1.The changes of IL-6 gene expression in acute repetitive hypoxia

Objective: This study was to observe the changes of IL-6 gene expression in acute repetitive hypoxia. Method: When the ratio of living cells was more than 95%, nerve cells cultured were passed through with mixed gases of 95% N_2+5%CO_2 for 3 min followed by another gases mixture of 95% O2+5% CO_2 for 10 min. After repeated the experiment as above, the alteration in the expression of IL-6 gene was measured using the RT-PCR method. The prod ucts of PCR were analyzed with computer Gel imaging and image analysis instrument. Result: After the first hypoxia IL-6 gene expression enhanced, and after the second hypoxia, it was still stronger than baseline although it declined slightly. There was no significant RNA disintegration. Conclusion:The IL-6 involves in the cellular defense in hypoxia adaptation.
2.Cloning and identification of human gene transactivated by hepatitis C virus core protein 1

Objective To screen and clone the target genes transactivated by hepatitis C virus (HCV) core protein. Methods The mRNA was isolated from HepG2 cells transfected with pcDNA3.1(-)-core and pcDNA3.1(-) empty vector, respectively, and suppression subtractive hybridization (SSH) method was employed to analyze the differentially expressed DNA sequences of the two groups. The obtained sequences were searched for homologous DNA sequence from GenBank. One of them was confirmed to be a new gene without homology with known genes in this database.Then, electric polymerase chain reaction was conducted for the cloning of the full-length DNA of the new gene, in conjunction with Kozak rule and the existence of polyadenyl signal sequence. The reverse transcription PCR (RT-PCR) was used to amplify the new gene with mRNA from HepG2 cell as the template. The coding sequence for the new gene was deduced according to the nucleotide sequence. Results A new gene with unknown function was named TAHCCP1.The nucleotide sequence of the new gene and its corresponding protein-encoding amino acid, which was 2 001nt and composing 667aa, have been determined. The sequence of the TAHCCP1 gene has been registered in GenBank with its accession number AY038359. Conclusion TAHCCP1 gene transactivated by HCV core protein was cloned and identified successfully by a combination of molecular biological technology and bioinformatics technique. The results are expected to pave the way for the study of the molecular mechanism of the transactivating effects of HCV core protein and the development of new therapies for chronic hepatitis C.
3.Possibility of the acellular bony microtubule used to construct tissue engineering ossicular prosthesis

OBJECTIVE To study the possibility of acellular bony microtubule used to construct tissue engineering ossicular prosthesis. METHODS To prepare the acellular bony microtubule, observe the change of its histocompatibility and biomechanics. The acellular bony microtubule carried with collagen/ BMP-2 was implanted into the middle ear of the animals. The ectopic osteogenesis was observed. RESULTS The acellular bony microtubule has good histocompatibility. There wasn’t significant change about biomechanics. After carried with collagen/BMP-2, the bony microtubule can induce osteogenesis in the animal’s middle ear. CONCLUSION The acellular bony microtubule carried with collagen/BMP-2 has the possibility to be used to construct tissue engineering ossicular prosthesis.
4.Clinical Observation of Xiaocuo Prescription for Acne Vulgaris:A Report of 325 Cases

[Objective] To investigate the effect of Xiaocuo Prescription (XP), a herbal medicine compound with the actions of clearing away heat and toxins, activating blood and removing blood-stasis, on acne vulgaris. [Methods] Four hundred and forty six cases of acne vulgaris were randomized to two groups: group A (325 cases) treated with oral use of XP (mainly composed of Flos Lonicerae, Herba Hedyotis Diffusae, Herba Taraxaci, Flos Chrysanthemi, Radix Scutellariae, Cortex Mori, Radix Salviae Miltiorrhizae, etc.) and external application of Acne Vulgaris Liquid (composed of 3% metronidazole and 2% erythromycin), group B (121 cases) treated with oral use of isotretinoin and external application of Acne Vulgaris Liquid. Skin lesions on the face as well as the side effects were observed before and after treatment. [Results] Ridit analysis showed that curative effect in group A was better that in group B ( u = 2.204 6, P .05). No obvious side effects were found in group A but in 5 cases of group B side effects such as skin dryness, nausea and vertigo were found. [ Conclusion ] XP is effective in treating acne vulgaris . Further research is needed for its therapeutic mechanism.
5.Hospital emergency response and contingency plan in catastrophic fire hazards and explosion:experiences and improvements

