2.Rapid and Sensitive Detection of PRRSV by a Reverse Transcription-Loop-mediated Isothermal Amplification Assay

A real-time monitoring reverse transcription loop-mediated isothermal amplification (RT-LAMP) assay was developed for the sensitive and specific detection of prototypic,prevalent North American porcine reproductive and respiratory syndrome virus (PRRSV)strains.As a higher sensitivity and specificity method than reverse transcription polymerase chain reaction (RT-PCR),the RT-LAMP method only used a turbidimeter,exhibited a detection limit corresponding to a 10-4 dilution of template RNA extracted from 250 μL of 105 of the 50% tissue culture infective dose (TCID50) of PRRSV containing cells,and no cross-reactivity was observed with other related viruses including porcine circovirus type 2,swine influenza virus,porcine rotavirus and classical swine fever virus.From forty-two field samples,33 samples in the RT-LAMP assay was detected positive,whereas three of which were not detected by RT-PCR.Furthermore,in 33 strains of PRRSV,an identical detection rate was observed with the RT-LAMP assay to what were isolated using porcine alveolar macrophages.These findings demonstrated that the RT-LAMP assay has potential clinical applications for the detection of highly pathogenic PRRSV isolates,especially in developing countries.
3.Application of recognizing bone species by touching blindly in experimental teaching of human anatomy

Objective:To propose the application of recognizing bone species by touching blindly (ARBTB) and verify its application effect in human anatomy experimental courses.Methods:The study included experiment 1 and experiment 2, and the research objects were 60 students majoring in clinical medicine in Batch 2019 and 60 students majoring in preventive medicine in Batch 2018, respectively. The research objects in each experiment were randomized into mainstream teaching method group and ARBTB group, with 30 students in each group. In experiment 1, after students studying for the same hours, both of two groups carried out the same test, and then the average scores of the two groups were compared. In experiment 2, the average time spent by the two groups of students in osteology learning was compared.Results:In experiment 1, ARBTB group's average score of the test was significantly higher than that of the mainstream teaching group ( P<0.001). In experiment 2, the ARBTB group took much less time in osteology learning than the mainstream teaching group ( P<0.001). Conclusion:Compared with the mainstream teaching method, ARBTB is more effective in osteology learning, which is worth popularizing.
4.Correlation between vitamin A and E concentrations during pregnancy and preeclampsia

Objective To investigate the relationship between vitamin A and vitamin E concentra-tions and preeclampsia. Methods A total of 160 pregnant women who were finished normal perinatal care and tested for serum vitamin A and vitamin E concentrations in the First Affiliated Hospital of Shenzhen Uni-versity (Shenzhen Second People's Hospital) from March 1, 2018 to February 28, 2019, including 80 pre-eclampsia cases and 80 healthy control cases were enrolled in the study. The concentrations of serum vita-mins A and E in early pregnancy ( <14 weeks) and late pregnancy (≥34 weeks) were determined by high performance liquid chromatography tandem mass spectrometry. The collected clinical and test data were sta-tistically analyzed. Results ⑴There was no significant difference in vitamin A concentration between pre-eclampsia group and control group during early pregnancy (P=0. 360) and there was no significant differ-ence in vitamin A concentration between preeclampsia group and control group during late pregnancy ( P=0. 450). ⑵ The concentration of vitamin E in early pregnant women in preeclampsia group was [(9. 51 ± 2. 82)mg/L], which was lower than that in healthy control group [(11. 19 ± 3. 04)mg/L], with significant statistically difference (t=8. 85, P=0. 01); the vitamin E concentration in late pregnant women in pre-eclampsia group was [(13. 52 ± 4. 40)mg/L], which was lower than that in healthy control group [(16. 20 ± 5. 00)mg/L], with significant statistically difference (t=10. 9, P=0. 01). ⑶Logistic regression anal-ysis showed that there was a correlation between vitamin E concentration in early pregnancy and late preg-nancy and preeclampsia. Conclusions There was no significant correlation between vitamin A concentra-tion and preeclampsia. Vitamin E may be involved in the pathogenesis of pre-eclampsia.
