1.Effect of DEC1 gene over-expression on proliferation and invasion abili-ties of human esophageal cancer ECA109 cells

[ ABSTRACT] AIM:To investigate the effect of DEC1 gene over-expression on the proliferation and invasion abili-ties of human esophageal cancer ECA109 cells.METHODS: ECA109 cells were transfected with plasmid pcDNA3.1 (-)/DEC1 (DEC1 group) or pcDNA3.1 (-) (vector group).The mRNA and protein levels of DEC1, cyclin D1 and MMP-9 were evaluated by real-time PCR and Western blot, respectively.The effects of DEC1 over-expression on the prolif-eration and invasion abilities of the ECA109 cells were evaluated by CCK-8 assay, colony formation assay and Transwell test respectively.RESULTS:The DEC1 expression level in ECA109 cells in DEC1 group was significantly higher than that in vector group (P<0.01), but the levels of MMP9 and cyclin D1 expression were opposite (P<0.01).However, both the proliferation and invasion abilities of ECA109 cells in DEC1 groups decreased significantly as compared with those in vector group (P<0.05).CONCLUSION:The over-expression of DEC1 significantly inhibits the proliferation and invasion of ECA109 cells, which may be involved in the expression of cyclin D1 and MMP9.
2.A retrospective study of lymphonode reactive hyperplasia

Objective:To summarize the experience in clinical diagno sis of lymphonode reactive hyperplasia (LRH). Methonds: 39 cases with the final pathological diagnosis of lymphonode reactive hyperplasia were retrospectively reviewed.Results: 39 patients had th e incidence at the age of 7~61 years old and the medical history of 14 days ~8 years.The patients consulted doctors because of unilateral or bilateral painles s masses in the superior neck and low fever.32 of them underwent repeated antib iotics treatment.On admission 9 cases were diagnosed with lymphoma,6 tuberculos is,6 inflammation of lymph node and 18 were without identified diagnosis.The f inal pathological diagnoses of all the 39 cases was LRH.Antibiotic treatment wa s effective in the patients.Conclusion:LRH was usually mis diagnosed as other diseases.Swelling of lymph nodes in superior neck with low fe ver is the main symptom of LRH.
3.Vertebroplasty with polymethyl methacrylate bone cement repairs elderly osteoporotic vertebral compression fractures

BACKGROUND:Percutaneous vertebroplasty with bone cement injection has the advantages of minimal invasion, short time, effectively restoring vertebral body height, preventing further colapse of the vertebral body and obvious analgesic effect, which has became an effective method for the treatment of elderly osteoporotic compression fractures. OBJECTIVE:To observe the therapeutic effect of percutaneous vertebroplasty with bone cement injection on elderly osteoporotic compression fractures. METHODS:Sixty-two patients with osteoporotic thoracolumbar vertebral compression fracture, including 22 males and 40 females, aged 55-92 years, involving 86 vertebrae, were included and subjected to percutaneous vertebroplasty with polymethyl methacrylate bone cement injection under C-arm X-ray fluoroscopy. During the postoperative folow-up of 12 to 36 months, visual analogue scale scores, Cobb angle and Oswestry disability index scores were compared before and after the treatment. RESULTS AND CONCLUSION: At 12 to 36 months after treatment, there were 11 cases of complications, including 7 cases of bone cement leakage, 2 cases of adjacent vertebral fractures, 1 cases of bone cement tailing and 1 case of unsatisfactory pain relief. In the final folow-up, Cobb angle, visual analogue scale scores, Oswestry disability index scores were significantly improved compared with those before treatment (P < 0.05).These results demonstrate that percutaneous vertebroplasty with polymethyl methacrylate bone cement injection in the treatment of elderly osteoporotic compression fractures can not only restore vertebral shape, reduce kyphosis, reconstruct spinal stability, but also significantly reduce the pain caused by fractures and improve the life quality of patients. The curative effects in short and medium term are positive.
4.Effect of blood pressure on perihematomal edema following intracerebral hemorrhage in spontaneously hypertensive rats

Objective To investigate the changes of hematoma and perihematomal edema in spontaneous hypertensive rats (SHRs),as well as the correlation between blood pressures and perihematomal edemaMethods A total of 24 6-month-old male SHRs were included.They were randomly divided into intracerebral hemorrhage 1-,3-,5-,and 7-day groups (n =6 in each group).Blood pressure was determined by tail-cuff sphygmomanometry.Collagenase Ⅳ was injected into caudate nucleus in order to induce a model of intracerebral hemorrhage.Magnetic resonance T2 weighted imaging was used to observe hematoma and perihematomal edema.Results On day 1 after modeling,the blood pressure was decreased significantly compared to that before modeling,and it was elevated gradually on day 3.Parallel profile test showed that the overall profile was parallel between the change rate of blood pressure and the change rate of perihematomal edema volume (F =2.820,P =0.063).The coincident profile test showed that both the overall profiles did not coincide (F =10.961,P =0.000).The change rate of systolic blood pressure was significantly positively correlated with the change rate of perihematomal edema volume (r =0.527,P =0.024) and the change rate of perihematomal edema volume (r =0.755,P =0.000) showed a significant positive correlation.Conclusions Brain edema appeared and expanded rapidly on the day of intracerebral hemorrhage in SHRs.It reached the peak on day 3,and then dissipated gradually.The blood pressure decreased significantly on day 1 after intracerebral hemorrhage,and it began to increase from day 3 till day 7.The changing trends of the blood pressure and perihematomal edema volume were parallel to each other,and the change rate of systolic blood pressure and the change rate of perihematomal edema volume showed a positive correlation.
5.MORPHOLOGICAL OBSERVATION IN VITRO MODEL OF BLOOD-TUMOR BARRIER OF GLIOMA

