1.The Study on Working Memory Impairment in Patients with Lacunar Cerebral Infarctions

Objective To study the characteristics of working memory impairment in patients with lacunar cerebral infarctions(LCI).Methods 42 patients proved by clinics were chosen,and divided into two groups:multiple and single LCI groups,versus left,right and double side infarction groups.Their working memory status were evaluated and compared with Digit Ordering Test,Corsi block task,House Visual Span Test and Stroop Digit Counting Test.All scores were analyzed statistically.Results(1)All scores of the four types of Tests in multiple LCI group were significantly lower than those in single group(P0.05).Conclusions Multiple LCI is closely related to working memory impairment.Left brain infarction may induce verbal working memory impairment,and right infarction may induce spatial working memory impairment,but object working memory impairment may related to double side of brain infarction.It suggested that in the physiological structure,different components of working memory regions had dissymmetrical effect in two sides of brain.

3.Amplification and Identification of Immature Dendritic Cells Resistant to Maturation from Human Peripheral Blood Monocytes

Objective To establish a method for in vitro induction and amplification of immature dendritic cells(DCs) with maturation resistance from human peripheral blood. Methods Mononuclear cells separated from peripheral blood were cultured with rhGM-CSF and rhIL-4 for 9 days, and rhIL-10 was added into medium at the 7th day. The suspending cells were examined with scanning electronic microscope and flow cytometry, and their ability for stimulating non-sensitized T lymphocyte proliferation was observed by mixed lymphocyte reaction(MLR). Cultured cells were stimulated with LPS and TNF-? for additional 2 days, respectively, and MLR was performed again. Results rhGM-CSF+rhIL-4-induced and IL-10-treated peripheral blood mononuclear cells(PBMC) exhibited typical morphological characteristics and immunological phenotype of DCs with high expression of CD1a and no expression of CD 83 on the cellular surface. Costimulating molecules CD 40 and CD 86 expressions were down-regulated.The capability of cultured cells for stimulating the proliferation of non-sensitized T lymphocyte was weak, and the same result was observed in cultured cells stimulated with LPS or TNF-?. Conclusion Immature dendritic cells with maturation resistance were obtained by culturing with IL-10,which might be a useful in the induction of immune tolerance of allogenic skin grafting for the major burn patients with deep burn wounds.
4.Expression of p38MAPK and TGF?_2 in retina of experimental diabetic hamster

Objective To explore the role of p38MAPK and TGF?2 in the development of diabetic retinopathy.Methods Fifteen Hamsters were induced into diabetic models by intraperitoneal injection of Streptozotocin(40 mg/kg once a day) for 3 d and 13 Hamsters were successfully established,whose blood glucose was over 13.5 mmol/L.Ten Hamsters as controls were intraperitoneally injected of physical saline of the same volume.At 16th week after induction,the total RNA of retina from all sacrificed Hamsters was collected.The mRNA expressions of p38MAPK,TGF?2 in retina were detected by semi-quantitative RT-PCR and their protein levels by Western blotting.Results The mRNA expressions of p38MAPK,TGF?2 in retina were of high tendency and their protein levels increased.Conclusion p38MAPK signal pathway may involve in the pathogenesis of diabetic retinopathy.
5.Relationship of compression area, time and weight with crush injury/crush syndrome in rats

Objective To investigate the relationship of compression area, time and weight as risky factors with local injury and systemic pathophysiological responses in rats so as to establish repeatable experimental model of crush syndrome. Methods A total of 144 male SD rats were divided into two groups, ie, mortality investigation group and biochemical indicator investigation group. Every group included the same 18 subgroups based on 18 kinds of combination with different levels of compression area (the right or both hind limbs), time (4, 6, 8 hours) and weight (2, 3, 4 kg). The circumference of the compressed hind limbs of all rats were measured and serum potassium (K+), serum creatine phosphokinase (CK), creatinine and carbamide were measured too before compression and three hours after decompression. Incidence of myoglobinuria of all rats was recorded. Muscles and kidneys were evaluated morphologically. Results The compressed hind limbs of all rats swelled significantly after three hours of reperfusion (P < 0. 05). All serum K + , CK, CR and BUN were increased significantly with the increase and prolongation of the compression area, time and weight (P < 0.05). Signs of direct cellular damage and ischemia-reperfusion injury were found in histology specimens of local compressed muscle.Hyperemia of glomeruli and renal tubule was found in the kidneys. Renal tubular necrosis and renal tubular cast were observed in group with compression weight ≥3 kg and compression duration ≥6 hours.Conclusions Increase and prolongation of the compression area, time and weight can aggravate the severity of crush injury. Compression area is more risky factor of severe crush injury. Both hind limbs ascompression area, compression weight ≥ 3 kg and compression duration ≥ 6 hours can be the effective experimental conditions for establishment of crush syndrome model in rats.
6.Analysis of the characteristics of medicine taking in patients with gastroesophageal reflux disease in Beijing

