2.A Clinical Study on Conservative Treatment of Incomplete Pharmaceutical Abortion

Objective To study the clinical effects of conservative treatment of incomplete pharmaceutical abortion. Methods 149 cases of incomplete pharmaceutical abortion were randomly divided into two groups. Group A(86 cases) was intramuscularly given progesterone or orally medroxyprogesterone. Group B (63 cases)was given methotrexate intramuscularly or orally. The therapeutic effects of two groups were observed. Results The successful rate in group A and group B was 79 07%(68/86) and 96 82%(61/63). Objective To investigate the effect of rh-BNP combined with sodium nitroprusside in acute decompensated heart failure patients with loss of blood flow dynamics, the effects on heart function and circulating factor levels.MethodsSelect Wenzhou Institute of Physiology from February 2014 to January 2016 admitted during the period of 120 cases of acute decompensated heart failure patients, according to random number table method to all of the patients were randomly divided into two groups, control group and test group with the control group patients were given conventional treatment of heart failure treatment, while the experimental group patients give tome sodium nitrate joint treatment with recombinant human brain natriuretic peptide, compare two groups of patients 12 h level of circulating factors, cardiac function and hemodynamics.ResultsTwo groups of patients after the hemodynamics after 24 h treatment was significantly lower than that after the treatment of 12h(P<0.05), the control group of patients after treatment of 12h and 24h after pulmonary capillary wedge pressure and right atrial pressure and central venous pressure index were significantly higher than those in the test group (P<0.05) for the treatment of 24h.After the control group of patients with left ventricular GSRa, GSRe, GSRs, ROTR, ROT, GCS, GLS levels were significantly lower than those in the test group (P<0.05).After treatment of 12h patients in the control group were hsCRP, ST2, IL-6 and blood The levels of cTn I and NT-proBNP were significantly higher than those in the experimental group (P<0.05).ConclusionRh-BNP combined with sodium nitroprusside in patients with acute decompensated heart failure treatment can effectively optimize the circulation of patients with factor level and heart function and hemodynamics, it is worth to be popularized in clinical use.
4.Clinical Observations on Acupoint Selection at Distant Places and Local Tenderness Points for Treatment of Migraine Without Aura

Objective To investigate the efficacy of acupoint selection at distant places and local tenderness points in treating migraine without aura.Method Sixty patients were randomly allocated, using a random number table, to two groups, 30 cases each. The treatment group received acupuncture at distant points selected along meridians and local tenderness points and the control group, acupuncture at conventionally selected points. The therapeutic effects were evaluated using the visual analogue scale (VAS) and the short-form McGill Pain Questionnaire (SF-MPQ).Result After treatment, the VAS pain score was lower in the treatment group than in the control group; there was a statistically significant difference (P<0.05). The total efficacy rate was 93.3% in the treatment group and 73.3% in the control group; there was a statistically significant difference between the two groups (P<0.05).Conclusion Both acupuncture at distant points selected along meridians and local tenderness points and acupuncture at conventionally selected points have a certain therapeutic effect on migraine, but the former produces a better effect than the later and can markedly relieve the pain and improve the quality of life in the patients. The outcome of this study provides a certain reference value for clinical treatment of this disease.
5.Internal mammary arterial infusion therapy for chest diseases

Objective To study the value of interventional therapy for chest diseases via internal mammary artery (IMA) and the features of internal mammary artery angiography. Methods 31 cases of different chest diseases were undertaken the angiography and interventional therapy through IMA. Results The lesions of 31 cases were supplied by internal mammary artery partly or totally. The good therapeutic effectiveness was achieved in 20 cases with pulmonary carcinoma. The masses and the enlarged lymph nodes were shrinked obviously in 4 cases of advanced breast carcinoma and one of them was cured with operation after internal mammary arterial infusion. One case of low invasive thymoma was cured by internal mammary arterial infusion combined with resection. The bleeding was stopped absolutely after IMA embolization in 6 cases of hemoptysis (bronchiectasis in 2 cases, pulmonary tuberculosis in 4 cases). Conclusions The interventional therapy via IMA is very important and necessary when the mass in the chest is supplied by IMA.
6.The correlation between cortactin differential expression and metastasis of colorectal cancer

