In presnt experiment, animal models for hyperthyroidism and hypothyroidism were made, and using radioimmunoassay, the immunoeactive leucine-enkephalin (Ir-LEK) levels of such discrete brain areas as hypothalamus, striatum, cerebral cortex(frontal lobe) and whole pituitary were determined in different groups. The results obtained indicated that both hyperthyroidism and hypothyroidism induced significant changes of concentration in CNS Ir-LEK. 2.INHIBITORY EFFECTS OF HUMAN CHOLANGIOCARCINOMA CELL LINE BY RECOMBINANT ADENOVIRUSES p16 AND p53 TRANSDUCTION

To evaluate the inhibitory effect of recombinant adenoviruses p16 (Ad p16) combined with Ad p53 on human cholangiocarcinoma cell line QBC939 in vitro. Expression of p16 was low and p53 was negative in QBC939 cells. High level of p16 and p53 expressions were observed in QBC939 cell line transfected with Ad p16 and Ad p53. The growth rates of the Ad p16 infected combined with Ad p53 infected QBC939 cells were inhibited, which was higher than those of the Ad p53 or close to Ad p16 transfection. Colony formation in Ad p16 with Ad p53 transfected cells greatly decreased versus Ad p16 transfected or Ad p53 transfected cells. The Ad p16, Ad p53 and Ad p16 with Ad p53 transfected cells manifested apoptosis and G 1 arrest, which were confirmed by the flow cytometry. The suppression effects mediated by expression of the exogenous p16 with p53 in tumor cell resulted mainly from apoptosis and G 1 arrest, which is higher than exogenous p16 or p53.

Objective To investigate the MRI manifestations and its pathologic foundation of adrenal myelolipoma.Methods MRI images of 13 patinets with adrenal myelolipoma confirmed by pathological assessment were analyzed retrospectively.MRI fingings were also studied in correlation with surgical pathology.Results The MRI diagnosis of adrenal myelolipoma in 13 patients were correct preoperative.Huge mass in the adrenal were found in 11 patients with T 1WI high signal intensity and mixed with conclamata and stripe low signal intensity.The high signal intensity was mature fatty tissue and the low signal intensity was myeloid tissue on pathologic examination.2 patients appeared as mixed signal intensity principle isointensity on T 1WI images,and the pathological findings was tumor hemorrhage.The high signal intensity changes to low signal intensity and slight higher than the signal of subcutaneous fatty tissue on T 1WI or T 1WI with fatty suppression.The low signal intensity of the myeloid tissue and the hemorrhage were high sinal intensity.No enhancement were found in all patients.Partly capsule enhancement were found in 2 patients with tumor hemorrhage and infection.The inferior cava vena was displaced to the anterior and internal side in all patients.Conclusion The characteristic manifestation of adrenal myelolipoma is high signal intensity change of the fatty tissue and no enhancement on contrast-enhanced MRI image.MRI could give correct diagnosis of the adrenal myelolipoma preoperative.
5.Imaging Diagnosis of the Lymphangiomyomatosis (A Report of 2 Cases)

Objective To improve the imaging recognition of the lymphangiomyomatosis.Methods The manifestations of 2 patients with lymphangiomyomatosis confirmed by pathological assessment were analyzed retrospectively and relevant literature were reviewed.Results Multiple,well-defined and thin-walled cysts were found on the chest HRCT.The size of the cystic airspace was different,and the wall of cysts ranged being fainted perceptible to near l mm in thickness.The retroperitoneal lymphangiomyomatosis were found in the 2 patients on CT images,and some lymphadens were confluence.The diameter of the largest lymphadens was 25 mm,no enhancement were found on contrast-enhanced CT scan.The right renal angioleiomyoma and retroperitoneal leiomyoma were presented in one patient,which were moderately homogeneous enhanced on contrast-enhanced CT scan.Conclusion Lymphangiomyomatosis is a rare lymphangial disease with smooth muscle abnormal hyperplasia.The diffuse cysts in bilateral lungs were its characteristic changes on CT images.Lymphangiomyomatosis can incorporate with renal angioleiomyoma,retroperitoneal leiomyoma and lymphangiomyomatosis.
6.The prognostic relative factors in treatment of tibial pilon fracture

