1.Study on base insertion/deletion of CAG-rich region in insulin receptor substrate-1 5′-regulatory sequence

To study the relationship of the polymorphism of the insulin receptor substrate-1(IRS-1) gene 5′-flanking regulatory sequence and Type 2 diabetes,the IRS-1 gene 5′-flanking regulatory sequence was scanned by PCR-SSCP in 78 healthy control subjects and 76 Type 2 diabetic subjects. Applying PCR-denatured polyacrylamide gel electrophoresis and silver staining, the insertion/deletion polymorphism of the CAG-rich region was analyzed. The genome DNA of the normal and variant subjects was amplified with high-fidelity pfu DNA polymerase. The purified and digested target fragments were then subcloned into the pCAT Basic vector. Each allele was identified according to the mobility by the restrictive endonuclease digestion of the recombinant combined with denatured polyacrylamide gel electrophoresis and silver staining, and finally the constructive plasmids containing different alleles were analyzed by DNA sequencing. Firstly, we found several insertion/deletion variations in the CAG-rich region of IRS-1 gene. Secondly,7 genotypes and 6 alleles(T1~T6) in this site were detected.Moreover, T5 and T6 were only observed in Type 2 diabetic group.
2.Error Analysis and Quality Assurance of Manufacture on Baffle Plate of Radiation Field

Objective To discuss error factor of the manufacture on baffle plate of radiation field and improve methods of quality assurance and quality control.Methods The errors on different types of baffles were analyzed and evaluated by combining the result of the practice of making baffle and testing of baffle.Results As the difference on the production process and the method of making baffle,there is different degree error on baffle plate of radiation field.The extreme consequence was influenced by quality grate of every manufactured link.Conclusion The technical level of personal,the accuracy of making methods,the equipment precision and the material quality can be produced varying degrees in making.The perfect quality control and quality assurance system can be ensured minimum error.
3.Uterine anterior and posterior wall transfixion suture, combined with intrauterine balloon compression and ligation of ascending branch of uterine artery for the treatment of massive hemorrhage in placenta previa and cesarean section

Objective To explore the curative effect of anterior and posterior wall of the uterus through suture combined with intrauterine balloon compression and uterine artery ascending ligation to treat the hemorrhage of the placenta.Methods In June 2015 to June 2017 in our hospital 60 cases of placenta previa during cesarean section hemorrhage patients, were randomly divided into treatment group and control group , the control of intrauterine balloon compression and ascending branch of uterine artery using the group of patients, patients in the treatment group in the control group based method on the back wall of the lower uterine segment through suture in the treatment of. The curative effect and operation of the two groups were compared.Results The total effective rate was higher in the treatment group than the control group(P<0.05). The intraoperative blood loss, postoperative 24h blood loss of the treatment group patients were lower than the control group, the treatment group patients with operation time, length of hospital stay were lower than the control group(P<0.05).Conclusion The lower uterine segment through anterior and posterior wall suture combined with intrauterine balloon compression and ascending branch of uterine artery ligation is an effective method in treatment of hemorrhage in cesarean section for placenta previa, facilitate clinical application.
4.Effects of Neoadjuvant Chemotherapy on Expressions of ER,PR,C-erbB-2 and p53 in Breast Cancer Tissues

0.05). CONCLUSION: Only partial of the breast cancer tissues were seen of expression changes in ER, PR, C-erbB-2 and p53 after neoadjuvant chemotherapy, but on the whole, the neoadjuvant chemotherapy has no significant influence on the expressions of ER, PR, C-erbB-2 and P53.

The paper reviewed the main methods for the determination of paralytic and diarrhoetic shellfish toxins,including mouse bioassay,immunoassay and high performance liquid chromatography coupled with multiple techniques and so on,and summarized the principles,advantages and disadvantages of these methods.Some newly developed techniques and the development tendency for the detection of paralytic and diarrhetic shellfish poisoning toxins were also described in this paper,in order to give some reference for the detection of marine food in our country.
6.Application of wrapping pancreatic duct-jejunum anastomosis in laparoscopic pancreaticoduodenectomy

Pancreaticojejunostomy is a difficult procedure in laparoscopic pancreaticoduodenectomy.How to improve the method of laparoscopic pancreaticojejunostomy and decrease the incidence of pancreatic leakage is a never-ending issue for clinicians.From November 2002 to March 2012,the digestive tract of 34 patients who received laparoscopic pancreaticoduodenectomy at the First Affiliated Hospital of Guangxi Medical University was constructed by wrapping pancreatic duct-jejunum anastomosis,and satisfactory outcomes were observed except for 2 patients with slight pancreatic leakage.The results of the study showed that wrapping pancreatic duct-jejunum anastomosis is a fast,safe and effective method for pancreaticojejunostomy.
7.Investigation of Microorganisms Contamination in Bathing Beach

