Objective:To study the optimum ethanol extraction process. Methods: The extraction rate of extractum and total flavonoids as well scutellarin was choosen as the assessment index. The optimum ethanol extraction process was selected with the orthogonal design. Results: The optimum ethanol extraction process conditions follow as: adding 10 fold of 75% ethanol, extracting for 2.0h and twice in all. Conclusion: The average extraction rate of extractum and total flavonoids as well scutellarin was 13.6%, 91.7%, 78.4%(n=3), respectively.
2.Effect and mechanism of chlorogenic acid in allergic rhinitis mice

Objective To study the effect and mechanism of chlorogenic acid in allergic rhinitis(AR) mice.Methods The model of AR mice was induced by intraperitoneal injection of adjuvant suspension on every other day for 7 times, and then by nasal drip of 10% OVA for 10 days.After administration with 50, 100, 200 mg/kg of chlorogenic acid for 30 min, AR mice were attacked locally for 10 days.Behavior analysis were observed and the effect of chlorogenic acid on the contents of cytokines, IgE and histamine by ELISA in AR mice were determined.Results Chlorogenic acid decreased the number of nose-scratching events in 10min, the weight of nosal draingage, the index of spleen, the content of IL-4 in nasal lavage fluid ( NLF) and the contents of cytokines, IgE and histamine in serum; while increased the index of thymus, the content of IFN-γin NLF, the contents of IFN-γin serum and the rate of IFN-γ/IL-4.The data showed a statistically significant up-regulation of IFN-γmRNA expressions and down-regulation of IL-4 mRNA, IL-5 mRNA, IL-10 mRNA expressions after adiminstration of chlorogenic acid.Conclusion Chlorogenic acid might exert the effect in AR mice through regulating the balance of Th1 and Th2.

Gene deletion vector pLJ04(pKC1139∷bkdF+bkdH)was used to disrupt bkdFGH in Streptomyces avermitilis 76-02-e,an industrial producer of anthelmintic avermectin.The disruptants were confirmed by PCR.Shake flask experiment and HPLC analysis showed that the mutant lost the ability to produce avermectins.As it is expected,the mutant,named S.avermitilis bkd76-3,could restore the ability of producing avermectins when the feeding of methylbutyric acid and isobutyric acid to its fermentations was carried out.The addition of cyclohexanecarboxylic acid(CHC)into fermentations of the S.avermitilis bkd76-3 allowed for production of four components,two of which was confirmed as CHC-B1 and CHC-A2 by LC/MS analysis,respectively.
4.Induction of specific anti-tumor immune responses against pancreatic cancer by transfected dendritic cells with pancreatic cancer MUC1 mRNA

Objective To investigate the induction of specific anti-tumor immune response by transfected dendritic cells (DCs) with MUC1 mRNA of human pancreatic cancer,and to provide the experimental evidences for the treatment of human pancreatic cancer with DC vaccine.Methods DCs were isolated and cultured from peripheral blood mononuclear cells (PBMCs),and then were identified by cell morphology and surface markers.After being transcripted and amplified,MUC1 mRNA was transfected into DCs by electroporation.The expression of MUC1 in DCs at different time points was detected by quantitative real-time PCR and Western blot.The survival rate of DCs before and after tramrfection was determined by MTT method.The induction of specific cytotoxic T lymphocyte (CTL) response by MUC1 mRNA transfected DCs was measured by 51Cr standard cytotoxicity test.The released amount of IFN-γ was evaluated by ELISA method.Results The cultured cells appeared typical characteristics with regard to morphology and phenotype (CD40 +,HLA-DR+,CD83 +,CD86 + ).After MUC1 mRNA transfection for 48 h,the expression of MUC1 mRNA of DCs reached the highest point ( 38.43 ) and the MUC1 protein expression also reached the highest point at 72 h.The survival rate of DCs was stabilized around 80% after transfection.The DCs transfected with MUC1 mRNA could effectively induce HLA-A2+/MUC1 + specific CTL immune responses.Stimulated by pancreatic cancer cell line Capan-2 cells or the DCs transfected with MUC1 mRNA,the IFN-γ released in 24 h by MUC1 specific CTL were ( 28.44 ± 4.96 ) U/m1 and ( 16.31 ± 2.54) U/ml,respectively.The difference between the two groups was statistically significant (P <0.05 ).Conclusions DCs transfected with human pancreatic cancer MUC1 mRNA could induce CTLs and produce specific anti-tumor immunity.
5.Prognostic value of SUVmax for locally advanced non-small cell lung cancer patients treated with synchronous cetuximab plus concurrent chemoradiotherapy