Objective To study the morphological characteristics of blood-tumor barrier(BTB)model of glioma in vitro. Methods After C 6 glioma cells/endothelia ECV 304 co-cultured mixed or in Transwell or on both sides of membrane of Transwell,the morphological characters of fenestra of endothelial cells,the junction between ECV 304 cells,the interface between C 6 cells and ECV 304 cells,and perivascular-end-feet of C 6 cells were observed under transmission electron microscope(TEM)and scanning electron microscope(SEM),and compared to BTB in 4 cases with human brain glioma. Results ECV 304 cells grown to confluence were not fenestrated,but with fomation of tight junction between ECV 304 cells after co-cultured with C 6 cells mixedly,in Transwell or on both sides of membrane of Transwell;It was not found that pseudopodia from C 6 cells as co-cultured in Transwell reaching into pore of Transwell;C 6 cells co-cultured on both sides of membrane of Transwell often sticked out to the ECV 304 cells,but with no pseudopodia from C 6 cells surrounded ECV 304 cells of penetrated the endothelia clefts.Perivascular-end-feet of C 6 cells were not integrant.These characters were similar to BTB in human brain glioma.Conclusion C 6 glioma cells/endothelia cell ECV 304 co-cultures on both sides of membrane of Transwell may simulate the morphological characters of BTB in vivo in some degree.;
6.A study of histopathological changes of small bowel allograft during acute rejection in rats

Objective To investigate the features and significance of pathologic changes in apoptosis of small bowel allograft during acute rejection in rats. Methods All 24 recipients were equally divided into four groups ; group A: nonoperative control; group B: allograft ; group C: isograft, group D: treatment control. The graft samples were harvested on day 3, 5, 7, 10 after transplantation, and subjected to histologic examination . Mucosal thickness, villous height and crypt depth were measured, and apoptotic cells of intestinal mucosa of grafts on day 3,5 and 7 after transplantation were examined. Results The mucosal structure was normal in group A; The degree of the inflammatory infiltrated cells ,intestinal mucosa cell apoptosis and structural injury of mucosa in group B were significantly severe compared with groups C and D. As the post-transplanted time increased, the number of musocal apoptotic cells and the degree of mucosal structural injury were significantly increased. The degree of mucosal structural injury in group C was milder than in group B. A few infiltrated cells and mild edema of mucosa occurred in group D , but no mucosal structural injury was found. Conclusions Inflammatory cell infiltration, mucosal epithelial cell apoptosis and mucosal structural damage are the main pathologic features of small bowel allograft during acute rejection. Dynamic observation of the pathologic changes and cell apoptosis of small bowel graft is of certain value in the diagnosis of acute rejection of small bowel graft and in assessment of the degree of small bowel injury.
7.The effect of the yang-warming and fluid retention-resolving method on AngⅡ, TNF-α and IL-6 expression in rats with chronic heart failure

Objective To observe the effect of the yang-warming and fluid retention-resolving mothed on neuroendocrine and inflammatory factors in chronic heart failure (CHF) rats. Methods A total of 50 healthy adult male SD rats were randomly divided into blank group (10), CHF group (40). In CHF group, the rat model of chronic heart failure was replicated by intraperitoneal injection of adriamycin (ADR). After the completion of the model making, doppler echocardiography was used to determine the cardiac function of rats. There rats were randomly divided into the model group, the yang-warming and fluid retention-resolving group, the Xuefu-Zhuyu group and the captopril group. After continuous administration for 28 d and 2 h after the last dose, the HE staining was used to observe the pathological morphological changes of myocardium in each group. The levels of AngⅡ, TNF-α and IL-6 in serum were by ELISA. Results Compared with model group, the content of serum AngⅡ(294.33 ± 25.45 pg/ml, 265.86 ± 27.80 pg/ml, 301.87 ± 41.67 pg/ml vs. 615.44 ± 22.31 pg/ml), TNF-α(117.15 ± 11.33 pg/ml, 114.02 ± 11.57 pg/ml, 133.35 ± 34.80 pg/ml vs. 185.72 ± 36.53 pg/ml), IL-6 (23.97 ± 4.77 pg/ml, 27.61 ± 7.01 pg/ml, 28.38 ± 5.78 pg/ml vs. 58.45 ± 6.36 pg/ml) in the yang-warming and fluid retention-resolving group, Xuefu-Zhuyu group and captopril group significantly decreased (P<0.01 or P<0.05). Compared with Xuefu-Zhuyu group, the content of serumAngⅡ (301.87 ± 41.67 pg/ml vs. 265.86 ± 27.80 pg/ml) in the captopril group significantly increased (P<0.05). Conclusions The yang-warming and fluid retention-resolving mothed can reduce early CHF rats through reducing the serum TNF-α, Ang and ILⅡ -6 content, which could be one of the mechanism of action of the treatment of CHF.
8.A neuritogenic compound from Tremella fuciformis.

