1.Preparation nucleic acid vaccine of brucella melitensis and its immune effect

Abstract Objective:To construct bmcella melitensis Br.melitensis genomic DNA expression library and study its immune effect.Meth-ods: Hind Ⅲ digested Br.melitensis genomic DNA fragments were cloned into pSV-?-Gal plasmds.The recombinant plasmids were transformedinto JM109 host bacteria. After large-scale isolation, the recombinant plasmids were inoculated into the quadriceps muscle of mice. In the 9thday after third boost immunization .agglutination antibodies against Br. melitensis and lymphocyte mitogenic ability of mice were determined. Re-sults : Expression library nucleic acid vaccine can induce the production of agglutination antibodies against Br. melitensis and stimulate the lym-phocyte porliferation primed by ConA. Conclusion:The genomic DNA expression library may provide a new vaccine against Br. melitensis infec-tion.
2.The clinical characteristics of colorectal cancer in Guangdong province

Objective To investigate the clinical characteristics of colorectal cancer and its trend in Guangdong province.Methods The data from 6501 patients with pathologically confirmed colorectal cancer of 5 hospitals from 1990 to 2004 was analyzed.Results The mean age of 6501 patients with colo- rectal cancer was 59 years old,with the predominant age ranged from 61 to 70 years old.The proportion of young patients decreased from 7.1% to 3.5%.and the ratio between male and female was 1.47:1, which was gradually increased with increasing of age.The prevelence reached a high peak(60 years old) and dropped after that as a single peak curve.The ratio between rectal cancer and colon cancer was 1.1: 1,and the descending colon cancer accounted for 27.6%.With ageing,the rectal cancers decreased while the right hemi colon cancer increased gradually.Tubular adenocarinoma was predominant with his- tological typing,which accounted for 76.0%.The progressing stage(Dukes B,C,and D)was found in 93.9% of all cases.Conclusions The onset age of patients with colorectal cancer become close to west- ern country.The proportion of young patients are decreasing.The ratio between male and female dis- plays a single peak curve from rising to dropping.A tendency of decreasing percentage in rectal cancer and the gradual increasing in right hemi colon cancer emphasize the importance of colonoscopy in colorec- tal caner screening.
3.Single-dose contrast-enhanced magnetic resonance aortography

Objective To probe the feasibility of single dose contrast enhanced magnetic resonance aortography(CEMRA) and evaluate its value in clinical application. Methods Transit time(TT) test and duration of peak enhancement of aorta(DPE) test in 28 healthy adults were performed. With 2 different injection rate of Gd DTPA at 2 ml/s and 3 ml/s, the TT,peak signal intensity of aorta(SPE),DPE and the interval between DPE and the time of injection(?T) were measured and compared. Two groups of healthy volunteers(each 10) underwent three diamentional fast imaging in static procession sequence (3D FISP) CEMRA with standard dosage of Gd DTPA with the injection rate at 2 ml/s and 3 ml/s respectively. The signal/noise ratio(SNR) of the source images and the image quality of CEMRA of both groups were compared. Fifteen patients with aortic disease did the same study as the voluteers with the injection rate only at 3 ml/s. Results With the injection rate of 2 ml/s and 3 ml/s,the TT,SPE,DPE,?T and the SNR of CEMRA in two groups were significantly different( P

The treatment plan of facial neuritis in Evidence-based Guidelines of Clinical Practice in Acupuncture and Moxibustion (2011 edition) is discussed, and case information of facial neuritis during the recent five years in department of acupuncture and moxibustion, PLA General Hospital, is retrospectively analyzed. In accordance with anatomy of the facial nerve to form the acupuncture prescription, the detailed diagnosis and treatment method for facial neuritis are introduced. The advantages of the diagnosis and treatment method for facial neuritis are summarized, hoping to establish a more comprehensive, standardized and unified treatment plan.

