1.Cloning and identification of the CDS region of the p55-v0 and p55-v3 gene in Pneumocystis carinii

This study sought to amplify the CDS region of p55-v0 and p55-v3 gene in Pneumocystis carinii (PC) and to analyze their sequences.The total RNA extracted from PC- infected lungs of rats were used as the template to amplify these genes by RT-PCR.The amplified products were connected to T-vector and transformed into E.coli DH5α.The recombinant T-vectors were selected in LB culture medium containing ampicillin.Then the positive clones of recombinants were identified by PCR and digested by restrictive endonuclease.After that the recombinant were sequenced and analyzed.It was found that the CDS region of p55-v0 gene contained 1245 bps,encoding 414 amino acids.The homology between this sequence and the one reported previously in nucleotide and amino acid were 99.8% and 100% respectively.Meanwhile,the CDS region of p55-v3 contained 1053 bps,encoding 350 amino acids.As compared with GenBank,the homology in nucleotide and amino acid was 99.9% and 100% respectively.However,the similarity in nucleotide between p55-v3 and p55-v0 was just 20.9%.In this study,the CDS regions of p55-v0 and p55-v3 gene were cloned successfully,and the high homology was found between p55-v0,p55-v3 and the one reported previously by sequence analysis,p55-v3 was different from p55-v0 in nucleotides and amino acids.This result would provide a basis for comparison of immunologic functions so as to explore the mechanism of action of the p55-v3 gene.
2.SIGIRR overexpression inhibits LPS-induced NF-κB activation in alveolar epithelial cells

AIM: To investigate the effect of single immunoglobin IL-1 receptor related protein (SIGIRR) on damage of alveolar epithelial cells in acute lung injury induced by lipopolysaccharide. METHODS: The acute alveolar epithelial cell injury model was constructed by stimulation of A549 cells with LPS. In order to over-express SIGIRR, the A549 cells were transferred with eukaryotic expression vector containing full length SIGIRR cDNA. The transcriptional activity of NF-κB was measured by dual-luciferase reporter assay system. The concentrations of IL-1β, TNF- α and IL-6 were detected by ELISA. The levels of these inflammatory factors between the transfected cells and untransfected cells were compared. RESULTS: The over-expression of SIGIRR inhibited the transcriptional activity of NF-κB. The increases in IL-1β, TNF-α and IL-6 concentrations in alveolar epithelial cells induced by LPS were observed. CONCLUSION: SIGIRR in alveolar epithelial cells inhibits TLR4 signals triggered by LPS and attenuates the inflammatory reactions in alveolar epithelial cells, which plays a protective role against the acute damage of the alveolar epithelial cells.
3.Preparation and Quality Control of Dexamethasone Acetate Borneol Cream

OBJECTIVE:To prepare dexamethasone acetate borneol cream and to establish its quality control method.METHODS:Dexamethasone acetate borneol cream was prepared by grinding method;dexamethasone acetate and phenol were identified by TLC,and the content of the principal agent was determined by ultraviolet spectrophotometry.RESULTS:The prepared cream fitted the description of China Pharmacopeia(2005 edition) in identification and tests etc.The linear range of dexamethasone acetate was 12.06~28.14 ?g?mL-1(r=0.999 7),and the average recovery rate of 98.52%(RSD=0.66%).CONCLUSION:The preparation is reasonable in compounding,simple and feasible in techniques,and its quality is stable and controllable.

5.Combined injection of medical ozone and collagenase for the treatment of lumbar disc herniation

Objective To observe the mid-term effectiveness of percutaneous lumbar puncture and injection of medical ozone and collagenase in treating lumbar disc herniation.Methods CT-guided percutaneous lumbar puncture and medical ozone injection into the intervertebral disc was performed in 78 patients with lumbar disc herniation, which was confirmed by imaging study or clinical manifestations.After 2-4 days, neucleolysis was carried out.Collagenase 12 000 U was injected via the anterior epidural space onto the surface or into the inside of the prominence of the protruded disc.Results The clinical results were evaluated in 6-25 months after the procedure.Of 78 patients treated with this technique, the result was excellent in 58(74.3%), good in 16(20.5%) and poor in 4(5.1%).Symptoms returned in 12 cases in 2 months after the procedure, and the reappeared symptoms were relieved in 9 of them within 2 months without giving any treatment.In 3 cases the symptoms disappeared after repeated injection of medical ozone.No serious complications occurred.Conclusion Percutaneous lumbar puncture and injection of medical ozone combined with collagenase is an effective, safe and minimally-invasive therapy for lumbar disc herniation.
6.Observation of curative effect of methylprednisolone with octreotide retrobulbar injection therapy on thyroid associated ophthalmopathy

AIM: To explore the curative effect of methylprednisolone with octreotide retrobulbar injection therapy on thyroid associated ophthalmopathy.
METHODS:Fifty- one cases ( 96 eyes ) with thyroid associated ophthalmopathy were treated with methylprednisolone ( 500mg/d ) for 3d, and then retrobulbar injection of octreotide (0. 1mg/mo) for 3mo in one course. Symptoms and signs of the patients were observed before and after the treatment course.
RESULTS: Symptoms of 49 cases were improved significantly. In all cases, the mean value of exophthalmus of the patients was significantly decreased after the treatment by 3. 1 ± 1. 4mm. The height of palpebral fissure was significantly decreased after the treatment by 2. 4 ± 0. 9mm, there were statistically significant difference before and after treatment (P<0. 05).
COCLUSION: Methylprednisolone with octreotide retrobulbar injection therapy can improve symptoms and signs of the patients with thyroid associated ophthalmopathy with high safety and no obvious side effect.
7.Modified acellular dermal matrix for chondrocyte implantation in repairing cartilage defects of rabbits