Background and purpose:We investigated whether lfuorine-18 lfuorodeoxyglucose (18F-FDG) maximal standard uptake value (SUVmax) of the primary tumor (SUV-T), SUVmax of the regional lymph nodes (SUV-N) or the overall loco-regional lesion SUVmax (SUV-TOTAL) was related to survival of patients with stage Ⅲ non-small cell lung cancer (NSCLC) who received Cetuximab and combined definitive chemoradiotherpay. Methods:From September 2009 to July 2012, seventeen patients with unresectable stageⅢNSCLC receiving cetuximab with cisplatin/vinorelbine (NP) followed by concomitant NP and intensity-modulated radiotherapy (IMRT) at the Fudan University Shanghai Cancer Center were enrolled onto a prospectively study. All patients received positron emission tomography/computerized tomography (PET/CT) scans within 2 weeks before enrolment. Univariate analysis were used to assess the correlation between SUV-T, SUV-N, SUV-TOTAL, gender, age, histology, tumour-node-metastasis (TNM) stage, performance status (PS) as well as smoking status and survival. The factors which showed statistical signiifcance entered into multivariate Cox-regression model. Survival functions of different populations were estimated by Kaplan-Meier method and compared by Log-rank test. Results:In the univariate analysis, SUV-T, SUV-N, SUV-TOTAL, PS and smoking status were prognostic factors. The best cut-off values for SUV-T, SUV-N and SUV-TOTAL were 11, 11 and 20, respectively. Multivariate analysis revealed that SUV-TOTAL (P=0.012), SUV-T (P=0.025), and SUV-N (P=0.033) were independent predictors of survival with hazard ratio (HR) of 14.7, 11.2, and 6.2, respectively. Conclusion:Local, regional and locoregional maximal SUVs deifned by 18F-FDG PET-CT scanning may have a strong correlation with survival in this patients setting, which merits further study.

Objective To study to take the oxidized Mannan?modified 786?0 in renal clear cell carcinom as tumor cells antigen to sensitize Dendrit?ic cells(DC)and to observe the its killing effect on renal clear cell carcinoma of CTLs induced. Methods Getting the peripheral blood mononucle?ar cells from the volunteers,and then to be stimulated to turn to be maturation by GM?CSF and IL?4 in vitro. Taking the clear renal carcinoma cell as the tumor antigen,and then making it to be modified by oxidized Mannan to acquire the tumor cell vaccine.Experimental groups include:blank group:DC?PBS group,control group:control?DC?786?0,experimental group:DC?Ox?Mannose?786?0 group. Taking the flow cytometry to detect the changes of DC phenotype,then taking the ELISA to detect the sencretion levels of supernatant of IL?12 of DC,then taking the CCK to detecte the cytotoxicity of lymphocytes(CTL)induced by DC of these experiment groups. Results Results by flow cytometry:the mature phenotype of DCs sensitized by Ox?Mannose?786?0 group included CD80,CD83,CD86 and HLA?DR expressed significantly higher than the other groups. As well as the secretion levels of IL?12. Meanwhile the cytotoxicity activity of lymphocytes(CTLs)induced by DCs which are sensitized by Ox?Mannose?786?0 increased more significantly than the other groups. Conclusion Glycosylated Antigens can be more effective in sensitizing antigen?presenting cells DC,and stimulating them to be maturation,while the killing effect to tumor cells also have noticeably improved.
7.Evaluation of left ventricular hemodynamic changes of patients with hypertension by vector flow mapping

Objective To investigate the left ventricular hemodynamic changes of patients with hypertension by echocardiography using vector flow mapping (VFM).Methods Twenty-eight patients with hypertension (hypertension group) and 26 normal subjects (control group) were involved.All the subjects underwent conventional transthoracic echocardiographic examination and VFM on apical 3 chambers view.The evolution of velocity distribution,stream line and vorticity line in left ventricle were observed.Left ventricle was divided into basal,mid and apical segments,in which vorticity parameter (Vmax) were quantitatively measured and analyzed.Its relations to conventional left ventricular parameters (E,A,E/A,EDT,E/e') were then analyzed.Results ①Compared with the control group,LV end-diastolic,end-systolic volumes and E/e' significantly increased.The evolution of velocity distribution,stream line and vorticity in the hypertension group was slightly irregular.② In mid-diastole phase,vorticity of the control group showed the trend (middle> base > apex)(P <0.05),which wasn't shown in hypertension group.In other phases,vorticity of both groups decreased from base to apex (P <0.05).③There were statistically differences in vorticity between the two groups in early diastolic phase,late diastolic phase of apex and late systolic phase of middle (P =0.045,0.004,0.015,respectively).④ There were negative correlations between Vmax in late diastolic phase of every segment and E/A (from basal segment to apex:r =-0.310,-0.334,-0.333;P =0.028,0.020,0.021).There was positive correlation between Vmax in isovolumic contraction time and E/e' (r =0.348,P =0.016).Conclusions VFM can be utilized to analyze left ventricular hemodynamics quantitatively and it may be a good supplement for assessing cardiac diastolic function.
8.Microenvironment changes induced by Endostar monotherapy in patients with non-small cell lung cancer:a pilot study

