1.Changes of Drug Transporters Expression in Brains of Chronic Epileptic Rats Induced by Lithium-Pilocarpine

Objective To explore the expression of drug transporter organic anion transporting polypeptide2(Oatp2) and P-glycoprotein(P-gp) in brains of chronic epileptic rats induced by lithium-pilocarpine.Methods Chronic epilepsy modles were elicited after status epilepticus(SE) induced by lithium-pilocarpine in adult Wistar rats.The expressions of Oatp2 and P-gp were detected by immuno-histochemistry(SABC) method.Results Positive expression of Oatp2 was predominantly in the plexus epithelial cells and(blood-brain) barrier endothelial cells.Weak staining for Oatp2 was also detected in the neurocyte.In contrast,apparent expression of(P-gp) was seen in brain capillary and neurocyte only.Compared with normal control rats,expression of Oatp2 on the plexus epithelial cells and brain capillary in chronic epileptic rats was significantly decreased(P
2.Experimental Study on Cold Preservation of Rat Liver with Panax Notoginseng Saponins

Objective To investigate the effects and mechanism of Panax Notoginseng saponins (PNGS) on rat liver during cold preservation. Methods Using isolated perfusion of rat liver model (IPRL), Fura 2 method was used to measur the concentration of calcium ion in hepatic cells which had been preserved in DMEM solution with different concentration of PNGS added and cold preserved for 2 hours. Liver function, metabolic products of oxygen free radicals, energy substance and aucount of biliary flow as well as morphological study were measured from liver tissue which had been cold preserved in lactate riuges’s solution containing different dosages of PNGS for 24 hours and 30 minutes reperfusion.Results The contents of intracellur calcium of the rat hepatocytes,MDA, AST, ALT, LDH were lower than those in the control group,but SOD, ATP, TAN, EC and bile production were higher than those in the control group( P 0.05). Conclusion PNGS relieve the injury of the rat liver during cold preservation. The mechanism might through inhibition calcium overload, improve the energy metabolism, play a role against free radical injury realize.
3.Intragastric administration of Radix Paeoniae Rubra Extracts versus Taohong Siwu Tang for the serum concentration of peonifiorin in mice

BACKGROUND: Taohong Siwu Tang is made up of Taoren, Honghua,Danggui, Chishao and Chuangxiong. Paeoniflorin is one of the major effective components in Radix Paeoniae Rubra Extracts and Taohong Siwu Tang, and it can promote blood circulation and remove blood stasis and has the effect of anti-acute myocardial ischemia.OBJECTIVE: To compare the serum concentration of paeoniflorin in mice after intragastric administration (i.g.) of Radix Paeoniae Rubra extracts and Taohong Siwu Tang.DESIGE: Rndomized controlled observation.SETTING: Department of Pharmacy and Department of Basic Medicine,Hebei North University.MATERIALS: The trial was performed in the first laboratory of Department of Pharmacy, Hebei North University during February to June 2005.Eighty involved Kunming mice of either gender and of clean grade, with body mass of 18 to 22 g, were purchased from Experimental Animal Center, Institute of Genetics and Developmental Biology, Chinese Academy of Sciences. Taoren, Honghua, Danggui, Chishao, Shengdi and Chuanxiong 6 Chinese herbs were all purchased from Zhangjiakou Institute of Traditional Chinese Medicine.METHODS: ①Blank serum was isolated from eight mice which were fasted for 12 hours. Control sample solution of paeoniflorin was added into blank serum. Acetonitrile was used to deposit protein, the solution was centrifugated and isolated ,and 20μL supernatant was taken to obtain regression equation. ② Seventy-two Kunming mice were randomized into Radix Paeoniae Rubra extracts group and Taohong Siwu Tang group, with 36 mice in each. After being fasted for 12 hours, the mice in each group were intragastric administration of Radix Paeoniae Rubra extracts and Taohong Siwu Tang, respectively and the concentration of paeoniflorin was 300 mg/kg.1 mL blood was taken respectively at 15, 30, 60, 90, 120 and 180 minutes after intragastric administration and prepared into serum sample (6 mice at each time point in each group). The time to peak and difference in peak concentration of serum paeoniflorin were compared between intragastric administration of Radix Paeoniae Rubra extracts and Taohong Siwu Tang.The concentration of paeoniflorin was measured with high-performance liquid chromatography (HPLC). Chromatographic condition: chromatographic column YWG-C18 (250×4.6 mm,10 μm). The mobile phase was methanolKH2PO4 (0.05 mol/L) (38∶62) at a flow rate of 1.0 mL/min. The detective wavelength was set at 230 nm. The column temperature was 25 ℃.MAIN OUTCOME MEASURES: Serum concentration of paeoniflorin after administration of Radix Paeoniae Rubra extracts and Taohong Siwu Tang in mice.RESULTS: Seventy-two mice were used for Radix Paeoniae Rubra extr acts and Taohong Siwu Tang tests, and all of them entered the stage of result analysis without deletion. ① Chromatographic isolation results: Paeoniflorin could be well isolated from other components. The linear range for detection of paeoniflorin was 5.40-646.0 μg/mL. The mean recovery rate was 93.6% and the limit of detection was determined as 1.08 mg/L. ②Comparison of concentration of serum paeoniflorin of mice between two groups: The concentration of serum paeoniflorin in the Radix Paeoniae Rubra extracts group and Taohong Siwu Tang group reached the peak at 30 and 60 minutes after intragastric administration, respectively. The peak concentration of paeoniflorin of Taohong Siwu Tang group was lower than that of Radix Paeoniae Rubraextracts group [(36.27±5.72) mg/L vs.(46.82±5.29) mg/L, P < 0.01 )].CONCLUSION: The concentration of serum paeoniflorin is significantly decreased after intragastric administration. It was further confirmed the decreased concentration of the crude paeonifiorin in the mice sera might result from promotion of its metabolic conversion by other chemical components in the complex prescription. This result needs further investigation.
4.Improvement of in Vitro Dissolution Rate of Daidzein by Solid Dispersion Technology

