1.Adenosine promotes bFGF protein and bFGF mRNA expression in human umbilical vein endothelial cells in vitro

AIM: To investigate the influence of adenosine on human umbilical vein endothelial cells (HUVEC) bFGF protein production and bFGF mRNA expression. METHODS: Immunohistochemistry staining was performed to detect bFGF protein. RT-PCR was performed to detect bFGF mRNA expression. RESULTS: Immunohistochemistry study demonstrated that there was only a small amount of bFGF positive cells and the color was weak in control group (without adenosine). In groups treated with 10~-4 mol/L and 10~-6 mol/L adenosine, bFGF protein was significantly higher than that in control group (P0.05). RT-PCR showed that in 10~-4 mol/L and 10~-6 mol/L adenosine groups, bFGF mRNA expression was higher than that in control group (P0.05). CONCLUSION: Adenosine may promote HUVEC proliferation and angiogenesis partly through inducing bFGF expression.
2.Comparison of total hip arthroplasty and hemiarthroplasty in treating fracture of femoral neck in elderly patients

Objective To compare the operation and clinical effects of total hip arthroplasty and hemiarthroplasty in the treatment of fracture of femoral neck in elderly patients.Methods 58 elderly patients( ≥85 ) with femoral neck fracture were divided into observation group(n=39) and control group(n=19).The observation group was treated by hemiarthroplasty and the control group was treated by total hip arthroplasty.The operation time,blood loss,hospital stay,postoperative hip functional score and complications were observed and compared between the two groups.Results The operation time and blood loss of the observation group were(71.6 ±9.3)min and (266.8±113.5) ml respectively,which were significantly less than those of the control group ( t=7.89,3.43,all P < 0.05 ).Therer was no significant difference of the hospital stay,postoperative hip functional score and incidence rate of complications between the two groups.Conclusion Hemiarthroplasty was an ideal rmethod in treating fracture of femoral neck in elderly patients,which was effective and simple.
4.Calcineurin signalling mechanisms in myocardial hypertrophy

Calcineurin dephosphorylates multiple serine residues near the N terminus of NFAT proteins enabling them to translocate from cytoplasm to nucleus, where they activate a subset of hypertrophic response genes. Transgenic mice over-expressing a constitutively active form of calcineurin or NFAT3, developed obviously hypertrophy and heart failure or sudden death proving its pathogenic role. Here we used literatures on MEDLINE (2000-2011), systematically reviewed the new development of calcineurin signaling pathway in myocardial hypertrophy.
5.Study on the changes of cardiac reserve function during normal vaginal delivery

Objective To investigate the trend of cardiac reserve function during the normal labor.Methods Sixty-three cases were chosen randomly from hospitalized maternal women in the First Affiliated Hospital of Chongqing Medical University from 2010 June to December ( six months).The digital technique of heart sound signal processing was used to analysize cardiac reserve function parameters including the heart rate ( HR),the ratio of the amplitude of the first heart sound to the second heart sound and the ratio of diastolic to systolic duration (D/S) of pregnant women.Results ( 1 ) Comparisons of cardiac reserve function between uterine contractions and relaxations during labor:①Latent phase of labor ( cervix dilation < 3 cm ):HR was ( 87.3 ± 14.0 ) beats/min in uterine contractions and ( 82.8 ± 12.5 )beats/min in uterine relaxations,the ratio of D/S was 1.14 ±0.27 in uterine contractions and 1.21 ±0.22 in uterine relaxations,the comparisons of the above two were statistically significant,P <0.05 ; But the ratio of S1/S2 was 2.19 ±0.82 in uterine contractions and 2.28 ±0.81 in ueterine relaxations,the comparison was not statistically significant,P > 0.05.② During early active stage of labor ( cervix dilation 3-6 cm):HR was (89.3 ± 15.4) beats/min in uterine contractions and (83.1 ± 13.5) beats/min in uterine relaxations,the ratio of D/S was 1.09 ± 0.30 in uterine contractions and 1.20 ± 0.27 in uterine relaxations,the comparisons of the above two were statistically significant,( P <0.05 ) ;But the ratio of S1/S2 was 2.42 ± 1.08 in uterine contractions and 2.29 ±0.83 in ueterine relaxations,the comparison was not statistically significant ( P >0.05 ); ③During late active stage of labor (cervix dilation 6-10 cm),HR was (95.4 ± 18.7 ) beats/min in uterine contractions and (86.2 ± 15.6) beats/min in uterine relaxations,the ratio of D/S was 1.01 ±0.25 in uterine contractions and 1.18 ± 0.25 in uterine relaxations,the comparisons of the above two were statistically significant,( P < 0.05 ) ; But the ratio of S1/S2 was 2.61 ± 1.26 in uterine contractions and 2.67 ± 1.19 in ueterine relaxations,the comparison was not statistically significant ( P > 0.05 ).④ The second stage of labor (cervical dilation ≥ 10 cm ):HR was (109.4 ± 19.7 ) beats/min in uterine contractions and (93.5 ± 16.7 ) beats/min in uterine relaxations,the ratio of D/S was 0.89 ± 0.23 in uterine contractions and 1.14 ±0.26 in uterine relaxations,the ratio of S1/S2 was 3.66 ± 1.37 in uterine contractions and (2.81 ± 1.07 ) in uterine relaxations,the comparisons of all were statistically significant (P<0.05).(2)Comparison of cardiac reserve function in uterine relaxations of each stage of labor:①Maternal heart rate gradually increased from latent stage of labor to the second stage of labor,and decreased postpartum,the comparison was statistically significant ( P < 0.05 ) ;② The ratio of S1/S2 of maternal gradually increased from latent stage of labor to the second stage of labor,and decreased postpartum,the comparison was statistically significant( P <0.05 ) ;③ The ratio of D/S gradually decreased from latency to the second stage of labor,and increased postpartum,the comparison was statistically significant( P <0.05 ).(3) Comparison of cardiac reserve function in uterine contractions of each stage of labor:① Maternal heart rate gradually decreased from latent stage of labor to the second stage of labor,the comparison was statistically significant( P < 0.05 ) ;② The ratio of S1/S2 of maternal gradually increased from latent stage of labor to the second stage of labor,the comparison was statistically significant( P < 0.05 ) ; ③ The ratio of D/S gradually decreased from latency to the second stage of labor,the comparison was statistically significant (P <0.05).Conclusions The maternal cardiac reserve function decreased in uterine contractions than relaxation during labor; With the progress of labor,the maternal cardiac reserve function declined,especially in the second stage of labor,and recovered in postpartum stage.

