR analysis. Then MSCs were transfected with lentiviral vectors pGC-FU-Sox9-EGFP. Enhanced green fluorescence protein (EGFP) expression and transfection efficiency were determined by fluorescence microscopy. The results demonstrated that EGFP caused no effect on the multilineage potential of adult MSCs. Sox9 gene expression of high level was maintained stable in the transfected MSCs and induced MSCs to differentiate into chondrocytes. Aggracan was positive in chondrogenic lineages and the expression of aggracan and type collagen II was significantly increased during MSCs chondrogenic differentiation. It was concluded that Sox9 gene-modified adult MSCs may be promising candidate cells for further studies on tissue engineering. EGFP facilitates the research on MSCs physiological behavior and application in tissue engineering during differentiation both in vitro and in vivo.
6.Gene expression profile of pulmonary tissues in different phases of lung ischemia-reperfusion injury in rats.

In order to provide us new clues to induce some endogenous protective molecular mechanisms, the changes in gene expression profile induced by ischemia-reperfusion in pulmonary tissues of rats were investigated and the dynamic mechanism of pulmonary ischemia-reperfusion injury was elucidated. Thirty male Wistar rats were randomly divided into 6 groups: 5 ischemia-reperfusion (I/R) groups (I/R 0-h, I/R 1-h, I/R 3-h, I/R 6-h, I/R 24-h) and control group (n=5 in each). An in situ ischemia-reperfusion lung injury rat model was established by occluded hilus of lung. The RatRef-12 Expression Beadchip (22 226 gene probes per array) was used to analyze the pattern of gene expression in all groups. The results showed that 648, 340, 711, 1279 and 641 genes were differentially expressed in I/R 0-, 1-, 3-, 6-and 24-h groups respectively. The differentially expressed genes were classified as following 7 functional categories: cytokine, adhesion molecule, growth factor and apoptosis-related factor, oxidation and antioxidation molecule, metabolic enzyme, ion channel and aquaporin, signal transduction molecule. It was suggested that gene chip technology was an effective and quick method for screening differentially expressed genes. Many differentially expressed genes with different functions interacted each other to result in pulmonary ischemia-reperfusion injury.

8.Study on Relationship Between Pathological Changes and TCM Syndrome Differentiation of Delayed Graft Function after Renal Transplantation

Objective To provide references for enriching and innovating patterns of TCM syndrome differentiation by exploring the relationship between pathological changes and TCM syndrome differentiation of delayed graft function (DGF) after renal transplantation. Methods Totally 57 patients with DGF after renal transplantation who were diagnosed by pathological biopsy were also diagnosed by TCM doctors to obtain TCM syndromes. And the pathological diagnoses were compared with the distribution of TCM syndromes to discuss the relationship between them. Results All the patients were classified into four TCM syndromes:spleen-kidney qi deficiency, liver-kidney yin deficiency, qi-stagnation and blood stasis, and deficiency of both qi and yin. The results of pathological biopsy were as follows:acute tubular necrosis, calcineurin inhibitor toxicity and acute rejection. Quantitative corresponding relation was found between the pathological diagnoses and the TCM syndromes. Conclusion There are inner links between pathological diagnosis and TCM syndromes of DGF, which can be references for diagnosis and treatment of TCM syndrome differentiation.
9.Establishment of orthotopic lung transplantation model in mice

ObjectiveTo investigate the establishment of stable mice orthotopic left lung transplant model MethodsForty male Balb/c mice randomly served as recipients and donors.Each couple had the similar weight. Three-cuff technique was used to establish mice orthotopic lung transplant model.The micro CT of chest and the artery blood analysis were detected at first month after transplantation.ResultsThe achievement ratio was 95%.More than 90% of the recipients survived after surgery.Cold ischemia time was (35.6±5.9) min,warm ischemia time was (25.3±7.2) min,donor lung back up time was (21.0±5.6) min,and the whole surgery time was (85±15)min.The long-term survival time achieved more than 30 days with functional lung graft.Micro CT showed clear left lung graft field one month after surgery,and left bronchus cuff was still open.The PaO2 of the artery blood was ( 106.9±5.8 ) mmHg before clipping right lung hlium,and after clipping that was (105.0±8.7) mmHg with the difference being not significant between them (P>0.05).Histologically,the lung graft appeared very similar to the right lung one month after surgery.The pulmonary alveoli were aerated well.ConclusionBased on our experience of the orthotopic lung transplantation model in mice,through practicing,we independently and successfully established orthotopic lung transplantation model in mice.Compared to the method established by the other country,our method is easier to perform and more stable.This will benefit the basic research related to lung transplantation.
10.Current situation and suggestions for ophthalmic medical work at a non-COVID-19 treatment hospital in later stages of epidemic in Wuhan, China

The epidemic of COVID-19 displays a declining trend in China with the enhanced containment and mitigation strategies, and public's normal demand for medical treatment is increasing sharply.The current focus is to ensure the daily medical services needed by non-COVID-19 patients while preventing and controlling the epidemic of COVID-19.Although ophthalmologists do not work on the front lines of COVID-19 outbreak, due to their area of expertise, a variety of situations, such as infection consultations or ophthalmic emergency treatments, can lead to the exposure of ophthalmologists to high-risk environments.This paper mainly analyzes the problems existing in work and life of ophthalmic medical staffs in a fixed-point treatment hospital for non-COVID-19 patients in later stages of the epidemic, and make recommendations and responses, which is helpful to us to obtain guarantee in both safety and quality of medical services, avoid cross-contamination in clinical practice, reduce infectious risk among ophthalmic medical staffs and lessen the mental and spiritual stress for medical staffs.
