1.cAMP induces axonal regeneration in spinal cord injury of rats

Objective cAMP promotes neurite outgrowth in vitro. The study is aimed to understand whether cAMP can repair spinal cord injury of rats. Methods 56 rats models of spinal cord hemisection were adopted and randomly distributed into six groups. Dibutyryl-cAMP or physiological saline was injected either once in the motor cortex with an amount of 6 ml of 50 mmol/L cAMP, or continuously infused through a polyethylene tube connecting with a micro-pump in the spinal lesion area or in the subarachnoid space with a total amount of 72 ml of 10 mmol/L cAMP for 72 h. The distribution of neurofilament (NF) in the lesion area was observed by immunohistochemistry. Corticospinal tracts (CST) and spinal axons regeneration were investigated by CST and spinal axons anterograde tracing with biotinylated dextran amine (BDA). The function of hindlimb movements were evaluated by BBB scales and as a reference to assess the repairing effect of treating strategy. Results NF were densely distributed and extended in the lesion area in the cAMP groups, but no connection could be found with the NF in the caudal. No axonal regeneration could be achieved when cAMP was input into the subarachnoid space. Many regenerated axons, including some CST axonal regeneration were presented in the lesion areas in cAMP groups though no continuous long regenerated axons traversed the lesion area, while there was no regenerated axon in the lesion areas in the control groups. All the rats restored to normally walk 4 to 5 weeks after operations, BBB scale exceed 20, and no significant difference between cAMP groups and control groups by comparing the BBB scales of hindlimb movements. Conclusion cAMP injected in the brain cortex or continuously infused in the spinal lesion area can induce the axonal regeneration and is beneficial to repair the spinal cord injury, but could not directly promote hindlimb movements recovering.
2.Pathogen distribution and resistance analysis of elderly patients with pulmonary infection

Objective Through To analyzing analyze the pathogen distribution and resistance analysis of elderly patients with pulmonary infection,in order to provide basis for clinical rational administration.Methods The clinical data of 285 elderly patients diagnosed as pulmonary infection were selected,and their sputum specimens cultivation were collected for statistical analysis,and the pathogenic bacteria distribution and drug resistance were analyzed.Results 292 strains of pathogenic bacteria were detected from 285 patients,of which 204 strains of gramnegative bacteria,accounting for 69.8%,mainly including pseudomonas aeruginosa,acinetobacter baumannii and klebsiella pneumoniae,while 84 strains of gram-positive bacteria,accounting for 28.8%,mainly including staphylococcus aureus and 4 strains of fungus,accounting for 1.4%.Different pathogens showed different degrees of antimicrobial resistance:pseudomonas aeruginosa,acinetobacter baumannii,klebsiella pneumoniae and e.coli had highly sensitivity for imipenem and meropenem,which were secondly sensitive to amikacin and highly resistant to ampicillin;Staphylococcus aureus and epidermis staphylococcus were all sensitive to linezolid and vancomycin,which were highly resistant to benzyl penicillin.Conclusion Gram-negative bacilli is the main bacteria for patients with pulmonary infection,which has higher antimicrobial drug resistant rate,and antimicrobial agents should be rationally used according to drug susceptibility results.

4.Evaluation on efficiency of 4 kinds of γ-interferon release assay detection kits for diagnosis of pulmonary tuberculosis

Objective To evaluate efficiencies of 4 kinds of γ‐interferon release assay(IGRA) detection kits in diagnosis of pul‐monary tuberculosis .Methods 4 kinds of IGRA reagents produced by Oxford Immunotec Ltd (Oxford) in British ,Shanghai Fuxing Changzheng Medical Science Co .,Ltd(Fuxing) ,Cellectis in Australia(Cellectis) and Haikou VTI Biological Institute(VTI) ,respec‐tively ,were used to determine release levels of peripheral bloodγ‐interferon which had antigenicity of Mycobacterium tuberculosis in 86 cases of patients with tuberculosis and 80 cases of healthy individuals ,and diagnostic efficiencies were evaluated .Results Among the 4 kinds of IGRA reagents ,including Oxford ,Fuxing ,Cellectis and VTI ,the sensitivity was 93 .02% ,88 .37% ,90 .70% and 91 .86% ,respectively ;the specificity was 92 .50% ,75 .00% ,82 .50% and 87 .50% ,respectively ;the positive predictive value was 93 .02% ,79 .17% ,84 .78% and 88 .76% ,respectively ;the negative predictive value was 92 .50% ,85 .71% ,89 .19% and 90 .91% ;the diagnostic accordance rate was 92 .77% ,81 .93% ,86 .75% and 89 .76% ,respectively ;the area under receiver operating charac‐teristic(ROC) curve was 0 .975 ,0 .892 ,0 .958 and 0 .963 .Conclusion There are no significant differences among Oxford ,Fuxing , Cellectis and VTI reagents ,and reagents could be selected according to clinical requirements .

