1.Effects of glial cell line-derived neurotrophic factor gene-activated matrix on repair of sciatic nerve defect of rats

Objective To evaluate the effects of glial cell line-derived neurotrophic factor(GDNF) gene-activated matrix(GAM) in bridging rat sciatic nerve defect.Methods The peripheral nerve extracellular matrix was harvested by chemical extraction.GDNF gene-activated matrix was comprised of the extracellular matrix and plasmids encoding GDNF.Ninety adult Wistar rats were randomly divided into three groups: group A(GDNF GAM conduits),group B(extracellular matrix conduits),group C (autografts).At the end of 12 weeks,rats from each group were subjected to walking tract analysis,electrophysiological and histomorphological studies.Results GDNF DNA could be retained in GAM,promoting transgene expression in the sciatic nerve,in GAM,and more importantly,in axotomized neurons in spinal cord for 12 weeks.The number of regenerated axons for the GAM grafts in the distal sciatic nerve was lower than that for autografts(2 117?294 vs 2 474?297) at 12 weeks.The sciatic nerve function index for the GAM grafts was lower than that for autografts [(30.92?2.98)% vs(26.27?2.71) %] at 12 weeks.Conclusion GDNF GAM may be a promising alternative to autografts for repair of rat sciatic nerve.
2.Calcium phosphate cement with rhVEGF and rhBMP-2 promotes revascularization of big section bone allograft in dogs

Objective To explore the ability of revascularization of big section deproteinized bone (DPB) allograft combining calcium phosphate cement (CPC) with recombinant human vascular endothelial growth factor (rhVEGF) and bone morphogenetic protein-2 (rhBMP-2). Methods Totally 36 adult hybrid dogs were inflicted to demi-articular defect models by exsecting 30 mm right femur inferior extremity near articular surface,and then randomly divided into 3 groups, group A, receiving material CPC/rhVEGF/rhBMP-2/DPB transplantation into the demi-articular defect, group B, material rhVEGF/rhBMP-2/DPB transplantation, and group C with simple material DPB transplantation. At 4, 8, 12 and 16 weeks after operation, vasculogenesis and new bone formation were assessed by X-ray examination, histological examination, transmission electron microscopy, vas capillare analysis by vaso-filling with ink and radionuclide bone imaging examination (ECT). Results The bone formation and vascularization was significantly superior in group A to that in group B and C. 3. The effect of AICAR on rat uterine smooth muscle

OBJECTIVE: To study the effect of AICAR on uterine smooth muscle of pregnant and nonpregnant rats. METHODS: The action of AICAR and dibutyryl-cAMP on the contractile function of isolated myometrium of pregnant and nonpregnant Sprague-Dawley rats was investigated. AICAR or dibutyryl-cAMP was used at cumulative doses from 10-9 to 10-4 mol · L-1 for myometrium of pregnant rats, and at 10-5 mol · L for myometrium of nonpregnant rats. The amplitude of the contractility of myometruim was recorded. RESULTS: For the myometrium of pregnant or nonpregnant rats, AICAR and dibutyryl-cAMP exerted strong inhibitory action on the contractile function in dose-dependent manners. For the myometrium of pregnant rats, AICAR or dibutyryl-cAMP showed inhibitory effect at a low concentration of 10-7 mol · L-1. AICAR and dibutyryl-cAMP also caused significant inhibition in myometrium from nonpregnant rats. CONCLUSION: AICAR exerted strong inhibitory action on the contractile function of myometrium from pregnant or nonpregnant rats, suggesting that AICAR may have inhibitory effect on spontaneous activity of myometrium of human.
5.Discussions on the management of health check-up nursing quality

Objective To identify the outcomes and effect of applying the health check-up nursing quality control system developed as supervised by JCI quality assessment standards.Methods The monitoring system for nursing quality in the health check-up department is established within the framework of the hospital quality improvement committee and in line with characteristics of the department.Quality improvement tools may be called into play for analysis and decision making to revolve critical problems found in health check-up nursing,upgrading nursing quality and customer satisfaction.Results Significant rise of health check-up quality and customer satisfaction for nurses,and the nurses are trained in quality control knowledge and get further involved in quality management of their department.Conclusion Health check-up nursing management system under the JCI standard is conducive to raising the nursing quality,and helps nurses with problem analysis and solution.