Objective To investigate the characteristics of medicine taking,the type and ratio of long-term medication and treatment satisfaction in patients with gastroesophageal reflux disease (GERD),to analyze the effects of GERD combined with functional bowel disease (FBD) on the treatment satisfaction,and to compare the changes of type of medication and treatment satisfaction in patients with GERD in recent years.Methods From April to June in 2011,the questionnaire survey was conducted in gastroenterology clinic of People's Hospital of Peking University.The incidence of typical GERD symptoms was investigated by a validated reflux disease questionnaire (RDQ),the score between one and five was considered there might be reflux symptoms and the score over 12 was diagnosed as GERD.FBD was diagnosed according to Rome Ⅲ criteria.The information of GERD related medication taking within one year before the survey,long-term medication taking (≥ 1 year)and the treatment satisfaction of patients whose RDQ score over zero was recorded and compared with the data collected in the same method in 2004.The chi-square test was performed for data statistical analyses with SPSS 17.0 software.Results Among 1074 patients who completed questionnaire survey in 2011,the percentage of patients with reflux symptoms was 32.7% (351/1074),the percentage of diagnosed GERD was 10.0% (107/1074),and GERD combined with FBD was 25.2% (27/107) of GERD.A total of 304 cases (86.6%) of patients with reflux symptoms had information of medication taking; 78.0% (237/304) of whom had taken GERD related medication within one year before the survey.The rate of GERD related medication taking especially proton pump inhibitor (PPI) and antiacid medication taking increased along with RDQ score (x2 =24.2,13.1 and 18.2,all P<0.05).A total of 104 cases of GERD patients had information of medication taking; 88.5 % (92/104) GERD patients had taken GERD related medication within one year before the survey,52.9% (55/104) GERD patients had taken PPI medication,30.8% (32/104) needed long-term medication and 20.2% (21/104) needed longterm PPI treatment.There was no significant difference in treatment satisfaction between different RDQ score ranges (x2 =3.3,P>0.05).Among GERD patients who appraised the effects of treatment,the percentage of satisfied,acceptable and not satified with the treatment was 37.1%(26/70),48.6% (34/70)and 14.3%(10/70),respectively.There was no significant difference in treatment satisfation between GERD with and without FBD (x2 =3.1,P>0.05).In 2011,the rates of medication taking (88.5%,92/104) and PPI taking (52.9%,55/104) in GERD patients significantly increased compared with those in 2004 (57.3%,98/171; 7.6%,13/171,x2 =29.4,71.4,both P<0.05).The percentage of treatment satisfaction in 2011(37.1%,26/70) increased compared with that in 2004(25.7%,18/70,x2 =12.8,P<0.01).Conclusions Some of patients with GERD symptoms need long-term medication,especially PPI treatment.In recent years,the rate of medication taking in GERD patients increased in outpatients department,PPI taking significantly increased and the treatment satisfaction also increased.Maybe the treatment satisfaction is not affected by GERD combined with FBD.
7.Endostar combined with pemetrexed for first-line treatment of advanced lung adenocarcinoma in elderly patients