ObjectiveTo explore the correlation between cortactin differential expression and metastasis of colorectal cancer by testing the expression of cortactin in cell sublines with different metastatic ability.MethodHuman colon cancer cell line SW1116, and sublines of fifth subcutaneous ( SW1116_5), first, fifth generation (CRCLM1 、CRCLM3 ) characteristic of different liver metastasis were implanted subcutaneously in BALB/c mouse.Tumors were transplanted into the colon, and liver metastasis was observed, Western blotting and real time-PCR was used to detect the varience of cortactin. Transwell assay was applied to evaluate four cell lines migration and invasion ability. Immunohistochemistry was used to analyze the relationship of cortactin and clinicopathological characterizing in 86 cases of colorectal cancer.ResultCRCLM3( 100% )has higher ability of metastasis than that of CRCLM1 ( 89% ), SW1116(40% )and SW1116_5 (44%) respectively. Expression of cortactin in the SW1116, SW1116_5, CRCLM1,CRCLM3 was gradually increased. There were significant differences between four cell lines by comparison between each others ( P < 0.05 ). Immunohistochemical expression of cortactin in 86 cases was positive in 57 cases(66% ), and negative in 29 cases (34%).ConclusionsDifferent expression of cortactin with colorectal cancer metastasis and clinical stage was positively correlated. Cortactin is a potential indicator for clinical staging and tumor metastasis of colorectal cancer.
7.Duction of immunoincompetence of T cells to oxidized-low density lipoprotein in vitro by CTLA4Ig

Objective To induce the immunoincompetence of T cells to oxidized-low density lipoprotein(ox-LDL)in vitro,in order to prevent immune injuries in atherosclerosis(AS)and to find new strategies to prevent AS.Methods Monocytes were separated from peripheral blood to induce dendritic cells (DC).DCs were treated with LPS(30 ng/m1),ox-LDL(10μg/m1)and LDL(10μg/m1)for 48 h,respectively.Then DCs were mixed with allogeneic T lymphocytes to earry out mixed lymphoeytes reaction (MLR).CTLA4Ig in different concentrations was added into the MLR of ox-LDL group.MTr method was used to assay the proliferation of T cells.The CD25 expression and apoptosis of T cells in MLR were tested by flow cytometry.And the excretion of IL-2,IFN-γ and IL-4 were assayed by ELISPOT method.Results SI of the ox-LDL group was higher than that of the LDL group significantly(DC:T=1:5,1.6717±0.3152vs 1.4250±0.2874.P<0.05;DC:T=1:10,1.5458±0.2748 vs 1.3352±0.2991,P<0.05),and CTLA4Ig inhibited the SI of the ox-LDL group in dose-dependence(CTLA4Ig 1.25μg/ml,0.96±0.30 vs μg/ml 1.29±0.28 vs 1.64±0.33 P<0.05).CTLA4Ig caused the decrease of CD25 expression(CTLA4Ig 1.25 μg/ml,11.26±0.58 vs 14.25±1.02,P<0.05;CTLA4Ig 10μg/rnl 8.42±0.45,P<0.01)and induced apoptosis of T cells in MLR(CTLA4Ig 1.25μg/ml,12.54±3.69 vs 6.09 4-2.24,P<0.05;CTLA4Ig 10μg/ml,26.87±5.06 VS 6.09±2.24,P<0.01).CTLA4Ig caused the decrease of ELISPOT counts of IL-2(CTLA4Ig 1.25μg/ml,386±42 VS 534±54,P<0.05;CTLA4Ig 10μg/rnl,230±27 VS 534±54,P<0.01)and IFN-γ(CTLA4Ig 1.25μg/ml,445±48 VS 672±46,P<0.05;CTLA4Ig 10μg/ml,193±39 VS 672±46,P<0.01),while that of IL-4 increased(CTLA4Ig 1.25μg/ml,401±32 VS 332±41,P<0.05;CTLA4Ig 10μg/ml,453±57 VS 332±41,P<0.05).Conclusion CTLA4Ig can induce T cens immunoin competence to ox-LDL in vitro by inhibiting T cells activation,inducing T cells apoptosis and TH 1/TH2 immune deviation.
8.Anti-proliferation and anti-angiogenesis of curcumin-K30 solid dispersion