7.Differential gene expression between Xinjiang Uygur and Han patients with ankylosing spondylitis

BACKGROUND:Genetic factors, environment, chronic infection, and autoimmune disorders are considered to be involved in the pathogenesis of ankylosing spondylitis. Genetic factors play an important role in the pathogenesis of the disease. Ethnic and regional diversity of differentialy expressed genes has become research hotspot because of family aggregation and ethnic diversity of ankylosing spondylitis.
OBJECTIVE:To screen differentialy expressed genes in Xinjiang Uygur and Han patients with ankylosing spondylitis by microarray screening and compare differences in gene expressions.
METHODS: Uygur and Han patients with active ankylosing spondylitis in department of rheumatology of our hospital were randomly colected with five patients for each. In addition, three healthy volunteers were selected as controls. RNA from peripheral blood was extracted and used for microarray hybridization after probe preparation to screen differentialy expressed genes in ankylosing spondylitis samples and the microarray results were verified by semi-quantitative RT-PCR analysis.
RESULTS AND CONCLUSION: Twenty differentialy expressed miRNAs were screened in Uygur and Han patients with active ankylosing spondylitis (P < 0.05). From relationship analysis of target genes and miRNAs, 15 target genes corresponding to the 79 miRNAs involved in human leucocyte antigens and interleukins which linked to human immunity system were found. These findings suggest that differentialy expressed genes can be screened from Uygur and Han patients with ankylosing spondylitis.
8.Therapic effect of lauromacrogol sclerotherapy on infantile hemangioma

Objective To investigate the effect of intralesional lauromacrogol therapy on infantile hemangioma.Methods 30 cases all received intralesional injection of lauromacrogol.The changes of tumor size,texture and color were monitored and recorded.The adverse effects after medication were observed and managed accordingly.The adverse effects after medication were observed.The times of treatment were determined according to degree of reduction of tumor volume.To assess the efficacy,a 4 scales system was adopted.Results All patients had completed the treatment and followup.The overall response was scale Ⅰ in 2 cases (6.7 %),scale Ⅱ efficacy in 4 patients (13.3 %),scale Ⅲ efficacy in 5 cases (16.7 %) and scale Ⅳ in 19 cases (63.3 %).The smaller the tumor,the fewer the times of treatment,and the better the results.The effects of treatment were better if patient could be treated by intralesional lauromacrogol therapy in early stage.No severe adverse effects were observed during 6 months of follow-up.Conclusions Intralesional injection of lauromacrogol sclerotherapy is a safe and effective way to treat infantile hemangioma.
9.Expression and its clinical significance of Oct4 gene in prostate cancer

Objective To study the expression and its clinical significance of Oct4 in prostate cancer.Methods The Oct4 mRNA expression levels in 2 normal prostate (NP) samples and 8 prostate cancer (Pca) samples were determined by reverse transcription polymerase chain reaction (RT-PCR) analysis.The Oct4 protein expression levels in 10 NP samples,10 benign prostate hyperplasia (BPH) samples and 45 Pca samples were detected by immunohisochemical analysis.Results The relative expression levels of Oct4 mRNA were 0.91,0.76,0.66,0.63,0.55,0.50,0.48,0.42 in 8 Pca samples and 0 in 2 NP samples,respectively.The positive rates of Oct4 protein expression was higher in Pca tissues than in NP and BPH tissues (91.1% vs.0.0%,91.1% vs.0.0%,both P<0.05).The level of Oct4 protein was increased with higher hostologic grade and clinical stage of prostate cancer.The strong positive rates of Oct4 expression were 72.7% and 30.0% in Pca tissues with and without lymph node metastasis respectively,and there was a significant difference in the strong positive rate between them (P<0.05).Conclusions The Oct4 expression is closely related to the differentiation and invasion of tumor.Oct4 might be an important index for the prognosis of prostate cancer.
10.Analysis of the UV absorbing constituents of the metabolites from UV-B tolerance bacteria and study on its anti-ultraviolet mechanism

Objective The anti-UV-B radiation mechanism of UV-B tolerance strain KFS-9 was studied from the profile of metabolites.Methods The compounds were separated by column chromatography and their structures were elucidated based on GC-MS,LC-TOF-MS,EI-MS and NMR analyses.Results Three unsaturated fatty acids(identified as 9-hexadecenoic acid,9,12-octadecadienoic acid and 11-octadecenoic acid) and 1,2-benzenedicarboxylic acid able to absorb ultraviolet were isolated from the petroleum ether extract of the fermentation liquid of Pantoea agglomerans KFS-9.Fraction(Ⅱ) was isolated from the ethyl acetate extract and was composed of 2,3-butanediol and a series of high unsaturated aroma compounds.Fraction(Ⅱ) had a wide absorption peak,and it could protect E.coli from UV-B damage in some sense.Conclusion Strain KFS-9 produced metabolites that were able to absorb UV to build a natural barrier and so improved the tolerance to UV radiation.The UV-B radiation protection test to the E.coli also showed fraction(Ⅱ) was not the only protector,and there definitely existedother materials and mechanism to protect the strain.