Objective:A study was conducted to observe and compare the efficacy and safety of endostar combined with peme-trexed in elderly patients with advanced lung adenocarcinoma. Methods:Sixty advanced lung adenocarcinoma (ⅢB-Ⅳ) patients who never received any therapy were included. The patients were divided into two groups. One group comprised endostar treatment com-bined with pemetrexed (26 cases of males, 15 cases of females, and 11 cases of individuals aged 65 years old to 78 years old), and the other group comprised pemetrexed only (34 cases of males, 20 cases of female, and 14 cases of individuals aged 65 years old to 78 years old). The two groups were treated for 4 to 6 cycles, and evaluation of treatments was performed every two cycles. Results:The endostar group was re-treated for 80 cycles, and the average cycle was 3.1. The group without endostar was re-treated for 115 cycles. The short-term effects are as follows. The total effective rates (RRs) in the experimental and control groups were 23.1%and 14.7%, re-spectively, and the difference was statistically significant (P<0.05). The disease control rate (DCR) was not significantly different (P>0.05). For pleural effusion, RR and DCR were significantly better in the experimental group than in the control group (P<0.05). In the experimental group, compared with PD, the microvessel density (MVD) in the DCR showed higher expression, and a statistically signif-icant difference (P=0.03) was observed. In the control group, compared with PD, the MVD in the DCR also showed higher expression, but no significant difference (P=0.73) was observed. The long-term effects were as follows: median progression-free survival (PFS), median survival, and side effects between the two groups were not significantly different (P>0.05). Conclusion: Endostar combined with pemetrexed showed increase in total efficiency in elderly patients with lung adenocarcinoma, and malignant pleural effusion was controlled without increasing the toxicity of chemotherapy. MVD can be used as a predictor of Endostar application.
8.Recent advances in the study of mechanism of APOBEC3G against virus.

APOBEC3 is a class of cytidine deaminase, which is considered as a new member of the innate immune system, and APOBEC3G belongs to this family. The research about APOBEC3G is a new direction of innate immune defense mechanism against virus. APOBEC3G has the restrictive activity on many viral replications, which deaminates dC to dU in the viral genome and then induces extensive hypermutation. APOBEC3G can also interrupt viral replication at several phases such as reverse transcription, replication, nucleocapsid and so on by non-deamination mechanisms. However, virus can encode viral proteins to counteract the restriction activity of APOBEC3G. Elucidation of the antagonistic interaction between APOBEC3G and the virus will be contributed to development of new antiviral drugs in the future.

9.Effects of recombinant human growth hormone on vascular endothelial growth factor expression of human gastric carcinoma xenografts in nude mice

Objective To investigate the effects of recombinant human growth hormone (rhGH) on tumor growth and vascular endothelial growth factor (VEGF) expression of human gastric carcinoma xenografts in nude mice with different expressions of growth hormone receptor (GHR). Methods Immunocytochemical method was used to pick out one GHR-positive and one GHR-negative cell line. Then the cells were subcutaneously injected into 24 nude mice separately. The nude mice bearing two different kinds of human gastric caicinoma were equalges of body weight and tumor volume of nude mice were recorded. Serum concentrations of VEGF in peripheral blood were analyzed by ELISA. VEGF protein and mRNA expression in tumor tissue were detected by immunohistochemistry and RT-PCR, respectively. Results We chose SGC-7901 as GHR positive group, and MKN-45 as the negative one. For nude mice bearing GHR + SGC-7901 xenografts, the tumor volumes were significantly larger in rhGH groups than in control group (P < 0.05), and the high-dose rhGH group revealed greater effect (P < 0. 05).Body weights were not significantly different among three groups (P > 0. 05). Serum VEGF concentration was (252.94 ± 15.32) ng/L in the high-dose rhGH group, which was significantly higher than that in control group [(49.94 ± 5.73) ng/L] and low-dose rhGH group [(167.60 ± 9.54) ng/L] (P < 0.05). Moderate positive staining with VEGF was observed in the control group, while VEGF staining was strong in rhGH administration groups. The relative expression of VEGF mRNA for the high-dose rhGH group was 0. 6470 ± 0. 0447, which was significantly higher than that in control group (0. 3230 ± 0. 0258)and low-dose rhGH group (0. 412 ± 0. 0351)(P < 0.05). While for nude mice bearing GHR-MKN-45 xenografts, the body weights of the rhGH-administrated groups were significantly higher than that of control group (P < 0.05), while tumor growth, serum VEGF concentration, and the expressions of VEGF mRNA and protein in tumor tissue were not significantly different (P > 0.05).Conclusions rhGH can promote tumor growth and increase the expression of VEGF in the GHR-highly-expressed SGC-7901 xenograft tumor model. However, such effects do not exist in GHR-negatively-expressed MKN-45 xenograft tumor model. The existence of GHR may be a key target where rhGH influences the secretion of VEGF.