Objective To evaluate the anti-proliferation and anti-angiogenesis effect of curcumin-K30 solid dispersion (Cur-K30) on tumors in vivo. Methods Growth inhibition rates of the tumor cells was measured with MTT method. Tumor inhibition was detected by tumors transplanted subcutaneously in mice treated with Cur-K30 [50, 100, and 200 mg/(kg·d)]. The expressions of CD34 and vascular endothelial growth factor (VEGF) were assessed by immunohistochemical study, and analyzed by Imageproplus software. Results Cur-K30 had inhibitory effect on different tumor cell lines in a dose dependent manner with IC50 values from 6.6 to 12.12 μg/mL. The in vivo study showed that the inhibitory rates of the 200 mg/(kg·d) Cur-K30 group on H22, B16, and SW480 were 43.2%, 53.1%, and 59.8%, respectively, which were all much higher than the inhibitory rates of curcumin suspension group with the same dose. Compared with the control group, the expression of CD34 and VEGF in SW480 tumors was down-regulated in the 200 mg/(kg·d) Cur-K30 group (P<0.01). Conclusion The proliferation inhibition of Cur-K30 is higher than curcumin in vivo, and the most significant effect is obtained in SW480 tumors transplanted subcutaneously in nude mice. Down-regulation of VEGF and decreased microvascular density may contribute to the anti-tumor effect of Cur-K30.
9.Correlation study between complement C3 level and degree of disease activity in patients with systemic lupus erythematosus

Objective To investigate the relationship between complement C3 level and the degree of disease activity in patients with systemic lupus erythematosus(SLE).Methods A total of 1 012 patients with SLE were enrolled in this study from January 2006 to December 2013 at department of rheumatology and immunology,the first affiliated hospital of Anhui medical university.Serum complement C3 level was detected by rate erythenephelometry assay.The relationship between reduction of complement C3 and SLE was analyzed.Results Serum complement C3 clearly decreased in 782 patients,accounted for 77.27%.There were significant differences concerning serum complement C3 level among different groups of disease activity (F =131.275,P<0.01).The low serum complement C3 level was correlated with the high degree of disease activity (r =-0.517,P <0.01).Patients with SLE had a severe disease activity (SLE disease activity index ≥ 15) when the level of serum complement C3 was less than 0.57 g/L.Serum complement C3 levels were positively correlated with serum complement C4 (r =0.845,P < 0.01),peripheral blood leukocyte count (r =0.115,P < 0.01),hemoglobin (r =0.069,P < 0.01),platelet count (r =0.177,P <0.01) and albumin level (r =0.091,P < 0.01).There was also a negative association of serum complement C3 with 24 h urinary protein (r =-0.228,P < 0.01).101 patients whose average level of serum complement C3 was(0.48 ±0.26)g/L,were treated with glucocorticoid pulse therapy as high degree of disease activity.Conclusions There is a close relationship between serum complement C3 level and the degree of disease activity in SLE.Serum complement C3 less than 0.57 g/L might be seen in SLE with severe disease activity.
10.in vitro Expansion and Differentiation of Cord Blood Hemotopoietic Cells

Objective: To explore the role of human hematopoietic growth factors (HGFs) in proliferation and differentiation of cord blood CD34+ cells. Methods: Human cord blood mononuclear cells (MNC) were cultured with different combinations of HGFs (including rhIL-1,rhIL-3, rhIL-6, rhG-CSF, rhGM-CSF and rhSCF). The expansion folds of MNC, the changes of cellular surface markers by FACS, and CFU-GM of hematopoietic cells were observed. Results: The total nucleated cells expanded by 44 folds after cultured with 6 cytokines for 20 days. The number of CFU-GM increased by 14 .74 folds after cultured in liquid for 8 days, but decreased heavily after 18 days. The total number of CD34+ cells increased by 127. 79 ~ 196 .40 folds at 6 to 8 day, but decreased to 101.51 folds at 18 day. Conclusion: Human hematopoietic growth factors can increase the expansion of cord blood CD34+ cells and CFU-GM significantly.