BACKGROUND:Acellular dermal matrix possesses good flexibility and simple trimming.The intracutaneous or subcutaneous injection of acellular dermal matrix powder has fibroblast migration and collagen deposition.It has been widely used in plastic and reconstructive surgery.OBJECTIVE:To explore the feasibility of reconstructed acellular dermal matrix as a scaffold for chondrocyte implantation.DESIGN,TIME AND SETTING:Comparative observation.The study was performed at the Peking University Medical Department and Beijing Jishuitan Hospital between August 2003 and February 2007.MATERIALS:Neonatal calf dermis was provided by Beijing Yuanheng Shengma Biology Technology Research Institute.A total of 24 healthy adult SD rats,weighing 250 g,regardless of gender,and 36 New Zealand rabbits,aged 3 months,were selected.METHODS:①Calf full-thickness back skin was incubated with cell free buffer or ABS/AES for decellularization,followed by surface modification using growth factors.②Three rectangle skin flaps at two sides of the spinal cord of rats were made,and implanted with acellular dermal matrix.The implants were harvested at 2,6,and 12 weeks postoperatively.③The rabbits were divided into experimental and control groups.The cartilage was obtained from the left articular facet to isolate chondrocytes.The chondrocytes were seeded on the acellular dermal matrix.The cartilage defect was made on the right hind limb of experimentalrabbits,and implanted with acellular dermal matrix containing autoiogous chondrocytes.Biogel wass dropped on the surface of carrier.In the control group,the cartilage defect was made on the right hind limb of rabbits and the wound was sutured.Two rabbits from control group and 5 from experimental group were selected respectively at 4,12 and 24 weeks postoperatively.MAIN OUTCOME MEASURES:Cross-linking effect comparison;repair effect of rabbit bone defects.RESULTS:①The acellular dermal matrix cross-linked by glutaraldehyde demonstrated an obvious inflammatory reaction with tissue bleeding and necrosis.Conversely,ADM treated with water-soluble cross-linking agent caused displayed good histocompatibility.②The cartilage defects were repaired completely;the attached cells survived and proliferated and the acellular dermal matrix was degraded after 24 weeks of surgery.CONCLUSION:The acellular dermal matrix decellularized with cell free buffer,digested with digestive buffer,Cross-linked by water-soluble cross-linking agent,and further decorated with growth factor exhibited good histocompatibility,and was suitable forcell attachment and growth.The acellular dermal matrix scaffold almost degrades in the rabbits,with no rejection,and the bone defects were repaired after 24 weeks.
8.Three-dimensional EH composite artificial template for repairing cranial defect in 53 cases

OBJECTIVE: To observe effects of three-dimensional EH composite template on repairing cranial bone defect. METHODS: A total of 53 inpatients were analyzed retrospectively at the Department of Neurosurgery, Taixing People's Hospital from July 2004 to May 2007, comprising 31 males and 22 females, aged 19-68 years. They were in accordance with cranial repairing indication. Reasons for skull defect: traumatic brain injury (41 cases), hypertension (7 cases), intracranial cancer (3 cases), intracranial aneurysm (2 cases). Thirteen patients developed bilateral cranial defect. The defect region: frontal (5 slices, comprising 2 with severe orbital part defect), temporal (9 slices), frontotemporal (42 slices), temporoparietal (3 slices), parietoocipital (4 slices),frontoparietal (3 slices). The smallest single-chip defect size was 4 cm × 6 cm, and the largest one was 12 cm×18 cm. All the patients were mended by three-dimensional EH composite template using spiral CT (≤5 mm thickness) scanning, CAD three dimensional reconstruction, quickly modeling technique. Infection of incisional wound, rejection and complications were observed. RESULTS: A total of 53 patients were followed up, with an average follow-up period of 18 months, and all of them were satisfied with the shapes (100%). Mild collection of the fluid under the scalp was found in 2 cases 4 days and 1 week following surgery, and disappeared after suction and pressure dressing. No other frequent complications were observed after cranioplasty. CONCLUSION: Three-dimensional EH composite template is an ideal material for cranial bone defect, because of its good biocompatibility, easy operation, good postoperative shape and less complications.

10.Study on purification of 2,3,5,4'-tetrahydroxy-stilbene-2-O-beta-D-glycoside from Polygonum multiflori.

OBJECTIVETo study the conditions and parameters of purifying 2,3,5,4'-tetrahydroxy-stilbene-2-O-beta-D-glycoside from Polygonum multiflori.
METHODAbsorption capacity of four resins for 2,3,5,4'-tetrahydroxy-stilbene-2-O-beta-D-glycoside was compared. With the adsorption ability as indexes, the process of absorbing and purifying 2,3,5,4'-tetrahydroxy-stilbene-2-O-beta-D-glycoside from P. multiflori with S-8 macroporous resin absorbent was selected by orthogonal design.
RESULTThe S-8 resin was the best of the four resins. The optimum process condition was 50% ethanol as eluting solvent, the flow rate at 1.5 mL x min(-1), pH at 7-8, and the solution concentration at 0.2 g x mL(-1). The absorption capacity by this process was 36.89 mg x g(-1).
CONCLUSIONThe process is simple and convenient and the regeneration of resin is easy, so this method of purification is advisable.
