1.The effects of hyperbaric oxygen therapy on promotion of intellectual rehabilitation in infants with severe hypoxic-ischemic encephalopathy

Objective To explore the long-time effects of hyperbaric oxygen(HBO) therapy on the promotion of intellectual rehabilitation in infants with severe hypoxic-ischemic encephalopathy(HIE). Methods Forty-seven infants with severe HIE were randomly divided into a HBO group(n=24) and a control group(n=23). All the infants were treated with routine therapy for three months,in addition to HBO treatment in the HBO group once a day for four courses of 10 d with the interval of 10~15 d. Neonatal Behavioral Neurological Assessment (NBNA)assay was employed at 7 and 28 d after born,and Bayley Scale of Infant Development (BSID)assay was got at two years in two groups, as well as Wechsler Preschool and Primary Scale of Intelligence(WPPSI)assay at five years, in order to evaluated the short-time and long-time effects of HBO on intellectual rehabilitation in infants with HIE. Objective To explore the possible mechanisms of the protective effect of leflunomide on kidneys by observing the effects of leflunomide on rat kidney tissue of focal segmental glomerulosclerosis (FSGS) expression level of transforming growth factor (TGF)-β1.Methods Twenty-four SD rats were randomly divided into 3 groups:normal control group (n =8),FSGS model group (n =8) and leflunomide treatment group (n =8).Unilateral nephrectomy 1 week after repeated injection of doxorubicin established FSGS model.Since 2 weeks after surgery,the treatment group had been given the leflunomide suspension 5 mg/(kg · d) orally,while normal control group and model group had been given the same amount of solvent orally.In the 8th week of the experiment,the rats were sacrificed and the specimens were collected,so serum creatinine,blood urea nitrogen,total cholesterol,albumin and 24 hours urinary protein were recorded; renal tissue was taken for pathological examination and calculation of glomerular sclerosis index (GSI) was made;immunohistochemical detection of TGF-β1 expression in the kidney was performed;The expression of TGF-β1 was examined by Western blot.Results Compared with normal control group,FSGS model group and leflunomide treatment group rats' 24 hours urinary protein excretion,serum creatinine,blood urea nitrogen,and cholesterol significantly increased while serum albumin significantly reduced severe renal pathological changes and TGF-β1 protein expression was significantly increased,and the differences were statistically significant (all P < 0.05) ; Compared with the FSGS model group,in the 8th weekend of the experiments,the treated rats' 24 hours urinary protein excretion,relevant serum biochemical indicators of renal pathological changes had different degrees of improvement in renal tissue and TGF-β1 protein expression was decreased,so the differences above were statistically significant(all P < 0.05).Conclusions Leflunomide may reduce the FSGS kidney tissue fibrosis by inhibiting the expression of TGF-β1,and thus protect the kidneys.

The anti-tumor activity of fermentation and the ethanol extraction of ten Polyprous fungi against human lung cancer cell NCI-H460 were determined by MTr method,the effect of medium and time for cultivation on the anti-tumor activity were studied.The results showed that the fermentation liquid of Onnia tomentosa and Ceizene unicolon and the mycelium ethanol extract of Formitopsis pinicola had obvious an- ti-tumor activity,the tumor inhibiting ratio of mycelium ethanol extract of Formitopsis pinicola cultivated in Potato-woodehipping medium was 88.87% at the concentration for 500?g/mL,and the inhibiting ratio had little change when Formitopsis pinicola cultivated in different medi- um and for different days.
4.The role of estrogen in the pathogenesis of lupus in NZB/w F1 mice by modulating the function of den dritic cells

Objective To investigate the effect of estrogen on dendritic cells (DCs) and its role in disease progression by comparing the effects of 17β-estradiol (E2) on spleen DCs in systemic lupus erythe matosus (SLE) murine modeI-(NZBxNZW) F1 (NZB/w F1 ) female mice before and after the disease onset.Methods Anti-CD11c antibody labeled magnetic bead was used to purify spleen DCs.Flow cytometry was utilized to detect the co-stimulatory molecules and intracellular cytokines of DCs.Mixed lymphocytes reaction (MLR) was used to measure the stimulatory activity of DCs to T iymphocytes.And semi-quantitative RT-PCR was employed to check the expression of estrogen receptor (ER).Results E2 could modulate the expression of surface molecule CD40,the production of cytokines IL-6,IL-10,IL-12 and TNFa,and the stimulatory a bility of spleen DCs in SLE model mice.Tamoxifen (TAM) could antagonize E2 effects and E2 could affect the estrogen receptor (ERα) level of DCs.These changes of DCs varied with age.Conclusion E2 may be in volved in the pathogenesis of SLE by modulating DCs by binding ERa.The effects of E2 vary in different stages of SLE progression.

