1.Citation analysis of published articles in the Chinese Journal of Dermatology from 2002 to 2007

Objective To learn the academic level and scientific impact of the Chinese Journal of Dermatology by using bihliometric methods. Methods Citation frequencies, first authors and their affiliation, citing journals of cited articles published in the Chinese Journal of Dermatology from 2002 to 2007 were statistically analyzed according to the database of Chinese Medical Citation Index (CMCI) by using bibliometric methods. Results From 2002 to 2007, a total of 2259 original papers were published in the Chinese Journal of Dermatology. Of them, 1224 were cited with a total citation frequency of 3963, citation rate of 54.1%, average citation frequency per paper of 3.2 and top citation frequency of 204. The number of first authors of cited papers amounted to 893, and 61 (6.63%) authors published 3 or more cited papers during this period. These authors were distributed in 30 provinces, municipalities, autonomous regions and 1 foreign country; Jiangsu, Beijing, Zhejiang, Guangdong, Shanghai and Shandong ranked in the top 6 regions with high total citation frequencies. There were 517 journals citing articles published in the Chinese Journal of Dermatology from 2002 to 2007;the top citing frequencies were observed in the Chinese Journal of Dermatology, China Journal of Leprosy and Skin Diseases, The Chinese Journal of Dermatovenereology, Journal of Clinical Dermatology and Southern China Journal of Dermato-Venereology, etc. Conclusions The articles in the Chinese Journal of Dermatology have a high academic quality and citation frequency. Chinese Journal of Dermatology is an important publication in the dermato- venereology field as well as a core periodical in medical field of China.

Proteins extracted from two varieties of Chinese roses leaves were separated by two- dimensional polyacrylamide gel electrophoresis (2-DE) with immobilized pH gradient (IPG). Many difference proteins were isolated with molecular weights ranging 10-30 kDa and pI5-6. Three proteins of high levels observed in a gel were excised and identified using peptide mass fingerprinting and MS-MS. A summary of the identified proteins and their putative functions are presented. They are identified as eIF-5A、LEA protein and Hsp17. 5. Functions of these proteins in plant tolerance to high temperature were discussed.
3.Value of two-dimensional,color Doppler ultrasound and real-time ultrasound elastography in diagnosing of Hashimoto's thyroiditis

Objective To investigate the different roles of two-dimensional,color Doppler ultrasound and real-time ultrasound elastography in the diagnosis of Hashimoto's thyroiditis(HT).Methods One hundred and forty-one patients clinically diagnosed as HT underwent traditional ultrasound and elastography.The patients were classified into three groups,the hyperthyroidism group,the hypothyroidism group and the euthyroidism group.One hundred and eight volunteers with normal thyroid functions were enrolled as control group.Ultrasonic characteristics (size,echo,accompanied nodules,blood flow distribution of thyroid) and thyroid elastic parameters were recorded and analyzed.Results ①Focal hypoechoic pattern was mainly seen in the group of HT hyperthyroidim,diffuse hypoechoic pattern was mainly seen in the groups of HT euthyroidism,diffuse hypoechoic pattern with heterogeneous linear hyperechoic and nodular changes were mainly seen in the HT hypothyroidism group.②Blood flow reduced according to the order by the hyperthyroidim group,euthyroidism group and hypothyroidism group.③Patients with HT hyperthyroidism had elastic image pattern Ⅰ.Patients with HT euthyroidism had pattern Ⅰ and pattern Ⅱ.Patients with HT hypothyroidism had pattern Ⅱ and pattern Ⅲ.④There were significant differences (P<0.05) between the mean elastic strain and the ratio of the blue area in each group.Conclusions Real-time ultrasound elastography is helpful for the diagnosis of HT.
4.Value of contrast-enhanced ultrasonography in the differential diagnosis of benign and malignant thyroid nodules under the background of Hashimoto's thyroiditis

Objective To evaluate the value of the differential diagnosis of benign and malignant thyroid nodules under the background of Hashimoto' s thyroiditis (HT) using contrast-enhanced ultrasonography (CEUS).Methods Ninety-three single thyroid nodules with HT confirmed by operations and pathology were given CEUS examination,and time-intensity curves were drawn by using TomTec analyzing software.The parameters as follows:rise time (RT),time to peak (TTP),mean transit time (mTT) and maximum intensity(IMAX) were compared by independent-samples t test,and the diagnostic value of parameters were analyzed by receiver operating characteristic (ROC) curve.Results In 93nodules,there were 56 benign thyroid nodules,and 37 malignant thyroid nodules.(1) After injection of contrast media,the enhancement and wash-out in benign nodules were mostly the same with peripheral gland,as well as the enhance intensity.Malignant nodules presented lower enhancement,with later rise time and earlier wash-out compared to peripheral gland.(2) mTT was longer,while IMAX was higher in benign nodules than those in malignant nodules (P <0.05).But there were no significant differences in RT or TTP between the two groups.(3) The cut-off value in diagnosis of benign and malignant thyroid nodules with HT assessed by ROC curve were mTT 21.70 s,IMAX 86.41%.When mTT,IMAX,and combination of the two parameters were used for the diagnosis of malignant thyroid nodules with HT,the sensitivity were 86.5 %,83.8 %,97.3 % and the specificity were 69.6 %,75.0 %,48.2 %,respectively.Conclusions CEUS is helpful to identify the benign and malignant thyroid nodules under the background of HT.