Objective To analyze prognostic relative factors in treatment by evaluating the surgical results of tibial pilon fracture retrospectively. Methods 32 cases with unilateral tibial pilon fracture were treated operatively between January 1997 and December 1999. Among them, 16 cases were found with unsatisfied results due to incisional complications, degenerative changes and stiffness of the ankle joint. Of 16 cases, there were 13 males and 3 females. The average age was 36.5 years (range, 25 to 55 years). The mechanisms of injury were fall in 7 cases, traffic accident in 5 cases, crash in 3 cases and other cause in 1 case. 6 cases with open fracture were treated in emergency. However, 10 cases with closed fracture were operated with an average interval of 8.1 days from injury to surgery(range, 2 to 25 days). According to the Ruedi-Allgower classification, there were type Ⅱ fracture in 7 cases and type Ⅲ fracture in 9 cases. Furthermore, there were 14 cases associated with fibular fracture. During operation, 7 cases were treated with open reduction and internal fixation with plates and screws, and 9 cases were treated with open reduced minimally and external fixation with a cast. Results The average period of follow-up was 24 months (range, 12 to 36 months). According to Mazurs criterion, the result of the treatment was evaluated as fair in 6 cases, 4 cases of which were type Ⅱ fracture, and 2 cases type Ⅲ fracture; poor in 10 cases, 3 cases of which were type Ⅱ fracture, and 7 cases type Ⅲ fracture. Complications included wound dehiscence in 1 case; infection in 4 cases, 2 of which with open fracture were superficial infection, and 2 cases with closed fracture were deep infection; posttraumatic arthritis of the ankle joint in 9 cases; and severe varus deformity in 2 cases.11 cases suffered from ankle stiffness. Conclusion Discouraged results of pilon fractures are significantly related to fracture type, reduction status and time of operation, reduction and fixation of fibular fracture, and bone grafting.
7.Research on spontaneous regeneration of optic nerve following various extent of incomplete injury in adult rats

Objective To examine and evaluate the ability of spontaneous regeneration following various extent of incomplete injury in adult rats. Methods The quantities and reduplicated various extent incomplete injured model of adult rats optic nerve was established using different wounding time with constant wounding force produced by across action forceps at 2mm behind eyeball. GAP-43 (growth associated protein-43) and its mRNA expressions were detected with immunohistochemistry (IHC) and reverse transcription polymerase chain reaction (RT-PCR). All data were analyzed by ANOVA. Results GAP-43 and its mRNA expression levels revealed that there was negative response at the distal area contrast to the strong positive response at the proximal in earlier period after injury until half mouth. One mouth later, the increased GAP-43 and its mRNA expression levels became more and more high and reached the climax at second month post injury. Then it decreased gradually. The result of RT-PCR showed there are significant difference among the various extent incomplete injured models and different time after injury. Conclusion Spontaneous regeneration of adult rat’s optic nerve can be detected and identified following incomplete injury, and the extent of regenerating ability is correlated with the extent of injury.
8.Effect of splenectomy plus pericardial devascularization on coagulation function in patients with cirrhotic portal hy-pertension

Objective To determine platelet count (PLT),mean platelet volume (MPV),and hemodynamic changes in patients with cir-rhotic portal hypertension (CPH)post splenectomy plus pericardial devascularization (SPPD),and to assess the surgical effect on coagula-tion function.Methods A retrospective analysis was performed on 83 CPH patients undergoing SPPD in our hospital from January 2008 to December 2012.Results Compared with preoperative levels,postoperative portal venous pressure decreased,blood flow was reduced,and portal vein diameter was significantly reduced;additionally,postoperative hepatic artery diameter was increased,and hepatic artery blood flow increased.Blood alanine aminotransferase,albumin,total protein,and fibrinogen levels,thrombin time,and MPV did not significantly change 30 days postoperatively versus 7 days preoperatively(P>0.05 );the corresponding total bilirubin,activated partial thromboplastin time and prothrombin time relatively declined(P<0.05),whereas PLT and prothrombin time activity increased(P<0.05).There was a significant increase in PLT (P<0.05)but no significant change in MPV 30 days postoperatively versus 7 days preoperatively (P>0.05). Conclusion SPPD can significantly improve liver functional reserve and coagulation function in patients with CPH.
9.Promoter methylation of RASSF1A gene in hepatocellular carcinoma and its clinical significance

Objective To explore the relationship between promoter methylation of RASSF1 A gene and clinico-pathological characteristics in hepatocellular carcinoma.Methods MS-PCR was used for analyzing the status of aberrant promoter methylation of RASSF1A in 100 primary HCC samples and adjacent noncancerous tissues,10 normal liver tissues,and six HCC cell lines.RT-PCR was used to assess reactivation of RASSF1A expression after HCC cell lines treated with demethylating agent 5'-aza-2' deoxycytideing.Results Abnormal promoter methylation of RASSF1A gene was found in 69(69%) cases of HCC,15 (15%) cases of adjacent normal tissues,and no abnormal promoter methylation of RASSF1A gene was found in normal liver tissues,and the difference was statistically significant (x2 =67.75,P <0.001).The methylation of RASSF1A gene was correlated to HBsAg (x2 =11.341,P < 0.05) and histological differentiation(x2 =10.575,P < 0.05).Four cell lines with abnormal CpG island methylation of RASSF1A gene were all re-expressed after treated with 5'-Aza-CdR.Conclusions RASSF1A gene promoter methylation was correlated to HBsAg and histological differentiation and this is one of the most important mechanism for low expression of RASSF1A in HCC.