Objective Daidzein solid dispersions were prepared by solid dispersion technology to improve in vitro dissolution rate. Methods Daidzein solid dispersions were prepared by solvent method using polyvinyl pyrrolidone K30 ( PVP K30) as carrier.The in vitro dissolution characteristics of solid dispersions were evaluated,and the properties were detected by IR and XRD. Results The dissolution rates of different mass ratio of daidzein-PVP solid dispersion were significantly improved compared with that of daidzein API.The cumulative dissolution of solid dispersion with mass ratio of 1∶6 within 30 minutes was up to 87.8%,equivalent to six times of API. The in vitro drug release kinetics were fitted mathematically to Korsemeyer-Peppas model. Conclusion Solid dispersion with PVP K30 as carrier could significantly improve dissolution rate of daidzein.
5.Effects of Rhamnocitrin in Oxytropis Falcata on Nonspecial Immune Function of ;Immunosuppressive Mice

Objective To study action mechanism of rhamnocitrin in Oxytropis Falcata on enhance the immune function. Methods Kunming mice were randomly divided into blank control group, model group, positive control group, traditional Chinese medicine high, medium, and low dose groups. All treatment groups received gavage with relevant medicine for 10 days. From the 8th day, blank control group received intraperitoneal injection with normal saline, while other groups received intraperitoneal injection with cyclophosphamide for 3 days. Through macrophages carbon clearance test, macrophage phagocytosis of chicken erythrocytes test, and immunosuppressive mice organ index detection, effects of rhamnocitrin in Oxytropis Falcata on nonspecific immune function of immunosuppressive mice were observed. Results Compared with model group and blank control group, traditional Chinese medicine high, medium, and low dose groups could improve the phagocytic activity of immunosuppressive mice, clearance index, and phagocytic coefficient (P<0.05), and increase the organ index of the thymus and spleen. Compared with the positive control group, the efficacy of traditional Chinese medicine high, medium, and low dose groups was low. Conclusion Rhamnocitrin in Oxytropis Falcata can enhance the immune function, improve the formation of spleen cells of mice serum hemolysin of chicken red blood cell immune.
6.A Research about Injection Project of Contrast Media During Enhanced CT Scanning

Objective To investigate a method that perfect imaging could be obtained and minimal contrast media was used. Methods The 5 cases with enhanced CT images were measured at each slice, then the Hu of aim-position was obtained and the Hu-time curve was got. At last, the valid enhanced period would be found. Results According to the experience, the enhancement threshold of aorta, portal vein and hepar were set at 70 HU. The onset and ending time of enhancement threshold were found. Conclusion The dose of contrast media should be extenuatory when new type spiral CT was operated, and the correlative parameters, such as flow rate, delay time etc. should be adjusted according to the objective condition.
7.Improvement of electrode fixing method in pediatric long-term electroencephalography