6.Construction of recombinant adenovirus harboring fusion gene NT4p53(C22)Ant and its killing effect on HepG2 tumor cells

ABSTRACT:Objective To construct a recombinant adenovirus vector harboring fusion gene NT4p53(C22)Ant and study its killing effect on HepG2 tumor cells.Methods Using molecular cloning technology,the rAVV-NT4p53(C22)Ant was produced by homologous recombination.Then we collected virus supernatant and measured its titer after it was amplified by PCR.The effect of this fusion gene on HepG2 tumor cells was evaluated by IHC, MTT assay,PI staining and flow cytometry.Results The recombinant adenovirus was successfully constructed. The p53 expression rate in rAAV-NT4p53(C22)Ant group was (44.88±2.45)%.MTT assay showed that rAAV-NT4p53(C22)Ant could strongly suppress the growth of HepG2 tumor cells.Flow cytometry showed that rAAV-NT4p53(C22)Ant could induce obvious apoptosis of HepG2 tumor cells.Conclusion The recombinant adenovirus vector encoding gene NT4p53(C22)Ant has been successfully constructed and expressed in this experiment,and it can inhibit proliferation and induce apoptosis of HepG2 tumor cells.
7.Effects of Prescription of Nourishing Blood and Stretching of Stoke on TXB2 and 6-Keto-PGF1αof Patients with Acute Ischemic Cerebrovascular Disease

Objective To investigate the effects of prescription of nourishing blood and stretching of stoke (PNBSS) on the levels of TXB2 and 6-Keto-PGF1αin serum of patients with acute ischemic cerebrovascular disease (AICD);To discuss its action mechanism in AICD treatment. Methods Ninety patients with AICD were randomly divided into trial group and control group, 45 cases in each group. The control group received western routine treatment, while the trail group received the western routine treatment plus PNBSS, one dose per day, for one week. Rating scale of neurologic deficit was employed to evaluate treatment effectiveness. Venous blood was collected before the treatment and on the 3rd and 7th days of treatment. Levels of TXB2 and 6-Keto-PGF1αin serum were detected respectively. Results The score of neurologic deficit of post-treatment in two groups apparently decreased compared with baseline (P<0.01), and score of neurologic deficit in trial group on 7th day was lower than that of control group (P<0.05). The total effective rate in trial group was 93.3%, which was apparently higher than that of control group (84.4%). The level of TXB2 in serum and ratio of TXB2/6-Keto-PGF1α (T/P) in two groups on 3rd and 7th days remarkably decreased compared with baseline (P<0.01), while the level of 6-Keto-PGF1α in two groups on 3rd and 7th days was higher than that of baseline (P<0.01). Meanwhile, the level of TXB2 and ratio of T/P in two groups on 7th day were apparently lower than that of 3rd day (P<0.01), and the level of 6-Keto-PGF1αon 7th day was higher than that of 3rd day (P<0.01). The level of TXB2 in serum and ratio of T/P on 3rd and 7th days in trial group were apparently lower than that of control group (P<0.01, P<0.05), while the level of 6-Keto-PGF1α on 3rd and 7th days in trial group was apparently higher than that of control group (P<0.01, P<0.05). Conclusion One of the mechanisms of PNBSS for AICD appears to inhibit overavtivity of thrombocyte, and regulate the misadjustment of ratio of T/P.
8.Effects of Najia Method of Midday-midnight Point Selection for NSE and S100B Protein in Acute Ischemic Stroke Rats

Objective To observe the effects of Najia method of midday-midnight point selection for acute ischemic stroke (AIS) model rats onthe contents of NSE and S100B protein in serum. Methods SPF SD male rats were chosen to establish the models by middle cerebral artery bolt method. Rats were divided into blank group, sham-operation group, model group, channel-point group, and Najia method group by random number table method. Blank group, sham-operation group, and model group were in the absence of treatment, while the channel-point group received acupuncture treatment according to differentiation syndrome. Najia method group used Najia method of midday-midnight point selection to conduct acupuncture treatment once a day. Improvement of neural function and cerebral infarction volume were observed. The contents of NSE and S100B protein in serum were detected. Results Compared with model group, neurological function score, infarct volume and infarct volume percentage, and the contents of NSE and S100B protein in serum decreased in Najia method group and channel-point group (P<0.05, P<0.01). The effects of Najia group were generally better than the channel-point group. Conclusion Najia method of midday-midnight point selection can decrease the content of NSE and S100B protein in serum of AIS model rats, so as to achieve the effects of neuroprotection and treatment.

10.Relationship between the antileukemic activity of L-asparaginase and Asn level around leukemic cells.

OBJECTIVETo study the antileukemic activity of L-asparaginase through determining the changes of 4 kinds of amino acids (Asn, Aspa, Glu and Gln) in cell culture medium.
METHODSFollowing L-Asp treatment with designed concentrations and duration, the IC50 (inhibitory concentration 50%) of 8 kinds of common leukemia cell lines (U937, HL-60, Jurkat, NB4, THP-1, Namalwa, Karpass299, K562) were determined by CCK-8 assay. The changes of the 4 kinds of amino acids mentioned above were detected by high performance liquid chromatography (HPLC).
RESULTSThe asparagines in cell culture medium were rapidly exhausted when treated with 0.01 U/mL L-Asp for 4 hrs or 1 U/mL L-Asp for 5 minutes. There were significant differences in the sensitivities to L-Asp of different leukemia cell lines. The sensitivities to L-Asp of various cell lines were dose-dependent. Low concentration of L-Asp resulted in a low IC50 and the IC50 increased following the L-Asp concentration increased.
CONCLUSIONSDifferent leukemia cell lines have different sensitivities to L-Asp, suggesting that exhaustion of asparagines around leukemia cells could not reflect the treatment efficacy of L-Asp. L-Asp antileukemic activity is dose-dependent, which suggests the importance of high-dose L-Asp on childhood acute lymphoblastic leukemia.