6.Clinical and pathological analysis of hepatitis B virus-associated glomerulonephritis in 20 cases

Objective To understand the clinical and pathological features of hepatitis B virus-associated glomerulonephritis(HBV-GN).Methods Twenty children with HBV-GN were analysed including 17 cases of nephritic syndrome (4 cases of simple nephrosis, 13 cases of nephritic nephrosis),2 cases of isolated proteinuria, 1 case of nephritic syndrome according to the clinical classification.Results On the basis of pathology of kidney biopsy, there were 15 cases of membranous glomerulonephritis(MN),3 cases of manbranoproliferative glomerulonephritis(MPGN),2 cases of mesangial proliferative glomerulonephritis(MsPGN).Eight of 20 cases were treated with recombinated human alpha-interfeon. The average follow-up time was 5.7 years. As the result, complete remission clinically was on 5 cases,partial remission on 2 cases and ineffectiveness on one case. Besides, serum HBsAg, HBeAg were converted to negative on 3 cases , HBeAg disappeared and anti-HBe appeared on one case.Conclusion MN is common in childhood with HBV-GN.It also indicates the prognosis of the associated nephropathies is quite favorable.
7.INDUCTION OF APOPTOSIS BY EXTRACT OF TOTAL FLAVONOIDS OF CHRYSANTHEMUM INDICUM ON ADJUVANT ARTHRITIS SYNOVIAL CELLS

Objective To study the effect of the extract of total flavonoids of chrysanthemum indicum(TFC) on adjuvant arthritis synovial cells. Methods 0.1ml of the complete Freund's adjuvant was subcutaneously injected into the right hind feet pads of the SD rats.24 days after immunity synovial cells in knee joint were treated with TFC and inhibition of proliferation was measured with MTT assay.DNA fragmentations were analyzed with DNA gel electrophoresis.Fluorescence staining of Hoechst 33258 to observe apoptotic body.Results The IC_50 of TFC on synovial cells was 112mg/L.DNA gel electrophoresis showed ladder-like strap.Apoptotic bodies were observed by Hoechst 33258.Conclusion TFC can inhibit proliferation and induce apoptosis in synovial cells,and exerts therapeutical effect on rheumstoid arthritis.
8.EFFECT OF TOTAL FLAVONOIDS OF CHRYSANTHEMUM INDICUM ON THE APOPTOSIS OF ADJUVANT ARTHRITIS RAT SYNOVIOCYTES AND THE EXPRESSION OF CASPASE-3

Objective To study the effect of the extract of total flavonoids of Chrysanthemum indicum(TFC) on adjuvant arthritis(AA) synoviocytes.Methods Totally 0.1ml of the complete Freund's adjuvant was subcutaneously injected into the right hind feet pads of 20 SD rats.24 days after immunity synoviocytes in the knee joint were treated with TFC.Cell morphology was examined with electron microscopy.Protein level of caspase-3 cleaved fragments was analyzed by Western blotting.The annexin V stain assay was applied to explore the effect of caspase-3 inhibitor on the amelioration in synovial cells apoptosis of AA rats.Results Typical morphology and biochemical feature of apoptosis in synovial cells of AA rats were observed with TFC.The protein level of caspase-3 cleaved fragments increased obviously and was related with the concentration of TFC in synovial cells of AA rats.The apoptotic cells positively stained with annexin V were markedly reduced by caspase-3 inhibitor.Conclusion TFC can induce apoptosis in AA rats synoviocytes,which may achieve therapeutical effects in AA.The activation of caspase-3 may be one of the main causations.
