1. Anti-inflammatory fraction and chemical constituents of Hosta plantaginea

Objective: To screen the active fraction with anti-inflammatory effect of Hosta plantaginea and study the chemical constituents of the active fraction. Methods: Different polar fractions were prepared by extraction with organic solvents. Their anti-inflammatory effects were evaluated by mice models treated by acetic acid-induced celiac capillary permeability and cotton pellets-induced granuloma. The constituents of active fraction were purified by chromatographic methods and identified by physicochemical properties and spectral data. Results: The ethyl acetate fraction could significantly inhibit the reinforce of celiac capillary permeability and the cotton pellets granuloma. Ten compounds were isolated from the ethyl acetate fraction and identified as docosanol (1), β-sitosterol (2), stigmasterol (3), (25R)-2α,3β-dihydroxy- 5α-spirostane-9(11)-en-12-one (4), daucosterol (5), (25R)-2α, 3β-dihydroxy-5α-spirostane-9(11)-en-12-one-3-O-{O-β-D- glucopyranosyl-(1→2)-O-[β-D-xylopyranosyl-(1→3)] -O-β-D-glucopyranosyl-(1→4)-β-D-galactopyranoside} (6), kaempferol 3-O-(2″-O-β-D-glucopyranosyl)-β-D-rutinoside (7), kaempferol 3-O-β-D-rutinoside-7-O-β-D-glucopyranoside (8), (25R)-2α,3β,12β-trihydroxy-5α-spirostane 3-O-{O-α-L-rhamnopyranosyl-(1→2)-β-D-galactopyranoside} (9), and (25R)-2β, 3β-dihydroxy-5α-spirostane 3-O-{O-β-D- glucopyranosyl-(1→4)-β-D-galactopyranoside} (10). Conclusion: The ethyl acetate extract was the anti-inflammatory fraction of H, plantaginea. Compounds 1-10 were isolated from H. plantaginea for the first time, and compound 10 is a new natural product.
2.Clinical study of tamsulosin in prevention of urinary retention after radical hysterectomy

Objective To study the clinical value of tamsulosin in prevention of urinary retention after radical hysterectomy.Methods Sixty-six patients after radical hysterectomy were enrolled in this study, and divided into two groups in random.Thirty-three cases in the treatment group were treated with tamsulosin 0.2 mg once every night,3 days before the catheter was drawn.While 33 cases in the control group only with placebo.Catheter keeping days, residual urine volume at first extraction,the rate of urinary retention and urinary tract infection,hospitalization time and drug adverse reaction were compared in two groups.Results Catheter keeping days and hospitalization time were shorter in the treatment group than those in the control group(P< 0.05).The residual urine volume at first extraction was(38.2 ± 5.6) ml in the treatment group and (168.5 ± 11.8) ml in the control group,there was significant difference between two groups (P< 0.05).The rate of urinary retention was 18.2%(6/33) in the treatment group and 36.4%(12/33) in the control group,there was significant difference between two groups (P <0.05).There was no adverse reaction resulted from tamsulosin in the treatment group.Conclusion Tamsulosin is an effective drug which could prevent and treat the urinary retention after radical hysterectomy.
3.Clinical analysis of 50 patients with T-cell non-Hodgkin’s lymphoma

