1.Experimental comparison of the antitussive and expectorant effects of seven traditional Chinese medicine compounds in mice

Objective In order to screen effective traditional Chinese medicine compounds to prevent and control porcine respiratory disease syndrome ( PRDC) , seven compound preparations of traditional Chinese medicine were tested and to analyze their antitussive and expectorant effects in mice .Methods Two hundred 6-week old ICR mice ( male∶fe-male=1∶1) were used in this study .Dextromethorphan and ammonium chloride were used as positive control drugs , and physiological saline was used as blank control .The antitussive and expectorant effects of the seven Chinese medicine com-pounds (groups 1, 2, 3, 4, 5, 6, 7) were observed by ammonia-induced cough model and tracheal phenol red secretion method in mice .Results The results showed that compounds 7 and 5 significantly prolonged the cough incubation period (P <0.05), and reduced the cough times within 5 min (P <0.05).Except for the group 4, tracheal phenol red excre-tion in the other groups was significantly lower than that of blank control group (P <0.05), and phenol red excretion in the mice of groups 7, 5 and ammonium chloride group was significantly lower than that in other treatment groups ( P <0.05).Conclusions The Chinese medicine compounds 5 and 7 show most evident expectorant effects , and worthy of fur-ther validation of them as a drug in the treatment of porcine respiratory disease syndrome .
2.Short-term effect of surgery on the health-related quality of life in children with intermittent exotropia

? AIM: To investigate health - related quality of life ( HRQOL ) state in children with intermittent exotropia using the Intermittent Exotropia Questionnaire ( IXTQ ) and research the effect of strabismus surgeries on HRQOL.?METHODS: In this prospective study, we chose 42 patients with intermittent exotropia ( aged 5-17 years) as case group, and 42 patients ( aged 5-17 years) as control group. The Chinese IXTQ was used to evaluate HRQOL at 1d preoperatively and 3mo postoperatively in the two groups, and the differences of the two groups before and after surgery and the effect of strabismus surgery on HRQOL were analyzed.?RESULTS:The scores of HRQOL in control group were statistically significant higher than that of case group ( P<0. 01 ). Every items showed a statistically significant difference except on “Kids tease me because of my eyes”and “ My eyes make it hard for me to make friends” ( P<0.05). At 3mo postoperatively ,the scores of HRQOL in case group significantly increased than that at 1d preoperatively(P<0. 01). Child IXTQ of case group was lower on every items than those of control group after surgery(P<0. 05).?CONCLUSION:Intermittent exotropia could affect the HRQOL in psychosocial and visual functional. The greatest HRQOL concerns for children with intermittent exotropia were shutting one eye when sunny, waiting for their eyes to clear up instead of taunts and friendship. The surgical treatment could improve HRQOL in children with intermittent exotropia.
3.Effect of intracellular-free Ca~(2+) concentration on transient magnesium-free treatment induced epileptic injury in developing cortical neurons of rats

Objective: To investigate the effect of intracellular free Ca 2+ concentration ([Ca 2+ ] i ) on injury following transient Mg 2+ free treatment in vitro in developing cortical neurons. Methods: Embryo cortical neurons of rats cultured for 6 d and 17 d were directly exposed to Mg 2+ free media, or pretreated with NMDA receptor antagonists or calcium channel antagonist before being exposed to Mg 2+ free media. MTT assay was used to study the injury of neurons. [Ca 2+ ] i were measured using fluo 3, a fluorescent calcium sensitive dye and laser scanning confocal microscope, and calculated by the fluorescent intensity. Results:Compared with control, MTT conversion rates decreased after transient (3 h) Mg 2+ free treatment in neurons cultured for 6 d and 17 d in vitro, (59.1?6.87)% and (51.2?5.90)%, respectively. In neurons pre and co treated with 10 ?mol?L -1 MK 801, 50 ?mol?L -1 AP 5 and 10 ?mol?L -1 nifedipine, MTT conversion rates were higher than those of neurons with only Mg 2+ free treatment. 4.A study on the promoter hypermethylation of TSHR and pl6 gene in human papillary thyroid carcinoma

Objective To study the expression of the tumor suppressor gene TSHR and pl6 in papillary thyroid carcinoma (PTC) and explore the relationship of the tumorigenesis and the promoter aberrant methylation of the two above genes. Methods RT-PCR was used to detect the mRNA expression of two tumor suppressor genes in 50 cases of PTC, 20 cases of nodular goiter and 12 cases of thyroid adenoma tissue. The promoter methylation status of the two genes were detected by methylation-specific PCR technique (MSP) (which of p16 by nested PCR). The promoter hypermethylation of the two genes was tested by randomly gene sequencing. Results Hypermethylation of promoter region were detected from 68.0 % (34/50) TSHR gene and 54.0 % (27/50) pl6 gene in PTC, while 21.9 % (7/32) and 15.60 % (5/32) in controls. The rate of promoter methylation in PTC was significantly higher than that in controls (χ2 = 16.61, P <0.05 vs χ2 =12.08 P <0.05). The relative mRNA expression of TSHR gene and pl6 gene were (0.41±0.11) and (0.51±0.17) in PTC, respectively, while those were (0.63 ±0.08) and (0.72 ±0.22) in controls, respectively. The mRNA expression of the TSHR gene and pl6 gene was obviously lower in PTC than that in controls (t = 3.86, P < 0.05 vs t =3.66, P <0.05). By the sequencing, it was confirmed that the CG in methylated promoter of the two genes was not changed, while the CG in unmethylated promoter was changed into TG. Conclusion Methylation of the TSHR gene and p16 gene in promoter region is a common molecule event and may be invovled in the genesis and development of human PTC.
5.Action of Erythrocytes Immuno - Adhering in Pathogenesis of Children with Mesangio - Proliferative Glomeru-lonephritis

