1.Determination of Moxifloxacin in Human Serum by RP-HPLC

OBJECTIVE:To establish a RP-HPLC method for the determination of the concentration of moxifloxacin in human serum.METHODS:The separation of sample was performed on Gemini C18 column with a fixed sample injection volume of 20?L.The mobile phase was methanol-water-triethylamine(112.5∶208.9∶0.06) at a flow rate of 1mL?min-1.The UV detective wavelength was set at 289nm;the column temperature was 25℃ and the sensitivity was 0.01AUFS.RESULTS:In the range of 0.312 5~10?g?mL-1(r=0.999 8),moxifloxacin showed a good linear relation in HPLC.The recoveries of moxifloxacin at three concentrations(10,2.5,0.625?g?mL-1) were 99.46%,103.12%,and 96.14%,respectively,with intra-day RSD at 3.27%,3.35%,0.92%,respectively and inter-day RSD at 7.19%,6.25%,6.68%,respectively.CONCLUSION:This method is simple,rapid,accurate and sensitive,and it can be used for the concentration determination and the pharmacokinetic study of moxifloxacin.
2.Perspiration on the Head, A Special Manifestation in Severely icteric Chronic Hepatitis, Report of 124 Cases,

Of the 411 hepatitis with severe jaundice admitted, 124 ( 30. 17%) had head perspiration. Based on Zhang Zhongjing's etiologic - pathogenic conception, 102 cases of such patients were treated by the principle of cooling and activating blood, benefitting Qi and Yin. 89 cases had jaundice eliminated, 13 cases had residual jaundice. All 22 cases of chronic severe hepatitis survived. Of them, 9 early stage cases were all survived, 7 of the 8 cases of intermediate stage survived, only 1 out of 5 cases of late stage survived.
3.Three-dimensional CT liver image segmentation based on hierarchical contextual active contour.

In this paper, we propose a new active contour algorithm, i. e. hierarchical contextual active contour (HCAC), and apply it to automatic liver segmentation from three-dimensional CT (3D-CT) images. HCAC is a learning-based method and can be divided into two stages. At the first stage, i.e. the training stage, given a set of abdominal 3D-CT training images and the corresponding manual liver labels, we tried to establish a mapping between automatic segmentations (in each round) and manual reference segmentations via context features, and obtained a series of self-correcting classifiers. At the second stage, i.e. the segmentation stage, we firstly used the basic active contour to segment the image and subsequently used the contextual active contour (CAC) iteratively, which combines the image information and the current shape model, to improve the segmentation result. The current shape model is produced by the corresponding self-correcting classifier (the input is the previous automatic segmentation result). The proposed method was evaluated on the datasets of MICCAI 2007 liver segmentation challenge. The experimental results showed that we would get more and more accurate segmentation results by the iterative steps and the satisfied results would be obtained after about six rounds of iterations.

4.Interventional effect of Chinese herbal compound digan oral liquid on cyclin genes and adherent molecules of hematopoietic stem cells and stromal cells in bone marrow of mice with irradiative injury