Background and purpose:T-cell non-Hodgkin’s lymphoma(T-NHL) has relatively high incidence in Asian countries, and the incidence rate showed an upward tendency. It has a dual characteristic of both ethnic and regional. We conducted this study in order to analyze the clinical characteristics, pathological type and survival of T-NHL in our hospital. Methods:Records of 50 cases with T-NHL treated from Jan. 2002 to Dec. 2012, were analyzed in terms of clinical characteristics, distribution of pathological type and survival. Results:Patients with T-NHL account for 24%of NHL cases in our study. Of the 50 cases, 37 were Han, 13 were Uygur;The most frequent type was NK/T cell lymphoma(NK/TCL)(16/50, 32%). We performed pathological type for each age group:4 cases with children’s group were all T-lymphoblastic leukemia/lymphoma(T-LBL)(4/4,100%), the most frequent type of youth and middle age group was NK/TCL(8/20, 40%;7/13, 53.8%), as the senile group was angioimmunoblastic T-cell lymphoma(AITL)(6/13, 46.2%). There was also a signiifcant difference in pathological type between the stage, with stageⅠ-Ⅱwere all NK/TCL(12/12, 100%), stageⅢ-Ⅳwere observed with peripheral T-cell lymphomas-unspeciifed(PTCL-U)(9/38, 23.7%) and AITL(9/38, 23.7%). The frequent type of Han was NK/TCL(14/37, 37.8%), as the Uygur was T-LBL(5/13, 38.5%). The median overall survival(OS) time was 12 (ranged 0-112) months. The 5 year OS rate was 39%. Both pathological type and age group were important factors influencing survival. The difference in outcome for the pathological type(P<0.05): NK/TCL and anaplastic large cell lymphoma(ALCL) had the better 3 year OS rates(71%, 61%), but PTCL-U and T-LBL had poor prognosis(19%, 7%). Age did a signiifcant effect on OS(P<0.05):children’s group had the worse 5-year OS rate (0%), middle age group had a better prognosis (67%), the OS of the youth group was 35%, for the senile group was 21%. Conclusion:T-NHL in Xinjiang region have their unique clinical characteristics:The overall incidence rate was similar with the domestic report, however, higher than the reports abroad. The incidence of Han was higher than Uygur. Pathological type showed in this study was different from that in European and American countries. There was a significant difference in distribution of pathological type in different age group, stage and nation. The long-term survival and prognosis of patients in Xinjiang region was poor. Both pathological type and age group were important factors inlfuencing survival.
4.Analysis of the pedigreed population genetics of SPF Yorkshire and Landrace pigs imported from Canada

Objective To understand the genetic background of the specific pathogen-free Yorkshire and Landrace pigs, imported from Canada by Harbin Veterinary Research Institute, Chinese Academy of Agricultural Sciences.Methods The population genetics were investigated by using 19 pairs of microsatellite primers.Results In the Yorkshire pigs and Landrace pigs, 84 and 89 alleles, respectively, were detected at 19 microsatellites loci.The average polymorphic informa-tion content and mean heterozygosity in the Yorkshire pigs were 0.5271 and 0.5877, and in the Landrace population were 0.5652 and 0.6066, respectively.Because of the significant ( P<0.01) differences of alleles in different loci such as S0155,S0143,S0178,Sw857 and Sw936, it made them possible to be used to identify Yorkshire and Landrace pigs′breed. F-statistics showed that the differentiation within the population was small and genetic structure was stable.Conclusions Compare with the domestic pedigree large White and Landrace pigs, the SPF pedigree pigs imported from Canada are more stable in genetic structure, and can be used as laboratory animal models in animal science research.
5.Significance of galectin-3 expression in differentiating benign and malignant thyroid neoplasms

Objective To investigate the clinical value of galectin-3 expression on fine needle aspiration (FNA) smearsandtissueslicesof thyroid tumors in distinguishing benign from malignant tumors. Methods Galectin-3 expression on FNA smears and tissue slices of thyroid tumors from 72 thyroidectomy specimens was detected by immunochemical method (SP method). Results Galectin-3 expression was high on FNA smears and tissue slices of malignant thyroid tumors, whereas there was no or low expression of galectin-3 on FNA smears and tissue slices of benign thyroid tumors. The difference of positive rates of galectin-3 expression between malignant and benign thyroid tumors was significant on both FNA smears and tissue slices (? 2=43.73 and ? 2=48.16, respectively, both P0.05). Conclusion Galectin-3 expression is different between benign and malignant tumors. Galectin-3 expression level is higher in FNA smears and tissue slices of malignant thyroid neoplasms than that of benign ones, suggesting that galectin-3 is a reliable molecular marker in distinguishing benign from malignant thyroid tumors. Galectin-3 expression with immunochemical method may be used in diagnosing benign or malignant thyroid tumors.
6.The relationship of Slit2 and bone marrow mesenchymal stem cells with the angiogenesis

