1.Determination of Substances with High Molecular Weight in Porcine Anterior Pituitary and Adrenal Cortex Extracts Injection

Objective:To determinate the substances with high molecular weight in porcine anterior pituitary and adrenal cortex extracts injection. Methods:The HPLC analysis was performed on a TSK-GEL G2000SWXL(7. 8 mm × 300 mm,5 μm) column with the mobile phase of trifluoroacetic acid-acetonitrile-water(0. 05:35:65). The flow rate was 0. 5 ml·min-1, and the detection wave-length was 214 nm. Results:The substances with high molecular weight in porcine anterior pituitary and adrenal cortex extracts injec-tion was below 1%. Conclusion:The method is simple and repeatable, which can be used for the control of the substances with high molecular weight in porcine anterior pituitary and adrenal cortex extracts injection.

Objective To explore the health effects of organic extracts from ambient air particulates with various diameters on the DNA damage of human peripheral blood lymphoctys. Methods Using a 5-stage air sampler, the ambient air particulats samples were collected in a residential area of Taiyuan city and were extracted with dichloromethane, acetone and methanol. The damage of DNA in human peripheral blood lymphocytes induced by organic extracts from ambient air particulates with various diameters at doses of 25, 50, 100 ,200 ?g/ml were detected by sigle cell gelelectrophoresis assay. Results The toxic efficiency of organic extraction of ambient air particulates increased with the decreases of the diameters of particulates. 4.Relationship between podocyte molecule’s distribution and expression,foot process morphology and proteinuria

Objective: To investigate the association between nephrin, podocin and ? actinin of the glomerular podocyte molecules, the morphometric change of podocyte foot process and the development of proteinuria. Methods: Puromycin aminonucleoside (PAN) nephrosis was established. Immunofluorescence staining, image analysis and real time quantitative PCR were employed to study the distribution and quantitation of glomerular expression of nephrin, podocin and ? actinin. Morphometric methods were applied to evaluate the morphology change of podocyte foot processes under electron microscopy. Results: (1) Before the onset of proteinuria, 2 days after PAN injection, the podocyte foot process became swollen;nephrin and podocin staining were changed into discontinuous pattern accompanied by the decrease of podocin staining intensity. The foot process became more swollen on day 5,and podocin intensity continued to decrease. Meanwhile, nephrin decreased significantly both in protein intensity and at mRNA level. (2) When heavy proteinuria [(130.8?30.7) mg/d, P =0.02]occurred, complete effacement of podocyte foot processes was revealed; both podocin and nephrin staining intensity decreased dramatically( P
5.The effects of low-frequency pulsed electromagnetic fields on microcirculation angiogenesis in acute hindlimb ischemia among diabetic rats

Objective To observe the effects of low-frequency pulsed electromagnetic fields (LFPEMFs) on microcirculation angiogenesis in the hindlimbs of diabetic rats with acute ischemia. Methods Models of acute hindlimb ischemia were established in 60 male Sprague-Dawley diabetic rats. The diabetes model was established using 60 mg/kg intraperitoneal injections of streptozotocin (STZ). Fasting blood glucose levels were greater than 300 mg/dL. The rats were randomly divided into experimental and control groups. The rats in the experimental group were exposed to low-frequency pulsed electromagnetic fields for 2 hours each day, while the control group was not given any treatment. Laser-Doppler perfusion was used to measure blood flow in the ischemic hindlimb on days 0,7, 14 and 28 after the operation. The immunofluorescence of rat endothelial cell antigen-1 ( RECA-1) was used to evaluate the changes in angiogenesis. The levels of vascular endothelial growth factor (VEGF) and fibroblast growth factor-2 (FGF-2) were determined by both Western blotting and ELISA, and VEGFR2 and FGFR1 levels in the ischemic skeletal muscle were determined by Western blotting on days 7, 14 and 28 after the operation. Results The average perfusion ratio was significantly greater in the experimental group at days 14 and 28 compared with the control group. RECA-1 density in the tissues had increased significantly in the experimental group at the 14th and 28th day. The same was observed for FGF-2 and its receptor, but there was no significant difference for VEGF or its receptor in either group. Conclusions LFPMEFs can promote angiogenesis in acute hindlimb ischemia of diabetic rats by up-regulating FGF-2. This suggests that LFPMEFs may be useful for preventing and treating lower limb ischemia in diabetic humans.
6.Radiation induced bystander effect on hepatoma HepG2 cells under hypoxia condition

Objective To investigate radiation induced bystander effect and its mechanism on hepatoma HepG2 cells under hypoxia condition. Methods Non-irradiated bystander hepatoma cells were co-cultured with irradiated cells or treated with the conditioned medium (CM) from irradiated cells, then micronuclei (MN) were measured for both irradiated cells and bystander cells. Results The MN yield of irradiated HepG2 cells under hypoxic condition was significantly lower than that under normoxia, the oxygen enhancement ratio of HepG2 cells of MN was 1.6. For both hypoxic and normoxic condition, the MN yield of bystander cells were obviously enhanced to a similar high level after co-culturing with irradiated cells or with CM treatment, and it also correlated with the irradiation dose. When the hypoxic HepG2 cells were treated with either DMSO, a scavenger of reactive oxygen species (ROS), or aminognanidine, an iNOS inhibitor, the yield of bystander MN was partly diminished, and the reducing rate of DMSO was 42.2%-46.7 %, the reducing rate of aminognanidine was 42 %. Conclusion ROS, NO and their downstream signal facets are involved in the radiation induced bystander effect of hypoxic HepG2 cells.
7.Application of dual energy cerebral arteries computed tomographic angiography in diagnosis of cerebral aneurysm