Current researches indicate that transplanted BM‐MSCs accompanies massive death ,and its effect is unsat‐isfactory .The miRNAs are a class of small non‐coding single‐stranded RNA molecules ,involve therapeutic course of bone marrow mesenchymal stem cell treating myocardial infarction in many respects .The present article made an o‐verview on effect of microRNAs in transplanted BM‐MSCs treating myocardial infarction via mainly illustrating bio‐logical function of microRNAs and its regulation effect on BM‐MSCs differentiation and paracrine secretion .

7.CT and MR imaging findings of acinic cell carcinoma in salivary gland

Objective To explore the CT and MR imaging findings of acinic cell carcinoma (ACC)in salivary gland and enhance the diagnosis of this rare disease.Methods The CT and MR imaging characteristics of 20 patients (7 males and 13 females,median age 44 years old) with pathologically proved ACC were retrospectively reviewed.CT and MR images were evaluated in relation to the following: location,size,morphology,margin,CT density/MR signal intensity and enhancement pattern.Results Twelve lesions were located in parotid gland,two lesions in maxillary Sinus,and one lesion in submandibular,parapharyngeal,infratemporal fossa,buccal,nasal cavity,and hard palate respectively.The size of lesions were 0.7~5.8 cm.Sixteen lesions less than 3 cm in size demonstrated round or oval round masses with well defined margin.Four lesions more than 3 cm in size demonstrated irregular masses with unclear margin.Among them,bony destruction of the wall of the maxillary sinus was noted in 3 cases,and the preauricular skin was invaded in 1 case.The densities (signal intensity) were homogeneous in 6 cases and heterogeneous in 14 cases.Lesions were slightly low density in 11 case and isodensity in 3 cases on CT plain scan.Lesions were homogeneous isointense in 2 cases and heterogeneous isointense in 4 cases on T1WI,heterogeneous hyperintense in all cases on T2WI.On post contrast images,lesions demonstrated remarkable enhancement in 14 cases,moderate enhancement in 2 cases and mild enhancement in 3 cases.Conclusions The imaging characteristic of ACC were nonspecific.CT and MR could accurately detect the extent of tumor involvement and was helpful to provide more comprehensive information for the strategy of clinic therapy.
8.Ultrasonographic characteristics of ovarian yolk sac tumor

Objective To investigate ultrasonographic features of primary and metastatic ovarian yolk sac tumors.Methods Ultrasonographic features of 19 primary lesions and 33 metastatic lesions in 35 patients were retrospectively analyzed.Results Primary tumors were sized (14.6±3.6)cm in maximum diameter,manifesting as cysti-solid masses.Solid components of primary tumors were mainly hypoechoic or isoechoic(16/19)with rich blood supplies.Thirty-three metastatic lesions were located in pelvoceliac cavity(26/33) and liver parenchyma(7/33),sized (9.4±4.5)cm,(9.2±4.9)cm and (5.6±1.6)cm in maximum diameter respectively.Metastatic lesions in pelvoceliac lesions mainly demonstrated as hypoechoic masses(21/26), however lesions in the liver were mainly hyperechoic(5/7).Anechoic regions could be found in 9/26 of the pelvoceliac lesions.Blood supply was found less rich in metastatic masses than that in primary ones.Elevated serum level of alpha fetoprotein (AFP) was observed in all patients; ranging from 217 to 211 682 μg/L.Conclusions Primary and metastatic lesions of ovarian yolk sac tumor have obvious ultrasonographic characteristics.Combined with serum AFP level,the accuracy of diagnosis could be improved.
9.Inhibitory effect of chlorogenic acid on senescence of human skin fibroblasts

