1.Research and Application of Analytical Technique onδ18Op of Inorganic Phosphate in Soil

Analytical technique on oxygen isotope composition of phosphate in soil has important applications in biogeochemical research. As the composition of the soil is complex with multiple oxygen sources, a liable phosphate enrichment and purification technology is important for phosphate oxygen isotope research. Here we presented a protocol on the analysis of the δ18 Op of inorganic phosphate in soil, and phosphate was purified with multiple mineral precipitations and finally precipitated as silver phosphate. The efficiency of NH4 OH purification method was examined and the purity of the product was verified through XRD and element analysis. The result showed that treatment of the Ag3 PO4 sample by recrystallizing with ammonia could remove the impurity efficiently without significantly modifying oxygen isotope compositions. The validity of sample processing and reliability of isotope analyses were confirmed by processing a KH2 PO4 standard in parallel with soil P sample. The analytical result of three soil types indicated that this method provided good precision of 0. 2‰. The differences of δ18 Op between three soils groups are significant ( From 15. 2‰ to 19. 5‰) and can be used to tracing P sources.
2.Effect of Mirror Therapy on Upper Extremity Motor Function and Activities of Daily Living after Stroke:A Meta-analysis

Objective To systematically summarize the effects of mirror therapy on upper extremity motor function and activities of dai-ly living after stroke. Methods The literatures about mirror therapy published between January, 2006 and May, 2016 were recalled from Medline, PubMed, CINAHL, OT seeker, PEDro, Wanfang and CNKI databases. The randomized control trials were included, and those didn't concern upper extremity motor function and activities of daily living were excluded. The data were extracted and analysed with Rev-Man 5.3. Results Fourteen trials were included. Mirror therapy could significantly improve Fugl-Meyer Assessment score (SMD=0.81, 95%CI=0.43-1.20, P<0.001, I2=64%). Subgroups analysis indicated movement based mirror therapy was more effective on mild to moderate hemiplegia (SMD=0.96, 95%CI=0.59~1.34, P<0.001, I2=51%) and the subgroup differences were also significant (P=0.004). Mirror therapy could improve score of Action Research Arm Test (SMD=0.33, 95%CI=0.01-0.64, P=0.040, I2=0), Box and Block Test (SMD=0.70, 95%CI=0.03-1.37, P=0.040, I2=62%) and Brunstrom stages (SMD=1.56, 95%CI=1.07-2.06, P<0.001, I2=41%) of affected upper extremities. It could also improve activities of daily living (SMD=0.93, 95%CI=0.62-1.24, P<0.001, I2=0). No evidence revealed mirror therapy could change muscle tone of flexors of affected upper limbs (SMD=-0.22, 95%CI=-0.73-0.28, P=0.890, I2=0). Conclusion Mirror therapy can signifi-cantly improve upper extremity motor function and activity of daily living.
3.Construction of the evaluation index system for the communication effect of nursing WeChat public accounts in hospital

Objective:To construct evaluation index system for the communication effectiveness of nursing WeChat public accounts in hospital, and to provide a scientific and systematic evaluation tool for the communication effectiveness of nursing WeChat public accounts in hospital.Methods:Based on Lasswell′s "5W" communication process model, the evaluation index system for the communication effectiveness of nursing WeChat public accounts in hospital was constructed by applying Delphi expert consultation method and analytic hierarchy process.Results:The the evaluation index system for the communication effect of nursing WeChat public accounts in hospital consists of 5 first-level index, which contained communicator, communication content, communication media, communication audience, communication environment, and 12 second-level index, which contained professional quality, function setting, public trust, using experience, internal environment, etc, and 52 third-level index.Conclusions:The evaluation index system for the communication effectiveness of nursing WeChat public accounts in hospital could provide nursing managers an systematic evaluation tool. It could promote the construction and development of nursing WeChat public accounts and ensure that patients receive the health information which were scientific, authoritative and standard.
4.Analysis of Volatile Organic Compounds by Specific Carbon Stable Isotope

Theδ13 C values of volatile organic compounds ( VOCs) in various emission sources and ambient air were analyzed by using thermal desorption coupled with gas chromatography and isotope ratio mass. The lowest sample concentration and peak shape quality needed for high precision and accurate analysis were investigated. Fuel evaporation ( gasoline and diesel) , vehicle exhaust, solvent evaporation, dining fumes and ambient air of different functional zones of Xiamen city were collected using Tenax TA tube, and the significant differences in δ13 C values of VOCs between these sources were observed. The δ13 C value of gasoline exhaust ( 97 # ) was heavier (-25 . 84‰) than that of dining fumes (-30 . 26‰) and theδ13 C values of fuel evaporation were heavier than that of vehicle exhaust after combustion. The average δ13 C value of atmospheric VOCs in Xiamen was at the level of -27 . 03‰ to -25 . 40‰, which was close to the δ13 C value of the evaporation and exhaust of gasoline and diesel, indicating that the VOCs in the atmosphere of Xiamen was highly influenced by transportation related sources.
5.Effects ofLiuwei Dihuang Decoction on cAMP and PDE3B in Adipose Tissues of Rats with Type 2 Diabetes