Background and purpose:Clinical data show that Endostar, a recombinant human endostatin, has the therapeutic beneift for patients with non-small cell lung cancer (NSCLC) while combined with chemotherapy or ra-diotherapy. However, the microenvironment changes induced by Endostar monotherapy in NSCLC is not yet clear. The purpose of this study was to prospectively study tumor vascular effects of Endostar monotherapy in patients with locally advanced or advanced NSCLC by dynamic contrast-enhanced perfusion computed tomography (CT perfusion, CT-p). Methods:Previously untreated patients with histologically or cytologically conifrmed locally advanced or advanced NSCLC were eligible. All patients received daily Endostar (7.5 mg?m2) for 14 days. CT-p scans were acquired at the baseline and post-treatment. CT-p parameters, such as blood lfow (BF), blood volume (BV) and permeability surface PS (area product), were measured in all patients.Results:Of all 7 patients enrolled, four were staged asⅢB and three as stageⅣ (2 with malignant pleural effusion, 1 with brain metastasis). The median BF, BV and PS values of baseline and post-treatment were 27.1/48.9 mL/100 mL/min, 86.8/84.8 mL/100 mL and 45.0/54.0 mL/100 mL/min, respectively. After administration of Endostar for 14 days , BF showed a signiifcant increase compared with that at baseline (P=0.028), whereas no signiifcant changes were found in BV (P=0.398) and PS (P=0.237) values.Conclusion:Our results suggest that Endostar monotherapy induces a signiifcant increase in BF whereas no signiifcant difference in BV and PS.
9.Imaging features and pathological analysis of calvarial cavernous hemangioma

Objective To investigate the imaging features of the calvarial cavernous hemangioma and the surgical efficacy to improve diagnosis and treatment of the calvarial cavernous hemangioma.Methods The clinical information,imaging materials and surgical efficacy from ten cases of calvarial cavernous hemangioma confirmed by pathology and the related literature was reviewed.Results The lesion was located in frontal bone in eight cases,in parietal bone in one case and in occipital bone in one case.The lesions were round-shape hypodensity with clear margin in X-ray.The lesions were hyperdensity or slightly hyperdensity on CT scan,and were osteolytic lesions with a characteristic honeycomb or starburst pattern on bone window.The MRI features were complicated and variable.The lesions were heterogenous and unevenly enhanced signal intensity.Nine patients underwent radical resections and reconstructed immediately by titanium mesh.The patient with tumor in occipital bone underwent radical resection only.Macroscopically,the pathologic bone was a huge purple-red blush mass protruding from the skull surface.Histological examination revealed the diploe with large,thin-walled,dilated blood-filled spaces lined by flattened endothelial cells without evidence of malignancy.No recurrence was noted in any case during a follow-up period from 3 to 24 months.All the patients survived well without recurrence.Conclusions The imaging features of calvarial cavernous hemangioma have a high value in the diagnosis and may provide guidance for the treatment.The radical resection and immediate reconstruction treatment for calvarial cavernous hemangioma is satisfied.
10.Functional changes of intestinal mucosal barrier in surgically critical patients

BACKGROUND: The gut is capable of inducing multiple organ dysfunction syndrome (MODS). In the diagnosis and treatment of critical ill patients, doctors should pay particular attention to the protection or recovery of intestinal barrier function. However, no reliable diagnostic criteria are available clinically. This study aimed to assess the changes of intestinal mucosal barrier function in surgically critical ill patients as well as their significance. METHODS: Thirty-eight surgically critical ill patients were enrolled as a study group (APACHE II>8 scores), and 15 non-critical ill patients without intestinal dysfunction were selected as a control group (APACHE II<6). General information, symptoms, physical signs, and APACHE II scores of the patients were recorded. The patients in the study group were subdivided into an intestinal dysfunction group (n=26) and a non-intestinal dysfunction group (n=12). Three ml venous blood was collected from the control group on admission and the same volume of plasma was collected from the study group both on admission and in the period of recovery. The plasma concentrations of endotoxin, diamine oxidase (DAO), D-lactate, and intestinal fatty-acid binding protein (iFABP) were detected respectively. The data collected were analyzed by the SPSS 17.0 software for Windows. RESULTS: The levels of variables were significantly higher in the study group than in the control group (P<0.01). They were higher in the intestinal dysfunction group than in the non-intestinal dysfunction group (DAO P<0.05, endotoxin, D-lactate, iFABP P<0.01). In the non-intestinal dysfunction group compared with the control group, the level of endotoxin was not significant (P>0.05), but the levels of DAO, D-lactate and iFABP were statistically significant (P<0.05). The levels of variables in acute stage were higher than those in recovery stage (P<0.01).The death group showed higher levels of variables than the survival group (endotoxin and D-lactate P<0.01, DAO and iFABP P<0.05). CONCLUSION: The plasma concentrations of endotoxin, DAO, D-lactate, and intestinal fatty-acid binding protein (iFABP) could reflect a better function of the intestinal mucosa barrier in surgically critical ill patients.