Objective To know the present sanitary status of bathing beach in Haikou,and provide the scientific data for improving the assessment of microorganisms contamination of bathing beach. Methods The routine water quality data and related meteorological data of the bathing beach were collected from 2007 to 2009. In July 8th-10th 2009,a high grade bathing beach and an ordinary one were selected,four sampling sites were selected where the contamination by feces were often seen,the number of swimmers was the largest,the samples were collected from the deep-water areas and sand beach respectively,and fecal coliform and enterococcus were examined. Results The concentration of fecal coliform was(1 085.8?538.8)cfu/L in water and(120 000.0?32 659.9)cfu/1 000 g in sand,and the E.faecalis was(32.5?19.1)cfu/L in water and(2 425.0?689.8)cfu/1 000 g in sand. 8.The effect of release and debridement with arthroscopy in the treatment of elbow malfunction

Objective To explore the effect of elbow release and debridment with arthroscopy for elbow malfunction. Methods The study was carried out on 15 patients (male 11, female 4; age 21-63 years old, average 40.1 years old) with the use of arthroscopy to brisement accretion and articular capsule from August 2001 to December 2003. The mean course was 55.2 months (range, 8-24 months). The flexion angle of joint preopration was 15?-60?, average 30.4?, the extension angle was -80?--20?, average -51.2?. The diagnosis was osteophyma and liberum in 4. The old fracture of radius capitulum was in 2; the old fracture of ulnar olecranon in 2; the old fracture of condyle of humerus in 3; degeneration in 4. The brachial plexus anesthesia,the elbow hung to traction, interna and extra-pathway, to cut synovium and accretion fibers with shver, removal liberum and milling osteophyma, meanwhile brisement articular capsule. Pathology manifestation in arthroscopy: there were a lot hyperemia synovium and fiber accretion. There was cartilage exfoliation in 8, hyperplasy and liberum in 5, ossification of cicatricle in 2. The motion range of elbow was reexamined, if the extension function was restricted, release was performed on anterior soft tissue and capsule. If the flexion function was restricted, release was carried out on posterior capsule via posterior straight approach(3 cm supra point of olecranon). Results All patients recovered daily life and occupation postoperative 7 to 14 days. Transient ulnar nerve paralysis occurred postoperatively in one case, which recovered three months later. There were no blood vessel and nerve injury. The mean follow-up period was 14.1 months (range, 7-20 months). At the final follow-up, the flexion of joint post operation was 70?-120?, to improve average 60.5?; the extension of joint post operation was -20?--5?, to improve average 37.6?. In accordance with HSS scoring system, excellent 7, good 5, fail 3. Conclusion Using of arthroscopy to release elbow joint have many advantages such as less trauma, quick recovery and less sequela. The application in release with arthroscopy is a good way for elbow malfunction.
9.Relationship between expression of RANTES and microvascular density in tissues of primary hepatocarcinoma

Objective To investigate the relationship between the expression of regulated-upon activation normal T expressed and secreted(RANTES)and microvascular density(MVD)in hepatocarcinoma(PHC)tissues,and explore its clinical significance. Methods The samples of PHC tissues and adjacent tissues in 47 patients were collected.The expression of RANTES in tissues was detected by immunohistochemistry,and MVD was calculated.The relationship among the expression of RANTES,MVD and clinicopathological features was analysed. Results The positive expression rate and expression score of RANTES in PHC tissues were significantly higher than those in adjacent tissues(55.32% vs 19.15%,P<0.01;1.89±1.77 vs 0.77±1.29,P<0.01).MVD in PHC tissues was significantly higher than that in adjacent tissues(67.30±13.68 vs 37.20±10.58,P<0.01).MVD of PHC tissues in patients with metastasis was significantly higher than that in patients without metastasis[(73.50±13.77)/HP vs (64.10±12.68)/HP,P<0.05],while there were no correlations among MVD,expression of RANTES and the other clinicopathological features of PHC.MVD was positively correlated with the expression score of RANTES in PHC tissues(r=0.386,P<0.05). Conclusion RANTES might be closely related to the angiogenesis in PHC.
10.Construction of recombinant plasmid of Pseudomonas aeruginosa exotoxin A and its secreting expression in E. coli

Objective To clone and to express the full-length Pseudomonas aeruginosa exotoxin A gene and to purify the expressed protein using affinity chromatography. Methods Exotoxin A gene was amplified from the recombinant plasmid pSK/PEA-T vector and subcloned into the pMAL-P2X vector. Then the recombinant plasmid pMAL- PEA was transformed to E.coli BL21. After induction with IPTG, the expressed protein was analyzed by SDS-PAGE and purified with affinity chromatography. Results The recombinants expressed the fusion protein at a level of about 20% of total cell proteins, and 80% of the fusion protein was secreted into the supernatant. Conclusion Successful expression and purification of PEA are of significance for in-depth study of the pathogenic mechanism of Pseudomonas aeruginosa and preparing immunotoxin.