Objective To explore the electrode fixing method in children long-term electroencephalography and to improve the traditional method by combining with the pediatric characteristics and by means of clinical practice .Methods The pediatric subjects were given the appropriate preinstallation preparation ,then the double-layer elastic bandage combined with adhesive tape fixing method was adopted to fix the electrodes .Results The new method was generally accepted by children subjects ,the electrodes were easily to be installed and demounted without toxicity and residue ,the record diagram was better in quality .Conclusion Compared with the traditional method ,the improved electrode fixing method has the advantages of simplicity ,rapidness ,safety ,stability and humanization ,and is worth popularizing .
8.A report of 18-month follow-up study on a case of classic maple syrup urine disease

Objectives To explore the long-term prognosis, treatment (especially dietary treatment) of classic maple syrup urine disease. Methods The complications and dietary treatment were observed by follow-up study of a classic MSUD patient. Results The patient have obvious damage in nervous system. However, reasonable dietary leucine tolerance therapy after the neonatal period can effectively reduce the metabolic crisis and complications. A mutation in BCKDHB gene was detected in the patient by genetic testing. Conclusion It is suggested that dietary restriction and monitoring of branched-chain amino acids are helpful to reduce the development of acute metabolic crisis and complications and improve the quality of life.
9.Investigation and analysis of hospital infection in childhood nephrotic syndrome

To find out the risk factors of hospital infection in childhood primary nephrotic syndrome(nephrosis) and prophylactico therapeutic measures, 190 cases of child patients hospitalized for nephrosis during the period lasting from 1991 to 1999 were collected. Then a retrospective analysis of the distribution of the various risk factors was made using the single factor analysis and the logistic multi factor regression model. The hospital infection rate was found to be 34.2%, with respiratory infection accounting for 69.7%. The single factor analysis indicated that the risk factors included quantity of urinary protein per kilogram of weight within 24 hours(urinary protein), length of stay, length of hormone usage, entities of antibiotics used and length of such usage. The multi factor analysis indicated that the risk factors included urinary protein, entities of antibiotics used and length of such usage. The authors present the view that strengthening the control of respiratory diseases, setting up clean wards, and ensuring the rational use of antibiotics are some of the key measures for reducing the rate of hospital infection. The quantity of urinary protein is a sensitive index of hospital infection in nephrosis. Once diagnosis of nephrosis has been confirmed, it is necessary to use ACH in full dose and for a complete course of treatment so as to reduce the loss of protein from the urine as soon as possible.
10.Renal sympathetic denervation for treatment of resistant hypertension: a meta-analysis

Objective To systematically evaluate the effectiveness and safety of renal sympathetic denervation (RSD) for treatment of patients with resistant hypertension.Methods English and Chinese literatures of controlled clinical trials on RSD in treatment of resistant hypertension were searched from the Cochrane Library,PubMed,Web of Science,Wanfang Database and CNKI up to February 2013.Metaanalysis was performed with the selected studies by using software Rev.Man 5.0.Results Seven studies involving 354 cases with RSD and 146 controls were included for analysis.Compared with control group,RSD significantly lower both systolic and diastolic blood pressure in patients with resistant hypertension (systolic,1 month:MD =-18.04,95% CI:-19.94--16.14,P <0.01; 3 months:MD =0.01,95%CI:-28.10--17.98,P<0.01; 6 months:MD=-25.59,95%CI:-34.08--17.11,P<0.01; diastolic,1 month:MD=-7.53,95% CI:-8.60--6.45,P<0.01; 3 months:MD =0.01,95%CI:-12.5--4.47,P < 0.01; 6 months:MD =-10.54,95% CI:-16.44--4.63,P =0.000 5).In addition,there was no significant difference in adverse reactions between RSD treatment group and control group (OR =1.13,95 % CI:0.34-3.76,P =0.84).Conclusions Renal sympathetic denervation can effectively reduce blood pressure in patients with resistant hypertension 1-6 months postoperatively,without increase of adverse reactions.The long-term efficacy and safety need to be further observed.
