2.Effect of human cementum extract prepared by guanidine on the attachment of gingival fibroblasts and osteoblasts

Objective: To study the bioactivity of human cementum extract . Methods: Cementum was harvested from freshly extracted heatlhy teeth and cementum extract was prepared by guanidine. Human gingival fibroblast and MC3T3 E1 osteoblast were cultured. The cells were incubated in DMEM with cementum extract concentration 0, 2.5, 5, 10 or 20 ?g/ml for an hour respectively, the cell attachment rate was measured by cell counting. The attachment rate of the cells in different incubation time(30 min,60 min, 90 min, 120 min) in DMEM containing 10 ?g/ml cementum extract was also assayed.Results: The extract increased the cell attachment rate in a concentration and time dependent effect. The extract at the concentration of 10 ?g/ml and with exposuse time of 90 min gave the most effective increase of cell attachment. Conclusion: Human cementum extract prepared by guanidine contains some bioactive proteins that promote the attachment of gingival fibroblasts and osteoblasts.
3.Change of Surface Shape and Distribution of Fibronectin Receptors on Aspergillus fumigatus Conidia

Objective To observe the changes of the outer ce ll wall in resting phase and during sw elling process of conidia of Aspergillus fumigatus,and the distribution of fibronectin receptors on conidia surface.Methods After being harvested with 3.5~4.0?m steel filters,the conidia were resuspended in RPMI 1640+10%FBS medium.The specimens were observed by the scanning electron microscopy after incubation for 0,2,4,6hours a t37℃respectively.The same amount of conidia was resuspendesd in PBS buffer,and put the suspension on 50?g /ml fibronectin coated microtitre plate,after incubation for 3hrs at 37℃,t he conidia were washed three times wi th PBS.Then rabbit antifibronectin antibody and fluorescein isothiocyanate(FITC)conjugated goat anti-rabbit immunoglob-ulin antibody were added,respectiv ely.After digesting with trypsin,t he conidia were washed again with PBS and examined by immuno-confocal microscopy.Results Acharacteristic echinulate pattern of protrusions was shown in the cell wall surface of resting coni dia.During swelling process,the ec hinulate layer of swollen conidia wa s gradually disintegrated,and almost complete ly shed 4hr later.Then,a smooth cell wall layer covered by some strands of fib-rillar material was shown.Binding o f fibronectin to the surface of conid ia was revealed by immuno-confocal m i-croscopy.FITC was homogeneously ad hered to the protrusions on the conid ial outer cell wall layer.Conclusion In resting phase the conidia of A.fumigatus express high levels of fibronectin r eceptors on their surface,they are h o-mogeneously distributed over the ex ternal echinulate layer of the conid ia,and shed gradually during the swe lling process.
4.Progress of anti-tumor mechanism of arsenic

The arsenic compounds including arsenic trioxide and arsenic sulfide have played a crucial role in the clinical treatment of hematologic malignancies and solid tumors. Arsenic agents can induce tumor cells differentiation, apoptosis and autophagy, eliminate leukemia-initiating cells, and directly bind to the target proteins. This paper reviews the mechanism progress of arsenic agents in various tumors to further understand the intricate anti-tumor mechanisms of arsenic agents and to expand its therapeutic spectrum.

6.Determination of Atractylenolide Ⅰ in Xiangshayangwei Pills by RP-HPLC

Objective: To develop a method to determine the content of a atractylenolideⅠ in Xiangshayangwei Pills. Methods: Reverse Phase HPLC was used to determine the content of atractylenolideⅠ in Xiangshayangwei Pills. The separation was performed on YWG ODS column with methol water (60∶40) as a mobile phase and the wavelength of UV detector was 220nm. Results: The linearity of this method was well with the average recovery of 99.54%. Conclusion: The method is simple, reliable, and sensitive. It also shows good separating degree. It can be used in quality controll of Xiangshayangwei Pills.
7.The effect of palate repair on middle ear function

Objective: To study the effect of palate repair operation on middle ear function. Methods: Cleft palate was repaired with double-opposing Z-plasty(DOZ) in 20 cases and with routin method (RM ) in 23 cases. Secretory otitismedia (SOM), high negative pressure (HNP), stapedial reflex(SR) and low acoustic compliance (LAC) were determined 3d before and 6 months after operation respectively. Results: Of the total 86 ears, before operation the persentages of SOM, HNP, SR and LAC were 61. 63, 75. 58, 29. 07 and 65. 15; after operation those were 31.39, 43.02, 66.28 and 33.72, respectively. Of the 40 ears of the cases treated with DOZ the persentages of those were 20.00, 27.50, 72.50 and 25.00) while of the 46 ears of the cases treated with RM those were 41.30, 56.52, 60.87 and 41.30 respectivellly. Conclusion: Middle ear function can be improved by palate repairs surgery, better results may be achieved with double opposing Z-plasty technique.
8.Visual acuity test for children under 5 years of age

The visual acuity testa were performed in 8219 children(16 438 eyes)under 5 years of age with sound and light toys,electric torch and a retinoscope.The general ocular examination and mydriatic optometry were also performed for the ametropia.In 8219 children 61 were found to have dominant strabismus,including 59 cases of concomitant strabismus and 2 paralytic strabismus.AbnormaJ refraction was detected in 404 children(739 eye)accounting for 4.92%,including 68 cases of anisometropia (0.83%).In 16 438 eyes 621(3.78%)were moderate or hyper-presbyopia;53(0.32%)were myopia;65 (0.40%)were mixed astigmatism.The vision acuity test described above is a simple and convenient method suitable for children under 5 who are not able to read the conventional test chart.
9.Distribution and Pharmacokinetics of Curcumin in Tumor-bearing Mice

Objective To study the distribution and pharmacokinetics of curcumin in tumor-bearing mice.Methods Curcumin injection(100 mg/kg)was injected to the tumor-bearing mice through tail vein.The liver,kidney,lung,heart,parenchymal tumor and ascitic fluid were acquired to abstract curcumin at 20,40,100 minutes after injection respectively.A HPLC method was established for determination of curcumin.Results Curcumin contents in the liver,kidney,lung,heart,parenchymal tumor and ascitic fluid were 6.40,0.98,0.13,0.06,0.05 and 0.02 ? g/g after injection of 20 minutes respectively.While after 40 minutes,the contents reduced to 0.20,0.06 and 0.02 ? g/g in the liver,kidney and lung respectively,and could not been detected in other tissues.After 100 minutes,curcumin was only detectable in the liver(0.04 ? g/g).Conclusion Curcumin distribution is focused in the liver of tumor-bearing mice,while the focal concentration is lower .Metabolism of curcumin in vivo is fast.
10.Pharmacokinetics of curcumin solid dispersion in mice

Aim To study the pharmacokinetics of curcumin-pvp solid dispersion ig administration in mice in comparison with the free curcumin suspension.Methods Drugs were administered at a dose of 300 mg?kg-1 via ig.The plasma concentration of curcumin was determined by HPLC,the pharmacokinetics were calculated by DAS ver1.0 program.Results The curcumin pharmacokinetics conforms to a two-compartment open model after a single ig dose of curcumin solid dispersion in mice.The parameters were as follows:T12? and T12? were 16.4 and 266.1 min,respectively.AUC was 89.6 mg?L-1?min-1,Vd was 763.9 L,V1 was 51.0 L,and CL was 1.99 L?min-1.The absorption rate of curcumin solid dispersion was 6.75 times as much as curcumin suspension.Conclusion The curcumin solid dispersion improves the absorption of curcumin in vivo in mice evidently.