A retrospective study and cause analysis on the emergency rescue and medical service during the 2015 Tianjin harbor explosion accident,probed into the defects of the contingency plan of the hospital.Problems found include impracticability or deficiency of the plan,lack of safety awareness,and absence of preventive measures.A cause analysis was thus made to propose the following improvements on the contingency plan:1.Development of specific standard operation procedures(SOPs)for medical emergency response,including SOP for emergency response organization and command,that for the procedure and responsibility of massive injuries examination,and that for emergency rescue;2. Development of the plan for data statistics,that for volunteer management,that for media communications and that for donation management.
6.Considerations about Achievements and Age

9.An event-related potential investigation of deficient inhibitory control in individuals with internet addiction disorder

Objective To investigate deficient inhibitory control in individuals with IAD using a visual go/no-go task by ERPs. Methods 26 individuals met YDQ criteria for IAD were enrolled as research group and marched sexual and age 26 healthy person enrolled as control group. BIS-11 was used for measures of impulsivity.A go/no-go task involved eight different two-digit numerical stimuli. The response window was 1000 ms and the ITIwas 1500 ms. EEG was recorded during participants performed the task. BESA 5.2.0 was used to perform data analysis and the no-go N2 amplitude was analyzed for investigation of inhibitory control. Results BIS-11 total scores, attentional key scores and motor key in IAD group were higher than that of control group. In the go/no-go task, false alarm rate of IAD group was higher and hit rate was lower than that of control group. A repeated measure ANOVA revealed a significant group, frontal electrode sites and group × frontal electrode sites main effect for N2amplitudes of no-go conditions ( for group: F= 3953, df= 1, P= 0.000;for frontal electrode sites: F= 541, df= 9, P= 0.000;for group × frontal electrode sites: F = 306, df = 9, P = 0. 000 ), and a significant group, central electrode sites and group × central electrode sites main effect for N2 amplitudes of no-go conditions ( for group: F=9074, df= 1, P = 0. 000;for central electrode sites: F = 163, df= 2, P = 0.000;for group × central electrode sites: F = 73, df= 2, P = 0.000). N2 amplitudes of no-go conditions were lower than those at control group. Conclusions Individuals with IAD were more impulsive than controls and shared neuropsychological and ERPs characteristics of compulsive-impulsive spectrum disorder, which supports that IAD is an impulse disorder or at leastrelated to impulse control disorder.
10.Clinical Observation of Bevacizumab Combined with FOLFIRI Plan in the Treatment of Advanced Colon Cancer

OBJECTIVE:To observe therapentic efficacy of bevacizumab combined with irinotecan+leucovorin+fluorouracil (FOLFIRI)plan in the treatment of advanced colon cancer,toxic reaction and patients'survival rate. METHODS:A total of 113 patients with advanced colon cancer admitted to the oncology department in our hospital from Jan. 2010 to Aug. 2014 were random-ized into observation group 1(40 cases),observation group 2(39 cases)and control group(34 cases). Three groups received FOLFIRI;observation group 1 and 2 were additionally given Bevacizumab injection 5 and 7.5 mg/kg 14 d as a treatment course, for 8 cycles. Clinical efficaices as well as the positive rate of VEGF-A,immune indexes(the proportion of CD3+,CD3+CD4+,CD3+CD8+ in T cell subset)before and after treatment,the incidence of toxic reaction,1-year and 2-year survival rates were compared among 3 groups. RESULTS:The total response rate of observation group 1 and 2 were significantly higher than control group,and the positive rate of VEGF-A in observation group 1 and 2 were significantly lower than control group,with statistical significance (P<0.05). The proportion of CD3+,CD3+CD4+ and CD3+CD8+ in 3 groups were significantly lower than before treatment,but ob-servation group 1 and 2 were significantly higher than control group,with statistical significance(P<0.05). The incidence of grade hypertension(grade 3-4)in observation group 1 and control group were lower than observation group 2;the incidence of leucope-nia(grade 3-4)in observation group 1 and 2 were significantly lower than control group,with statistical significance(P<0.05). There was no statistical significance in 1-year survival rate among 3 groups(P>0.05). 2-year survival rate of observation group 1 and 2 were significantly higher than control group,with statistical significance(P<0.05). CONCLUSIONS:For advanced colon cancer,different doses of bevacizumab combined with FOLFIRI have significant synergistic effect,can effectively inhibit VE-FG-A,play a role of immune protection and anti-toxic side effects,and prolong the survival time. The incidence of hypertension in patients treated with low-dose bevacizumab is relatively lower and the safety is better.