[Abstract ] Objective To evaluate the inhibitory effect of chlorogenic acid on senescence of human skin fibroblasts (HSFs). Methods Fibroblasts isolated from human foreskin were treated with 1 mmol/L glyoxal in vitro to develop a model for cellular senescence. In order to select effective concentrations of chlorogenic acid, some HSFs were treated with 1 mmol/L glyoxal alone or in combination with chlorogenic acid at different concentrations (5, 10, 20, 40, 80 μmol/L)for 3 days, with those receiving no treatment serving as the blank control group. Then, methyl thiazolyl tetrazolium (MTT)assay was performed to evaluate the proliferative activity of HSFs. Some HSFs were divided into 5 groups to be cultured alone(blank control group), or treated with 1 mmol/L glyoxal(glyoxal group)or the combination of 1 mmol/L glyoxal and chlorogenic acid at effective concentrations of 10, 20 and 40 μmol/L (glyoxal + chlorogenic acid groups). Senescence associated β-galactosidase (SA-β-gal)staining and real-time fluorescence-based quantitative PCR were conducted to determine the percentage of senescent cells and expression level of p16INK4a mRNA respectively. Statistical analysis was carried out by one-way analysis of variance followed by the least significant difference(LSD)-t test. Results Compared with the blank control group, the glyoxal group showed significantly decreased cellular proliferative activity of HSFs (55.65% ± 2.00% vs. 100% ± 6.90%, P < 0.01), while chlorogenic acid increased the proliferative activity of HSFs in a dose-dependent manner, and the increase reached a peak at 40 μmol/L. Concretely speaking, the glyoxal + 10-, 20-, 40-, 80-μmol/L chlorogenic acid groups all significantly differed from the glyoxal group in cellular proliferative activity (60.75% ± 1.32%, 67.65% ± 1.90%, 75.71% ± 3.25% and 75.69% ± 2.38% vs. 55.65%± 2.00%, all P < 0.05), but no significant difference was observed between the glyoxal group and glyoxal + 5-μmol/L chlorogenic acid group or between the glyoxal + 40-μmol/L chlorogenic acid group and glyoxal + 80-μmol/L chlorogenic acid group (both P > 0.05). Therefore, 10 - 40 μmol/L was selected as the effective concentrations of chlorogenic acid. The glyoxal group showed significant increases in the percentage of senescent (SA-β-gal-positive)cells (35.65% ± 2.24% vs. 13.00% ± 2.22%, P < 0.01)and expression level of p16INK4a mRNA (2-ΔΔCt: 1.00 ± 0.06 vs. 0.26 ± 0.05, P <0.01)compared with the blank control group, while the glyoxal + 10-, 20-, 40-μmol/L chlorogenic acid groups showed significantly decreased percentage of senescent cells (31.50% ± 2.13% , 22.31% ± 3.11% and 19.32% ± 3.01%respectively)and expression level of p16INK4a mRNA (2-ΔΔCt: 0.88 ± 0.08, 0.73 ± 0.06 and 0.68 ± 0.04 respectively) compared with the glyoxal group (all P < 0.05). Additionally, the percentage of senescent cells decreased with the increase in chlorogenic acid concentrations in the glyoxal + chlorogenic acid groups. Conclusion Chlorogenic acid can protect HSFs from glyoxal-induced senescence.
10.Effect of anemia on quality of life in elderly patients with advanced cancer

Objective To observe the prevalence of cancer-associated anemia and its effect on elderly patients with advanced cancer.Methods Functional Assessment of Cancer Therapy-Anemia Version 4 (FACT-An) was used to investigate the quality of life (QOL) of 86 elderly patients with advanced cancer.The patients were divided into anemia group and non-anemia group.The influence factors for QOL of the elderly patients with advanced cancer were examined,and a multivariate regression was used.Results The prevalence of cancer-associated anemia in elderly patients with advanced cancer was 61.63 % (53/86).The scores in physical well-being (16.03±4.14 vs 12.47±4.68,P =0.001),emotional well-being (11.30±3.98 vs 9.45±4.04,P =0.041),functional well-being (13.61±3.74 vs 10.30±4.02,P < 0.001),additional concerns (57.39±7.28 vs 40.06±10.45,P < 0.001) and FACT-An (111.70±13.19 vs 84.34±18.95,P < 0.001) of the non-anemia group were all significantly higher than those of the anemia group.The multivariate regression analysis results indicated that hemoglobin (P < 0.001),comorbidities (P =0.002),performance status (P =0.018),age (P =0.030) were the influence factors for QOL of elderly patients with advanced cancer.Conclusion The morbidity of anemia in elderly patients is increased and it affects the QOL of senior patients with advanced cancer.