BACKGROUND: Chinese herb digan oral liquid promotes manufacture of hematopoietic cells. During irradiative injury, how are the therapeutic effects of intervention with Chinese herbal compound on expressions of stromal cellular adherent molecule (SCAM) and cyclin gene that are closely related to hematopoietic cells?OBJECTIVE: Mouse model with irradiative injury was established to assay the effects of Chinese herbal compound-digan oral lipid on the expression of medullary cyclinD1mRNA and the expressions of hematopoietic stem cells (HSCs), stromal cellular adherent molecular CD45 (SCAMCD45) and vascular cellular adherent moldecular-1 (VCAM-1).DESIGN: Randomized controlled experiment was designed.SETTING: Department of Integrated Traditional Chinese and Western Medicine, Wuhan General Hospital of Guangzhou Military Area Command of Chinese PLA; Institute of Hematology of Tongji Medical College and Institute of Hepatology of Hubei College of Traditional Chinese Medicine.MATERIALS: The experiment was performed in Institute of Hematology of Tongji Medical College and Institute of Hepatology of Hubei College of Traditional Chinese Medicine from January 2001 to June 2002, in which,48 outbred Kunming mice were employed, aged varied from 8 to 12weeks, mass-weighted varied from 18 to 22 g, of either sex. Drugs: 100% digan oral lipid is composed of shudihuang (radix rehmanniae preparata) and zhigancao (radix glycyrrhizae preparata). At ratio of 2/1, water decoction was prepared and concentrated as 100% oral liquid (containing raw herb 1 g/mL). METHODS: Totally 48 mice were randomized into 4 groups, 12 mice in each one. Normal group: no any management. Oral lipid group: Digan oral lipid was applied for gastric perfusion in model with irradiative injury,0.02 ml/g, twice a day. Model group: After modeling, physiological saline was used for gastric perfusion with the same dosage as the above. Batilol group: After modeling, batilol suspension was used for gastric perfusion (0.1 g/kg). Gastric perfusion was done continuously for 9 days in each group. On the 10th day, hybridization in situ was used to assay cyclinD1 mRNA expression of medullary mononuclear cell (MNC), Immuniohisto chemical SABC method was used to assay medullary VCAM-1 expression and flow cytometry (FCM) was used to assay CD45 content.MAIN OUTCOME MEASURES: CyclinD1 mRNA expression of medullary mononuclear cell (MNC) and expression of medullary stromal cellular VCAM-1 and CD45 antigenRESULTS: After modeling, there were 12 mice in normal group and 10 mice in rest groups. In result analysis, 5 mice were randomized in each group. ① In oral liquid group, the expression of medullary cellular cyclinD1 mRNA and VCAM-1 were higher significantly than model group[oralliquid group: (27.34±4.49), (53.94±8.67)% and model group: (16.06±4.57),(31.92±5.01)%, P< 0.01]. ② Expression of CD45 antigen iroral liquid group was higher significantly than model group [(41.38±5.76), (19.24±4.98)%, P < 0.01]. Both digan oral liquid and batilol improved the expressions of cyclinD1,VCAM-1 and CD45 antigen.CONCLUSION: Chinese herbal compound-digan oral liquid enhances the expression of adherent molecule of hematopoietic stem cells, promotes the expression of stroma] cellular adherent molecule and improves the transcription of medullary cellular cyclinD1, which induces hematopoietic cells to be differentiated more maturely and to be transported peripherally so as to supplement blood.

To elucidate Bcl 2 and P53 protein expression in renal cell carcinoma(RCC) and their patho clinical significance, the expression of Bcl 2 and P53 proteins was evaluated immunohistochemically and compared with the patho clinical factors from 113 patients. BACKGROUND:During the outbreak of severe acute respiratory syndrome(SARS),certain people had been isolated by various reasons and appeared a series of psychological, physical and behavioral reactions. OBJECTIVE:To understand the different defensive features in people with different level of mental health in the isolated population, and explore the relationship between defensive style and mental health. DESIGN:An in investigative study taking the isolated population during outbreak of SARS as the subjects. SETTING:A psychiatric department of a military hospital. PARTICIPANTS:Totally 187 people of different sex,age and education, who were isolated during April and May 2003 due to SARS outbreak in a city of northwest China,were selected as the subjects. INTERVENTIONS:The 187 subjects,who were isolated due to SARS outbreak,were evaluated by using the symptom checklist(SCL 90) and defense style questionnaire(DSQ). RESULTS:About 36.4% people of this population had distinct mental or physical health problems that were characterized by anxiety,horror,depression,hostility and compulsion.There was difference in defensive styles between the high symptom group and low symptom group,among which the score of DSQ factors in the immature type of high and low symptom groups were 5.72± 1.56 and 4.35± 0.96 respectively while the scores in the intermediate type defense were 4.98± 1.44 and 3.72± 0.89 respectively(P< 0.01).Mental health problem was positively correlated with the application of immature defensive style,but had negative correlation with application of mature defensive means. CONCLUSION:There is difference in the defensive styles among the isolated people with different mental health status,and their defensive strategies are closely related to the mental health.
7.Relationship between diabetes mellitus and hypothalamic-pituitary-adrenal axis:A systematic review

