1.Lack effect of metabotropic glutamate receptors on neurotoxicit y of PC12 cells induced by 6-OHDA

AIM: To explore if metabotropic glutamate receptors (mGluRs) i nduces neuroprotection against 6-hydroxydopamine (6-OHDA) neurotoxicity in PC1 2 cells. METHODS: The alteration of the glutamate in extracellul ar fluid of PC12 cells was detected by high performance liquid chromatography (H PLC) with a fluorescent detector. The cytotoxic activity of PC12 cells was assay ed by means of MTT colorimetric method. RESULTS: 6-OHDA dose-d ependently increased glutamate release and decreased cell activity of PC12 cells , and the mGluRs ligands had no significant effect on that. CONCLUSION: The mGluR3 ligands has no protective effect on neurotoxicity of PC12 cel ls induced by 6-OHDA.
2.The analysis of the efficacy of patients with allergic rhinitis treated with specific immunotherapy.

OBJECTIVE:
To evaluate the efficacy and safety of standardized subcutaneous immunotherapy for dust mite in patients with allergic rhinitis.
METHOD:
Using self-control methods, 35 cases with allergic rhinitis were treated with specific immunotherapy for 2 years. Symptom score and visual analogue scale (VAS) score were observed before treatment and 1 years, 2 years after treatment respectively.
RESULT:
The symptoms, signs and VAS score of the 35 patients who were treated with specific immunotherapy after 1 year were significantly reduced than that before treatment, the differences were statistically significant (P < 0.05). The symptoms, signs and VAS score of the patients who completed 2 years' treatment, compared with that of pretreatment and 1 year treatment were significantly reduced, the differences were statistically significant (P < 0.05).
CONCLUSION
Standardized immunotherapy for dust mite is a safe, effective method for patients with perennial allergic rhinitis, which can be used as a routine treatment for allergic rhinitis. To further improve the therapeutic effect, immunotherapy should be continued for at least 2 years.

3.The Screen for the Upper GI Diseases in Residents of Jiangsu Dongtai

Objective To know the situation of upper GI diseases among the residents in Dong tai city. Methods With three levels examination methods of Occult Blood Bead Detector,Gastroscope and pathological examination,residents aged over 30 yrs in Dongtai were screened during Aug 2005 to Sept 2006. Results 70 809 residents,about 6.13% of whole city population,were taken the screen with occult blood bead detector. There were about 28 392 people positive; the positive rate was 40.10%. Among the positive people,there were 8 121 people taking gastroscope inspection,and while 2 294 of them also took the pathological examination. The gastroscope results showed that 4 636 people suffered from chronic superficial gastritis,1 431 rotten gastritis,565 esophagus gastritis,339 chronic & atrophic gastritis,24 suspicious cancers,53 stomach cancer,44 esophagus cancer. Of these 97 cases with cancers diagnosed by gastroscope,87 cases were confirmed by pathological examination,the false positive rate of gastroscope examination was about 10.31%; Among 2 197 of the non-cancer subjects diagnosed by gastroscope,there were 25 cases with cancer diagnosed by pathological method,the false negative rate of gastroscope was 1.14%. For those 112 cancer cases diagnosed by pathology,there were 83 cases (74.11%) who were in the early or middle phase of cancer. Conclusion The prevalence of upper GI diseases in Dongtai City was higher. It should strengthen the education and the improvement of the health situation.
4.Effectiveness and Safety of Oral Administration of Type Ⅱ Collagen on Patients with Active Rheumatoid Arthritis

Objective To assess the effectiveness and safety of oral administration of type Ⅱ collagen(C Ⅱ) versus placebo in patients with active rheumatoid arthritis(RA).Methods Seventy patients with active RA were randomly divided into receive placebo(n=35) or bovine CⅡ(n=35) at 0 1mg/day for 12 weeks,in a double-blind study.The cumulative response rates were analyzed by utilizing the American College of Rheumatology(ACR) criteria for improvement in RA,and the requirement for ≥50% improvement of outcome measures in ACR criteria.Results After the 12-week course of treatment,much more outcome was significantly improved in C Ⅱ group than in placebo group.The rates of improvement and remarkable improvement in CⅡ group were increased significantly,as compared with placebo group(54 55% vs 12 12% and 18 18% vs 3 13%,respectively,P0 05).Conclusions Oral administration of CⅡ in treating RA was significantly effective and has slight side effects.Further investigation in the field is worthwhile.
5.Effects of alendronate on trabecular bone structural properties

Objective To evaluate alendronate with regards to its effect s on trabecular bone structural properties. Methods Alendronate was administered at a daily p.o. dose of 0.5 mg?kg-1?d-1 over a 12 weeks period in Hound do gs (n=8 + 8 for control and treated group), the structural indices of the lumba r vertebral (L 1 and L 2) trabecular bone were assessed directly from 3 D image s. Results Treatment with alendronate increased bone volume fraction by 9.5% a nd 7.7% respectively, in the L 1 and L 2. Trabecular thickness significantly in creased after alendronate treatment, whereas trabecular separation remained cons tant. The degree of anisotropy for the alendronate treated group was decreased compared with that of the control group. Bone surface to volume ratio declined s ignificantly in the alendronate treated group, whereas alendronate induced a hi gher value of bone surface density. Conclusion Alendronate increased the struct ural properties of canine trabecular bone after short-term treatment at a dose of 0.5 mg?kg-1?d-1. Thus, alendronate could increase the structural properti es of healthy trabecular bone in a short-term period.
6.Collagen synthesis in distraction osteogenesis in alveolar cleft