Objective To study the value of dual energy cerebral arteries computed tomographic angiography (CTA) in diagnosis of cerebral aneurysm.Methods As a gold standard of digital subtraction angiography (DSA) findings,the dual energy cerebral arteries CTA imaging features of 30 cases confirmed by DSA for cerebral aneurysm were retrospectively analyzed.The diagnosis accuracy of cerebral aneurysm between dual energy cerebral arteries and DSA was compared.Results Dual energy cerebral arteries CTA could display the direct signs of cerebral aneurysms,it played better in the signs of aneurysm of the siphon segment of internal carotid artery than that by conventional cerebral arteries CTA.The measurement of the size and neck by dual energy cerebral arteries CTA had good agreement to DSA,there was no statistical difference (P > 0.05).The radiation dose of dual energy cerebral arteries CTA was 19.1-25.4(21.7 ± 0.5)mSv.Conclusion The dual energy cerebral arteries CTA can show the size and neck of cerebral aneurysm clearly,has high value in clinical application of the diagnosis of cerebral aneurysm.
8.Advances in research of the role of microglia polarization in the mechanisms of pathogenesis of multiple sclerosis

Multiple sclerosis ( MS) is an autoimmune disease of the central nervous system ( CNS) characterized by demyelination and inflammation lesions.MS predominantly affects young adults with a high incidence of disability. However, the exact pathogenesis of MS is still not clear.Studies found that microglia polarization tending to pro-inflammatory M1-like state during the onset of MS, causing the M1/M2 ratio imbalance, forming pro-inflammatory microenvironment state, and which further leading to nervous tissue damage ultimately.Microglia polarization may be considered as the initiator of pathologic alterations by releasing pro-inflammatory cytokines and secondarily trigger the initial microglia response.Given the pivotal role of imbalanced microglia polarization in MS initiation, a critical review of microglia polarization is presented here, in order to elucidate the pathogenesis of MS and highlight the noteworthy candidate therapeutic targets for clinic treatment.
9.Plasma levels of leukocyte cell-derived chemotaxin 2 in patients with newly diagnosed type 2 diabetes complicated with non-alcoholic fatty liver disease and its association with insulin resistance

Objective To observe the changes in levels of plasma leukocyte cell-derived chemnotaxin 2 (LECT2) in patients with newly diagnosed type 2 diabetes (T2DM) complicated with non-alcoholic fatty liver disease (NAFLD),and to investigate the clinical significance.Methods A total of 137 subjects were enrolled in the study,including 50 patients with newly diagnosed T2DM complicated with NAFLD,47 patients with newly diagnosed T2DM with non-NAFLD and 40 healthy subjects.The level of plasma LECT2 was determined by ELISA.Anthropometric data and other biochemical indicators were measured in three groups.The body mass index (BMI) and waist-to-hip ratio (WHR) were calculated.Insulin resistance and pancreas β-cell function were determined by homeostasis model assessment (HOMA-IR,HOMA-%β).Results Plasma concentration of LECT2 in patients with newly diagnosed T2DM complicated with non-NAFLD was higher than that in type 2 diabetic patients with non-NAFLD [(32.95 ± 10.11 vs 29.08 ± 7.54) ng/mL,P < 0.01].Plasma LECT2 levels in both groups were significantly higher than that in normal control group [(22.38 ± 4.40) ng/mL,P < 0.01].Plasma LECT2 level was positively correlated with BMI,FPG,FINS,C peptide,HbA1c,GGT,TG and HOMA-IR,while negatively with HDL-C and HOMA-% β (all P < 0.01).Multivariate regression analysis showed that levels of BMI,FPG and HDL-C were important factors affecting plasma LECT2 level.Conclusions Plasma LECT2 concentration significantly elevates in patients with newly diagnosed T2DM complicated with non-NAFLD.Plasma level of LECT2 is closely correlated with insulin resistance and glycolipid metabolism.LECT2 may play an important role in the patho genesis of insulin resistance and T2DM.
10.Development of intelligent NO delivery system cooperating with ventilator

This paper develops an intelligent NO delivery system cooperating with the ventilator to make NO inhaled safely, reliably, conveniently and economically, which is based on correlation between the parameter setup of the ventilator and the concentration of NO. The circuit control system consists of a mass flow controller and a CPU mainly. A mixing unit is designed to mix NO with the gas from the ventilator uniformly. Clinical experiments show that the NO from the intelligent delivery system can decrease PH and increase SaO2 with the concentration of generated NO2 less than 5ppm.