7.Malignant inferior vena cava obstruction involving right atrium: palliative treatment with self expandable metallic stent

Objective To investigate the safety and efficacy of placement of self expandable metallic stent within right atrium and inferior vena cava (ICV) in patients with malignant ICV obstruction involving right atrium. Methods There were 5 male patients with advanced hepatocellular carcinoma, aged from 42 to 65 years (mean 56 3 years). The malignancies invaded right atrium and ICV simultaneously, and caused obstruction of ICV. These patients presented symptoms and signs of obstruction of ICV such as hepatomegaly, ascites, edema of lower extremities. “Z” type stainless steel stents (7 5~10.0 cm in length and 2 5 cm in diameter) were selected for these patients. Part (1.0~3.0 cm in length ) of stent was placed in right atrium and the rest was in ICV. Results All of stents were placed successfully and obstructed ICV reopened. The symptoms of obstruction of ICV relieved or disappeared. There were no recurrence of symptoms of obstruction of ICV and any cardiac complication related to placement of stents in follow up period (67~188 days). Conclusion Placement of self expandable metallic stent within right atrium and ICV in treatment of patients with malignant ICV obstruction involving right atrium is one of the safe and effective methods.
8.The biomechanical study and clinical application of an improved tension band wire for the treatment of supracondylar fractures of the humerus

In the bending test, in order to give rise to the fragmental displacement of 2.30? induced by bending load, the bending force of 3.3Nm was needed in ITBW fixation, and was 1.9 Nm greater than CW fixation, so stiffness resistant to bending of ITBW was 2.3 times of that of CW. The patients were followed up from 6 to 78 months, with an average of 38 months. There were no complications of nerve and vessels injury, infection and wire or nails loosening. Varus elbow happened in 2 cases, the mean varus angel was less than 5?. 2 cases and All the cases treated with ITBW healed uneventfully with good functions, there were excellent 32 cases and good 12 cases according to the Flynn evaluation system. Conclusion The fixation effect of ITBW is much better than that of CW. The stability provided by ITBW makes external fixation unnecessary and allows for early mobilization of the elbow. The ITBW technique is in satisfactory agreement with biomechanical principle. It is valuable for widely clinical application.
9.Evaluation of the effect of KAI1 gene and ME491 gene suppressing metastasis on pancreatic cancer.

Objective To study the effect of suppressing metastasis between KAI1 and ME491 on pancreatic cancer.Methods Normal pancreases and pancreatic cancers were analyzed using Northern blot,in situ hybridization and immunohistochemistry.Results The expression of KAI1and ME491 was higher in pancreatic cancer than that in normal pancreases,KAI1 expression was higher in pancreatic cancer without metastasis than that in pancreatic cancer with metastasis(P0.05).Conclusion The gene of KAI1 may be involved in the metastasis of pancreatic cancer,ME491 gene may have no effect on the metastasis of pancreatic cancer.
10.Preliminary study of microsatellite instability and loss of heterozygosity in thymic squamous cell carcinoma

Objective To investigate the frequency of micresatellite instability (MSI) and loss of heterozygosity (LOH) and select sensitive loci for studying microsatellite DNA imbalance in thymic squamous cell carcinoma. Methods 5 microsat-ellitc polymorphism markers and extrated DNA were selected from 9 specmiens of paired thymie squamous cell carcinoma/nor-real tissues. MSI and LOH in the specmiens of thymic carcinoma and relevant pericancerou tissues were detected by polymerase chain reaction (PCR) followed by 6% polyacrylamide gel electrophoresis(PAGE) with silver staining. Results MSI or LOH was detected in 9 thymic carcinoma tissues. The frequency of MSI or LOH was 66.7% (6/9) at loci of D6S1708, 33.3% (3/9) at TP53, 33.3% (3/9)at DM, 33.3% (3/9)at D11S988 and 0% (0/9)at D8S136, LOH at D6S1708 (5/6) was a common genetic alteration. DI1S988 had only LOH alteration. Conclusion D6S1708, TP53, DM, and D11S988 are sensi-tive loci for studying microsatellite DNA imbalance in thymic squamous cell carcinoma. Microsatellite DNA imbalance may play a certain role in occurrence and development of thymic squamous cell carcinoma, and the relationship between MSi or LOH.The linicopathological features of thymic squamous cell carcinoma needs further investigation.