Objective To observe the effects ofLiuwei Dihuang Decoction on cAMP and PDE3B in adipose tissues of rats with type 2 diabetes;To explore its mechanism.MethodsTotally 80 SD rats were randomly divided into two groups:control group and molding group. Rats in the molding group established the type 2 diabetic models by feeding high sugar and fat diet combined with intraperitoneal injection of Streptozotocin. The successful modeling rats were divided into model group, Rosiglitazone group, andLiuwei Dihuang Decoction group. Administration groups were given relevant medicine for gavage, while model group and blank group were given the same amount of normal saline for 30 days. FBG levels were detected, and cAMP and INS levels were detected by ELISA. The protein expressions of PDE3B in adipose tissues were determined by Western blot. The mRNA expressions of PDE3B in adipose tissues were detected by RT-PCR.Results Compared with the model group, the FBG and INS levels in type 2 diabetic rats were reduced in Rosiglitazone group andLiuwei DihuangDecoction group (P<0.05,P<0.01). InLiuwei Dihuang Decoction group, the protein and mRNA expressions of PDE3B in the adipose tissues were higher, and cAMP levels were lower than the model group (P<0.01).ConclusionLiuwei Dihuang Decoction can increase PDE3B expression, and decrease cAMP level, which may be one of the mechanisms for improving insulin resistance in type 2 diabetic rats.
6.Establishment of a mouse model of kidney deficiency induced by oral administration of hydrocortisone and evaluation of related factors

Objective To establish a mouse model of kidney-yin and kidney?yang deficiency after oral administra?tion of hydrocortisone, and to explore the related evaluation factors. Methods The model was established by oral adminis?tration of hydrocortisone to induce kidney?yin and kidney?yang deficiency in mice. The survival and body weight of the mice were observed. The serum content of adrenal cortical hormone ( ACTH) , cortisol ( Cor. ) in the hypothalamic?pituitary?ad?renal (HPA) axis, and thyroid stimulating hormone (TSH), triiodothyronine (T3), thyrox (T4) in the hypothalamic?pi?tuitary?thyroid (HPT) axis, follicle?stimulating hormone (FSH), estradiol (E2), testosterone (T) in the hypothalamic?pituitary?gonadal ( HPG) axis were determined by radioimmunoassay. Results The body weight of kidney?yin and kidney?yang mice were decreased, the serum ACTH, Cor, TSH, T3, T4 contents were decreased, the serum FSH, E2, T contents were increased in the kidney?yang deficiency model mce ( P<0. 01 ) , and those parameters in the kidney?yin deficiency model mice were changed in opposite direction. Conclusions It is found that the hormone levels of ACTH, Cor, TSH, T3, T4, FSH, E2 and T in kidney deficiency mice are changed, and cortisol can be used as an important index to evaluate the model of kidney deficiency induced by glucocorticoid.
7.Regulatory effect of electroacupuncture on blood sugar and blood lipoprotein(a)in rats with diabetes mellitus

BACKGROUND: Many study results proved that electroacupuncture can cause change of endogenous opioid peptide system.OBJECTIVE: To observe the change of blood sugar and blood lipoprotein (a) in rats with diabetes mellitus after electroacupuncture therapy and probe into the effect of opioid peptide system in the treatment for diabetes with electroacupuncture.DESIGN: Randomized controlled trial with experimental animals as the subjects.SETTING: The experiment was conducted in the Department of Pathophysiology, Medical College of Shandong University.MATERIALS: The experiment was completed in the Department of Pathophysiology, Medical College of Shandong University from July 2004 to December 2004. Totally 140 male Wistar rats were involved.METHODS: Ten Wistar rats were collected randomly as the blank control group. The other 130 rats were given 20 g/L streptozotocin 50 mg/kg intraperitoneally and the rats in blank control group were given the same volume of saline intraperitoneally. Three weeks later, the tails were cut off and the blood sugar was measured 2 hours after food intake. The rats with obvious rapid sugar tolerance were given foods with high calories for 28 days as the model rats. There were 64 rats were successful models and they were classified into 3 groups randomly, namely model control group( n = 20), electroacupuncture therapy(n=22) and electroacupuncture with naloxone groups( n = 22) . The blood sugar and blood lipoprotein(a) were detected after treatment for the diabetic rats with streptozotocin.MAIN OUTCOME MEASURES: Blood sugar level and blood lipoprotein(a) of each rat after the final therapy.RESULTS: Totally 74 rats entered the result analysis. Electroacupuncture therapy could reduce the blood sugar and blood lipoprotein(a) of diabetes mellitus rats, while rats with naloxone injection before therapy were insensitive to electroacupuncture. [The blood sugar value were(13.91 ± 3.79),( 10.33 ± 2.36), (12.96 ± 2.23)mmol/L in the model control group, electroacupuncture therapy group, electroacupuncture with naloxone group respectively; while the level of blood lipoprotein(a) were(13.75 ±2.69),( 10.81 ± 2.96), (13.01 ± 3.23)mg/L respectively].CONCLUSION: Electroacupuncture therapy can reduce the level of blood sugar and the risk of atherosclerosis in rats with diabetes mellitus. Although the mechanism can not be explained through the meridian theory, according to the experimental result that blood sugar and blood lipoprotein(a) were no longer intervened by electroacupuncture therapy after injecting naloxone, it is presumed that electroacupuncture therapy can exert its intervention effect on blood sugar and blood lipoprotein(a)through opiate system.
8.Construction and identification of expressing human c-Jun N-terminal kinase(JNK)recombinant adenovirus