OBJECTIVETo search for low-molecular-weight neuritogenic compounds from the traditional Chinese medicine (TCM).
METHODAn extract library of TCM was prepared. Targeted isolation guided by biological screening led to the discovery of compound 1, and its structure was elucidated by analysis of spectroscopic methods and comparison of spectroscopic data with these reported from the literature.
RESULTA neuritogenic compound 1, 3-O-beta-D-glucopyranosyl-22E, 24R-5alpha, 8alpha-epidioxyergosta-6, 22-diene, was isolated and identified from the methanol extract of T. fuciformis. This compound showed a significant neuritogenic activity against PC12 cells at 3 micromol x L(-1)).
CONCLUSIONMethonal extract of T. fuciformis and targeted compound 1 both showed significant neuritogenic activity against PC12 cells. These results suggested that the extract and compound 1 might be used to prevent and treat neurodegenerative disease such as Alzheimer's disease.

9.Effect of Taichi footwork assisted routine rehabilitation training on the influence of limb balance disorder of patients with hemiparesis after stroke in the recovery period

Objective:To evaluate the effect of Taichi footwork assisted routine rehabilitation training on the influence of limb balance disorder in patients with hemiparesis after strokein the recovery period.Methods:A total of 60 patients with hemiparesis after stroke in our hospital from March 2017 to May 2018, who met the selection criteria, were divided into 2 groups by random number table method, 30 in each group. The conventional rehabilitation group took the conventional rehabilitation training method, and the Taichi footwork group took Taichi footwork training method based on conventional rehabilitation method. Both groups were treated for 4 weeks. The Berg Balance Scale (BBS) was used to evaluate the patient's balance ability, the Holden Walking Aability Classification (HWAC) was used to evaluate the lower limb walking ability, and the Fugl-Meyer Motor Function Rating Scale (FMS) was used to assess the limb function.Results:After the treatment, the BBS score (44.4 ± 7.9 vs. 38.7 ± 9.9, t=4.885), the HWAC grade (3.6 ± 1.0 grade vs. 2.7 ± 1.0 grade, t=5.152) and FMS score (12.3 ± 1.4 vs. 10.4 ± 1.4, t=4.785) of the Taichi footwork group were significantly higher than those of the conventional rehabilitation group ( Ps<0.01). Conclusions:Taichi footwork assisted conventional rehabilitation training can improve the walking ability of patients with hemiparesis after stroke in the recovery period, which helps the patientsregain limb function.
10.Study on the Proportion Regularity of 8 Kinds of Ginsenoside to Ginsenoside Rg 1 in Panax ginseng of Different Growth Years in Jilin Province

OBJECTIVE: To investigate the proportion of 8 kinds of ginsenoside to ginsenoside Rg1 in Panax ginseng and the regularity of growth year in Jilin province, and to provide reference for the identification of growth year. METHODS: The samples of garden ginseng, wild-cultivated ginseng and wild ginseng were collected from different growth years (3-30 years) in Jilin province. The contents of 8 ginsenoside (ginsenoside Rg1, Re, Rf, Rb1, Rc, Rb2, Rb3, Rd) in P. ginseng were determined by HPLC. The contents of saponins as well as the proportion of 8 kinds of ginsenoside to ginsenoside Rg1 were calculated; the relationship of the proportion with growth year was investigated. RESULTS: As the increase of growth year, the proportion of 8 kinds of ginsenosides in garden ginseng to ginsenoside Rg1 as well as that of ginsenoside Re, Rb1, Rc, Rd to ginsenoside Rg1 were decreased gradally (P<0.001); the proportion of ginsenoside Re to ginsenoside Rg1 in wild-cultivated ginseng decreased first and then increased(P<0.001); the proportion of 8 kinds of ginsenosides to ginsenoside Rg1 as well as the proportion of ginsenoside Re and Rb1 to ginsenoside Rg1 were increased gradually in wild ginseng (P<0.001); the proportion of ginsenoside Rf, Rb3 to ginsenoside Rg1 in garden ginseng, wild-cultivated ginseng and wild ginseng had no significant difference(P>0.05). CONCLUSIONS: Garden ginseng, wild-cultivated ginseng and wild ginseng contain 8 kinds of ginsenosides. The growth year can be predicted preliminarily according to the proportion of ginsenoside Rg1 and ginsenoside Re, Rb1 to ginsenoside Rg1.