Objective To provide objective evidences for early screening of the hypothalamic-pituitary-adrenal(HPA)axis function in diabetic patients, the relationship between HPA axis function and diabetes mellitus was systematically reviewed. Methods PubMed, Ovid, Embase, Web of science Databases were retrieved from articles reporting the relationship between HPA axis function and diabetes mellitus. Meta-analysis were carried out by Stata 12. 0 software for heterogeneity test and data merged among articles for the inclusion and exclusion criteria. Results Our meta-analysis showed the following results: Compared with control group, the basal plasma cortisol level in total diabetes group, T1DM group, T2DM group, diabetes patients(with or without chronic complications)combined with hypertension was significantly higher. Compared with control group, postdexamethasone cortisol level was markedly higher in diabetes group. There was no significant difference in 24 h urinary free cortisol(24h UFC)and ACTH levels between diabetes group and control group. Our qualitative evaluation shows the following results: the differences of awaking salivary cortisol and cortisol awakening response(CAR)between the two groups were inconsistent among all included studies. Most studies showed that CAR was lower in diabetic patients. Conclusion The present analysis has demonstrated that HPA axis dysfunction exists along with diabetes mellitus, and diabetic comorbidity may further worsen the HPA axis dysfunction.
8.Determination of triptoquinone H in Tripterygium wilfordii and its Tablet by HPLC

Objective To develop an HPLC method for determination of triptoquinone H in Tripterygium wilfordii and its Tablet preparations. Methods An external method with Agilent Zorbax SC-C_(8)(250 mm?4.6 mm, 5 ?m) column as fixed phase and methanol-water (75∶25) as mobile phase was adopted. The detective wavelength was 258 nm and the flow rate was 1.0 mL/min. Results The linearity range of triptoquinone was 6.28 — 100.5 ?g/mL (r=0.999 7). The average recovery of T. wilfordii was 96.94%, RSD was 1.57% (n=9) and the average recovery of T. hypoglaucum Tablet was 100.02%, RSD was 1.74% (n=9). Conclusion The method is accurate and sensitive. It is adoptable for quantity analysis of triptoquinone H in T. wilfordii and its Tablet.
9.Determination of tripterine in Tripterygium wilfordii and its tablets by HPLC

Objective To determine the content of tripterine in Tripterygium wilfordii and its tablets by HPLC.Methods An external standard method by HPLC with Zorbax C_(18)column as fixed phase and methanol-1% HAc(87∶13) as mobile phase was adopted.The detection wavelength was 425 nm and the flow rate was 1.0 mL/min.Results The linear range for tripterine was 40.96~204.8 ?g/mL(r=(0.999 6).) The average recovery of Chinese medicinal materials was 98.37% and RSD was 1.01%(n=9);the average recovery of preparation sample was 98.59% and RSD was 1.18%(n=9).Conclusion The method is simple and accurate,which can be adoptable for quantitative analysis of tripterine in the plants of Tripterygium L.
10.Using modified crushing-cercariae escaping method to detect infected snails

Objective To evaluate the detection rate and the efficiency of the modified crushing-cercariae escaping method.Methods The detection rates of the modified crushing-cercariae escaping method and the crushing methods were compared by using a double-blind control experiment with the latter as a gold standard.meanwhile the number of the cercariae was quantified.The efficiency of the two methods aforementioned and the cereariae escaping method were compared in field.Results The detection rate of the modified crushing-cercariae escaping method was 100%.the average number of cercariae in each infected snail was (4 778±1 157);and the number in certain volume of water sample was positively correlated with the number of infected snails.The efficiency of the modified menthed Was 18.2 times and 17.3 times as high as those of the crushing method and cereariae escaping method,respectively.Conclusions The modified crushing-cercariae escaping method Can detect the infected snails quicky and Can quantify the number of infected snails and cercariae,and is suitable for the detection of infected snails in large number.