BACKGROUND:Bone marrow mesenchyme stem cells are important non-hematopoietic stem cells in the bone marrow, which can stimulate angiogenesis. While, Slit can also stimulate angiogenesis, as many studies have proved. OBJECTIVE:To review the biological functions, clinical application and effects of bone marrow mesenchymal stem cells and Slit2 on promoting angiogenesis. METHODS:A computer-based online research of CNKI and PubMed databases was performed to col ect articles published between 1980 and 2014 with the keywords“MSCs”and“Slit2”in Chinese and English. There were 436 articles after the initial survey. Final y, 65 articles were included according inclusion and exclusion criteria. RESULTS AND CONCLUSION:Both bone marrow mesenchymal stem cells and Slit2 play an important role in promoting angiogenesis, but the relevance of bone marrow mesenchymal stem cells and Slit2 is stil controversial. If assuming that bone marrow mesenchymal stem cells secrete Slit2, more researches should be done to reveal whether bone marrow mesenchymal stem cells promoting angiogenesis is relevant to Slit2 and through which signaling pathway Slit2/Robo functions to adjust bone marrow mesenchymal stem cells thus to promote angiogenesis. If relevant, the transplantation of the Slit2 and bone marrow mesenchymal stem cells wil be a promising treatment of cerebral infarction and other central nervous injuries.
7.Sulodexide prevents diabetic nephropathy through inhibiting renal NF-κB activation and MCP-1 expression

Objective To study the effects of sulodexide on renal NF-κB activation and monocyte chemotactic protein 1 (MCP-1) expression in diabetic rats and elucidate the possible mechanism of sulodexide in preventing diabetic nephropathy (DN). Methods Wistar rats were fed with high-sucrose-high-fat diet and injected with a low dose of STZ (streptozotocin,35 mg/kg) into abdominal cavity to induce diabetes.DM rats were randomly divided into non-treated group of treatment,blood glucose (BG),triglyceride (TG),cholesterol,serum creatinine (Scr),urea nitrogen (BUN),24 h urinary albumin excretion (UAE) were measured.HE staining was performed in renal tissues for light microscopy examination of mean glomerular volume (MGV).MCP-1 expression was detected by immunohistochemical method.NF-κB activation was determined by Western blot. Results Compared with NC group,DM group and DMS group had significant elevated BG,TG and TC levels (all P<0.01).There were no significant differences of BG,TG or TC levels between DM group and DMS group.Compared with NC group,DM group and DMS group had significant increased Scr,BUN,UAE levels (all P<0.01).Scr,BUN,UAE levels were significantly lower in DMS group than those in DM group [(39.1±0.88) μmol/L vs (41.0±2.16) μmol/L,(9.12±1.06) mmol/L vs (9.87±0.19) mmol/L; (19.92±0.96) mg/24 h vs (25.99±0.52)mg/24 h,all P<0.05].Compared with NC group,the MGV of DM group was significantly increased [(7.47±1.11)×105 μm3 vs (4.22±1.09)×105 μm3,P<0.01].Compared with DM group,the MGA of DMS group was significantly reduced [(6.64±0.71)×105 μm3 vs (7.47±1.11)×105 μm3,P<0.05],but was still increased compared with that of NC group (P<0.01).Compared with NC group,the MCP-1 expression of DM group was significantly higher [(12.17±1.94)/HPF vs (1.19±0.70)/HPF,P<0.01].MCP-1 expression in DSM group was significantly lower than that of DM group [(9.22± 1.61)/HPF vs (12.17±1.94)/HPF,P<0.01],but still higher than that of control group (P<0.01).Compared with NC group,the NF-κB activity was significantly higher in DM group [(0.89±0.07) vs (0.24±0.03),P<0.01].Compared with DM group,NF-κB activity of DMS group was significantly lower [(0.27±0.01) vs (0.89±0.07),P<0.01].There was no significant difference of NF-κB activity between DMS group and NC group. Conclusion Sulodexide has protective effects on diabetic nephropathy,and one of the mechanisms may involve the inhibition of NF-κB activation as well as the suppression of MCP-I expression.
8.Discoursing on Soxhlet extraction of ginseng using association analysis and scanning electron microscopy

Soxhlet extraction is a common method of sample preparation. However, there has been no discussion about the efficiency of Soxhlet extraction from different batches and the factors that cause content fluctuation. In this study, Panax ginseng was selected as a model sample. Soxhlet extraction by means of a water bath, which has always been neglected, was identified as a novel key factor in the poor repeat-ability in different batches of Soxhlet extraction, as it can affect the siphon times and reflux time, which have been positively correlated with the ginsenoside contents. By substituting round bottom flasks in the same column, the relative standard deviation of the most fluctuated compound, ginsenoside Rb1, was decreased from 24.6% to 5.02%. Scanning electron microscopy analysis confirmed that the breakdown of the surface of the ginseng powder in the Soxhlet extraction led to a better dissolution of ginsenosides, indicating that chloroform may promote the extraction of ginsenosides by disrupting the cell structure. Moreover, 70% methanol was regarded as the better solvent for extracting the ginsenosides. Overall, this work offers a practical and effective protocol for improving the accuracy and repeatability of Soxhlet extraction methodology for ginsenosides and other analytes.
9.Clinical application of miniature tissue transplantation in hand surgery

