1.The effects of XIAP siRNA and embelin on TRAIL-induced growth inhibition and apoptosis in hepatocarcinoma cells

ObjectiveTo investigate the effects of X-linked inhibitor of apoptosis (XIAP) siRNA or XIAP antagonist Embelin on the growth inhibition and apoptosis of hepatcellular cancer cell line HepG2,which was treated by tumor necrosis factor-related apoptosis (TRAIL). Methods HepG2 cells were tranfected either by XIAP siRNA or a negative control,followed by treatment with TRAIL,Embelin or a combination of the two.XIAP expression,cell growth,and caspase-3 activity were determined by Western blot,MTT assay and fluorescent caspase-3 assay,respectively.Cleaved PARP expression levels of were measured by Western blot.ResultsThe XIAP protein expression was significantly downregulated by transfection with XIAP siRNA.Compared with the negative control,the XIAP siRNA significantly inhibited the growth of HepG2 cells treated by 100ng/ml(6.8% ±1.2% vs.11.8%±4.0%,P<0.05)and 1000 ng/ml (18.9% ±2.0% vs.26.6% ±1.5%,P<0.01)by TRAIL.TRAIL-induced caspase-3 activation and PARP cleavage were also increased significantly by XIAP siRNA transfection.In addition,Embelin also significantly inhibited cell growth (P<0.01),activation of caspase-3 (P< 0.01) and TRAIL-induced PARP cleavage.Conclusions Both XIAP siRNA and Embelin may potentially contribute to clinical treatment of hepatocellular carcinoma.
2.The Value of 3D-Reconstruction Technique with Spiral CT in Acetabulum Fracture

Objective To evaluate the usefulness of the transect imaging and 3D-reconstruction with spiral CT in acetabulum fracture.Methods 23 cases of acetabulum fracture proved by clinical plain radiograph,transect imaging and 3D-reconsection were reviewed retrospectively.3D-reconsection were obtained on the basis of spiral transect imaging with use of 2~3 mm collimation.3 pitch and 1.2~2 mm reconstruction interval.Results Fractures were demonstrated definitely with transect imaging.3D-reconstruction in all patients the transect imaging displayed the fractures and the displacement of the burden opex of acetablulum,the disruption within joint,the direction of fracture.Displacement of detached bone fragments and capitis femoris were display stereoscopically by 3D-reconstruction.Conclusion The transect imaging and the 3D-reconstruction of spiral CT technique with spiral CT could afford the more valuable messages.Surgeon will benefit greathy from their usefulness in the choice of the surgical intervention and operative planning.
3.Effect of leech injection on thrombosis and activity changes of plasma t-PA and PAI in mice

Objective: To investigate the effect of leech injection on thrombosis and plasma tissue plasminogen activator (t PA) and its inhibitor (PAI) in the thrombosis model of mice. Methods: The different dose leech injection of different doses were given to mice in different groups for 7 days respectively. Thrombosis was induced in mice by carrageenan. The plasma t PA and PAI activity were detected by chromogennic substracte analysis.Results: The average thrombosis length of tail in the groups of leech injection were significantly shorter than the length in the groups of thrombosis control. Compared with the group of thrombosis control, the plasma PAI activity in the leech injection groups was lower. Conclusions: The leech injection can reduce thrombosis in mice. The mechanism might be associated with reducing the release of PAI.
4.Effects of tripter gium wilfordi on urine mononeuclear cells and CD44 expression in patients with IgA nephropathy

Objective:To study the possible therapeutic mechanisms of triptergium wilfordi in treating glomerulonephritis.Methods:Thirty seven patients with IgA nephropathy were detected urine mononeuclear cells densities with flow cytometry during taking triptergium wilfordi tablets,30 healthy people were used as normal control.Results:The patients urine CD3 +?CD4 +?CD8 +?CD14 + and CD44 + cells densities were much higher than normal control's,but obviously decreased after triptergium wilfordi treatment.CD4/CD8 ratio was much lower in patients than in normal control.It increased markedly after treatment.Patients who reached remission initially had a lower CD4 + cell percentage and higher CD14 + cells percentage than those showed no response to triptergium wilfordi treatment.Conclusion:Triptergium wilfordi could modulate kidney immune cell function and adhesive molecule CD44 expression.Its effects were associated with renal immunity.