Objective: To observe collagen synthesis in the different phase of distraction osteogenesis(DO) in alveolar cleft.Methods:8 dogs were used to make experimental model of alveolar cleft and underwent the closure of the cleft by distraction osteogenesis.The animals were sacrificed on day 0, 14, 28 and 63 after completion of DO respectively. The samples were examined by histological observation and the expression of typeⅠ,Ⅱ,Ⅲ collagen was studied by immunohistochemistry staining. Results:On day 0, a lot of type Ⅲ collagen fibers were observed in the distraction area. On day 14, there was trabecula bone, collagen Ⅰexpression was stronger than collagen Ⅲ. On day 28, trabecula bone developed into network-like structure with type Ⅰ collagen expression. On day 63,lamellated bone with type Ⅰ collage expression filled the distraction area. No obvious expression of collagen Ⅱ was observed.Conclusions:The formation of new bone is mainly intramembrane osteogenesis along the distraction vector.
7.The change of intracelluar Ca~(2+) level regulated by endothelial protein-activated by acetylcholine in cultured bovine aorta endothelia cell

AIM To examine the change of intracelluar Ca 2+ level regulated by EPA in bovine aorta endothelium of different stages of culture and determine the time that cultured endothelium loses functional EPA. METHODS As fluorescence probe, Fluo 3 and Fura 2 were used in assaying [Ca 2+ ] i in endothelium by confocal microscopy and fluorophotometry. RESULTS Ach could activate EPA and elicite the [Ca 2+ ] i change of endothelium in certain periods of culture, especially in primary stage of cul ture. The rises of [Ca 2+ ] i exhibited as oscillation characterized by iso timing and immediate tolerance. The functional EPA was lost in passage 13. CONCLUSION EPA can be activated by Ach, which mediated the rise of intracelluar Ca 2+ in bovine aorta endothelia cell. This character of EPA is lost during passaging.
9.Time-invariant CTA assessment of collateral circulation of patients with acute ischemic stroke

Objective To explore the value of time-invariant CTA in assessing collateral circulation of patients with acute ischemic stroke and assisting clinicians in predicting clinical outcomes.Methods The score of collateral circulation was compared between single-phase and time-invariant CTA.NIHSS score was calculated at admission and two weeks after admission.A 50% or greater decrease in NIHSS score over two weeks was considered as major neurologic improvement,which showed good clinical outcome;otherwise,it indicated bad outcome.The predictive ability of time-invariant CTA for clinical outcomes was assessed based on ROC curves.Results Compared with single-phase CTA,more collateral vessels could be viewed on time-invariant CTA.The average score of collateral circulation on time-invariant and single-phase CTA was 1.50±0.69 and 1.15±0.49 respectively (P=0.006<0.05 ).Time-invariant CTA had the moderate predictive ability for clinical outcomes in patients with acute ischemic stroke (AUC=0.810;P=0.032<0.05). Conclusion The time-invariant CTA showed potential value in assessing collateral circulation of patients with acute ischemic stroke and assisting clinicians in predicting clinical outcomes.
10.Simultaneous measurement of magnesium isoglycyrrhizinate and glycyrrhetic acid in plasma of dog by HPLC

AIM: To quantify magnesium isoglycyrrhizinate(MGL) and glycyrrhetic acid in the plasma of dog by develop a simple, rapid, sensitive high-performance liquid chromatography (HPLC-UV) method. METHODS: HPLC-UV methods with wavelength 252 nm were used for the quantitation of MGL and GA in plasma. The concentration of MGL and GA were assayed on a Kromasil ODS-1 C18 column with the column temperature 25 ℃. The mobile phase was a gradient system with 0.1 % diethylamine in water (pH 4.60 ) and acetonitrile at a flow rate of 1.0 ml?min~ -1 . RESULTS: There was a good linear response range of 0.2 -2.5 mg?L~ -1 and 2.5 -100 mg?L~ -1 , while the limit of quantification was 0.2 mg?ml~ -1 . The relative recovery rate of MGL was 94.3 %-101.9 %,GA 96.4 %-101.9 % (n=5);the absolute recovery rate was MGL 78.7 %-87.0 %, GA 77.5 %-87.7 % (n=5). The intra-day and inter-day variations were all less than 15% (n=5). The plasma samples preserved in refrigerator is stable at -20 ℃ for 14 days, freeze thawing 3 times and at room temperature for 10 h without degradation. CONCLUSION: This method is shown to be specific, sensitive, reliable and suitable for the quantitative determination of magnesium isoglycyrrhizinate following oral administration in biological sample.