Objective To investigate the mechanisms of estrogen in the pathogenesis of systemic lupus erythematosus(SLE),we compared the effects of 17β-estradiol(E2)on bone marrow(BM)-derived dendritic cells (BMDCs)in(NZBxNZW)F1(NZB/w F1)female mice befbre and after the disease onset.Methods Immature,BMDCs were differentiated from BM monocytes cultured with rmGM-CSF,rmlL-4,E2 and estrogen receptor (ER)modulator-tamoxifen(TAM)for 7 days.Then immature DCs were stimulated by LPS for 24h to get mature BMDCs.Flow cytometry was utilized to detect the production of costimulatory molecule CD40 and intracellular IL-6,IL-10,IL-12 and TNFα of DCs.Mixed lymphocytes reaction was used to measure the stim-ulatory activity of DCs on spleen T lymphocytes.Results E2 mainly increased the expression of CD40 on im-mature BMDCs but reduced its expression on mature BMDCs.E2 enhanced the stimulatory activity of immature BMDCs,but weakened the activity of mature BMDCs.E2 decreased the production of cytokines of BMDCs in young mice,but increased them in old mice.Conclusion E2 modulates the functions of BMDCs of lupus model mice by binding with ER.The modulation varies depending on disease progression and cell maturation status.

The tongue and its surrounding structures have special anatomical and functional characters and how to restore its shape and function after glossectomy (especially total glossectomy) still remains a difficult problem. The authors devised a new method of repair by using a forearm free flap with results in 16 out of 17 patients. The selection and shaping of the flap, the evaluation of the recipient site and its blood vessels, the prevention of the oral infection,the monitoring of the circulatory status of the flap and the evalution of the operative result were discussed.
8.Expression of miR-483-5p in epithelial ovarian cancer and its effects on cisplatin resistance in epithelial ovarian cancer cells

Background and purpose:Although cisplatin-based chemotherapies are used as the first-line treatment for ovarian cancers, the majority of patients eventually progress with platinum-resistant disease. miR-483-5p is overexpressed in lung cancer. However, the research on miR-483-5p in epithelial ovarian cancer (EOC) is still unclear. This study aimed to investigate the expression of miR-483-5p in EOC and its effects on cisplatin resistance in EOC cells.Methods:This study analyzed the expression of the miR-483-5p by real-time lfuorescent quantitative polymerase chain reaction (RTFQ-PCR) in EOC tissues, normal ovarian tissues, and EOC cells. The role of miR-483-5p in EOC was evaluatedin vitro by lentivirus-mediated knockdown of miR-483-5p or overexpression of miR-483-5p in EOC cell lines. Drug sensitivity assay was carried out by CCK-8 kit.Results:miR-483-5p was upregulated in EOC tissues as compared with normal tissues (P<0.01). Furthermore, miR-483-5p expression in advanced stage (Ⅲ–Ⅳ) EOC was significantly higher than that in early stage (Ⅰ–Ⅱ) EOC (P<0.05). Interestingly, miR-483-5p expression was higher in cisplatin-resistant A2780/CP cells than other cells. Increased miR-483-5p expression caused EOC cell resistance to cisplatin and downregulated the expression of p21 and Bcl-2, whereas reduced miR-483-5p expression induced its sensitivity and upregulated the expression of p21 and Bcl-2.Conclusion:The results suggest that miR-483-5p is highly expressed in EOC and contributes to cisplatin resistance. Thus, miR-483-5p is a potential therapeutic target for ovarian cancer.

NT-3 can protect typeⅠspiral ganglion cell from glutamate neurotoxicity even applied 60 minutes after the occurrence of glutamate induced cochlear impairment. The result indicates that NT-3 may be an ideal candidate for the treatment of a series of acute neural hearing loss.It may lead to a better outcome when NT-3 gene therapy is actilized.