Diminished apoptosis,due to overexpression of antiapoptotic proteins, is involved in tumorigenesis and chemoresistance. Antisense oligodeoxynucleotide (ASODN) has been used to inhibit the expression of antiapoptotic proteins(f.g bcl 2, bcl x L , survivin),aimed to induce apoptosis or enhance chemosensitivity.The combination of ASODN with conventional treatment is possibly a reasonable approach in furture cancer therapy. Objective To evaluate the clinical value of bronchofiberscope and catheter intervention in treatment of multi-drug resistant pulmonary tuberculosis.Methods Thirty-five patients with multi-drug resistant pulmonary tuberculosis were treated by injecting drug gel,it was composed by rimifon and rifampin and aldinamide and capreomycin through bronchofiberscope and catheter in addition to chemotherapy,while thirty-five controls were treated by chemotherapy only.Results At the end of the treatment,the sputum bacterial conversion to negative rate was 88 5%,radiographic improvement rate was 82 9% and cavity closing rate was 31 4% in the treatment group,all of which were higher than in the controls(51 4%,45 7% and 11 4% respectively)(P
7.Clinical Observation of Carboprost Tromethamine in the Treatment of Postpartum Hemorrhage with Two Routes of Administration

OBJECTIVE:To observe therapeutic efficacy and safety of carboprost tromethamine in the treatment of postpartum hemorrhage with two routes of administration. METHODS:285 patients with postpartum hemorrhage were randomly divided into control group(143 cases)and trial group(142 cases). Control group was given Carboprost tromethamine injection 250 μg on del-toid of arm;trial group was given same dose of Carboprost tromethamine injection via cervix uteri. Both groups received medicine after third stage of labor. The therapuetic efficacy,the amount of endometrorrhagia and colporrhagia within 2 h,the incidence of ADR after labor were observed in 2 groups. RESULTS:After treatment,the amount of postpartum hemorrhage was smaller than 400 ml in 2 groups and didn't exceed the standard,without statistical significance(P>0.05). The amount of endometrorrhagia in trial group was significantly higher than in control group,with statistical significance(P<0.05);the amount of colporrhagia in tri-al group was significantly lower than in control group,with statistical significance(P<0.05). There was no statistical significance in the incidence of ADR between trial group(4.90%)and control group(6.31%)(P>0.05). CONCLUSIONS:The intramuscular injection is selected for the prevention of postpartum hemorrhage caused by uterine inertia;the cervical injection is selected for the prevention of postpartum hemorrhage caused for the other patients.
8.Antibiotic Resistance and Genotype of Escherichia coli in Biliary Tract Infection

OBJECTIVE To investigate the drug resistance of Escherichia coli in biliary tract infection and genotype of ESBLs.METHODS Antimicrobial susceptibility test was performed using Kirby-Bauer method.The genotypes of ESBLs were detected by PCR.RESULTS The detection rate of ESBLs producing bacteria was 32.4%(23 strains),and the drug-sensitivity test indicated that E.coli was sensitive to imipenem,meropenem and cefoperazone/subactam,but these ESBLs producing strains were resistant to other antimicrobial agents.ESBLs were identified as TEM type in 16 strains,SHV type in 4 strains,CTX-M type in 12 strains and OXA type in 5 strains.CONCLUSIONS ESBLs-producing E.coli in biliary tract infections show strous resistance.TEM and CTX-M are the most common ESBLs genotypes.
9.Effect of estrogen on the osteogenic ability of osteoblast

Objective To verify the effects of different conc entrations of auecifemine on the function of osteoblast cell expression and oste ogenic ability in vitro . Methods Iliac spongy bone specimens were obtained from twel ve adult patients scheduled surgery. After the bone pieces were treated with col lagenase-trypsin, osteoblasts were obtained from cancellous bones. Then the ost eoblasts were purified and cultivated. They were cultured in medium with various concentrations(1?10 -11 , 1?10 -9 , 1?10 -7 , 1?10 -6 , 1 ?10 -5 , 5?10 -5 mol?L -1 ). The alkaline phosphatase (ALP) acti vity, osterocalcin and osteogenic ability were examined by biochemical assay and radioimmunologic assay (RIA), respectively. Results The co rrelation of auecifemine of different concentrations with ALP activity and osteo calcin secretion was positive. Auecifemine of lower concentrations (1?10 -11 mol?L -1 , 1?10 -9 mol?L -1 and 1?10 -7 mol?L -1 ) s howed the stimulating effect on osteogenic ability and but not higher concentrat ion. Conclusion Estrogen can increase osteoblast ALP activi ty, osteocalcin production, and osteogenic ability.
10.Randomized controlled trial of sodium hyaluronate for degenerative disorders of the knee joint

Objective To assess the effect of sodium hyaluromate (HA) on degenerative disorders of the knee joint (KJ). Methods A prospective randomized controlled clinical trial was conducted. The experimental group received injections in the compartments of the involved KJ with 1% HA 20mg, whereas the control group received prednisolone (PS) of 75mg once a week with five injections as one course. One week before and after the treatment , clinical symptoms, amount of interleukin-6 (IL-6) and total protein of synovial fluid were measured and compared. Results Both drugs could relieve the clinical symptoms of KJ degenerative disorders. In HA group, marked improvement rate was 52.78% and failure rate was 2.86%, whereas marked improvement rate was 39.29% and failure rate 17.86% in PS group. The declined levels of IL-6 in synovial fluid were notably greater in HA group than those in PS group. Conclusion Intra-articular injection of HA is effective and safe in treating KJ dgenerative disorders with mild adverse reactions.