Objective To explore the concept,denomination and technique of miniature tissue transplantation in hand clinic. Methods In 537 cases,local skin or composite tissue defect in 756 thumbs or fingers were repaired with 4 kinds of miniature island flap and 10 kinds of miniature vascularized composite tissue.Some of them were created in clinic practice.The precess,main points and indication of the operation were expounded and compared. Results In 639 fingers covered with miniature island flap,the transferred flaps were surviverd completely in 621 fingers(97.2%),survivered partly in 11 fingers(1.7%)and necrosised in 7 fingers(1%).In 117 fingers repaired with miniature vascularized composite tissue,all the transplanted tissues were survivered.In all the injuried fingers,beautiful outlook were recovered and no fat and clumsy were found,two-point difference was 6-8 mm,effctive sensibility and good function were restored,and all the patients were satisfied. Conclusion Miniature tissue transplantation were good methods to cover the local defect in fingers or thumbs,worth of generalization and aplication in hand clinic,and have an important significance for improve the trauma repair quality and treatment level in hand surgery.
10.Correlation between serum levels of sTim-3, Galectin-9 and inflammatory factors IFN-γ, IL-4 in patients with brucellosis

Objective:To explore the correlation between serum soluble T cell immunoglobulin-mucin molecule-3 (sTim-3), galactose lectin-9 (Galectin-9) and inflammatory factors interferon-γ (IFN-γ), interleukin-4 (IL-4) in patients with brucellosis.Methods:Ninety patients with brucellosis admitted to Yantai Yeda Hospital from January 2016 to December 2018 were selected as the observation group, of whom 49 cases in acute stage and 41 cases in chronic stage. Another 50 healthy subjects who underwent health examination during the same period were selected as the control group. Five milliliter of fasting venous blood samples were collected from patients with brucellosis and healthy subjects, and enzyme-linked immunosorbent assay (ELISA) was used to detect sTim-3 and Galectin-9 levels, and cytometric bead array (CBA) to detect IFN-γ and IL-4 levels; Pearson correlation test was used to correlate serum levels of sTim-3, Galectin-9, sTim-3/Galectin-9 and inflammatory factors IFN-γ, IL-4 in patients with brucellosis.Results:The levels of sTim-3 [(18.28 ± 5.38), (7.01 ± 1.97) ng/L], Galectin-9 [(2.79 ± 0.94), (1.67 ± 0.23) ng/L], sTim-3/Galectin-9 (6.55 ± 2.96, 4.20 ± 0.67), IFN-γ [(5.99 ± 1.94), (10.09 ± 2.03) ng/L], IL-4 [(7.34 ± 3.13), (3.71 ± 1.76) ng/L] were significantly different between the observation group and control group ( t = 14.271, 8.276, 5.527, 11.785, 7.556, P < 0.05). In the observation group, the levels of sTim-3 [(22.09 ± 3.13), (14.21 ± 3.42) ng/L], Galectin-9 [(2.98 ± 0.67), (2.41 ± 0.74) ng/L], sTim-3/Galectin-9 (7.41 ± 1.07, 5.90 ± 1.72), IFN-γ [(3.78 ± 1.65), (7.02 ± 1.73) ng/L], and IL-4 [(9.17 ± 2.04), (5.67 ± 1.54) ng/L] were statistically significant difference between patients with acute and chronic brucellosis ( t = 11.402, 3.833, 5.084, 9.075, 9.037, P < 0.05). Pearson correlation test showed that the levels of sTim-3, Galectin-9, and sTim-3/Galectin-9 in patients with brucellosis were negatively correlated with IFN-γ level ( r = - 0.568, - 0.531, - 0.519, P < 0.05), and positively correlated with IL-4 level ( r = 0.517, 0.524, 0.536, P < 0.05). Conclusion:The expressions of sTim-3 and Galectin-9 in serum of brucellosis patients are high, and they are significantly correlated with IFN-γ and IL-4 levels, suggesting that sTim-3 and Galectin-9 maybe related to the occurrence and development of brucellosis.