Objective To construct replication deficient recombinant adenovirus expressing human c-Jun N-terminal kinase by homologous recombination.Methods The linearized recombinant shuttle vector pAdTrack-CMV-WT-JNK was co-transformed with backbone vector pAdEasy-1 into bacteria BJ5183 for recombinant adenoviral vector.The recombinant adenoviral vector was transfected into HEK293 packing cells to construct replication deficient recombinant adenovirus,and then the recombinant adenovirus was detected by PCR and DNA sequencing.Results JNK recombinant adenoviral vector was effectively transfected into HEK 293 cells and was successfully packed by intracellular enzyme.The expression of green fluorescent protein(GFP)was observed on the 5th day after transfection.The fragment of JNK gene was amplified by PCR and identified by sequencing.The animal experiment confirmed that Ad-WT-JNK was effectivety expressed in liver tissue. Conclusion The research successfully constructed recombinant adenoviral vector and recombinant adenoviral particle.And the achievement laid a foundation for further investigation of the function and application of JNK.
9.Construction and identification of dominant-negative c-Jun N-terminal kinase(DN-JNK)recombinant adenovirus

Objective To construct and identify replication deficient recombinant adenovirus expressing human c-Jun N-terminal kinase(JNK)by homologous recombination adenovirus dominant-negative type JNK(Ad-DN-JNK).Methods The linearized recombinant shuttle vector pAdTrack-CMV-DN-JNK Was co-transformed with backbone vector pAdEasy-l into bacteria BJ5183 for recombinant adenoviral vector.The recombinant adenoviral vector was transfected into HEK293 packing cells tO construct replication deficient recombinant adenovirus,and then the recombinant edenovirns WaS detected by PCR and DNA sequencing.Western blot analysis was utilized to detect the Cxpression of Ad-DN-JNK and the level of insulin receptor substrate l Serine307 phosphorylation.Results JNK recombinant adenoviral vectorcould be effectively transfeeted into HEK 293 cell and successfully packed by intracellular enzyme.The expression of green fluorescent protein(GFP)Was observed on the 5th day after transfection.The fragment of JNK gene waS amplified by PCR and identified by sequencing.The titer of the prepared Ad-DN-JNK is 2.5×1010 pfu/ml.The animal experiment confirmed that constructed Ad-DN-JNK could be effectively expressed in liver tissue.Conclusion The research successfully constructed recombinant adenoviral vector and recombinant adenoviral particle.Animal experiment demonstrated the Ad-DN-JNK could effectively mediated the expression of DN-JNK gene and down-regulated the level of IRSlscfine307 phosphorylation.The achievement laid a foundation for further investigation of the function and application of JNK.
10.Construct a rat model of nonalcoholic fatty liver disease with insulin resistance by feeding ruts fat-rich diet

Objective To construct a rat model of nonalcoholic fatty liver disease (NAFLD) by feeding rats fat-rich diet and analyze the effect of insulin resistance (IR)in the development of NAFLD. Methods Male SD rats were randomly divided into normal diet group (NG, n =24) and fat-rich diet group (FG, n =24). At the end of feeding for2 weeks, 4 weeks, 6 weeks or 8weeks, 6 rats in NG and FG were randomly took out. Their weight were recorded, then the serum fasting blood sugar, fasting insulin, triglyceride, total cholesterol, ala-nine aminotransferase and aspartate aminotransferase were measured, and the fasting insulin resistance index and liver index (liver weight (g)/body weight(g) × 100%)was calculated. Then liver tissues were homogenized, and muleic dialdehyde and superoxide dismutase were determined. The hepatic steatosis in all rats was assessed according to the results under light microscope. Results The body weight of rats in NG increased faster than those in FG after six weeks. The liver index of rats in FG was markedly higher than that in NG since the second weekend. The rats in FG began to have hepatocyte steatosis from the second weekend, had insulin resistance, hyperlipidemia, dysfunction of liver and lipid peroxide of liver from the fourth weekend, suffered mild fatty liver from the sixth weekend, and developed to moderate fatty liv-er from the eighth weekend. Conclusions NAFLD with IR model was successfully developed by feeding SD rats with an improved rich-fat diet for 6 weeks. IR may play an important role in the development of NAFLD.