Objective To determine the role of erythrocytes immunoadhering in the pathogenesis of the patients with mesangiopro-liferative glomerulonephntis (MsPGN). Methods The immunoadhering functions of erythroeytes and leukoeytcs were measured in 31 patients with no- IgAN,24 patients with IgAN and 30 normal children by rosette tests:RCR,RICR,TRR,TNR and TLR. Results 1. The immunoadhering functions of erythroeytes (RCR and TRR) of the patients with no- IgAN and IgAN were obviously decreased compared to those of control group,but RICR showed no significant difference;2. The immunoadhering functions of leukocytes (TNR and TLR ) of the patients with no- IgAN and IgAN were obviously decreased compared to those of control group;3 The degree of the decreased immunoadhering functions of erythroeytes was correlated respectively with that of the immunoadhering functions of leukoeytes. Conclusions The decrease of the function of erythroeyte and leukocyte immunoadhering plays an important role in the pathogenesis in patients with MsPGN. These rosette tests may be used in clinic.
6.Association of CD40 gene polymorphisms and serum CD40 levels with the pathogenesis of systemic lupus erythematosus

Objective To explore the association of CD40 gene single nucleotide polymorphisms (SNPs) and haplotypes with the susceptibility to systemic lupus erythematosus (SLE),as well as the association of serum levels and genotypes of CD40 with the occurrence of SLE.Methods A multiplex PCR single-base extension assay (PCR-SBE) and DNA sequencing were performed to analyze 4 SNPs of the CD40 gene,including rs1883832 C/T,rs13040307 C/T,rs752118 C/T and rs3765459 G/A,in 205 patients with SLE (SLE group) and 220 healthy human controls (control group).Enzyme-linked immunosorbent assay (ELISA) was conducted to measure serum levels of CD40 in these subjects.Results Compared with the control group,the SLE group showed significantly increased serum levels of CD40 (P < 0.05).There were significant differences in genotype and allele frequencies of the SNP rs1883832 C/T in the CD40 gene between the SLE group and control group (all P< 0.01).Relative risk analysis showed that the risk of developing SLE in rs1883832 T allele carriers was 1.517 times that in rs1883832 C allele carriers (OR =1.517,95% CI:1.157-1.990,P=0.003).Moreover,serum levels of CD40 were significantly higher in rs1883832 T allele carriers than in rs1883832 C allele carriers (P < 0.01).The risk of developing SLE was significantly increased in TCCA haplotype carriers compared with the healthy controls (OR =2.322,95% CI:1.181-4.564,P=0.012).Conclusion The CD40 gene rs1883832 C/T polymorphism and its TCCA haplotype were both associated with the occurrence of SLE,and the rs1883832 T allele may be a gene predisposing to SLE.

Objective To study the expression of VEGF,Cox2 and mPGES in colon cancer.Methods VEGF,Cox2 and mPGES mRNA expression in 32 paired samples(tumor and adjacent normal tissue)were de- termined by using real time RT-PCR.Results VEGF was overexpressed in 19 of 32(59.3 %)tumor tissues compared with that in 6 of 32(18.7 %)adjacent normal tissue;COX2 was overexpressed in 20 of 32(62.5 %) tumor tissues compared with that in 5 of 32(15.6 %)adjacent normal tissue;mPGES was overexpressed in 24 of 32(75 %)tumor tissues compared with that in 9 of 32(28.12 %)adjacent normal tissue.Conclusion Our result suggested that VEGF165,mPGES and COX2 overexpressed in colon cancer.
8.Detection and Partial Characterization of γ-glutamyltranspeptidase from Cordyceps sinensis Mycelia

γ-glutamyltranspeptidase was detected from the cultured mycelia of Cordyceps sinensis (CSGT). Km and Vmax of CSGT was 2.54×10-4 mol/L and 0.1808 mol/L·min respectively when L-glutamic acid 5-(4-nitroanilide) (GpNA) and glycyglycine was used as its substrate. CSGT was stable from pH 8.0 to 11.0 and at or below 20℃. It was optimally active at pH 9.0~10.0 and 30℃. A series of reducing reagents could activate CSGT, and metal cations such as Zn2+, Cu2+, Hg2+ , Mn2+ inhibited strongly activity of the enzyme, but K+, Ca2+, Mg2+ and Na+ at high concentrations had no effect on its activity, indicating that its active center could contain -SH.
9.Neutral alpha-glucosidase activity is correlated with the location of epididymal obstruction in azoospermia men.

OBJECTIVETo evaluate the correlation of neutral alpha-glucosidase in seminal plasma with the location of epididymal obstruction in azoospermia men.
METHODSWe detected neutral alpha-glucosidase activity in the seminal plasma of 59 men with obstructive azoospermia followed by determining the location of epididymal obstruction by scrotal exploratory surgery. Then we analyzed the correlation between neutral alpha-glucosidase and the location of epididymal obstructive azoospermia.
RESULTSAmong the total number of patients, there were 25 cases of bilateral cauda epididymal obstruction, 15 bilateral corpus, 12 bilateral caput, 4 unilateral caput-opposite cauda, and 3 unilateral corpus-opposite cauda. The neutral alpha-glucosidase levels in the seminal plasma of bilateral cauda, corpus and capus epididymal obstructions were (4.1 +/- 1.9), (13.8 +/- 4.4) and (46.8 +/- 19.3) mU per ejaculate, respectively, with statistically significant differences among the three groups (P < 0.05).
CONCLUSIONNeutral alpha-glucosidase activity is significantly correlated with the location of epididymal obstruction in azoospermia men, which helps to locate epididymal obstruction, evaluate surgical prognosis and reduce the time of scrotal exploratory surgery.
